In vitro cleavage of mdrl mrna fragment by synthetic ribozyme (rz) and transcribed ribozyme from tna-trimming plasmid

Abstract:The failure of chemotherapy to cure more than a minority of tumors is predominantly due to drug resistance. This is most often accompanied by the overexpression of a membrane transport protein, P-glycoprotein, and its encoding mRNA. In order to reverse the resistant phenotype, we constructed a RNA-trimming plasmid pRG5mR3. In the plasmid three Rz genes, GR5 (5'-cis-Rz gene), mR (trans-Rz gene) and GR3 (3'-cis-Rz gene) are arranged in the order from 5' to 3' downstream from the T7 promoter. In vitro transcription of this plasmid shows that the trans-Rz can be trimmed to definite length by the cis-Rzs on both sides of the trans-fit. The transcribed Rz and synthetic Rz all possess catalytic activity that cleaves the 3'-end of the GUC sequence in codon 880 of the mdrl mRNA. We demonstrated that these constructed ribozymes are able to cleave a synthetic mdrl mRNA under physiological conditions in a cell-free system.
