Abstract:Background S100P is a member of the S100 family of calcium-binding proteins, and it participates in pathophysiological events, such as tumor growth and invasion. Based on the striking similarities between trophoblast cells and tumor cells with regard to proliferative and invasive properties, we raised the question of whether and how S100P expresses in trophoblast cells during development. This study aimed to investigate the expression pattern of S100P in the human placenta during pregnancy development. Materials and Methods In this experimental study, we collected 16 first-trimester placental tissues, 10 second-trimester placental tissues, and 12 term placentas. The mRNA expression levels of S100P were detected by reverse-transcription-polymerase chain reaction (RT-PCR) and quantitative real-time PCR, the protein expression levels were detected by western blot, and the localization of S100P was measured by immunohistochemical staining. The values obtained from PCR and western blot analysis were expressed as the mean ± SD. Levene’s test was used to test equal variances, and one-way analysis of variance (ANOVA) was used to evaluate differences between groups. Results Protein and mRNA expression of S100P could be detected in placenta during pregnancy, with minor higher levels in first-trimester (p > 0.05). Immunohistochemical staining revealed that S100P protein was strongly expressed in syncytiotrophoblasts, and moderate expression was detected in villous cytotrophoblasts and cytotrophoblast columns. The S100P protein was localized to both cytoplasm and nuclei in syncytiotrophoblasts, while it only existed in the cytoplasm of cytotrophoblasts. Conclusion S100P was strongly detected in human placenta during pregnancy. The specific expression and distribution of S100P in human placenta throughout gestation suggested that S100P function might vary with its location in the placenta.

Abstract:ABSTRACT Objective: To study the localization and quantification of laminin (LN) and laminin receptor (LNR) in the human placental villi during early, middle and term pregnancy. Methods: LN and LNR in the human placental villi was measured using immunohistochemical ABC method and Q500 image analysis system. Results: LN positive immunostaining was found in placental villous epithelial basement membranes and vascular endothelial basement membranes. LNR positive immunoreactive substance was localized in the villous epithelial cellular plasma (including cytotrophoblast and syncytiotrophoblast ) and syncytiotrophoblastic cellular surfaces, but not in the nucleus. Along with placental development, the amount of LN and LNR decreased gradually. Conclusion: LN and LNR exist in the human placental villi, and their relative content decreased following the placental growth.

Methods: Immunohistochemical ABC method was used to the experiment.Results: Both syncytiotrophoblast and cytotrophoblast,stromal cells, capillary endothelium, lymphocytes in capillarycavity,and all cells of blood islet in early human placentaswere all shown SPR immunoreactivity in cytoplasm withnegative nuclei. Conclusion: SP produced by placental villimay take part in regulation of synthesizing and releasing ofplacental hormone, and regulation of capillary developmentand microcirculation in placental villi, and regulation of fetalimmune fuction.

Abstract:The localizations of mRNAs encoding LHβ and FSHβ in porcine pituitary were investigated by insitu hybridization technique. Biotinylated porcine LHβ and FSHβ cDNA probes were used on frozen sections of paraformaldehyde-fixed pituitary specimens. Hybridizations to both mRNAs were observed specifically in cytoplasm with unstained nuclei. Furthermore, cells hybridized for LHβ mRNA were demonstrated to be identical to those for FSHβ mRNA. This study provided the first morphological evidence that both gonadotropin β genes are expressed in the same cell.
Studies on the localization of 5-hydroxytryptamine and its receptors in human placenta

Abstract:The localization of 5-hydroxytryptamine (5-HT) and its receptors in human placenta was studied under light and electron microscopy using immunohistochemistry and in situ hybridization. The syncytiotrophoblast, cytotrophoblast, stromal cells and decidual cells in human placentae all appeared to be 5-HT immunoreactive. The 5-HT immunoreactive material was distributed in cytoplasm with negative nuclei. The 5-HT immunoreactive material was also found in capillary endothelium. Trophoblast cells cultured in serum-free medium also showed 5-HT immunoreactivity in the cytoplasm. Fetal white blood cells and both syncytiotrophoblast and cytotrophoblast in human placenta showed 5-HT receptor immunoreactivity and 5-HT,A receptor mRNA hybridized signal was also detected in cytoplasm. The stromal cells and capillary endothelium in placental villi and maternal decidual cells all showed 5-HT receptor immunoreactivity in cytoplasm. The small flattened vesicles and large dense cored vesicles within trophoblast cells showed electron-dense 5-HT receptor immunoreactivity using immunoelectron microscopy. These results suggest that human placenta may not only produce 5-HT but also be a 5-HT target organ, and that 5-HT may not only play roles in placental development and pregnancy maintenance by paracrine and autocrine interactions but may also take part in regulating fetal development. Abstract:The localization of IL-6 at light microscopic levels and its quantitative analysis in human placentas were studied by using immunohistochemistry. Both syncytiotrophoblast and cytotrophoblast in placental villi, white blood cells in villose capillary cavity stromal cells and capillary endothelium present IL-6 immunoreactivity in cytoplasm. Using a quantitative immunohitochemical method, the amounts of IL-6 in placenta were low at the 6th week of gestation, increased saliently and peaked at the 7th, then reduced gradually during the rest of the gestation period and maintained the same level as that in the 6th week of gestation. This changing trend was paralleled with that of GnRH in our previous studies. These results revealed that IL-6 could be produced by the human placenta and may take part in the regulation of placental hormone releasing.
[Expression and Significance of STOX1 in Villi and Placenta at Different Stages of Normal Gestation].

Abstract:Objective: To investigate the expression and significance of STOX1 in different stages of gestation villi and placenta. Methods: Totally 137 cases of normal villi and placenta of pregnant women were collected from the Department of Obstetrics of Shanghai Pudong Hospital from October 1(st) 2015 to February 28(th) 2018, including 64 cases of early pregnancy (early pregnancy group) which consists of 32 cases of 5-7(+6) weeks gestation (early pregnancy group A) and 32 cases of 8-11(+3) weeks gestation (early pregnancy group B), 28 cases of 14-26 weeks gestation(middle pregnancy group) and 45 cases of 37-41 weeks gestation (late pregnancy group). The expression and localization of STOX1 mRNA and protein in placenta were evaluated by RT-qPCR, Western blotting and immunohistochemistry. Results: (1)STOX1 was positively expressed in the cytotrophoblasts and syncytiotrophoblasts as well as interstitial and vascular endothelial cells of all groups. (2)STOX1 mRNA expression in each group was significantly different (P<0.05), the lowest was in the early pregnancy group A(0.007 8±0.000 4), which increased along with the progression of gestational age(P<0.05),and reached the highest level in the third trimester(0.064 4±0.001 3). (3)The protein level of STOX1 in different stages of normal pregnancy was 0.53±0.20 in early pregnancy group A;0.62±0.37 in early pregnancy group B;0.70±0.03 in middle pregnancy group and 0.81±0.04 in late pregnancy group respectively; which was positively related with the progression of gestational age (P<0.05). Conclusion: The expressions of STOX1 is gradually increasing along with the normal pregnancy progression, suggesting that it might be involved in proliferation, differentiation and infiltration and (or) apoptosis of trophoblast cells and the development of the placenta.

Abstract:Objective:To investigate the cell source secreting the granulocyte monocyte and granulocyte colony stimulating factors (GM CSF, G CSF)in human placenta. Methods:The frozen sections of placental tissue from 19 health pregnant women were analyzed by using anti GM CSF and anti G CSF monoclonal antibodies and immunohistochemical technique. Nasal polyps were negative control. Results:The placental trophoblastic cells had strong positive reactions while the control groups showed negative reactions. Conclusion:Trophoblastic cells in the placental tissue may be the main production source of GM CSF and G CSF in human cord blood.

Abstract:SWAP-70 has been demonstrated as a multiple functional signaling protein involved in formation of membrane ruffling induced by signal cascade of tyrosine kinase growth factor receptors. In the present study, the spatial and temporal expression pattern of SWAP-70 on human fetomaternal interface was investigated using specimens collected from tubal and normal pregnancies by in situ hybridization, immunohistochemistry, and Western blotting. Data showed an intense expression of SWAP-70 in trophoblasts at weeks 3-6 of fallopian implantation and at weeks 6-7 of normal pregnancy. The most intense expression was exhibited by those highly motile and invasive extravillous trophoblasts. From gestational week 8 on, the level of SWAP-70 in trophoblasts decreased significantly, and the signal was restricted in villous cytotrophoblast cells. In the in vitro cultured human trophoblast cell line, B6Tert-1, colocalization of SWAP-70 with F-actin was verified. Data in human placenta were similar to what we recently reported on rhesus monkey fetomaternal interface. Our results suggest that SWAP-70 may be involved in regulating migration and invasion of trophoblast cells during the processes of embryonic implantation and placentation in primates.

Abstract:Objective: To prove the expression of chemokine CCL2 and its receptor CCR2 in human first-trimester placental villi. Methods: The transcription and translation of chemokine CCL2 and its receptor CCR2 in the first-trimester placental villi were analyzed by semi-quantitative RT-PCR and immunohistochemistry, respectively. Results: CCR2 and its ligand CCL2 were expressed moderately in human first-trimester placental villi. Conclusion: CCR2 and CCL2 are co-expressed by human first-trimester villous tissue.

Abstract:Somatostatin (SST) receptors (SSTRs) belong to the typical 7-transmembrane domain family of G-protein-coupled receptors. Five distinct subtypes (termed SSTR1-5) have been identified, with SSTR2 showing the highest affinity for natural SST and synthetic SST analogs. Most neuroendocrine tumors (NETs) have high expression levels of SSTRs, which opens the possibility for tumor imaging and therapy with radiolabeled SST analogs. A number of tracers have been developed for the diagnosis, staging, and treatment of NETs with impressive results, which facilitates the applications of human SSTR subtype 2 (hSSTr2) reporter gene based imaging and therapy in SSTR negative or weakly positive tumors to provide a novel approach for the management of tumors. The hSSTr2 gene can act as not only a reporter gene forin vivoimaging, but also a therapeutic gene for local radionuclide therapy. Even a second therapeutic gene can be transfected into the same tumor cells together with hSSTr2 reporter gene to obtain a synergistic therapeutic effect. However, additional preclinical and especially translational and clinical researches are needed to confirm the value of hSSTr2 reporter gene based imaging and therapy in tumors.

Abstract:AIM　To study whether gonadotropin releasing hormone receptor (GnRHR) mRNA exists in rat pancreas and its cellular localization. METHODS　In situ hybridization with digoxigenin-labeled oligonucleotide probes was adopted. RESULTS　Pancreatic endocrine islet cells, exocrine glandular cells and exocrine duct epithelial cells were all found to have GnRHR mRNA hybridization signals. The signals distributed in cytoplasm of all positive cells with negative nucli. CONCLUSION　Both of the exocrine and endocrine cells of the pancrea can produce GnRH recepor. GnRH is also a kind of digestive hormone produced by pancrea and is responsible for modulating the exocrine and endocrine function.

Abstract:Aim: To establish a powerful technique as a whole-mount in situ hybridization for determination of the temporal and spatial gene expression patterns in early developmental-stage embryos.Methods: Murine blastocysts were collected and fixed.Whole-mount in situ hybridization was performed as:preparation of VASP and IL1R2 gene specific digoxigenin-labeled cRNA probe,pre-treatment and whole-mount in situ hybridization of the blastocysts,visualization of the labeled cRNA and treatment after visualization,microscope analysis.Results: Expression of VASP and IL1R2 was detected in both inner cell mass and trophoblast of blastocysts.Conclusion:The present protocol described here is suitable for detection of the gene expression patterns in early developmental-stage embryos.

Abstract:Abstract The significance of the placenta in success pregnancy outcome has been generally acknowledged. Despite our growing understanding of placenta cellular compositions and function, the heterogeneity among billions of nuclei within the syncytiotrophoblast (STB), a multinucleated entity primarily responsible for placental function, remains largely understudied. To better understand the identity of nuclei within STB, we applied integrated single-nucleus RNA-seq (snRNA-seq) and single-nucleus sequencing assay for transposase-accessible chromatin (snATAC-seq) on human placentas at early and late pregnancy. We inferred dynamic STB nuclear-heterogeneity, lineage-determining enhancer gene-regulatory networks and differentiation trajectories of STB nuclei and revealed their crucial biological features. Intriguingly, we found that nascent STB nuclei were involved in diverse biosynthetic processes that might be coordinated by MAPK and Erbb siganling cascades, and STBFLT1 represented mature nuclei exhibited high expression of genes associated with oxygen response and extracellular matrix organization. Gene expression in STBPAPPA represented ones was associated with a wide variety of hormone production. We identified mature lineage biased master TFs such as JUNB and STAT5A. Through STAT5A deletion in the hTSCs, we verified the regulatory roles of it in down-regulation of STBPAPPA nucear associated gene expressions. Our data paves the way for charting human STB development and its related diseases.

Abstract:AIM:To partially isolate and identify hepatic stimulator substance mRNA from human fetal liver tissues.METHODS:The poly (A)mRNA was extracted from human fetal liver tissues of 4-5 month gestation, fractionated by size on sucrose gradient centrifugation, translated into protein from each fraction in vitro and then its products were tested for HSS activity.RESULTS:Twenty-two 500 total RNA was obtained from human fetal liver tissues and pooled.mRNA of 420 was yielded,processed by oligo(dT)-cellulose column chromatography, then was size-fractionated by ultracentrifution on a continuous sucrose density gradient (5%-25%), and separated into 18 fractions.Translated products of mRNA in fraction 8 and 9 could produce a two-fold increase in the incorporation of( 3)H-TdR into DNA of SMMC-7721 hepatoma cells and in a heat resistant and organ-specific way.CONCLUSION:The partially purified HSS mRNA was obtained and this would facilitate the cloning of HSS using expression vectors.
Localization and Mrna Expression of Dopamine Receptor Subtype D_4 and D_5 in Human Stomach and Duodenum

Abstract:Objective To investigate the localization and mRNA expression of dopamine receptor subtype D_4 and D_5 mRNA in human stomach and duodenum by in situ hybridization. Methods The oligonucleotide probe was synthesized and labeled with digoxin, and then hybridized with human gastric and duodenal samples, which were stored in liquid nitrogen, fixed in hypothermia, and embedded in paraffin. The results were recorded and quantitatively analyzed using the pathological image computer analysis system. Results Dopamine receptor subtype D_4 and D_5 mRNA both expressed in human stomach as well as duodenum. They distributed mainly in the interstitial of mucosal glands and the lamina propria near lamina muscularis mucosa. The mRNA expression value of D_4 was higher than that of D_5 in both stomach and duodenum. In stomach, the number of positive target cells was 41.29±5.06 vs. 26.25±5.82 ( P 0.01 ) and the density was 4.68± 0.59 vs. 2.97±0.65 ( P 0.01); similarly, in duodenum, the number of positive target cells was 30.71±10.06 vs. 22.18±4.96 ( P 0.01) and density was 3.48± 1.13 vs. 2.52±0.57 ( P 0.01). D_4 mRNA expression in stomach was higher than that in duodenum( P 0.01). Conclusions Dopamine receptor subtype D_4 and D_5 can both express in human stomach as well as duodenum. The value of D_4 mRNA expression may be higher than that of D_5 in both stomach and duodenum. The values of D_4 and D_5 mRNA expression may be higher in stomach than in duodenum respectively.

Abstract:The human placenta has a vital role in ensuring a successful pregnancy. Despite the growing body of knowledge about its cellular compositions and functions, there has been limited research on the heterogeneity of the billions of nuclei within the syncytiotrophoblast (STB), a multinucleated entity primarily responsible for placental function. Here we conducted integrated single-nucleus RNA sequencing and single-nucleus ATAC sequencing analyses of human placentas from early and late pregnancy. Our findings demonstrate the dynamic heterogeneity and developmental trajectories of STB nuclei and their correspondence with human trophoblast stem cell (hTSC)-derived STB. Furthermore, we identified transcription factors associated with diverse STB nuclear lineages through their gene regulatory networks and experimentally confirmed their function in hTSC and trophoblast organoid-derived STBs. Together, our data provide insights into the heterogeneity of human STB and represent a valuable resource for interpreting associated pregnancy complications.

Expression of somatostatin receptor in human colorectal cancers and normal mucous tissues

Abstract:Objectives: The study was designed to determinate whether human colorectal cancers (CRC) would express somatostatin receptor and whether somatostatin could be used in human CRC. Methods: Forty-two specimens of CRC and normal colorectal mucous membrane were taken in patients who suffered CRC. Immunohistochemistry (IHC) technique was used to demonstrate somatostatin receptor expressions. The relationship between expression of SSTR and clinical or pathological factors was analyzed. The expression of SSTR,proliferative index (PI) and apoptosis index (AI) of the CRC were measured. Results: SSTR was expressed in human CRC(35/42, 83.3%) and the normal mucous tissue (39/42, 92.9%, P 0.001 ). Sharply contrasting with SSTR-negative tumors, the normal colorectal mucous membrane was positive. A significant correlation was found between SSTR expression and tumor stage (P0.001) and tumor differentiation (P0.001). And there was no statistically significant relationship between the expression of SSTR and the age, gender of patients and the tumor size (P=0.682, P=0.789, P= 0.342 ). Conclusions: Somatostatin receptor is expressed in human colorectal cancer and appears to be down-regulated compared to the normal mucous tissue. Somatostatin and its analogues can be used in colorectal cancer cases which express somatostatin receptor proteins.

Abstract:Objective To investigate the mRNA and protein expression of somatostatin receptors in hepatocellular carcinoma HCCLM3 cell lines and to explore the mechanism by which somatostatin effects on hepatocellular carcinoma. Methods RT-PCR., immunocytochemistry and MTT were used to detect mRNA and protein expression of somatostatin receptors in hepatocellular carcinoma cells and evaluate the antiproliferative effect of somatostatin. Results The effect of somatostatin on the cellular proliferation was verified. Immunocytochemistry study revealed a mainly intracellular distribution of all SSTRs with unique patterns for each of them. mRNA expression of all 5 subtypes of somatostatin receptors was different, SSTR2 and SSTR1 mRNA expressions were significantly higher than SSTR3, SSTR4 and SSTR5 ( P < 0. 05 ). Conclusion All 5 subtypes of somatostatin receptors exist in hepatocellular carcinoma HCCLM3 cell lines. Cell proliferation is reduced by somatostatin and SSTR2,3,5 via a protein tyrosine phosphatase dependent mechanism.
Expression and Localization of Mesothelin in Developing Rat Pancreas.

Abstract:To define a genetic network that regulates development of the pancreas, we used high-density microarray (Affymetrix) to generate transcriptional profiles of rat pancreas from five biologically significant stages of development: embryonic day 12.5 (E12.5), E15.5, E18.5, postnatal day 0 (P0) and adult. Many genes were notably highly expressed in the later gestation when islet architecture and function are gradually forming. The expression and localization of mesothelin, one of these genes, was further examined. Reverse transcription-polymerase chain reaction and Western blot analysis revealed that mRNA and protein levels of mesothelin were high from later gestation to 2-3 weeks after birth, and with relatively low but detectable expression levels in adult rat pancreas. Immunolocalization indicated that mesothelin localized not only in islet beta-cells but also in the mesenchyme of developing rat pancreas. Transient mesothelin expression was concomitant with the development of islets architecture formation, remodeling and maturation. These findings indicate that mesothelin is dynamically expressed in the developing rat pancreas and that mesothelin might be involved in some developmental events during development of rat pancreas.
