Expression Of Hyaluronan Receptor Cd44 And Rhamm In Human Gastric Cancers

Abstract:Hyaluronan receptors, CD44 and RHAMM, are known to be important in invasion and metastasis of some cancers but their expression patterns in human gastric cancers have not yet been well characterized. To address this issue, monoclonal antibodies against variant types of CD44 and antisera recognizing wild type or mutant type of RHAMM had been prepared and immunocytochemical characterization of CD44 and RHAMM molecules were performed on 58 cases of gastric cancer specimens, preneoplastic tissues and their normal counterparts. It is revealed that expression of standard CD44 (CD44s) and CD44 with variant exons 5 and 7 (v5 and v7) are common in normal gastric mucosa (68% and 76%), but less frequent in cancer cells (44% and 48%); v8vl0 are almost absent in all types of samples so far checked; v6 is negative in normal mucosa (0%), 1/2a (4.4%) in preneoplastic changes but 12/58 (21%) in cancers; among v6 positive cancers, 10 are poorly- or un-differentiated ones. The positivity of wild type RHAMM are 34% in normal, 6% in preneoplastic and only 3% in cancer samples, mutant type RHAMM can be detected more frequently in tumors (91%) and preneoplastic changes (82%) than in normal tissues (32%). Our results indicate that there is no distinst differentce of CD44v expression between the normal and the malignant except v6, which seems linked to less differentiated status of stomach cancers. Unlike CD44v, mutant RHAMM is more abundent in preneoplastic and neoplastic cells, suggesting its potential usefulness as a malignant biomarker for gastric cancers.
