Abstract:Chirality is a common phenomenon in nature, including the dominance preference of small biomolecules, the special spatial conformation of biomolecules, and the biological and physiological processes triggered by chirality. The selective chiral recognition of molecules in nature from up-bottom or bottom-up is of great significance for living organisms. Such as the transcription of DNA, the recognition of membrane proteins, and the catalysis of enzymes all involve chiral recognition processes. The selective recognition between these macromolecules is mainly achieved through non covalent interactions such as hydrophobic interactions, ammonia bonding, electrostatic interactions, metal coordination, van der Waals forces, and π-π stacking. Researchers have been committed to studying how to convert this weak non covalent interaction into macroscopic visualization, which has further understood of the interactions between chiral molecules and is of great significance for simulating the interactions between molecules in living organisms. This article reviews several models of chiral recognition mechanisms, the interaction forces involved in the chiral recognition process, and the research progress of chiral recognition mechanisms. The outlook in this review points out that studying chiral recognition interactions provides an important bridge between chiral materials and the life sciences, providing an ideal platform for studying chiral phenomena in biological systems.

Abstract:A molecular mechanics field, Alchemy II, was utilized to model the chiral recognition between S-N-acetyl-alpha-methyl-alpha-naphthylamine and (R, S)-N-(3, 5-dinitrophrnyl)-alpha-methyl-benzeneacetamide and between beta-cyclodextrin and (R, S)-fenoprofen. Some preliminary results have: been obtained to sustain the three-point action models and the induce-fit action in enantiorecognition.

Abstract:Chirality is a universal phenomenon in molecular and biological systems, denoting an asymmetric configurational property where an object cannot be superimposed onto its mirror image by any kind of translation or rotation, which is ubiquitous on the scale from neutrinos to spiral galaxies. Chirality plays a very important role in the life system. Many biological molecules in the life body show chirality, such as the "codebook" of the earth's biological diversity-DNA, nucleic acid, etc. Intriguingly, living organisms hierarchically consist of homochiral building blocks, for example, l-amino acids and d-sugars with unknown reason. When molecules with chirality interact with these chiral factors, only one conformation favors the positive development of life, that is, the chiral host environment can only selectively interact with chiral molecules of one of the conformations. The differences in chiral interactions are often manifested by chiral recognition, mutual matching, and interactions with chiral molecules, which means that the stereoselectivity of chiral molecules can produce changes in pharmacodynamics and pathology. Here, the latest investigations are summarized including the construction and applications of chiral materials based on natural small molecules as chiral source, natural biomacromolecules as chiral sources, and the material synthesized by design as a chiral source.

Abstract:Molecular interactions are the key to understanding the structure and properties of materials. Also, it is critical of chiral recognition. Since the molecular interactions at play in chiral discrimination is difficult to investigate under traditional experiment conditions. We have undertaken a computer experiment of van der Waals interaction between chiral molecules and a polymer, which works for chiral separation. Building molecular models, choosing proper force field and Monte Carlo method, we have calculated the difference of interaction energy between isotactic poly-(triphenylmethyl methacrylate) and (+)-hexahelicene or (-)-hexahelicene. It is finally found about 1.379 4 kJ/mol between the polymer and the two chiral hexahelicenes. Reasonably, this difference is regardd as the main reason which induced chiral separation. Furthermore, we compared the results from computer smulation with that from experiment. It can be seen that some details during chiral separation process, such as trailing effect, can also be predicted, and be consistent with the experiment results. Finally, the effective structureal characteristics of the recognition site are pointed out.

Abstract:Molecular chirality has traditionally been viewed as a binary property where a molecule is classified as either chiral or achiral, yet in recent decades, mathematical methods for quantifying chirality have been explored. Here, we use toy molecular systems to systematically compare the performance of two state-of-the-art chirality measures: (1) the Continuous Chirality Measure (CCM) and (2) the Chirality Characteristic (χ). We find that both methods exhibit qualitatively similar behavior when applied to simple molecular systems such as a four-site molecule or the polymer double-helix, but we show that the CCM may be more suitable for evaluating the chirality of arbitrary molecules or abstract structures such as normal vibrational modes. We discuss a range of considerations for applying these methods to molecular systems in general, and we provide links to user-friendly codes for both methods. We aim for this paper to serve as a concise resource for scientists attempting to familiarize themselves with these chirality measures or attempting to implement chirality measures in their own work.

Abstract:The construction of chemical sensors that can distinguish molecular chirality has attracted increasing attention in recent years due to the significance of chiral organic molecules and the importance of detecting their absolute configuration and chiroptical purity. The supramolecular chirality sensing strategy has shown promising potential due to its advantages of high throughput, sensitivity, and fast chirality detection. This review focuses on chirality sensors based on macrocyclic compounds. Macrocyclic chirality sensors usually have inherent complexing ability towards certain chiral guests, which combined with the signal output components, could offer many unique advantages/properties compared to traditional chiral sensors. Chirality sensing based on macrocyclic sensors has shown rapid progress in recent years. This review summarizes recent advances in chirality sensing based on both achiral and chiral macrocyclic compounds, especially newly emerged macrocyclic molecules.

Abstract:Chiral substances occur naturally in abiotic and living systems. The recognition and detection of chiral substances in the natural environment or their analysis and detection in biological systems are crucial. Chiral recognition is a research hotspot in clinical medicine, pharmacology, biochemistry, and other fields. Indeed, many researchers have developed various sensors with different functionalized materials for detecting and analyzing enantiomers. Supramolecular systems have important applications in the development of molecular recognition technologies, and the development of supramolecular chemistry is closely related to research on molecular devices. Therefore, this review summarizes the principle of chiral supramolecular sensors for the detection of enantiomers from the perspective of various sensor types, including optical, electrochemical, electrochemical luminescence, photoelectric, and supramolecular chemical sensors. This review also summarizes the relevant reports on chiral supramolecular sensors in the last five years. Finally, we highlight the prospects of supramolecular chiral sensors in future research.

Abstract:Calixarens are an important class of host compounds and have extensive applications in host-guest recognition. Especially, chiral calixarene is one of the most interesting research fields in calixarene chemistry because of its excellent chiral recognition, enantiomer separation and asymmetrical catalysis. In the study of chiral recognition, NMR spectrum is the most widely investigated measurement for determining ee values and assigning absolute configurations of chiral compounds due to its advantages of convenience, intuition and marked effect, etc. Herein, this paper deals with the latest research progress in the synthesis of chiral receptors based on calixarenes and their applications in the chiral recognition by NMR spectroscopy. In addition, the application prospects of chiral calixarenes are also discussed.

Abstract:The stereospecific analysis of chiral molecules is an important issue in many scientific fields. In separation sciences, this is achieved via the formation of transient diastereomeric complexes between a chiral selector and the selectand enantiomers driven by molecular interactions including electrostatic, ion‐dipole, dipole‐dipole, van der Waals or π‐π interactions as well as hydrogen or halogen bonds depending on the nature of selector and selectand. Nuclear magnetic resonance spectroscopy and molecular modeling methods are currently the most frequently applied techniques to understand the selector‐selectand interactions at a molecular level and to draw conclusions on the chiral separation mechanism. The present short review summarizes some of the recent achievements for the understanding of the chiral recognition of the most important chiral selectors combining separation techniques with molecular modeling and/or spectroscopic techniques dating between 2020 and early 2024. The selectors include polysaccharide derivatives, cyclodextrins, macrocyclic glycopeptides, proteins, donor‐acceptor type selectors, ion‐exchangers, crown ethers, and molecular micelles. The application of chiral ionic liquids and chiral deep eutectic solvents, as well as further selectors, are also briefly addressed. A compilation of all published literature on chiral selectors has not been attempted.

Abstract:Chirality is ubiquitous in nature with primary cellular functions that include construction of right-/left-handed helix and selective communications among diverse biomolecules. Of particularly intriguing are the chiral peptide-based materials that can be deliberately designed to change physicochemistry properties via tuning peptide sequences. Critically, understanding their chiral effects are fundamental for the development of novel materials in chemistry and biomedicine fields. Here, we review recent researches on chirality in peptide-based materials, summarizing relevant typical chiral effects towards recognition, amplification, and induction. Driven forces for the chiral discrimination in affinity interaction as well as the handedness preferences in supramolecular structure formation at both the macroscale and microscale are illustrated. The implementation of such chirality effects of artificial copolymers, assembled aggregates and their composites in the fields of bioseparation and bioenrichment, cell incubation, protein aggregation inhibitors, chiral smart gels, and bionic electro devices are also presented. At last, the challenges in these areas and possible directions are pointed out. The diversity of chiral roles in the origin of life and chirality design in different organic or composite systems as well as their applications in drug development and chirality detection in environmental protection are discussed.
Prediction on Separation Factor of Chiral Arylhydantoin Compounds and Recognition Mechanism Between Chiral Stationary Phase and the Enantiomers.

Abstract:Chiral hydantoins display potential antidepressant and anticancer activities. The quantitative structure property relationship (QSPR) study on chiral compounds plays a key role in predicting the retention factor and the separation factor even the elution order of the enantiomers. In this study, the structures of chiral arylhydantoin compounds have been built to compute molecular structural parameters using VolSurf program. The satisfying models were established between the parameters and the retention factor as well as the separation factor. Analysis on the variables shows that the large difference values of volume, hydrophilic regions and hydrogen bond acceptor and donor regions at some energy levels will result in the large separation factor. The differences of hydrophobic regions and the unbalance between the centre of mass and the barycentre of the hydrophobic region at high energy levels are also favorable to the separation. The differences of surface area, hydrophilic regions and hydrogen bond acceptor and donor regions at other energy levels as well as hydrophobic regions and the unbalance at low energy levels between the enantiomers are disadvantageous to the separation. The analysis on the chiral recognition mechanism demonstrates that the differences of the interaction energies between the enantiomers are mainly affected by the hydrogen bond, π-π interaction and CH3-π interaction. The retention factor and the separation factor, especially the elution order of the enantiomers can be easily predicted using the models. The study provides a helpful guidance for studying other chiral compounds.

Abstract:Inherently chiral calixarenes, whose chirality is based on the absence of a planar symmetry or an inversion center in the molecules as a whole through the asymmetric array of several achiral groups upon the three-dimensional calix-skeletons, are challenging and attractive chiral molecules, because of their potential in supramolecular chemistry. The synthesis and optical resolution of all varieties of inherently chiral calixarenes are systematically discussed and classified, and their applications in chiral recognition and asymmetric catalysis are thoroughly illustrated in this review.

Abstract:The self-assembly and chiral recognition are significant for various fields. In this report, we studied the self-assembly of bisbenzocoumarins (DB) and their recognition with three kinds of small chiral molecules by spectroscopical and morphological methods. The results showed that low in solvent polarity could facilitate the aggregation of DB. Moreover, the chiral recognition of DB to 1-Phenylethanol (PE) occurred, which was confirmed by the circular dichroism (CD) spectra. The method of simulation chemistry was used to assist in understanding the recognition mechanism. It is supposed that the increased DB dihedral angle was caused by the non-covalent interaction between DB and PE. This work highlights the fundamental understanding of self-assembly of chiral bisbenzocoumarins and the chiral recognition to small molecules thereof.

Abstract:Chiral analysis is of great importance to fundamental and applied research in chemical, biological and pharmaceutical sciences. Due to the superiority of mass spectrometry (MS) over other analytical methods in terms of speed, specificity and sensitivity, chiral analysis by MS has attracted much interest in recent years. Chiral analysis by MS typically involves introduction of a chiral selector to form diastereomers with analyte enantiomers, and comparison of the behaviors of diastereomers in MS. Chiral differentiation can be achieved by comparing the relative abundances of diastereomers, the thermodynamic or kinetic constants of ion-molecule reactions of diastereomers in the gas phase, the dissociation of diastereomers in MS/MS, or the mobility of diastereomers in ion mobility mass spectrometry. In this review, chiral recognition and determination of enantiomeric excess by these chiral MS methods were summarized, and the prospects of chiral analysis by MS were discussed.

Abstract:Chirality is a fundamental and ubiquitous feature of living organisms in nature. Magnetic materials, in particular magnetic nanoparticles (MNPs), show some interesting properties such as large specific surface area, easy surface modification, magnetic responsivity and separation ability. Integrating MNPs with chirality in a single material will undoubtedly create a large number of advanced multi-functional materials. Despite the great advancements made in this area, there have been no review articles to summarize the relevant studies. The present work reviews the major progress recently made in constructing chiral magnetic hybrid materials (CMHMs) using macromolecules, which are classified based on the primary chiral macromolecular organic components, namely, biological polymers and synthetic polymers, and the applications of the resulting chiral hybrids in chiral research fields, including asymmetric catalysis, enzymatic resolution, chromatographic separation, enantioselective crystallization and enantioselective adsorption, are also summarized. The challenges and prospects of related research fields are proposed in the last section.
