Abstract:A new indole alkaloidal glucoside together with three known compounds aurantiamide acetate (2), eleutheroside E (3) and 1-O-caffeoyl-β-D-glucopyranoside (4) has been isolated from ethanol extract of the aerial parts of Clematis terniflora DC. On the basis of their spectroscopic and chemical evidence, the new compound was elucidated as (6-O-β-D-glucopyranosyl-1H-indol-3-yl) carboxylic acid methyl ester (1). Compounds 1 and 3 showed significant cytotoxicity against human ECA-109.

Abstract:Investigation on the whole plant of Ceratostigma willmottianum for bioactive compounds has led to the identification of 15 compounds. Two of them were characterized as new compounds, plumbolactones A (5-(2,3-dihydroxyphenyl)dihydro-4-methyl-2(3H)-furanone, 1) and B (3-(1,2-dihydroxypropyl)-5-hydroxy-1(3H)-isobenzofuranone, 2), while the other 13 were known compounds, plumbagic acid (3), isoshinanolone, epiisoshinanolone, plumbagin (4), N-trans-caffeoyltyramine, N-trans-feruloyltyramine, myricetin, quercetin, tricetin 3',5'-dimethyl ether, vanillic acid, syringic acid, caffeic acid, and 6,7-dihydroxycoumarin. The structures of the new compounds were assigned by 2D-NMR techniques.

Abstract:Two new tartrate derivative glucosides, coelovirin C (1) and D (2), were isolated from rhizomes of Coeloglossum viride ( L.) Hartm. var. bracteatum ( Willd.) Richter (Orchidaceae). Their structures were elucidated as (2R, 3S)-2-beta-D-glucopyranosyl-2-isobutyltartrate-l-(4-beta-D-glucopyranosyloxybenzyl) ester 1 and (2R, 3S)-2-beta-D-glucopyranosyl-2-isobutyltartrate-4-(4-beta-D-glucopyranosyloxybenzyl) ester 2 by means of chemical and spectroscopic methods.

Abstract:To discover anti-pancreatic cancer cells growth inhibitor, a new water-soluble glucan, WTMA, was isolated from the rhizome of Gastrodia elate Bl. The mean molecular weight of the polysaccharide was estimated to be 7.0 x 10(5) Da by high performance gel permeation chromatography (HPGPC). The structure of WTMA was characterized by GC, GC-MS, NMR and MALDI-TOF. WTMA was deduced as an alpha-(1 --> 4)-glucan with alpha-(1 --> 4) linked branches attached to O-6 at branch points. Bioactivity tests in vitro indicated that WTMA and its partial acid hydrolysis-derived mixture product, WTMA-AD-O, could significantly inhibit the proliferation of pancreatic cancer cells. However, WTMA-DE, the degraded polysaccharide without branch chain by isoamylase, did not inhibit the proliferation of pancreatic cancer cells. Thus side chain on WTMA was deduced to be required for the anti-cancer cell activity. Further study demonstrated that WTMA and all of its hydrolysis products had no inhibition effect on liver LO2 cells. (C) 2011 Elsevier Ltd. All rights reserved.

Abstract:A new xanthone glycoside, 1,8-dihydroxyl-2,5-dimethoxy-xanthone-6-O-β-D-glucoside (1), along with two known xanthone glycosides and two flavonoid glycosides were isolated from the aerial parts of Lomatogonium rotatum (L.) Fries es Nym. The structure of 1 was elucidated by analysis of its spectroscopic data, including UV, IR, HR-ESI-MS and extensive 1 D and 2 D NMR techniques. In vitro test, compound 1 behaved similarity to swertianolin against α‑glucosidase and more potent inhibitory effects than the positive control, acarbose.

Abstract:From a China mangrove plant Acanthus ilicifolius Linn., a new compound, 5,11-epoxymegastigmane glucoside (1) was isolated along with one known compound of megastigmane glucosides, (6S, 9S)-roseoside (2). The structure of compound 1 was determined on the basis of 1D- and 2D-NMR techniques (DEPT, COSY, HMBC, HMQC, NOESY) and HR-FABMS. The structure of 1 was established as (1S, 3S, 5R, 6R, 7E)-5,11-epoxy-3-hydroxymegastigman-7-en-9-one-3-O-beta-D-glucopyranoside.

Abstract:As a part of our interest in biologically active germacranolides from the genus Carpesium (Compositae), we have investigated the constituents of Carpesium cernuum. This paper reports the isolation and structural elucidation of a new germacranolide, cemolide A (Compound 1), from the herb. The structure of Compound 1 was determined as 2α,3β-dihydroxy-9-angeloxygermacra-4-en-6,12-olide on the basis of spectral evidence. The skeleton of Compound 1 was elucidation by IR, MS, 1H and 13C NMR, COSY, HMQC and HMBC experiments. The stereochemistry of Compound 1 was deduced by ROESY spectral data. Finally, the procedures of extraction and isolation were described in detail.

Abstract:Two new compounds armochaetoglosin A1 ( 1 ) and armochaetoglosin A2 ( 2 ), together with three known compounds ( 3 – 5 ), were isolated from Chaetomium sp. Strain . Their structures were elucidated based on spectroscopic methods, including extensive NMR and MS spectra. Compound 5 showed potential inhibitory activity on α -glucosidase with the IC 50 value of 38.81 ± 8.08 μM. Molecular docking studies confirmed our result on compound 5 α -glucosidase inhibitory properties.

Abstract:From Ceratophyllum demersum growing in Vietnam, twelve compounds were isolated and structurally elucidated, including six previously undescribed compounds, demersones A-D (1-4), acetylvelutins A and B (8 and 9), together with six known compounds, (+)-cyclocolorenone (5), 1-hydroxycyclocolorenone (6), 10-hydroxycyclocolorenone (7), retusin (10), betulinic acid (11), and lupeol (12). The chemical structures and stereochemistry of 1-12 were identified through analysis of spectroscopic data (1D and 2D NMR and HRESIMS), ECD data, and DFT calculation. Notably, this is the first time that humulene-type (1 and 2), guaiane-type (3), and aromadendrane-type (4-7) sesquiterpenoids have been reported in this genus. Compounds 8 and 9 are the first examples of 8-acetoxyflavones found in nature. Upon evaluation of the alpha-glucosidase of 1-3 and 5-12, it was found that 12 exhibited the highest potential with an IC50 value of 15.4 ± 1.1 μM. The molecular docking of 3 and 8 was further studied.

The structures of these new products were established by spectral and chemical methods as 6'-(2,3-dihydroxybenzoyl)sweroside (1) and 6'-(2,3-dihydroxybenzoyl)swertiamarin (2), respectively.

Abstract:Objective: To study a glucan (GB II) isolated from Gastrodia elata. Methods: The glucan was obtained with water extraction, alcohol precipitate, DEAE-Sepharose Fast Flow column and Sepharose CL-6B column chromatography; sugar composition analysis, IR and NMR were used to determine the structural feature. Results: The molecular weight of the glucan was 4 300 dalton estimated by HPGPC; it contained 27 glucose residues, which mainly composed of alpha-D-(1-->4)-glucose with little glucuronic acid and branch O-6 points. Conclusion: The glucan was a new glucan for the first report from Gastrodia elata.

Abstract:In the course of our ongoing phytochemical investigation on the n-butanol extract of the fruits of Tetradium ruticarpum (Rutaceae), three new compounds, ruticarpsides A–C (1–3), were obtained and their structures were elucidated by a comprehensive analysis of NMR and MS data. Compound 3 showed a weak inhibition effect on nitric oxide production in BV-2 microglial cells stimulated with lipopolysaccharide.

Abstract:A new isocoumarin glycoside, 3R-(+)-5-O-[6'-O-acetyl]-α-D-glucopyranosyl-5-hydroxymellein (1), and a new phenylethanol glycoside, (−)-phenylethyl-8-O-α-L-rhamno-pyranoside (2), were isolated from the ethyl acetate extract of the fungus Xylaria sp. cfcc 87468, together with five known steroids, β-sitosterol (3), stigmast-4-en-3-one (4), ergosterol (5), (22E)-cholesta-4,6,8(14),22-tetraen-3-one (6), and 4α-methyl- ergosta-8(14),24(28)-dien-3β-ol (7). The structures of compounds 1 and 2 were elucidated by MS, extensive 1D and 2D NMR spectroscopy, and the circular dichroism (CD) spectroscopy.
