Corticosterone inhibits interleukin-1 production by receptor-mediated mechanism

Abstract:The effect of physiological concentration of corticosterone (B) on interleukin-1 (IL-1) production and its molecular mechanism at receptor level were studied. IL-1 production by peritoneal macrophages of C57BL/6 mouse was inhibited by B in a dose-dependent manner when its concentration ranging from 10 -11 mol/L to 10 -7 mol/L. The Scatchard plot of the specific binding of 3H-dexamethasone to the macrophages showed that Kd was 3.17 �� 0.44 nmol/L, and Ro 5628 �� 312 binding sites/cell. RU38486, a competitive antagonist of glucocorticoids (GC), not only antagonized the specific binding of 3H-dexamethasone to macrophages, but also blocked the inhibitory effect of B on IL-1 production. Both the antagonizing and blocking effects of RU38486 were concentration-dependent. Our results strongly suggested that B inhibited IL-1 production through its specific receptor.
