Pharmacokinetics, tissue distribution and excretion studies of ginkgolic acid (17:1) in rats

Abstract:A rapid and sensitive method for the determination of ginkgolic acid (17:1) in rat plasma, tissues, urine, feces and bile was developed using high performance liquid chromatography tandem mass spectrometry (HPLC-MS/MS). The biological samples were processed with ethyl acetate extraction and separated on an Elite hypersil BDS C-18 column (2.1 x 100 mm, 3 mu m) and eluted with acetonitrile-water (93:7, v/v, containing 0.1% formic acid). Detection of the analytes was achieved using negative ion electrospray ionization (ESI) in the multiple reaction monitoring (MRM) mode. The MS/MS ion transitions monitored were m/z 373 -> 329 and m/z 345 -> 301 for GA (17:1) and internal standard, respectively. The analyte showed good linearity over a wide concentration range (r(2) > 0.995) and the lower limit of quantification was 10 ng/mL. The method was successfully applied to the pharmacokinetics, tissue distribution and excretion studies of GA (17:1) in rats. The pharmacokinetic investigation demonstrated that GA (17:1) was rapidly absorbed with an absolute bioavailability approximate 19.5%. The tissue distribution result indicated that liver and kidney were the major accumulation tissues for GA (17:1) in rats. In addition, GA (17:1) could hardly penetrate the blood-brain barrier. The excretion study suggested that feces was the main excretion pathway,while high proportion of GA (17:1, > 35%) was excreted as the unchanged form.
