Botulinum toxin time-dependently inhibits gastrocnemius twitch induced by electric stimulating sciatic nerves

Abstract:Objective To analyze the effects of botulinum toxin A(BoNT-A) on gastrocnemius twitch induced by electric stimulating sciatic nerve(ESSN) in toad.Methods Toads were randomly divided into two groups,suxamethonium chloride(SC) treated group(n = 44),which was consisted of Ringer′s solution(RGS,0.2 mL) injection(the left gastrocnemius) and SC injection(0.04 mg,the right side),BoNT-A treated group(n = 42),which was consisted of RGS injection(the left gastrocnemius) and BoNT-A injection(0.75-5.0 U,the right side).The tension test model was set up by a gastrocnemius with sciatic nerve separated from body after toads treated with various dose BoNT-A,SC and RGS for 1,4,7,10,24 h and 48 h.The isometric contractile force generated from gastrocnemius was induced by ESSN(0.5 Hz,2 V) for twitching,and then the contractile tension was recorded with polygraphs.Results SC suppressed twitching contraction of gastrocnemius induced by ESSN for 1 h,but was ineffective for 4,7,10,24 h and 48 h compared with RGS.The dosage of 1.25,2.5,5.0 U BoNT-A injection significantly reduced the contraction of gastrocnemius from 1 to 48 h(P < 0.05 or P < 0.01) compared with RGS control.The inhibitory effects of BoNT-A from 10 to 48 h was greater than that from 1 to 7 h(P < 0.05).The effective dosage of BoNT-A was 1.25 U.Conclusion BoNT-A time-dependently inhibits gastrocnemius twitch induced by ESSN,and its effective dosage is 1.25 U.
