Abstract:4-Acetoxy-2-methyl-2-butenal,a key intermediate of vitamin A,was synthesized from methylglyoxal dimethyl acetal via condensation with chlorovinyl magnesium to give the corresponding tertiary alcohol,followed by acylation to afford 1,2-diacetoxy-2-methyl-1-methoxy-3-butene,which was subjected to hydrolysis and rearrangement with an overall yield of about 49%.

Abstract:Abstract We describe the versatile and concise total synthesis of all stereoisomers of tarchonanthuslactone from (R)- or (S)-3-hydroxybutyrate via eight steps. The key steps of the synthesis include a highly diastereoselective chelation-controlled Mukaiyama aldol reaction of a p-methoxybenzyl-protected aldehyde and a Yamaguchi lactonization of a δ-hydroxy-trans-α,β-unsaturated carboxylic acid.

Abstract:3-(4-t-Butylphenyl)-1-(4-methoxyphenyl)-2-propen-1-one(1) in yield of 91% was synthesized by condensation reaction of 4-t-butylbenzaldehyde with 4-methoxyacetophenone using sodium methoxide as a catalyst.4-t-Butyl-4′-methoxydibenzoylmethane(2) in overall yield of 81% was prepared by addition reaction of 1 with bromine,and then debromination in presence of sodium methoxide.The structure of 2 was confirmed by UV and 1H NMR.

Abstract:The effective method of synthesis of 3-methyl-2-butenoic acid,the cheap 2-Methyl-3-butyn-2-ol was taken as starting material,by Meyer-Schuster rearrangement to 3-Methyl-2-butenal.After oxidized 3-Methyl-2-butenal by air,the title compound was obtained.The conditions of Meyer-Schuster rearrangement and oxidation reaction were investigated in detail.The result showed that using liquid paraffin as solvent,rearrangement reacting at 115～125℃ for 3 hrs,3-methyl-2-butenal 85% yield with more than 98% purity was obtained.Using traces CuCl as catalyst,blowing air into 3-methyl-2-butenal below 35℃,3-methyl-2-butenoic acid 90% yield with more than 97.5% purity was obtained.The method was considered a good one with advantages such as short procedure,high yield,starting material cheap and easy to get and environmentally friendly.

Abstract:Objective: To design a new route to prepare 1-chloro-4-methylsulfinyl- butane. Methods: The target compound was synthesized successfully from DMSO by two steps. Results and Conclusion: The structure of target compound was confirmed by IR, 1HNMR and the total yield was 65. 6%. This procedure has advantages of simple starting material and convenient operations.

Abstract:A new way to synthesis of (S)-tert-butyl 6-benzyloxy-5-hydroxy-3-oxohexanoate was investigated. The target compound was successfully prepared via several mildly reaction processes by using R-epichlorohydrin, which is an inexpensive and easily available starting material. Through screening the amounts of BF3 center dot Et2O and comparing two different synthetic methods, this route becomes more suitable for large-scale preparation for its high yields, mildly reacting-conditions, easily available materials and facilely purification of products.

Abstract:A new method for the synthesis of the 4-methoxy-2,3,6-trimethylbenzaldehyde,a key intermediate of acitretin,was a-chieved from Hagemann' s ester in four steps including methylation at C-3 position,aromatic isomerization,reduction and oxidation. The obtained compounds were characterized through 1 H NMR,13 C NMR and MS. The overall yield of the synthetic route was 49%. The raw materials used in this synthetic route were cheap and readily available while the reaction conditions were relatively mild.

Abstract:The α-methylene-γ-butyrolactone motif is a widely encountered unit in many natural products and pharmaceutical compounds. Herein, a practical and efficient synthesis of α-methylene-γ-butyrolactones from readily available allylic boronates and benzaldehyde derivatives was developed with chiral N,N'-dioxide/AlIII complex as the catalyst. The key success of this transformation was the kinetic resolution of allylboration intermediate via asymmetric lactonization. This protocol enabled to assemble all of four stereoisomers from the same set of starting materials upon variable lactonization. Taking advantage of the current method as the key step, catalytic asymmetric total synthesis of eupomatilones 2, 5, and 6 was accomplished. Control experiments were carried out to probe into the tandem reaction as well as the origin of stereoselectivities.

Abstract:In this paper, 2-methyl-3-butyn-2-amine has been synthesized by ammonolysising 3-chloro-3-methyl-l-butyne in liquid amine, which was converted by 2-methyl-3-butyn-2-ol as raw material. And we have explored a suitable method for industrial synthesis of 2-methyl-3-butyn-2-amine through optimized the synthesis conditions. 2-Methyl-3-butyn-2-amine was primarily identified by IR and GC-MS.

Abstract:The asymmetric total synthesis of (3S,6R)-3,6-dihydroxy-10-methylundecanoic acid and its trimer was accomplished from commercially available 1-bromo-3-methylbutane in 11 steps in a 27.5% overall yield and 14 steps in a 24.5% overall yield, respectively. The key steps were asymmetric allylboration via allyldiisopinocampheylborane and Yamaguchi's esterification reaction.

Abstract:Methyl 4-(1-methylpyrrolidin-2-yl)-3-oxobutanoate and hygrine are important biosynthetic intermediates for tropane alkaloids. We have developed a concise method to synthesize these two compounds from the key intermediate N-methylpyrrolinium cation. Methyl 4-(1-methylpyrrolidin-2-yl)-3-oxobutanoate and hygrine were obtained in four and six steps from commercially available 4,4-diethoxybutylamine with overall yields of 42% and 25%, respectively.

Abstract:General enantioselective routes to 3,4-cis-dialkyl substituted gamma-lactones and 4,5-cis-dialkyl substituted delta-lactones using TiCl(4)-mediated Evans asymmetric aldolization as the key step are reported. The syntheses are exemplified with two natural fragrant molecules, cis-3-methyl-4-decanolide (1) and aerangis lactone (2). The (R,R) steroegenic centers were established using (S)-phenylalanine-derived 2-oxazolidinone or thiazolidinethione as chiral auxiliary, whereas the (S,S) ones were constructed with auxiliary prepared from (R)-phenylglycine. NaBH(4)/CaCl(2)/THF in the presence of a small amount of EtOH was introduced as a new effective method for reductive cleavage of chiral oxazolidinone auxiliaries. Previously unknown, tricky concentration effects were observed during the monotosylation of diol 7 and BOM protection of Evans aldol 23.

Abstract:A new synthetic method for ((4R,6S)-6-hydroxymethyl-2,2-dimethyl-1,3-dioxinan-4-yl)-acetic acid tert-butyl ester was accomplished by a chemoenzymatic approach. Racemic 4-(4-methylbenzyloxy)-3-hydroxybutyronitrile (3) was firstly synthesized via condensation and epoxy opening reactions. (S)-4-(4-Methylbenzyloxy)-3-hydroxybutyronitrile (4) was obtained from resolution of racemic 3 by the lipase from Artgribacter sp. with a high enantiomeric excess (98% ee) in the mixture solvent of acetonitrile/hexane (V : V= 1 : 4). Dihydroxy ester 7 was synthesized by a syn-selective triethylborane mediated reduction with a de value of 97%. After acid-catalyzed protection of 7 and hydrogenolysis of the benzyl protecting group, the objective 9 was obtained with a high optical purity.
