Construction of a cDNA library of human hepatoma cells with apoptosis induced by adriamycin

Abstract:In order to identify some new genes related to apoptosis in hepatana cells, an expressing cDNA library was constructed based on the establishment of the model on apoptosis in human HCC-9204 hepatoma cell line. The apoptosis was induced by adriamycin at 20 uM/L for 3 hours in hepatoma cells, which could be significantly inhibited by actinomycin D at 0.5 ug/ ml. It suggests that some new large molecular proteins related to apoptosis were actively synthesized during the process of apoptosis in hepatoma cells. In brief, mRNAs were dirctly extracted from hepatoma cells treated by both adriamycin and actinomycin D as mentioned above, then ss- and ds-DNA were synthesized successfully. After removing the short cDNA fragments less than 200bp by spin-column chromatography, EcoR I adaptors were added on the remaining cDNA and they were recombined with phosphorylated lambda gt11-EcoR I arms. The recombined cDNAs were packaged in vitro and used to infect Y1090 host for amplification. The recombinant account was over 5 �� 105/ug in the library. It was confirmed that all randomly selected white plaques contained cDNA inserts which were over 1 Kb. This cDNA library laid a very important foundation for screening and identifying some new genes related to apoptosis in hepatoma cells.
