Nitric oxide mediates the protective effect of ramiprilat against damages of rabbit aortic endothelium by oxygen free radicals

Abstract:Ramiprilat (Ram) was shown to protect against damages induced by oxygen free radicals on aortic endothelium. The aim of the present study was to investigate whether this protection of Ram is mediated by the facilitation of release of nitric oxide. The superfusion cascade bioassay of rabbit thoracic aorta was used. Oxygen free radicals were generated by electrolysis of Krebs solution. The content of cyclic guanylic acid (cGMP), inorganic nitrite, and malondialdehyde (MDA) in the donor vessel was assayed biochemically. Oxygen free radicals generated in electrolytic Krebs solution caused inhibition of vasodilator responses of rabbit thoracic aorta to acetylcholine (ACh), decrease in levels of cGMP and inorganic nitrite as well as elevation of MDA content in the rabbit aortic segment. The protective effect of Ram on ACh-relaxation was concentration-dependent. L-Arginine also significantly prevented damages induced by free radicals. The protective effects of Ram and L-arginine were abrogated by treatment with nitro-L-arginine (30 ��mol �� L-1), while indometacin (10 ��mol �� L-1) did not prevent the protective effect of Ram on vasodilator responses to ACh. The present study suggests that Ram could protect rabbit aortic endothelium against damages induced by oxygen free radicals, and that the effect of Ram may be due to facilitation of the release of nitric oxide resulting in reduction of lipid peroxidation.
