Abstract:The title compound, 3,7,11-trimethyl-10-oxo-dodecanoic acid, a new sesquiter pene isolated from Caulerpa recemosa was synthesized through eight steps starting from geraniol in an overall yield of 18.5%. The key step was the regioselective alkylation of methyl isopropyl ketone.

Abstract:The asymmetric total synthesis of (3S,6R)-3,6-dihydroxy-10-methylundecanoic acid and its trimer was accomplished from commercially available 1-bromo-3-methylbutane in 11 steps in a 27.5% overall yield and 14 steps in a 24.5% overall yield, respectively. The key steps were asymmetric allylboration via allyldiisopinocampheylborane and Yamaguchi's esterification reaction.

Abstract:The marine peptides have been a fertile source for new bioactive molecules, and many possess potent, activity, across a broad spectrum of targets. Their diversity in both biological activity and in chemical complexity has made these secondary metabolites being the focus of much work in recent years. As a fragment, (R)-2-methyloctanoic acid-7-yne was always found in those nature materials. In this article, (R)-2-methyl-7-octynoic acid was synthesized with pentane-1, 5-diol as the start material. Stereogenic carbon(C-2) was obtained through alkylation induced with Evans' chiral auxiliary, and alkyne 13 was got by use of Ohira-Bestmann reagent. Through 13-step reactions the target molecule was prepared with over yield of 17. 1%, and e. e. beyond 99%.

The key steps involved the catalytic asymmetric allylation of hexanal and the coupling reaction of a chiral alkyne and a protected bromide in the presence of t-BuLi.

Abstract:The asymmetric total synthesis of (-)-retigeranic acid A was described, which relies on a crucial reductive skeletal rearrangement cascade for the controllable assembly of diverse angular triquinane subunits. Taken together with an intramolecular Michael/aldol cyclization, an ODI-[5 + 2] cycloaddition/pinacol rearrangement cascade, a Wolff ring contraction and a stereoselective HAT reduction, our synthetic approach has enabled the access to (-)-retigeranic acid A in a concise and practical manner.

Abstract:3,7-Dimethyl-7-hydroxy-2-octen-1,6-olide (1) and 3,7-dimethyl-2,6-octadien-1,6-olide (2), the natural bioactive compounds isolated from the fruit of Litsea cubeba and the liverwort Plagiochila rutilans, were totally synthesized using easily available cis-geraniol as raw material in short, convenient, and low-cost, five-step reactions including three steps of oxidation, cyclization, and dehydration, with an overall yield of 47.5% and 37.3%.

Abstract:The total synthesis of 3,7-dimethyl-7-hydroxy-2-octen-1,6-olide and (E)-6,7-dihydroxy-3,7-dimethyl-2-octenic acid were carried out though Wittig reaction, hydrolyzation, epoxidation and intramolecular cyclization or ring-opening in overall yields of 6.4% and 23.7% using 6-methyl-5-hepten-2-one as raw materials, and their structures were characterized by H-1 NMR, C-13 NMR and ESI-MS spectral data.

Abstract:The tropone sesquiterpene phaeocaulisin D, isolated from the rhizomes of Curcuma phaeocaulis, has previously been shown to inhibit nitric oxide production in macrophages. A total synthesis of phaeocaulisin D was accomplished by using an intramolecular cyclization-dearomatization as a key step. The highlights of the synthesis are effective formation of the 5-7 fused tropone system, and selective methylation of a late-stage intermediate.

Abstract:A total synthesis of the title sesquiterpene 4 is described that starts with the chiral, non-racemic cis-1,2-dihydrocatechol 10 obtained through the whole-cell biotransformation of p-iodotoluene. Compound 10 is elaborated over seven steps, including Negishi cross-coupling and intramolecular Diels-Alder (IMDA) cycloaddition reactions, to ketone 7 that engages in a photochemically promoted 1,3-acyl migration and so affording cyclobutanone 6. Compound 6 was converted over further steps into the title compound 4.

Abstract:The first total synthesis of guajavadimer A, a dimeric caryophyllene-derived meroterpenoid featuring an unprecedented 4-9-6-6-6-9-4-fused ring system, is reported. Key to the approach is the construction of the pyrano[4,3,2-de]chromene core via a cascade of double hetero-Diels-Alder reactions. Practically, a 4-substituted-2,6-dihydroxybenzaldehyde dimethyl acetal serves as an effective surrogate for ortho-quinone methide, which is generated from the corresponding aldehyde and trimethyl orthoformate, with beta-caryophyllene undergoing cycloaddition to generate pyrano[4,3,2-de]chromene derivatives with excellent regioselectivity and stereoselectivity in one pot under mild conditions.

Abstract:Objective To prepare(2E,6E)-octadiene-3,7-dimethyldiol-1acetate.Methods The target compound was synthesized by five steps(acetylating,addition-elimination,replacement,oxidation and rearrangement)with geraniol.Results The desired product was obtained in over all yield of 49.7% and its structure was confirmed by 1H-NMR and IR.Conclusions A new route for the synthesis of (2E,6E)-octadiene-3,7-dimethyldiol-1acetate was explored.

Abstract:The Caulilide which shows a potent cytotoxicity was synthesized from 2,6,6-trimethylcyclohex-2-ene-1,4-dione via selective ketalization,Wittig reaction,deprotection,hydrolysis-lactonization and reduction in 6.0% total yield,in which wittig reaction was the key step.The difficulty of this reaction came from the strong blocking effect,many reaction conditions were optimized,a better yield of this reaction was gained in 57.8%.The synthesis of caulilide was completed by reduction of a lactone with NaBH_(4),the crude product was separated by high performance liquid chromatography to give the target compound.

Abstract:There are two methods in the literature for the preparation of 12-oxo-1. 15-pentadecanolide(1) from α-ni-trocyclododecanone: four-step reaction sequence and four-step one pot reaction sequence. The total yield of the latter is higher than the former. The reason for lower total yield of the four-step reaction sequence was studied. On this basis , 1 was synthesized by four-step two pot reaction sequence from α-nitrocyclododecanone in total yield of 77%.
