Abstract:Nasal administration of acetylcholine receptor(AChR) to experimental animals prior to immunization with AChR plus complete Freund's adjuvant (CFA) could effectively prevent clinical signs of experimental autoimmune myasthenia gravis (EAMG). We demonstrated that the suppression of AChR-specific lymphocyte proliferation responses from mononuclear cells (MNC) of popliteal and inguinal lymph nodes (PILN) in nasal AChR tolerized rats were more obvious than that in non-tolerized EAMG control rats after immunization; the suppression of AChR-specific CD4+/CD8- clonal proliferation from PILN of nasally tolerized rats were obvious too, the statistically significant differences were found as compared to the non-tolerized control rats; the AChR-specific skin delayed-type hypersensitivity (DTH) responses were markedly decreased in nasally tolerized rats. The results suggest that the downregulation of specific T lymphocyte responses induced by nasal administration of AChR might be the basis of clinical myasthenia suppression.
