Abstract:OBJECTIVE:To investigate the influence of Auricularia Auricular polysaccharide (APP) on acute cerebral injury induced by ischemia/reperfusion in rats and its underlying mechanism.METHODS:Adult male SD rats were intragastrically pretreated with AAP at a low (50 mg/kg) or high (100 mg/kg) dose once a day for 20 days before operation. Rats intraperitoneally injected with ginkgo biloba extract (EGb671) were taken as positive control. Focal ischemia was achieved by middle cerebral artery occlusion (MCAO) on the right side for 60 min. After 24 hrs of reperfusion, the nerve function defects were recorded by Longa's score and the brain infarct sizes were measured by 2,3,5-Triphenyl-tetrazolium-chlor (TTC) staining. Apoptotic neurons were detected by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) staining after 48 h of reperfusion. The levels of oxidative stress was determined via the mitochondria-generated reactive oxygen species (ROS).RESULTS:AAP treatment decreased Longa's score, brain infarct size, apoptotic neurons and mitochondria-generated ROS in a dose-dependent manner. AAP at 100 mg/kg gave a better performance compared with EGb671 on all parameters examined.CONCLUSION:AAP treatment protected rat brain from focal ischemia/reperfusion injury by its anti-oxidative effect and worked better than EGb671.
Verapamil Abolished the Enhancement of Protein Phosphorylation of Brainstem Mitochondria and Synaptosomes from the Hens Dosed with Tri-O-cresyl Phosphate

Abstract:To explore the changes of the endogenous phosphorylation of brainstem mitochondrial and synaptosomal proteins in adult hens dosed with tri-o-cresyl phosphate (TOCP) following the development of organophosphate-induced delayed neurotoxicity (OPIDN). Verapamil (7 mg/(kg day), i.m.) was given for 4 days. A dose of TOCP (750 mg/kg, p.o.) was administrated in second day after verapamil. Phosphorylation of the proteins from brainstem mitochondria and synaptosomes was assayed in vitro by using [γ-32P]ATP as phosphate donor. Radiolabeled proteins were separated by SDS-PAGE and visualized by autoradiography. The results showed that TOCP administration enhanced the phosphorylation of the cell organelle proteins (mitochondria: 60, 55, 45, and 20 kDa; synaptosomes: 65, 60, and 20 kDa), while verapamil abolished the enhancement induced by TOCP. Additionally, the reaction for the phosphorylation is catalyzed by the calcium/calmodulin protein kinase. Therefore, TOCP can enhance the phosphorylation of the brainstem mitochondrial and synaptosomal proteins from the hens with OPIDN; however, protection from the enhancement of the phosphorylation should be involved in the mechanisms of the amelioration of TOCP-induced delayed neurotoxicity by verapamil.
Effect of verapamil on phase 3 of action potential in guinea pig monocyte

Abstract:PURPOSE: To investigate effect of verapamil in guinea pig myocyte. METHODS: With the standard microelectrode technique. RESULTS: After 45 minutes perfusion of Tyrode solution with 6 ��mol/L verapamil, APD20 shorted from 86��16 to 71��14 ms (n= 7, P<0.01). APD50 from 152��24 to 140��26 ms (n=7, P<0.01), APD50 from 182��28 to 183��26 ms (n=7, P>0.05); After 40 minutes perfusion with nicorandil APD50 from 154��33 to 122��34 ms (n= 7, P<0.01). APD50 from 186��37 to 154��36 ms (P<0.01), and late perfusion with verapamil and nicorandil after the upper perfusion of nicorandil made APD50, APD90 further decreased, APD50 from 121��35 to 96��32 ms (P<0.01), APD90 from 153��37 to 115��34 ms (P<0.01). CONCLUSIONS: The results indicate verapamil prolong phase 3 duration of action potential in guinea pig myocyte relatively, and nicorandil suppress this effect.
Effect of verapamil on electrobiological activity and seizure behavior in phenytoin-carbamazepine resistant rats

Abstract:Objective To invesgate the effect of P-glycoprotein(PGP)inhibitor,verapamil,on electrobiological activity and seizure behavior in phenytoin-carbamazepine(PHT-CBZ)resistant rats.Methods The model of medically intractable epilepsy was established by kindling of amygdale. Verapamil was applied to PHT-CBZ resistant rats,followed by the observation on after discharge threshold (ADT),after discharge duration(ADD)and seizure activity.Results Compared with the control group, the ADT was higher in PHT-CBZ resistant rats peritoneally injected with verapamil((238.0±32.2)μA vs (177.0±23.3)μA,P<0.05);verapamil shortened the ADD,reduced the Racine′s behavior degree (P<0.05).Conclusions Specific inhibitors of PGP could improve the electrobiological activity and seizure behavior in PHT-CBZ resistant rats.It is benefit to inhibit PGP for multidrug resistant medically intractable epilepsy.
Comparison of Converting Efficacy and Safety of Intravenous Ibutilide and Propafenone in Chinese Patients with Atrial Fibrillation and Atrial Flutter

Abstract:Objective: To compare converting efficacies of intravenous ibutilide and propafenone on atrial fibrillation(AF) and artial flutter(AFL) in Chinese patients in a prospective study.Methods: We investigated 200 AF and AFL patients treated with ibutilide(1 mg,or 0.01 mg·kg-1 if body weight 60 kg,iv) or propafenone(70 mg,iv,repeated if necessary) in the randomized and controlled study.A second infusion was performed 10 min later if first infusion failed.Results: Overall,ibutilide had a higher cardioversion rate(55%,55/100) than propafenone(21%,21/100).Ibutilide converted more patients to sinus rhythm in patients with either AF [50%(40/79) vs 24%(21/90)] or AFL [71%(15/21) vs 0%].The RR intervals were not statistically different between AF and AFL during conversion.One torsade de pointes(TdP,1%) developed in ibutilide group and was converted by direct current electrical conversion.Conclusion: Ibutilide is more effective than propafenone in converting AF or AFL in Chinese patients.Adverse events are not significantly different between ibutilide and propafenone.
Effects of Cyproheptadine on cerebral ischemia-reperfusion injury and nerve cell apoptosis

Abstract:AIM:To study the protection and mechanism of Cyproheptadine(CYP) on focal cerebral ischemia-reperfusion in rats. METHODS:The models with cerebral ischemicre-reperfusion were induced by intraluminal middle cerebral artery occlusion(MCAO) with a nylon monofilament suture,2 h occlusion followed by 3,6,24,48 h reperfusion.The volume of cerbral infarction was observed using 2,3,4-triphenyltetrazolium chloride(TTC) dyeing,apoptosis of the neurocytes was detected by TUNEL technique,the expression levels of Caspase-3 proteinum in brain tissue were detected by immunohistochemistry technique. RESULTS:Compared with model group,the volumes of cerbral infarction were reduced significantly in the CYPI,IIgroups(P<0.01),the increased LDH had been decreased(P<0.01),apoptosis of the neurocytes of ischemia area in the brain tissue was depressed by CPY,the ratio of Caspase-3 proteinum expression was decreased(P<0.01). CONCLUSION:The CPY can prevent cerebral ischemia reperfusion injury by reduce the number of nerve cells apoptosis and the level of Caspase-3 in brain.
EFFECTS OF QUERCETIN ON MEMBRANE FLUIDITY OF INJURED VASCULAR ENDOTHELIAL CELLS WITH HYPOXIA AND THE LACK OF GLUCOSE

Abstract:Objective: To study the effects of different concentrations of Quercetin on nitric oxide (NO) production and membrane fluidity of the injured human umbilical vein vascular endothelial cell line (ECV-304) with hypoxia and the lack of glucose. Methods: The experiments were performed in the culture of ECV-304 injured with hypoxia and the lack of glucose in vitro. The releases of intracellular lactate dehydrogenase (LDH) of ECV-304 was measured with automatic biochemistry analysis. NO level of ECV-304 was monitored with colorimetry. The membrane fluidity of ECV-304 was measured with the fluorescence polarization method. Results: After ECV-304 was cultured in hypoxia and the the lack of glucose for 24 hours, the release of LDH and the membrane fluidity were increased significantly; NO level was decreased. Preincubation of ECV-304 with 20, 80, 160 μmol · L -1 of Quercetin for 24 hours reduced LDH activity, membrane fluidity and increased the level of NO in hypoxia and the lack of glucose induced ECV-304. Conclusion: These results demonstrate that Quercetin can produce the protective effect on hypoxia and the lack of glucose induced injury of ECV-304 by increasing release of NO and changing membrane fluidity.
Quercetin relieves chronic lead exposure-induced impairment of synaptic plasticity in rat dentate gyrus in vivo

Abstract:Increasing evidence suggests that lead (Pb) produces impairments partly through oxidative stress. Though many researchers have investigated protective effect of some antioxidant nutrients against Pb toxicity, little information is available about the effect of antioxidants on Pb-induced impairment of synaptic plasticity. Quercetin, a strong antioxidant and radical scavenger, is the representative natural flavonoid molecule abundant in fruits and vegetables. Previous studies have found that quercetin was neuroprotective in many cases. This study was designed to evaluate the effect of quercetin on chronic Pb exposure-induced impairment of synaptic plasticity in adult rat dentate gyrus (DG) area in vivo. The input/output (I/O) functions, paired-pulse reactions (PPR), excitatory postsynaptic potential (EPSP), and population spike (PS) amplitude were measured in the DG area of different groups of rats in response to stimulation applied to the lateral perforant path. The results showed that the depressed I/O, PPR, and long-term potentiation (LTP) of Pb-exposed group were significantly increased by quercetin treatment. In addition, hippocampal Pb concentration was partially reduced after quercetin treatment. These findings suggest that quercetin treatment could relieve chronic Pb exposure-induced impairment of synaptic plasticity and might be a potential therapeutic intervention to cure cognitive deficits induced by Pb.
Effect of Quercetin in the 1-Methyl-4-phenyl-1, 2, 3, 6-tetrahydropyridine-Induced Mouse Model of Parkinson's Disease

Abstract:In this paper, the protective effect of the bioflavonoid quercetin on behaviors, antioxidases, and neurotransmitters in 1-methyl-4-phenyl-1, 2, 3, 6-tetrahydropyridine-(MPTP-) induced Parkinson's disease (PD) was investigated. Quercetin treatment (50 mg/kg, 100 mg/kg and 200 mg/kg body weight) was orally administered for 14 consecutive days. The results show that quercetin treatment markedly improves the motor balance and coordination of MPTP-treated mice. Significant increases were observed in the activities of glutathione peroxidase (GPx), superoxide dismutase (SOD), and Na + , K + -ATPase, AchE, the content of dopamine (DA) in the quercetin plus MPTP groups compared to those in the MPTP group. Significant reduction the 4-hydroxy-2-nonenal (4-HNE) immunoreactivity in striatum of brains was observed in the quercetin plus MPTP groups in comparison to the MPTP group. Taken together, we propose that quercetin has shown antiparkinsonian properties in our studies. More work is needed to explore detailed mechanisms of action.

Comparative Study on Effects of QuChuWaiFeng and YiQiHuoXue Recipes on Experimental Ischemic Stroke

Abstract:Objective:To sutdy the respective therapeutic effects of QuChuWaiFeng recipe(QCWFR) and YiQiHuoXue recipe(YQHXR) in treating ischemic stroke,further make comparative research. Methods:30 SD male rats were randomly divided into 3 groups,the model group,the QCWFR group and the YQHXR group.Six point neuro score and body weight loss were observed after either 24 or 48 hours of reperfusion followed by 90 minutes ischemia.Histomorphometry of the ischemic brain damage infarct volume,was qualified at 48 hours of reperfusion by use of TTC staining.Result:Infarct volumes and neuro scores of both QCWFR and YQHXR treating groups significantly decreased in contrast with the model group. In addition,there were obvious improvements in body weight loss of QCWFR treating group comparing with two other groups. Conclusion:QCWFR and YQHXR have definite therapeutic effect on ischemic stroke.

Abstract:Objective The experiment was designed to observe the chemosensitive and reversal effect of verapamil( Ver) on non?p?gp overexpressed cells.Method 2,3 bis(2 methoxy 4 nitro 5 sulfophenyl) 5 〔(phenylamino)Carbonyl〕 2H tetrazolium hydroxide(XTT) bioassay was used.Results IC 50 of verapamil alone to HL?60 and HL?60/Adr cells was about 26.13 mg·L -1 and 39.00?mg·L -1 ,respectively.IC 50 of etoposide(VP16) alone to HL?60 and HL?60/Adr cells was about 247.17?μg·L -1 and 872.97?μg·L -1 which were reduced to 1/1.73 and 1/4.19,respectively,when incubated with Ver 5?mg·L -1 .Conclusion Ver has a chemosensitive and reversal effect on non p gp overexpressed cells.

Abstract:Objective To study the curative effect of different regimens on diabetic peripheral neuropathy (DPN). Methods 96 cases ofpatients with DPN treated in our hospital fromOctober 2015 to February 2017 were selected as study subjects, andthey were divided into A, B and C three groups by random number table method, with 32 cases in each group. Group A were treated with mecobalamin on the basis of routine treatment. Based on the treatment for group A, group B were addition-ally treated with αlipoic acid, and group D were additionally treated with Mudan granules and α lipoic acid. The blood glucose, blood lipids, vibration sense, pressure sense, temperature sense, ankle jerk, symptom score and adverse reactions were com-pared among the three groups before treatment and after 10 days of treatment. Results After 10daysof treatment, the vibration perceptionthresholds (VPT) of left and right lower limbs of group C [ (15.24±2.95) V, (15.07±2.75) V] were significantly lower than those of group A and group B, while VPT of left and right lower limbs of group B [ (17.39±3.63) V, (17.11±3.21) V] were significantly lower than those of group A [ (19.62±3.14) V, (19.35±2.87) V]. The abnormal rates of ankle jerkin group B and group C after 10 days of treatment (18.8% and 15.6%) were significantly lower than that in group A (43.8%). Conclusion Mudan granules combined with α lipoic acid and mecobalamin can promote the repair of damaged nerve tissues andrelieve symptoms of DPN.
Protective Effects of Querectin against MPP+-Induced Dopaminergic Neurons Injury via the Nrf2 Signaling Pathway

Abstract:Background: Parkinson's disease (PD) is a common selective and progressive neurodegenerative disorder of nigrostriatal dopaminergic (DA) neurons. Quercetin is a bioflavonoid with antioxidant, anti-inflammatory, anti-aging and anti-cancer properties. However, the exact mechanism by which quercetin exerts its protective effect on DAergic neurons remains unclear. Purpose: To investigate the underlying molecular mechanism of quercetin's protective effect on DA neurons using 1-methyl-4-phenylpyridinium (MPP+)-induced PD ferroptosis model in vitro. Methods: MPP+ was used to induce cytotoxicity in SH-SY5Y/primary neurons. Cell viability and apoptosis were assessed by CCK-8 assay and flow cytometry. The expression levels of ferroptosis-related proteins (NCOA4, SLC7A11, Nrf2, and GPX4) were determined by Western blotting. Malondialdehyde (MDA), iron, and GPX4 levels were assesed using corresponding assay kits. Lipid peroxidation was assessed by C11-BODIPY staining. Results: In the MPP+-induced ferroptosis model of SH-SY5Y cells, the expressions of SLC7A11 and GPX4 were inhibited, and the expression of NCOA4 protein was increased, causing the overproduction of MDA and lipid peroxidation. Quercetin can reduce the above changes caused by MPP+, that is, reduce the protein expression of NCOA4 in SH-SY5Y cells, increase SLC7A11 and GPX4 partially inhibited by MPP+, and reduce MDA overproduction and lipid peroxidation to protect DA neurons. Nrf2 inhibitor ML385 could inhibit quercetin-induced increase of GPX4 and SLC7A11 protein expression, indicating that the protective effect of quercetin was mediated through Nrf2. Conclusions: The results of this study suggest that quercetin regulates ferroptosis through Nrf2-dependent signaling pathways, thereby inhibiting MPP+-induced neurotoxicity in SH-SY5Y/primary neurons.
Systematic Evaluation of Propafenone and Verapamil in the Treatment of Paroxysmal Supraventricular Tachycardia

Abstract:OBJECTIVE:To evaluate the efficacy and safety of propafenone and verapamil in the treatment of paroxysmal supraventricular tachycardia(PSVT) systematically.METHODS:Literature analysis based on evidence-based medicine was performed.The databases of clinical controlled trials including CMB,CNKI,Medline,EMBase and Cochrane library were retrieved to collect clinical research documents on the efficacy and safety of propafenone and verapamil in the treatment of PSVT.RESULTS:12 studies were included,involving 6 302 patients.Jadad scores of 5 studies were more than 3 points and included in high quality researches.The Meta-analysis showed:compared with verapamil group,propafenone group had longer conversion time [SMD=1.08,95%CI(0.12,2.03),P=0.03],but less adverse drug reaction [OR=0.48,95%CI(0.29,0.82),P=0.007].There was no significant difference in the effective rate between 2 groups [OR=1.10,95%CI(0.73,1.66),P=0.66].CONCLUSION:Therapeutic efficacy of propafenone group for PSVT is similar to that in verapamil group and good conversion rate have been obtained.In addition,verapamil has shorter effectual time than propafenone,but propafenone is more safety than verapamil.We should choose drugs correctly according to the needs of patients and pros and cons.It should be noted that there is limitation in protocol of the research and well-designed large-scale sample study is necessary.
Cerebral Protective Effect of Carbamylated Erythropoietin on Epileptic Model Mice

Abstract:Aim: To explore the protective effect of carbamylated erythropoietin on hippocampus neurons in the C57/B6 mice with status convulsion. Methods: The mice were randomly divided into three groups: normal group, CEPO-treated group, and saline(NS)-treated group. The status convulsion model was produced by pilocarpine in the latter two groups. In CEPO-treated group, CEPO was administered intraperitoneally at 2 hours before status convulsion, 2 and 24 hours after convulsion, while NS was injected instead at the same time points in NS-treated group. NeuN immunohistochemical staining was applied to evaluate neuron loss in the hippocampal CA1, CA2, CA3 and CA4 regions of the mice. The mortality, the latency of status convulsion and hippocampal cell loss were observed and compared among the groups. Results: There was no significant difference in mortality between CEPO and NS-treated groups. However, the latency of status convulsion produced by pilocarpine in CEPO-treated group were longer than that in NS-treated group(P<0.01). In addition, the NeuN immunohistochemistry demonstrated that neuron loss in hippocampus in CEPO group was significantly less compared with NS-treated group. Conclusion: CEPO can reduce the neuron apoptosis in the hippocampal after status convulsion in C57/B6 mice, which implies that CEPO might have the anti-apoptosis effect.
Comparison Between the Effects of 2-Selenium Bridged Β-Cyclodextrin and Ebselen on Treating SHRsp Stroke

Abstract:A glutathione peroxidase(GPX) mimic, 2-selenium bridged beta-cyclodextrin(2-SeCD), was synthesized. In order to examine its role and mechanism in treating stroke we chose stroke-prone spontaneously hypertensive rats(SHRsp) as animal model. 56 SHRsps of 8-week olds were randomly divided into several groups: test groups (low, moderate, high dose of 2-SeCD) and control groups(positive and negative). After onset of the stroke, the rats in test groups were orally administrated with different amounts of 2-SeCD, the positive control group with ebselen, and the negative control group with drinking water. The treatment lasted two weeks, followed by observation of the rats for 10 days, meanwhile blood pressure, biochemical parameters of plasma, and the contents of nitric oxide(NO) and malondialdehyde(MDA) in plasma and brain were determined. The results show that there were significant differences in contents of NO and MDA in plasma and brain between the test groups(high, moderate dose of 2-SeCD) and negative control group. The NO contents of the test. groups were obviously higher than that of the negative control group (P < 0.01). The MDA contents of the test groups(high, moderate dose of 2-SeCD) were obviously lower than that of the negative control group(P < 0.01). The mechanism of 2-SeCD in treating stroke was discussed, which maybe related to the increase of NO and the decrease of MDA in plasma and brain tissue, but the exact mechanism should be further studied. Moreover, the tendencies of changes in systolic blood pressure, contents of NO and MDA, and other physiological parameters for the test groups were shown to be much better than the corresponding parameters for the positive group(the group with ebselen)(P < 0.05), indicating that the treatment effect of 2-SeCD is better than that of ebselen.

Abstract:OBJECTIVE To investigate the possibility and mechanism of quercetin and kaempferol,flavonoid molecules,for reversing multidrug resistance(MDR) in K562/A02 cells.METHODS K562 and K562/A02 cells were cultured in vitro with the flavonoids as single and in combination respectively.Cell growth inhibition and adriamycin(ADM) sensitivity were detected by MTT assay.Intracellular ADM concentration was determined by flow cytometry when K562/A02 cells was cultured with ADM and the flavonoids for 2 h.Cell apoptosis was examined by Annexin V/PI staining method.Moreover,the expression of related genes for drug transporters and apoptosis was also tested by real time PCR array.RESULTS The MTT assay showed that quercetin and keampferol inhibited the proliferation of K562 and K562/A02 at the concentrnation of 5-160 mol·L-1.They also increased the sensitivity of K562/A02 cells to ADM at a low toxicity concentration.Flow cytometry showed quercetin increased intracellular ADM in a dose dependent manner.but keampferol showed trivial effect on intracellular ADM accumulation.However,quercetin+keampferol treatment showed a similar effect to that of quercetin alone.Cell apoptosis experiment showed both compounds had a dose dependent effect on K562 and K562/A02 cells.Furthermore,PCR array showed that some of the genes related to drug transporters such as ATP-binding cassette(ABC) and solute carrier(SLC) and some of the genes related to apoptosis such as Bcl-2,TNF and TNFR families were regulated.CONCLUSION The experiment indicated that quercetin and kaempferol may be used as reveratrol in leukemia chemotherapy,but their interaction and difference should be noticed.
Comparison of the effects of hydroalcoholic extract of Capparis spinosa fruit, quercetin and vitamin E on monosodium glutamate-induced toxicity in rats

Abstract:Capparis spinosa L. has many biological effects such as antioxidant properties. In the present study, we compared the effects of the hydro-alcoholic extract of Capparis spinosa fruit, quercetin (Q), and vitamin E (Vit E) on monosodium glutamate (MSG)-induced toxicity. The following groups were designed: Control groups (normal saline and/or corn oil); MSG group (4.00 g kg-1 MSG); MSG + low dose extract group (4.00 g kg-1 MSG with 100.00 mg kg-1 extract); MSG + high dose extract (HDE) group (4.00 g kg-1 MSG with 300.00 mg kg-1 extract); MSG + Q group (4.00 g kg-1 MSG with 10.00 mg kg-1 Q); MSG + Vit E group (4.00 g kg-1 MSG with 200.00 mg kg-1 Vit E). All chemicals were orally administered for 14 consecutive days. Tissue specimens from the heart, kidney, and liver tissues and blood samples were collected for histopathological and biochemical evaluations. The results showed that the MSG-induced tissue edema, congestion, and inflammatory cell infiltration were resolved by HDE, Q, and Vit E treatments. These chemicals also restored tissue malondialdehyde level and superoxide dismutase activity. Besides, alterations induced by MSG in serum levels of aspartate transaminase, alanine aminotransferase, urea, lactate dehydrogenase, and creatine kinase-MB were also resolved. It is concluded that Capparis spinosa fruit extract, Q and Vit E can produce approximately similar protective effects on tissue function through oxidative stress alleviation and antioxidant mechanisms restoration.
Comparison of the cardiac electrophysiological effects of sodium ferulateto amiodarone on rabbit right ventricular myocardium

Abstract:Objective:The acute effects of sodium ferulate on the prolongation of ventricular repolarization and refractoriness were investigated in rabbit's right ventricular myocardium in order to study its antiarrhythmic mechanism in vivo. Method:Sixteen rabbits were randomly divided into two groups: the sodium ferulate and the amiodarone group. Suface electrocardiogram and the monophasic action potential (MAP) of right ventricular endocardium were simultaneously recorded before and after intravenous administration. The sinus cycle length (SCL), amplitude of MAP(MAPA), the duration of QRS, MAP at 90%(QRSD, MAPD 90 ) and effect refractory period (ERP) were evaluated. Result:Sodium ferulate caused significantly time dependent prolongation of the ERP and MAPD 90 but did not change the ratio of ERP to MAPD 90 just like amidarone. After 15 minutes administration of amiodarone, the ERP and MAPD 90 were prolonged from ( 125.00 ± 16.26 )ms and ( 143.12 ± 15.80 )ms to ( 199.00 ± 21.62 ) and ( 216.88 ± 23.14 )ms respectively(P 0.01 ). Similarly, after 25 minutes administration of soduim ferulate, the ERP and MAPD 90 were prolonged from ( 122.50 ± 8.13 )ms, ( 140.00 ± 6.25 )ms to ( 143.75 ± 6.87 )ms and ( 164.38 ± 7.19 ) ms respectively (P 0.01 ).Conclusion:Sodium ferulate is a potassium channel blocker with characteristic of class Ⅲ antiarrhythmic agents just like amidarone.
Effects of Quercetin on the Efficacy of Various Chemotherapeutic Drugs in Cervical Cancer Cells

Abstract:Purpose: This study aimed to investigate the effects of quercetin on the efficacy of various chemotherapeutic drugs in cervical cancer cells. Methods: All drug experiments were performed in HeLa and SiHa cells. The cell viability was detected by Cell Counting Kit-8 assay, and cell proliferation was estimated by bromodeoxyuridine assay. CompuSyn software was utilized to calculate the combination index (CI) and evaluate the synergistic or antagonistic effect of quercetin with cisplatin, paclitaxel, 5-fluorouracil and doxorubicin on cell viability. Cell migration and invasion abilities were detected by transwell assays, and cell apoptosis was measured by flow cytometry. The expression levels of matrix metallopeptidase 2 (MMP2), ezrin, P-glycoprotein (P-Gp) and methyltransferase-like 3 (METTL3) protein treated with various drugs were analyzed by Western blotting. Results: Quercetin inhibited the viability of HeLa and SiHa cells in a dose- and time-dependent manner. The CI values of quercetin with cisplatin, paclitaxel, 5-fluorouracil and doxorubicin were <1, >1, >1 and >1, respectively. The effect of combination of quercetin and cisplatin on cell proliferation was stronger than their individual effects. Co-treatment group could inhibit more cell migration and invasion in contrast to single-drug group. Besides, quercetin combined with cisplatin group induced more cell apoptosis in contrast to single-drug group. The results of Western blotting showed that the expression levels of MMP2, ezrin, P-Gp and METTL3 in co-treatment group were lower than in cisplatin group, respectively. Conclusion: Quercetin and cisplatin had synergistic inhibitory effect on cervical cancer cells. Quercetin might enhance the antitumor effect of cisplatin via inhibiting proliferation, migration and invasion and elevating apoptosis through weakening MMP2, ezrin, METTL3 and P-Gp expression of cancer cells.
