Abstract:It is well established that luhulin and dynein are involved in many essential functions in animal cells, hul it not well known in plant cells. In the present paper. lubulin and dynein-like protein were investigated in the ungerminated pollen of Ginkgo hiloba. a typical gyrnnosperm. Tubulin was purified by a combination of acetone powder preparation and chromaiography from ginkgo pollen The average yield of tubuhn was 2 mg per 100 g of polien grain The purified lubulin was eleclrophoretically homogeneous II was composed of two subunits and was resolved as two spots on 2D-electrophoresis. The apparent molecular weighi of the two subunits was about 54 KO and 52 KI) respectively. It "as also demonsirated that lubulin from ginkgo pollen was immunochemically related-with animal brain luhulin. The purified tubulin could be polymerized lo microtubular aggregates in the presence of taxol and GTP in vitro. On western blots of Ihe total soluble pollen proteins, the 400 KI) polypeptide could be recognized by the polyclonal antibody, a mouse antiserum lo a synthetic polypeptide reproducing the sequence of ihe most conserved ATP-binding site (GI'AG TGKT) among dynein heavy chains . showing that Ihe 400 KD polypeptide contained the similar AIPhinding site as that in animal cells. The sedimentation analysis demonslraled that the sedimentation coefficient of the polypeptide was about 12 S. the fraction rich in 400 KD polypeplide. obtained from a 5-25% sucrose gradient sedimentation, had ATPase activity which could he stimulated 1.5-fold by addition of bovine brain microtubules The results showed that tubulin and dynein-like proteins were present in ginkgo pollen with the similar biochemical properties as that in animal cells.
