Abstract:Titanocene alanine complex, Cp2Ti（O2CCH（CH3）NH3+Cl-）2, has been found to have in vitro antitumor activity against Ehrlich ascites tumor cells. Study of the interaction between Cp2Ti（O2CCH（CH3）NH+Cl-）2 and DNA by UV-VIS spectra, fluorescence spectra, agarose gel elec-trophoresis reveals that Cp2Ti（O2CCH（CH3）NH3 Cl-）2 forms a stable compound with DNA in molar ratio 1:1 and reduces the migration rate of ccc-DNA band and oc-DNA band. However, unlike cis-platin, the complex does not induce nicking of DNA even after a long incubation at a high ratio of the complex to DNA. The results suggest that titanocene-based antitumor agents have different mechanism of action from cisplatin-like antitumor agents.

Abstract:Dioxycyclobutenedione-(1,2-cyclohexanediamine)platinum(II), (R,R-DC-Pt) was found to have stronger cytotoxicity against six cancer cell lines than cisplatin and its DNA interactions was studied by calorimetric measurements, (13)C NMR. The binding specificity study of DNA base with R,R-DC-Pt was conducted by HPLC. To understand the molecular mechanism of R,R-DC-Pt with stronger cytotoxicity than that of cisplatin, we studied R,R-DC-Pt interaction with an oligonucleotide, d(ACCACGTGGT)(2), which contained c-H-ras gene encoding GGT by NMR spectroscopy. The oligomer DNA double helix was destroyed almost completely upon the R,R-DC-Pt binding. However under the same condition, the cisplatin binding with DNA was not so affected, and instead another conformation was formed, which suggests that larger damage to DNA can be induced by R,R-DC-Pt complex than that by cisplatin.

Abstract:Whereas most conventional DNA probes are flat disklike aromatic molecules, we explored the possibility of developing quadruplex sensors with nonplanar conformations, in particular, the propeller-shaped tetraphenylethene (TPE) salts with aggregation-induced emission (AIE) characteristics. 1,1,2,2-Tetrakis[4-(2-triethylammonioethoxy)phenyl]ethene tetrabromide (TPE-1) was found to show a specific affinity to a particular quadruplex structure formed by a human telomeric DNA strand in the presence of K(+) ions, as indicated by the enhanced and bathochromically shifted emission of the AIE fluorogen. Steady-state and time-resolved spectral analyses revealed that the specific binding stems from a structural matching between the AIE fluorogen and the DNA strand in the folding process. Computational modeling suggests that the AIE molecule docks on the grooves of the quadruplex surface with the aid of electrostatic attraction. The binding preference of TPE-1 enables it to serve as a bioprobe for direct monitoring of cation-driven conformational transitions between the quadruplexes of various conformations, a job unachievable by the traditional G-quadruplex biosensors. Methyl thiazolyl tetrazolium (MTT) assays reveal that TPE-1 is cytocompatible, posing no toxicity to living cells.
Anticancer activity and mode of action of titanocene C

Abstract:Titanocenes constitute a class of metal-based anticancer agents that seem to display a mode of action distinct from that of platinum complexes and to be more tolerable with a differing spectrum of activity. In the present study, titanocene C (bis-(N,N-dimethylamino-2(N-methylpyrrolyl)-methyl-cyclopentadienyl) titanium(IV) dichloride) was shown to exhibit antiproliferative activity against human tumor cell lines with a mean IC₅₀ value of 48.3 ± 32.5 µM. In particular, high activity was found against small cell lung cancer (SCLC) cell lines with a profile different from cisplatin. Titanocene C induced cell cycle arrest at the G1/0-S interphase. Cross-resistance to either cisplatin or oxoplatin, respectively, was low for titanocene C and absent for titanocene Y in variant HL-60 cell lines. Alterations in gene expression of NCI-H526 SCLC cells induced by titanocene C were investigated using genome-wide expression arrays. Downregulation was found for genes coding for topoisomerases I and IIα, histones of the HIST1H4 cluster, enzymes involved in glycolysis, components of the cytoskeleton and vesicular transport, among others. In contrast, expression of genes involved in apoptosis, stress response, particularly members of the metallothionein gene cluster 1, DNA damage and growth factors was upregulated following exposure to titanocene C. Approximately 50% of those genes downregulated by titanocene C and cisplatin were concordant, including the previously identified markers of cisplatin-sensitivity, tubulin and stathmin, indicating partial overlap of the pathways affected by these metal complexes. The present findings point helicases/topoisomerases and HIST1H4 core histones out as targets of titanocene C and metallothioneins as putative main effectors of drug resistance.
Titanium(IV) Targets Phosphoesters on Nucleotides: Implications for the Mechanism of Action of the Anticancer Drug Titanocene Dichloride

Abstract:Reactions between the anticancer drug titanocene dichloride (Cp2TiCl2) and various nucleotides and their constituents in aqueous solution or N,N-dimethylformamide (DMF) have been investigated by 1H and 31P NMR spectroscopy and in the solid state by IR spectroscopy. In aqueous solution over the pH* (pH meter reading in D2O) range 2.3–6.5, CMP forms one new species with Ti(IV) bound only to the phosphate group. In acidic media at pH*<4.6, three species containing titanocene bound to the phosphate group of dGMP, AMP, dTMP and UMP are formed rapidly. The bases also appear to influence titanocene binding. Only one of these Ti(IV)-bound species can be detected in the pH* range of 4.6–6.5 in each case. The order of reactivity towards Cp2TiCl2(aq) at pH* ca. 3 is GMP>TMP≈AMP>CMP. At pH*>7.0, hydrolysis of Cp2TiCl2 predominated and little reaction with the nucleotides was observed. Binding of deoxyribose 5′-phosphate and 4-nitrophenyl phosphate to Cp2TiCl2(aq) via their phosphate groups was detected by 31P NMR spectroscopy, but no reaction between Cp2TiCl2(aq) and deoxyguanosine, 9-ethylguanine or deoxy-D-ribose was observed in aqueous solution. The nucleoside phosphodiesters 3′,5′-cyclic GMP and 2′,3′-cyclic CMP did not react with Cp2TiCl2(aq) in aqueous solution; however, in the less polar solvent DMF, 3′,5′-cyclic GMP coordination to {Cp2Ti}2+ via its phosphodiester group was readily observed. Binding of titanocene to the phosphodiester group of the dinucleotide GpC was also observed in DMF by 31P NMR. The nucleoside triphosphates ATP and GTP reacted more extensively with Cp2TiCl2(aq) than their monophosphates; complexes with bound phosphate groups were formed in acidic media and to a lesser extent at neutral pH. Cleavage of phosphate bonds in ATP (and GTP) by Cp2TiCl2(aq) to form inorganic phosphate, AMP (or GMP) and ADP (or GDP) was observed in aqueous solutions. In addition, titanocene binding to ATP was not inhibited by Mg(II), but the ternary complex titanocene-ATP-Mg appeared to form. These reactions contrast markedly with those of the drug cisplatin, which binds predominantly to the base nitrogen atoms of nucleotides and only weakly to the phosphate groups. The high affinity of Ti(IV) for phosphate groups may be important for its biological activity.

Abstract:Three new transition metal complexes [Mn2(DCA)2(bipy)2]·5H2O (1), [M2(DCA)2(bipy)2(H2O)]·10H2O(M=Ni(II)(2);Zn(II)(3)), (DCA=demethylcantharate, 7-oxabicyclo[2,2,1]heptane-2,3-dicarboxylate, C8H8O5) were synthesized and characterized by elemental analysis, molar conductance, infrared spectra and X-ray diffraction techniques. Each metal ion was six-coordinated in complexes. Complex 1 has a Mn2O2 center. Complexes 2 and 3 have asymmetric binuclear structure. Great amount of intermolecular hydrogen-bonding and π-π(*) stacking interactions were formed in these complex structures. The DNA-binding properties of complexes were investigated by electronic absorption spectra and viscosity measurements. The DNA binding constants Kb/(Lmol(-1)) were 1.71×10(4) (1), 2.62×10(4) (2) and 1.59×10(4) (3) at 298 K. The complexes could quench the intrinsic fluorescence of bovine serum albumin (BSA) strongly through static quenching. The protein binding constants Ka/(L mol(-1)) were 7.27×10(4) (1), 4.55×10(4) (2) and 7.87×10(4) L mol(-1) (3) and binding site was one. The complexes bind more tightly with DNA and BSA than with ligands. Complexes 1 and 3 had stronger inhibition ratios than Na2(DCA) against human hepatoma cells (SMMC-7721) lines and human gastric cancer cells (MGC80-3) lines in vitro. Complex 3 showed the strongest antiproliferative activity against SMMC-7721 (IC50=29.46±2.12 μmol L(-1)) and MGC80-3 (IC50=27.02±2.38 μmol L(-1)), which shows potential in anti-cancer drug development.

Abstract:A new cobalt(II) complex, [Co(C(3)H(4)N(2))(C(8)H(8)O(5))(H(2)O)(2)]center dot 2H(2)O, of demethylcantharate (7-oxabicyclo[2,2,1]heptane-2,3-dicarboxylate, C(8)H(8)O(5)) with imidazole has been synthesized from cobalt chloride, demethylcantharidin (NCTD) and imidazole. The complex was characterized by elemental analysis, IR, and X-ray single crystal diffraction. The complex crystallized in the monoclinic crystal system and P2(1)/m space group with a=0.634790(10)nm, b = 0.963030(10)nm, c = 1.221770(10)nm, alpha=90 degrees, beta=95.9700(10)degrees, gamma=90 degrees, V=0.742844(15)nm(3), M(r)=383.22, D(c)=1.713 g cm(-3), Z=2, F(0 0 0)=398, mu=1.206 mm(-1), the final R=0.0291, and wR=0.0837 [I>2 sigma(I)]. The interaction of the complex with deoxyribonucleic acid (DNA) was studied by electronic absorption spectra, fluorescence spectra, and viscosity measurements, which indicate that the complex binds to calf thymus DNA through a partially intercalative mode. The binding constant K(b) for the complex was 2.62 x 10(4) L mol(-1). The antiproliferation activity test showed that the complex has high antiproliferative ability against human hepatoma cells SMMC7721 (with IC(50) being 42.8 +/- 0.9 mu mol L(-1)) and human lung cancer cells A549 (with IC(50) being 65.1 +/- 3.2 mu mol L(-1)). The inhibition rates of the complex are much higher than those of NCTD.

Abstract:In this study, a thiosemicarbazone derivative (E)-2-((1,4-dihydroxy-9,10-anthraquinone-2-yl)methylene)-N-(4-fluorophenyl)hydrazinecarbothioamide (DAFPT) was synthesized, and the interaction of DAFPT with calf thymus DNA (ctDNA) was explored using ethidium bromide (EB) as a fluorescence probe. The binding mode between DAFPT and ctDNA was investigated by UV absorption spectroscopy, fluorescence spectroscopy and molecular docking. The fluorescence quenching mechanism of EB-ctDNA by DAFPT might be a combined quenching type. Thermodynamic parameters showed that the reaction was spontaneous. According to ionic strength, fluorescence polarization and melting temperature (Tm) curve results, DAFPT-ctDNA interaction was groove binding. The molecular modeling results indicated that DAFPT could slide into the A-T rich region of ctDNA.
Interaction Between DNA and Terbium(III)-Ciprofloxacin Fluorescence Complex and New Tissue Staining Method

Abstract:A new lanthanide complex that gives off green fluorescence upon exciting by UV radiation was synthesized by using terbium chloride and ciprofloxacin. The complex binds firmly on DNA and induces conformation changes of DNA based on the analysis of FTIR and fluorescence spectra. Strong fluorescence from terbium ions can be observed from the resultant composite under UV light. Thus, it is possible that the terbium ciprofloxacin complex can be developed as a fluorescent dye in biological and pathological studies. The results indicate that the complex is concentrated on the nuclei regions in the sections of oral tissues and the nuclei are highlighted under fluorescent microscope.

Abstract:The complex of a derivative of P-cyclodextrin, that is mono[6-deoxy-6-(2-butenedinitrile-2,3-dimercapto sodium salt)]-beta-eyelodextrin (6-mnt-beta-CD), with titanocene (titanocene di[mono [6-deoxy-6-(2-butenedinitrile-2,3-dimercapto)]-beta-cyclodextrin], Cp2Ti[6-mnt-beta-CD](2)) has been synthesized and characterized by IR spectroscopy, UV spectroscopy, elemental analysis, thermogravimetry, H-1- and C-13-NMR spectroscopy. Abstract:The new trinuclear tridentate Pt-II complex [Pt3Cl3(hptab)](3+) (1; hptab = N,N,N',N',N '',N ''-hexakis(2-pyridylmethyl)-1,3,5-tris(aminomethyl)benzene) exhibits promising cytotoxic effects in human and mouse tumor cells including those resistant to conventional cisplatin (Dalton Trans. 2006, 2617; Chem. Eur. J. 2009, 15, 5245). The present study is focused on the molecular pharmacology of 1, in particular on its interactions with DNA (which is the major pharmacological target of platinum antitumor drugs), to elucidate more deeply the mechanism underlying its antitumor effects. Results obtained with the aid of methods of molecular biophysics and pharmacology reveal new details of DNA modifications by 1. Complex 1 binds to DNA forming in the absence of proteins and molecular crowding agents mainly bifunctional intrastrand cross-links. In these DNA adducts all three Pt-II centers of 1 are coordinated to DNA base residues, which leads to extensive conformational alterations in DNA. An intriguing aspect of the DNA-binding mode of this trinuclear Pt-II complex 1 is that it can cross-link proteins to DNA. Even more interestingly, 1 can cross-link in the presence of molecular crowding agent, which mimics environmental conditions in cell nucleus, two DNA duplexes in a high yield-a feature observed for the first time for antitumor trinuclear platinum complexes. Thus, the concept for the design of agents capable of forming intramolecular tridentate DNA adducts, DNA-protein and interduplex DNA-DNA cross-links based on trinuclear tridentate Pt-II complexes with semirigid aromatic linkers may result in new compounds which exhibit a variety of biological effects and can be also useful in nucleic acids research.
Fluorescence Characteristic of Terbium-Ciprofloxacin Complex and Its Application

Abstract:On binding to deoxyribonucleic acid, the complex of terbium-ciprofloxacin (Tb(3+)-CIP) increases its fluorescence quantum efficiency. Based on this, an easy, rapid and sensitive method for the determination of DNA was developed. Like ethidium bromide (EB), the complex can be intercalated into DNA bases, but it is non-poisonous. Determination can be made at pH 7.0, where the native structure of DNA is not disrupted. The maximum emission is at 545 nm with excitation at 325 nm. This method has good sensitivity (2.8 x 10(-9) mol x L(-1) of ctDNA), high selectivity and a wide linear range (4.3 x 10(-7)-3. 0 x 10(-5) mol x L(-1) of ctDNA). This complex was also employed for the clinical examination of DNA in whole blood of tumor patients. Ten tumor patients and 10 normal persons were enrolled in this study. It was showed that significant difference existed between control group and tumor group (p < 0.05).

Abstract:The design of multinuclear Pt(II) complexes with novel structural feature is very important in the search for new anticancer agents. In this work, a dinuclear platinum(II) complex [Pt-2(DTBPA)Cl-2] (II) [DTBPA = (2,2'-(4,11-dimethyl-1,4,8,11-tetraazacyclotetradecane-1,8-diyl)bis(N-(2-(pyridin-2-yl)ethyl)acetamide))] was synthesized via two different methods and characterized by NMR, IR, electrospray mass spectrometry and elemental analysis. It binds to calf thymus DNA (CT-DNA) and induces its conformational changes. Gel electrophoresis data show that complex II leads to a clear decrease of migration rate of the negatively supercoiled band (form I) of supercoiled pUC19 plasmid. The cytotoxic activity of the complex II was tested against human cervical cancer cell line (Hela) and human ovarian carcinoma cell line (Caov-3) and compared with cisplatin. Abstract:A complex of Tb(3+), acrylic acid (AA), and 1, 10-phenanthroline (Phen) was synthesized. The structure and fluorescence of Tb(AA)(3)Phen center dot H(2)O was characterized with elemental analysis, FT-IR, and fluorescence spectroscopy. A novel copolymer containing rare earth complex, poly (METAC-co-NIPAm-co-Tb(AA)(3)Phen center dot H(2)O) (PMNTb), was prepared by free radical copolymerization in methanol with azodiisobutyronitrile (AIBN) as initiator. (1)H NMR, fluorescence spectroscopy, UV-vis spectroscopy, and TEM were used to characterize this copolymer. The interaction of PMNTb with DNA was studied by fluorescence spectroscopy, UV-vis spectroscopy, and agarose gel electrophoresis. The results of fluorescence, UV-vis absorption, and agarose electrophoresis indicated that the PMNTb could interact with DNA in an electrostatic bonding mode and the bonding constant was 3.4 x 10(5) L mol(-1). The TEM observation showed that the PMNTb could form micelles in water solution; the efficient complexation of PMNTb with DNA occurred. The results of in vitro cytotoxicity indicated that PMNTb had good biocompability. These results laid the foundation for the further study in potential gene detective reagent and gene delivery carrier.

Abstract:The DNA binding and cross-linking modes of a trinuclear platinum Complex [Pt3Cl3(hptab)][ClO4](3) (1; hptab = N,N,N',N',N '',N ''-hexakis(2-pyridylmethyl)-1,3,5-tris(aminomethyl)benzene) and its dinuclear analogue [Pt2Cl2(m-tpxa)]Cl-2 (2; m-tpxa = N,N,N',N'-tetra(2-pyridylmethyl)-m-xylylene diamine) are reported and compared. The adducts of 1 and 2 with 18-mer duplex N1, 5'-d(GAAGAAGTCA-CAAAATGT)-3'center dot 5'-d(ACATTTTGT-GACTTCTTC)-3', have been characterized by means of denaturing polyacrylamide gels, Maxam-Gilbert sequencing, and MALDI-TOF mass spectrometry combined with enzymatic degradation to obtain insights into Structural features responsible for the differences in their antitumor activities. The cytotoxic-active complex 1 readily forms various DNA adducts, such as through 1,3- and 1,4-intrastrand cross-links, and in particular, the unique and unprecedented interstrand cross-linked triadducts. In contrast, the cytotoxic-inactive complex 2 preferentially forms 1,4-intrastrand rather than 1,3-intra- and -interstrand cross-links. Digestion of the DNA adducts of 1 shows that the cleavage is completely blocked at one nucleotide before the cross-linked guanine residues on the opposite strand, a feature that appears to be unprecedented in antitumor platinum complexes. In the case of 2, the cleavage bypasses the first platinated guanine site and stops at one nucleotide prior to the second platinated site, confirming that very few 1,3-intrastrand cross-links are formed by 2. These results are supported by molecular-modeling studies of intra- and interstrand cross-links Of duplex NI with 1 and 2. The remarkable differences between 1 and 2 in DNA binding and cross-linking provide mechanistic insights into their different cytotoxicity against the tumor cell lines; these insights are useful for designing future antitumor agents.

Abstract:The DNA binding ability and binding mode of platinum complexes are crucial factors that govern their cytotoxic activity. In this work, circular dichroism spectroscopy, gel electrophoresis and MALDI-TOF MS spectrometry combined with enzymatic degradation have been used to elucidate the role of bridging ligands in DNA-binding ability and cross-linking patterns of two dinuclear antitumour active platinum(II) complexes, {[cis-Pt(NH(3))(2)Cl](2)L1}(NO(3))(2) (1, L1= 4,4'-methylenedianiline) and {[cis-Pt(NH(3))(2)Cl](2)L2}(NO(3))(2) (2, L2 = alpha,alpha'-diamino-p-xylene). Although both complexes have two cis-diammine-Pt(II) moieties (1,1/c,c), complex 1 exhibits much higher DNA-binding ability than complex 2. The former readily forms both 1,3- and 1,4-intrastrand cross-links with DNA oligonucleotides, while the latter preferentially forms 1,4- rather than 1,3-intrastrand cross-links. Cytotoxicity studies against a human non-small-cell lung cancer cell line (A549) demonstrate that complex 1 has higher activity than 2. These results show that the linker properties play a critical role in controlling the DNA-binding and cross-linking abilities and in modulating the cytotoxicity of dinuclear platinum complexes.

Abstract:The DNA interstrand cross-links of the antitumor drug {[cis-Pt(NH3)(2)Cl](2)(4,4'-methylenedianiline)}(2+) (1) play a prevalent role in its antitumor effects. Complex 1 forms DNA long-range interstrand cross-links uniquely in the 3'-3' direction. Conformational distortions induced by these interstrand cross-links in DNA represent a potential structural motif for recognition by high-mobility-group proteins.
