Abstract:The reaction of B-hexoses, D-pentose and organic pentacoordinate 1, 3, 2-dioxaphospholene were investigated. The observation showed that there wa e obvious differences of reaction ratio, intermediate, and final products in the reactions between hexoses and pentose via pentacoordinate phosphorus compound. The results should give significant clue to the metabolic mechanism of carbohydrate phosphate in nature.

Abstract:Total synthesis of 4d-deoxy Lewisx pentasaccharide is described. O-(2,3,6-Tri-O-benzoyl-4-deoxy-α-d-xylo-hexopyranosyl)trichloroacetimidate was condensed with a diol of glucosamine to give regioselectively a β1→4 linked disaccharide, which was further fucosylated to a protected deoxy Lewisx trisaccharide. A highly stereo- and regioselective glycosylation of this trisaccharide onto a lactoside diol was performed to provide a pentasaccharide in excellent yield. After deprotection, this pentasaccharide afforded the target oligosaccharide.

Abstract:C-oligosaccharides are pharmacologically relevant because they are more hydrolysis-resistant than O-oligosaccharides. Despite indisputable advances, C-oligosaccharides continue to be underdeveloped, likely due to a lack of efficient and selective strategies for the assembly of the interglycosidic C-C linkages. In contrast, we, herein, report a versatile and robust strategy for the synthesis of structurally complex C-oligosaccharides via catalyzed C(sp3 )-H activations. Thus, a wealth of complex interglycosidic (2→1)- and (1→1)-C-oligosaccharides becomes readily available by palladium-catalyzed C(sp3 )-H glycoside glycosylation. The isolation of key palladacycle intermediates and experiments with isotopically-labeled compounds identified a trans-stereoselectivity for the C(sp3 )-H glycosylation. The glycoside C(sp3 )-H activation manifold was likewise exploited for the diversification of furanoses, pyranoses and disaccharides.

Abstract:A new additive, methyl(phenyl)formamide (MPF), is introduced for the glycosylation of 2-azido-2-deoxyglucose building blocks. A linear α-(1,4)-glucosamine tetrasaccharide was assembled to prove the utility of MPF. Next, a hexasaccharide fragment of the Pseudomonas aeruginosa exopolysaccharide Pel was assembled using a [2 + 2 + 2] strategy modulated by MPF. The used [galactosazide-α-(1,4)-glucosazide] disaccharide building blocks were synthesized using a 4,6-O-DTBS protected galactosyl azide donor.

Abstract:Two novel tricoordinate phosphorous saccharides, methyl 4,6-O-benzylidene-alpha-D-glucopyranoside 2,3-cyclic phosphite ethyl ester 3 and its mannoside analogue 5 were synthesized by the reaction of protected pyrannosides (1 and 4) with ethyl dichlorophosphite 2. Addition of 2,3-butanedione to 3 resulted in the formation of pentacoordinate phosphorous compound 6.

Abstract:AbstractC‐Oligosaccharide sind pharmakologisch relevant, weil sie hydrolysebeständiger sind als O‐Oligosaccharide. Trotz unbestreitbarer Fortschritte sind C‐Oligosaccharide nach wie vor wenig erforscht, meist aufgrund des Mangels an effizienten und selektiven Strategien für den Aufbau von C‐C‐Bindungen. Im Gegensatz dazu veröffentlichen wir hier eine vielseitige und robuste Strategie für die Synthese strukturell komplexer C‐Oligosaccharide via einer katalytischen C(sp3)‐H‐Aktivierung. Auf diese Weise ist eine Fülle von komplexen interglykosidischen (2→1)‐ und (1→1)‐C‐Oligosaccharide mittels einer Palladium‐katalysierter C(sp3)‐H‐Glykosid‐Glykosylierung einfacher zugänglich. Die Isolierung von Schlüsselintermediaten des Palladacyclus und experimentell isotopisch markierten Verbindungen ergaben eine trans‐ Stereoselektivität für die C(sp3)‐H‐Glykosylierung. Der glykosidische C(sp3)‐H‐Aktivierungsmechanismus wurde ebenfalls für die Diversifizierung von Furanosen, Pyranosen und Disacchariden angewandt.

Abstract:The amphiphilic random copolymer poly(β-d-glucose-co-1-octyl)phosphazene (PGOP) can undergo continuous morphological transitions in DMF-water mixed solvents. In this study, the ratio of glucose moieties to octyl moieties was controlled via a two-step thiol-ene reaction. As a result, polyphosphazenes with glycosyl functionalization degrees of 58.1% (PGOP-1), 74.1% (PGOP-2) and 87.0% (PGOP-3) were obtained. These amphiphilic polyphosphazenes self-assemble in both water and water-DMF mixtures. Several self-assembled morphologies including spheres, rods and vesicles were formed though careful control of the water content (WC) in the DMF solvent as well as of the hydrophilicity or hydrophobicity of the copolymers. We also found that an increase in the hydrophobic proportion led to faster morphological transitions at a constant WC. The thermodynamics of micellization were also studied by Isothermal Titration Calorimetry (ITC), and the strong hydrophobic interactions in PGOP-1 were demonstrated by their highly exothermic nature. These self-assemblies have potential applications in biosensing, lectin adsorption and drug loading with controlled release.

Abstract:Ophiopogonis Radix polysaccharides with various bioactivities have caught people's attention in the pharmaceutical and functional food industries. It is necessary to reveal their structures, chain conformations, and solvent behaviors. A neutral polysaccharide named ORP-1 with molecular weight of 3667 Da was obtained from Ophiopogonis Radix. It was composed of d-fructofuranose and d-glucopyranose in the ratio of 0.85:0.15. Methylation, FT-IR and NMR analysis indicated ORP-1 consisted of 2,6-linked-Fruf units as the main chain and 1-linked-Glcp residue at the end. Congo red assay showed ORP-1 had no triple-helix structure. The observation of TEM and AFM found ORP-1 could self-assemble to form colloidal aggregate in water. This phenomenon was verified using CMC determination and MD simulation. Furthermore, intermolecular hydrogen bonds and hydrophobic interactions would be the main forces driving the aggregate. These results provided reference for the study of the chain conformation and behavior of polysaccharides in aqueous solution.

Abstract:Polysaccharide is a kind of biological macromolecule with a complex structure, which plays an irreplaceable role in the organism. Due to the complex purification process of natural polysaccharide, chemical synthesis is an important means to obtain it. In this work, a green and efficient solid-phase polymerization method was used to synthesize glucan polysaccharide from unprotected glucose in one step under mild solid-phase conditions, using Hβ zeolites as acidic catalyst. The final polysaccharide product was characterized by high-performance liquid chromatography (HPLC) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) with a conversion of 35.0% and a maximum degree of polymerization of 26. More importantly, it was speculated that the glycosylation of unprotected glucose catalyzed by Hβ zeolites conforms to the Fischer glycosylation mechanism. In addition, the results of methylation analysis and one-dimensional, two-dimensional nuclear magnetic resonance (1D, 2D NMR) showed that the resulting polysaccharides were mainly linear, containing 10 different glycosidic linkages, and (1,6-Glcp), (1-Glcp), and (1,4-Glcp) were the most abundant linkage types among them.

Abstract:Abstract Attaining complete anomeric control is still one of the biggest challenges in carbohydrate chemistry. Glycosyl cations such as oxocarbenium and dioxanium ions are key intermediates of glycosylation reactions. Characterizing these highly-reactive intermediates and understanding their glycosylation mechanisms are essential to the stereoselective synthesis of complex carbohydrates. Although C-2 acyl neighbouring-group participation has been well-studied, the reactive intermediates in more remote participation remain elusive and are challenging to study. Herein, we report a workflow that is utilized to characterize rhamnosyl 1,3-bridged dioxanium ions derived from C-3 p -anisoyl esterified donors. First, we use a combination of quantum-chemical calculations and infrared ion spectroscopy to determine the structure of the cationic glycosylation intermediate in the gas-phase. In addition, we establish the structure and exchange kinetics of highly-reactive, low-abundance species in the solution-phase using chemical exchange saturation transfer, exchange spectroscopy, correlation spectroscopy, heteronuclear single-quantum correlation, and heteronuclear multiple-bond correlation nuclear magnetic resonance spectroscopy. Finally, we apply C-3 acyl neighbouring-group participation to the synthesis of complex bacterial oligosaccharides. This combined approach of finding answers to fundamental physical-chemical questions and their application in organic synthesis provides a robust basis for elucidating highly-reactive intermediates in glycosylation reactions.

Abstract:4-Deoxypentenosides (4-DPs) are versatile synthons for rare or higher-order pyranosides, and they provide an entry for structural diversification at the C5 position. Previous studies have shown that 4-DPs undergo stereocontrolled DMDO oxidation; subsequent epoxide ring-openings with various nucleophiles can proceed with both anti or syn selectivity. Here, we report the synthesis of α- and β-linked 4'-deoxypentenosyl (4'-DP) disaccharides, and we investigate their post-glycosylational C5' additions using the DMDO oxidation/ring-opening sequence. The α-linked 4'-DP disaccharides were synthesized by coupling thiophenyl 4-DP donors with glycosyl acceptors using BSP/Tf2O activation, whereas β-linked 4'-DP disaccharides were generated by the decarboxylative elimination of glucuronyl disaccharides under microwave conditions. Both α- and β-linked 4'-DP disaccharides could be epoxidized with high stereoselectivity using DMDO. In some cases, the α-epoxypentenosides could be successfully converted into terminal l-iduronic acids via the syn addition of 2-furylzinc bromide. These studies support a novel approach to oligosaccharide synthesis, in which the stereochemical configuration of the terminal 4'-DP unit is established at a post-glycosylative stage.

Abstract:A base-catalyzed O-phosphorylation of aldehyde/ketone with R P -menthyl phenylphosphinate afforded phosphonates. The mechanism was proposed as an intermolecular SN@P reaction of α-hydroxy phosphinate. In an intermediate, the C-P bond was transferred from the equatorial to axial position via Berry pseudorotation, then was cleaved with the leaving of a carbon anion. The P-retention and C-racemization could be satisfactorily explained by the mechanism.

Abstract:There is an urgent need to develop reliable strategies for the rapid assembly of complex oligosaccharides. This paper presents a set of strategically selected orthogonal protecting groups, glycosyl donors modified by a (S)-phenylthiomethylbenzyl ether at C-2, and a glycosyl acceptor containing a fluorous tag, which makes it possible to rapidly prepare complex branched oligosaccharides of biological importance. The C-2 auxiliary controlled the 1,2-cis anomeric selectivity of the various galactosylations. The orthogonal protecting groups, 2-naphthylmethyl ether (Nap) and levulinic ester (Lev), made it possible to generate glycosyl acceptors and allowed the installation of a crowded branching point. After the glycosylations, the chiral auxiliary could be removed using acidic conditions, which was compatible with the presence of the orthogonal protecting groups Lev and Nap, thereby allowing the efficient installation of 1,2-linked glycosides. The light fluorous tag made it possible to purify the compounds by a simple filtration method using silica gel modified by fluorocarbons. The set of building blocks was successfully employed for the preparation of the carbohydrate moiety of the GPI anchor of Trypanosoma brucei, which is a parasite that causes sleeping sickness in humans and similar diseases in domestic animals.

Abstract:The reactive intermediates of pentacoordinated phosphoranes with amino acids (P(5)-AA) as building blocks, which were obtained by the reaction of O-phenylene phosphorochloridate with N,O-bis(trimethylsilyl)amino acids, were linked to a solid-phase support containing a hydroxymethyl polystyrene functional group. The first amino acid residue was coupled to the solid-phase support after washing the resin with organic solvent. Repeating the procedure led to oligopeptides linked on the resin. A series of free oligopeptides including tetra-Gly, di-Val, tri-Val, di-Leu, di-Phe, and Phe–Leu were obtained after cleavage from solid-phase support. The structure of these oligopeptides were determined by IR, 1H NMR, FAB-MS, and HPLC.
