Abstract:Two new beta-dihydroagarofuran sesquiterpene polyesters were isolated from Celastrus paniculatus. The structures were deduced using various 2D NMR techniques (H-H COSY, C-H COSY, COLOC, NOESY), NOE difference spectra and mass spectra. The positions of the ester moieties were located by 2D heteronuclear correlated spectra via long-range coupling, or combined with NOE experiments. Their stereochemical structures were studied and the complete assignments of the H-1 and C-13 chemical shifts were made.

Abstract:Three new beta-dihydroagarofuran sesquiterpene esters, 6 alpha-acetoxy-9 beta-benzoyloxy-1 beta-cinnamoyloxy-8 beta-butanoyloxy-beta-dihydroagarofuran (1), 6 alpha-acetoxy-9 beta-benzoyloxy-1 beta-cinnamoyloxy-8 beta-(2-methylbutanoyloxy)-beta-dihydroagarofuran (2), and 6 alpha-acetoxy-1 beta,8 beta-dibenzoyloxy-9 beta-hydroxy-beta-dihydroagarofuran (6), together with three known compounds (3-5) were isolated from the fruits of Celastrus orbiculatus THUNB. Their structures were elucidated on the basis of spectroscopic methods. Compound 4 showed moderate activity in inhibiting LPS-induced nitric oxide production in murine macrophage RAW264.7 cells with an IC(50) value of 43.7 microM.

Abstract:Two new insecticidal sesquiterpene polyol esters with a β-dihydroagarofuran sesquiterpene skeleton, celangulatin A (1), B (2), and two known compounds, celangulin IV (3) and V (4), were isolated from the leaves of Celastrus angulatus by bioassay-guided fractionation. Their chemical structures were elucidated mainly by analyses of the NMR and MS spectral data. Their insecticidal activities against Mythimna separate were demonstrated with the KD50 value of 68.5 and 215.8 μg g−1, respectively.

The new compounds were identified as 1 beta-picolinoyloxy-2 beta,6 alpha-diacetoxy-8 beta,13-diisobutanoyloxy-9 alpha-benzoyloxy-4 alpha-hydroxy-beta-dihydro- agarofuran (I), 1 beta-picolinoyloxy-2 beta,6 alpha-diacetoxy-8 beta,13-diisobutanoyloxy-9 alpha-furanoyloxy-4 alpha-hydroxy-beta-dihydroagarofuran (2), and 1 beta,6 alpha-diacetoxy-2 beta-nicotinoyloxy-8 beta,13-diisobutanoyloxy-9 alpha-benzoyloxy-4 alpha-hydroxy-beta-dihydro- agarofuran (3) on the basis of spectroscopic methods. Compounds 1-11 were all possessed cytotoxic activities against three human tumor cell lines by MTT assay.

Abstract:Three new sesquiterpene polyol esters with a beta-dihydroagarofuran skeleton, NW12 (1), NW27 (2), and NW31 (4), together with a known compound, NW30 (3), were isolated from the root bark of Celastrus angulatus. Their chemical structures were elucidated by analyses of MS and NMR spectral data. Preliminary insecticidal and antitumor activities of these compounds were evaluated. Compounds 1-4 showed moderate stomach toxicity against Mythimna separata, and their KD50 values were 673.6, 1121.3, 1720.0 and 548.6 microg/g, respectively. Only 1 and 3 exhibited slight antitumor activity against human breast cancer cell line (Bcap-37), and their IC50 values were all above 50 microM.
