Abstract:An efficient and stable Agrobacterium-mediated genetic transformation system for Brassica oleracea var.acephala was established through investigating the factors influencing the transformation efficiency.The results showed that the highest transformation frequency was obtained when the explants were pre-cultivated for 2 days,followed by Agrobacterium infection for 5 min with an OD600 of Agrobacterium of about 0.3,co-culture for 2 days and detention screening for 3 days.Results of PCR proved that the transgene was successfully integrated into the B.oleracea var.acephala genome.

Abstract:Brassica oleracea is an economically important species, including seven cultivated variants. Agrobacterium -mediated transformation of B. oleracea crops, mainly via hypocotyl and cotyledon, has been achieved in the past. However, previously established transformation methods showed low efficiency, severe genotype limitation and a prolonged period for transformants acquisition, greatly restricting its application in functional genomic studies and crop improvement. In this study, we have compared the shoot regeneration and genetic transformation efficiency of hypocotyl, cotyledon petiole and curd peduncle explants from twelve genotypes of cauliflower and broccoli. Finally, an Agrobacterium -mediated transformation method using curd peduncle as explant was established, which is rapid, efficient, and amenable to high-throughput transformation and genome editing. The average genetic transformation efficiency of this method is stable up to 11.87% and was successfully implemented in twelve different genotypes of cauliflower and broccoli and other B. oleracea crops with low genotype dependence. Peduncle explants were found to contain abundant cambial cells with a strong cell division and shoot regeneration ability, which might be why this method achieved stable and high genetic transformation efficiency with almost no genotype dependence.

Abstract:Rapeseed ( Brassica napus ) is an important oil crop that supplies a considerable amount of global vegetable oil production. Genetic transformation system is important to gene functional analysis and molecular breeding. Here, an efficient Agrobacterium -mediated transformation protocol using hypocotyl of rapeseed as explants is described. To develop this protocol, we compared several essential factors that would affect the transformation efficiency, such as Agrobacterium strains, selection marker genes, and genotypes of rapeseed. Comparison of different Agrobacterium strains showed that the GV3101 had higher transformation efficiency than that of C58C1 and EHA105. HPTII , NPTII , and RePAT were used as selection marker genes in tissue culture. The results showed that the transformation efficiency was 3.7–4.8%, 2.2–22.5%, and 1.6–5.9% when the hypocotyl of Westar was infected by GV3101 and screened under hygromycin, kanamycin, and basta, respectively. The transformation efficiency of Westar was the highest and ZS11 was the lowest when five different genotypes of rapeseed (Westar, ZS9, ZS11, GY284, and WH3417) were infected by GV3101. Using this protocol, it will take 8–10 weeks to obtain transgenic plants. This protocol has been used to study gene function in several genotypes of rapeseed in our laboratory. These results indicate that it is efficient to obtain transgenic plant of rapeseed using this protocol.

Abstract:We report here a seed transformation of sonication-assisted, no-tissue culture to rapidly produce transgenic Brassica napus plants. This method comprises the steps of treating seeds by ultrasonic wave, inoculating Agrobacterium tumefaciens with a recombinant ChIFN-a gene and germinating directly of treatment seed on wet filter papers. Abstract:Efficient genetic transformation approaches play pivotal roles in both gene function research and crop breeding. However, stable transformation in mustard, particularly for different horticultural types, has not been systematically studied and well-established so far. In this study, we optimized the key factors in the genetic transformation of mustard, including the optical density value of Agrobacterium suspension, the age of explants, and the combination of phytohormones at different concentrations. As a result, the optimal conditions for the genetic transformation of leaf and stem mustard included hypocotyl explants derived from 4-day-old seedlings, infection by 0.8 OD600nm Agrobacterium suspension, and then re-differentiation on a medium containing 4 mg/L trans-zeatin (tZ) and 0.1 mg/L indoleacetic acid (IAA) for leaf mustard, and for stem mustard, re-differentiation on a medium containing 2 mg/L tZ and 0.4 mg/L IAA, with positive rates of 4.74% and 5.26%, respectively. Those for root mustard were hypocotyl explants derived from 8-day-old seedlings, infection by 0.2 OD600nmAgrobacterium suspension, and a medium containing 2 mg/L tZ and 0.1 mg/L IAA, with a positive rate of 4.42%. Overall, this work provides an effective tool for both the theoretical study and genetic improvement of Brassica juncea.

Abstract:The Δ6-fatty acid desaturase gene from Oenothera biennis was introduced into the explants of Brassica napus in this study,and postive transgenic plantlets were confirmed with PCR which contribute to provide the primary basis for the future application of gene engineering to γ-linolenic production.This study used cotylendon node explant to optimize Agrobacterium-mediated Brassica napus genetic transformation system.The optimal pre-culture condition for the cotylendon node is two days on MS medium+6-BA 2.0 mg/L+NAA 0.2 mg/L.Agrobacterium was transformed with binary vector pPZP221 harboring 35S promoter-driven ObD6D gene,which was used to infect brassica napus cotylendon node with optimal concentration OD600=0.4 and time 5 min.Infected explants were cultured on differential selective medium with gentamicin and the resisitant explants were subsequently transfered to root-promoting medium.Postive transgenic plantlets were detected by PCR and transfered to soil.In total,we got twelve transgenic plantets.

Abstract:Leaf piece explants of five Brassica juncea (L.) Czern. cultivars were transformed with an Agrobacterium tumefaciens strain EHA105 harboring the plasmid pCAM-BIA1301, which contains the -glucuronidase (uidA) and hygromycin phosphotransferase (hpt) genes under the control of cauliflower mosaic virus 35S (CaMV35S) promoter. Transgenic plants were regenerated on Murashige and Skoog (MS) medium fortified with 8.87 M 6-benzylaminopurine, 0.22 M 2,4-dichlorophenoxyacetic acid, and 20 M silver nitrate in the presence of 30 mg/l hygromycin. When co-culture took place in the presence of 100 M acetosyringone, the efficiency of stable transformation was found to be approximately 19 in the T0 generation, with the transgenic plants and their progeny showing constitutive GUS expression in different plant organs. Southern blot hybridization of uidA and hpt genes confirmed transgene integration within the genome of transformed plants of each cultivar. Inheritance of hpt gene for single copy T-DNA inserts showed a 3:1 pattern of Mendelian segregation in progeny plants through germination of T1 seeds on MS medium containing 30 mg/l hygromycin. The protocol described here reports superior transformation efficiency over previously published protocols and should contribute to enhanced biotechnology applications in B. juncea.
A high-efficiency PEG-Ca2+-mediated transient transformation system for broccoli protoplasts

Abstract:Transient transformation of plant protoplasts is an important method for studying gene function, subcellular localization and plant morphological development. In this study, an efficient transient transformation system was established by optimizing the plasmid concentration, PEG4000 mass concentration and genotype selection, key factors that affect transformation efficiency. Meanwhile, an efficient and universal broccoli protoplast isolation system was established. Using 0.5% (w/v) cellulase R-10 and 0.1% (w/v) pectolyase Y-23 to hydrolyze broccoli cotyledons of three different genotypes for 3 h, the yield was more than 5×106/mL/g, and the viability was more than 95%, sufficient to meet the high standards for protoplasts to be used in various experiments. The average transformation efficiency of the two plasmid vectors PHG-eGFP and CP507-YFP in broccoli B1 protoplasts were 61.4% and 41.7%, respectively. Using this system, we successfully performed subcellular localization of the products of three target genes (the clubroot resistance gene CRa and two key genes regulated by glucosinolates, Bol029100 and Bol031350).The results showed that the products of all three genes were localized in the nucleus. The high-efficiency transient transformation system for broccoli protoplasts constructed in this study makes it possible to reliably acquire high-viability protoplasts in high yield. This research provides important technical support for international frontier research fields such as single-cell sequencing, spatial transcriptomics, plant somatic hybridization, gene function analysis and subcellular localization.

Abstract:According to the principle that phosphoenolpyruvate carboxylase gene(PEPC)controls rapeseed protein and fatty acid synthesis,we constructed the PEPC gene fragment interference expression vector ihpRNA that contained the palindrome structure of a forward and reverse PEPC gene fragment sequence.The vector structure which expresses RNA interference function(ihpRNA),in the process of cloning in E.coli was easy to cut by restriction enzymes,and splice site is random.Through repeated experiments,we finally got a vector structure which was consistent with our expectations that the forward and reverse sequence of PEPC gene.The construction of interference expression vector ihpRNA was successfully accomplished.The expression vector of PEPC gene was transferred into Brassica napus.L by the transformation method of floral-dip,and then through the resistance screening,PCR-specific amplification,PCR-Southern blot and cloning and sequencing of the obtained transgenic plants were confirmed,and the transformation rate reached 0.69 %.The data will serve to illustrate that the floral-dip method could effectively transform the genetic expression vector ihpRNA.

Abstract:Based on the established regeneration system,an efficient transformation and screening system of Brassica parachinensis mediated by Agrobacterium was developed.Four-day-old cotyledon served as excellent explants for transformation,and the factors affecting the transformation frequency,such as the kinds and concentrations of antibiotics to inhibit the growth of Agrobacterium,Agrobacterium strains,preculture time of cotyledon explants,concentration of Agrobacterium,infection time and co-infection time etc.were also discussed by GUS coloration analysis.2 days preculturation,15 min Agrobacterium(OD_(600)=0.8) infection with 100 μmol/L acetosyringone(AS),and 2 days' co-infection would lead to an increase in transformation efficiency.The differential medium with kanamycin 15 mg/L and Ticarcillin 100 mg/L was found to be most optimal for transgenic plantlets selection.Plantlets resistant to kanamycin were obtained and 8 transgenetic plants were obtained.Stable integration of GUS gene in these plants were confirmed by PCR and Southern blotting analysis.

Abstract:A reliable and efficient regeneration protocol from the cotyledons and hypocotyls is successfully established using three cauliflower genotypes. Thidiazuron (TDZ) is found to be an effective inducer of shoot formation. Both the hypocotyl and cotyledon segments are highly responsive (100%) to shoot formation and produce a maximum of 15.8 shoots per hypocotyl explant on MS salts with B5 vitamins medium containing TDZ at 0.2 mg L-1 and -naphthaleneacetic acid at 0.01 mg L-1. Using this efficient regeneration protocol, the reporter -glucuronidase (GUS) under the constitutive cauliflower mosaic virus (CaMV) 35S promoter is introduced into cauliflower cells of hypocotyls and cotyledons with Agrobacterium tumefaciens strain EHA105. By optimizing the parameters on the procedure of Agrobacterium-mediated transformation, a high transformation efficiency is demonstrated. The transformation efficiencies are 21.6% and 18.3% for cotyledons and hypocotyls, respectively. Integration of the transgenes in the plant genome is demonstrated by PCR and Southern hybridization analysis. Expression of the transgenes is confirmed by RT-PCR analysis and GUS staining. Compared with previous works, the present study provides higher regeneration and transformation efficiencies and establishes a highly efficient transformation system in cauliflower.

Abstract:The synthetic gene encoding hirulog was transformed into Brassica oleracea Vapitata. The plant expression vector of hirulog gene was constructed. It was transformed into the cabbage mediated by Agrobacterium tumefaciens. Regenerative plants was screened by kanamycin. The hirulog gene into cabbage genome DNA was confirmed by PCR assay. Transgenic cabbage of hirulog gene was obtained.

Abstract:Two double-low rapeseed cultivars westar and Zhong Shuang No.4 (Brassica napus L.)were used as recipient materials and the C-terminal fragment of CRY1, CCT1, fused to GUS (GUS-CCT1) was transferred with Agrobacterium-mediated floral-dip method. The transformation was performed at initial blossom stage. The effects of raceme position and surfactant Silwet-77 on the transformation frequency were investigated. The results showed that the best branches for dipping treatment were the top inflorescence and the uppermost two first-order branches in the raceme. Three consecutive dippings were conducted on every another day within a 7 day period. The best concentration of Silwet-77 was 0.05% (v/v) in the transformation buffer. The dipped racemes were bagged for self pollination and the seeds harvested were screened with Kanamycin in culture medium. The Kanamycin-resistant plants were further assayed by GUS reporter gene, and then by PCR test with a pair of specially designed primers based on the sequence of CCT1. The special DNA band from PCR was reclaimed, cloned and sequenced. The sequence of DNA was compared to CCT1 on NCBI internet and the homogeneity of sequence was 100%. It was certified that the foreign gene CCT1 was successfully transferred into the genome of the recipient rapeseed. The results showed that the Agrobacterium-mediated floral-dip method can be successfully applied to genetic transformation in rapeseed.

Abstract:Kanamycin-resistant Brassica napus cv.Xiangyou 13 were obtained by agrobac-terium tumefaciens-mediated genetic transformation at the highest frequency of 8.3% when cotyledonary petioles were immersed into the agrobacterium suspension for 3～5 min and co-cultured for 60～72 h, whereas by ovary-injecting genetic transformation it was succecded at the highest frequency of 12.8% when 0.5～1.5 μg of exogenous DNA was injected into the middle of an ovary from 20th hour to 30th hour after pollination.Southern blotting analysis of the total DNA extracted from transgenic plants indicated that Bt insecticidal protein gene had been integrated into the genome of oilseed rape．Transgenic plants were obtained by agrobacterium tumefaciens-mediated and ovary-injected at frequency of 1.18%，2.66% respectively. Ovary injection was practical and efficient for oilseed rape transformation.
