Effect of intracerebroventricular or intrathecal injection of orphanin FQ on pain and acupuncture analgesia in rats

Abstract:Orphanin FQ (OFQ) is a newly discovered 17-amino-acid peptide which has been identified as an endogenous ligand of 'orphan receptor', a new member of the opioid receptor family. The role of OFQ in pain modulation is distinct from the classical endogenous opioid peptide (EOP). In the present study, the effects of OFQ on electrical stimulus-induced pain and acupuncture analgesia (AA) in rats were observed using tail-flick test. The results showed that intrathecal (ith) or intracerebroventricular (icv) administration of 0.1 ��g (0.055 nmol) OFQ had no effect on pain threshold, while 1��g OFQ (0.55 nmol) produced a distinct decrease. OFQ at both doses (0.1 or 1.0 ��g) antagonized AA whether given ith-ly or icv-ly with the latter route of administration being more prominent. When the rats were repeatedly treated with antisense oligonucleotide to block the synthesis of OFQ receptor, the pain threshold increased significantly; or the anti-AA effect was held in beyance when OFQ was administered and combined with acupuncture.
