An experimental study on the transplantation of human fetal brain cells into monkey models of Parkinson's disease

Abstract:Unilateral administration of methyl-phenyl-tetrahydropyridine (MPTP) into the common carotid artery of the induced hemiparkinsonism model in monkeys. The cell suspensions of substantia nigra alone or those of substantia nigra mixed with caudate nucleus, obtained from human fetuses of 11~12 weeks of gestation, were injected stereotaxically into the head of the caudate nucleus or substantia nigra of the lesion side in the hemiparkinsonism model. After the transplantation procedure, the recipient were immunosuppressed by taking cyclosporine A. During the period of 18 weeks after transplantation the parkinsonian symptoms of the recipients were improved. Apomorphine-induced motor asymmetry was reduced significantly. With the use of the avidin-biotin peroxidase complex (ABC) method of immunohistochemistry, tyrosine hydroxylase (TH) staining examination revealed that there were TH-immunoreactive cell bodies surviving and sprouting in the caudate nucleus.
