Influence of medium pH and polyethylene glycol on the artificial pollen germination in Brassica

Abstract:Pollen of Brassica campestris var. purpurea (a vegetable crop) was cultured in a medium developed by Roberts et al (1983)containing 15% sucrose. The effect of pH value of the medium ranging from 5. 16 to 8. 97 was studied. The results showed that the best germination was obtained at pH8. 47. Complete substitution of sucrose by 20% PEG exhibited excellent effect on both pollen germination (up to 70. 8%) and tube growth (prolonged for 20 hours). Then this modified R medium composed of Roberts' salts and 20% PEG, pH8. 5, was adopted to the pollen of other Brassica species. Similar positive results were obtained in B. campestris (a rape seed plant) and in B. chinensis, but were not ideal in B. napus. Pollen incubated in this modified R medium, if pollinated onto the cut surface of style, could germinate well and their pollen tubes could grow down the stlye into the ovary. This might provide a biotechnological means for Brassica species using hydrated pollen as the carrier of foreign genes. In order to compare the artificial with natural condition of pollen germination, a microelectrode was used to test the pH value of pollen and stigma surface. The mean pH value was 6.12 in the former case and 5. 87 in the latter. This means that the optimal alkaline condition for artificial pollen germination was not a simple mimic of natural condition. The mechanisms of the influence of pH value and PEG on pollen germination are discussed.
