Abstract:Patients with ovarian cancer are at increased risk of venous thromboembolism (VTE), and the cumulative incidence is high, particularly at advanced stages of this disease. Nevertheless, it is challenging to investigate the molecular mechanisms of ovarian cancer-associated VTE (OC-VTE), mainly due to the lack of a well-developed animal model for this disease. We generated a mouse model for developing OC-VTE using ovarian cancer cell injection in combination with the inferior vena cava stenosis method. The rate of thrombosis in the OC-VTE group was 50%, compared with 0 in the control group. Moreover, we conducted a proteomic analysis using platelets from these models and revealed differentially expressed proteins between the OC-VTE and control groups, including upregulated and downregulated proteins. Gene Ontology analysis revealed that these differentially expressed proteins were mostly enriched in the biological process of negative regulation of fibrinolysis and the cellular component of the fibrinogen complex, both of which play key roles in thrombosis. In conclusion, this study lays the foundation for further investigation of the underlying mechanisms of how ovarian cancer promotes VTE formation.
The Establishment of a Spontaneous Metastatic Model of Rat Liver Cancer in Nude Mice

Abstract:Objective To establish a spontaneous metastatic model of rat liver cancer in nude mice.Methods The rat liver cancer cell line C5F was subcutaneously implanted in 4-week-old male and 7-week-old female BALB/cA nude mice respectively.Subcutaneous tumor formation was observed by measuring the MTD weekly.6-7 weeks thereafter,the moribund mice were sacrificed and macroscopic metastases were checked by autopsy.Serial section of the fixed lung tissue was done to grade and count lung metastases.Histological examination was performed for suspicious organs.Results The rate for the subcutaneous tumor formation was 100% (10/10).In the female nude mice,100% (6/6) overt metastases were observed.The median numbers of gross and microscopic metastatic foci were 45 and 755 per nude mouse respectively.Metastases were not found in abdomen and its organs.In contrast,neither macroscopic nor microscopic metastasis was observed in male nude mice.Conclusion This cell line can optimally demonstrate its spontaneous metastatic potential in female BALB/cA nude mice,with the lungs being the preferential target organ.The spontaneous metastatic behavior of C5F in BALB/cA nude mice could be used as a model for functional localization of metastatic suppressor genes for liver cancer on human chromosomes.
Molecular Imaging-Monitored Radiofrequency Hyperthermia-Enhanced Intratumoral Herpes Simplex Virus-Thymidine Kinase Gene Therapy for Rat Orthotopic Ovarian Cancer.

Abstract:Objective: To establish the technique of intratumoral combination therapy of radiofrequency hyperthermia (RFH) with herpes simplex virus-thymidine kinase/ganciclovir (HSV-TK/GCV) gene therapy for rat ovarian cancers. Material and methods: This study consisted of three parts: (1) in vitro experiments to establish the 'proof of principal' that combination of RFH and HSV-TK gene therapy has the synergistic effect on human ovarian cancer cells; (2) creation of bioluminescence imaging-detectable rat ovarian cancer model; and (3) in vivo experiments using this rat model to validate the technical feasibility of the combination therapy. Cells and nude rats were divided into four groups: (i) combination therapy (HSV-TK/GCV + RFH); (ii) RFH; (iii) HSV-TK/GCV; and (iv) phosphate-buffered saline (PBS). Data were analyzed using Dunnett t-test or Kruskal-Wallis test. Results: Cell proliferation assay demonstrated significantly greater reduction in viable cells with the combination therapy [0.52 (0.43, 0.61)] compared to other treatments [RFH 0.90 (0.84, 0.96), HSV-TK/GCV 0.71 (0.53, 0.88), PBS 1 (1, 1); p < .05]. For 24 rat models with bioluminescence imaging-detectable orthotopic ovarian cancer (n = 6 per group), optical imaging demonstrated significantly decreased relative bioluminescence signal with the combination therapy [0.81 (0.52, 1.08)] compared to other treatments [RFH 3.60 (2.34, 4.86), HSV-TK/GCV 2.21 (1.71, 2.71), PBS 3.74 (3.19, 4.29); p < .001]. Ultrasound imaging demonstrated the smallest relative tumor volume with the combination therapy [0.78 (0.45, 1.11) versus 3.50 (2.67, 4.33), 2.10 (0.83, 3.37), 3.70 (1.79, 5.61); p < .05]. Conclusion: The feasibility of intratumoral RFH-enhanced HSV-TK/GCV gene therapy was established on a unique rat model with molecular imaging-detectable orthotopic ovarian cancer.
C57BL/6 mouse model of epithelial ovarian cancer and its biological characteristics

Abstract:Objective To establish epithelial ovarian cancer of peritoneal metastasis model and subcutaneous tumor model in immunocompetent C57BL/6mice , thus to provide basis of diagnosis , treatment and prevention-related basic research for ovarian cancer.Methods Cultured inbred C57BL/6mice poorly differentiated ovarian adenocarcinoma cell line ID-8in vitro.SPF grade C57BL/6female mice (6～8weeks) were inoculated with logarithmic phase of ID-8cells with 1×10~7 , 5×10~6 , 1×10~6 and l×l05cells / dose intraperitoneally and subcutaneously into the flank.They were invided into 8groups with 6in each.Tumor formation rate , ascites , metastasis and survival rate were recorded.An additional set of 6animals were injected intraperitoneally (5×10~6 ID-8cells) and then sacrificed at different time intervals for necropsy and pathologic analysis.The remaining animals were sacrificed and examined just before they died.Results The tumor formation rate was 100%.In peritoneal tumor model group , the average survival time was (141±6.7) d ,(122.8±4.5) d ,(83.4±7.2) d ,(74.4±4.5) d in 1×10 ~5 , 1×10~6 , 5 ×10~6 and l×10~7 ID-8cells group , with the tumor cell load increased , the survival time was significantly shorter (P<0.05) .In subcutaneous tumor group , nodule formation time was about 1week in l×10~7 cells group ; 1week in 5×10~6 cells group ; 3weeks in 1×10~6 cells group and 6weeks in 1×10~5 cells group.As the tumor cells inoculation load increased , the diameter and volume of tumor increased significantly (P<0.05) .Conclusions C57BL/6 mice intraperitoneal ovarian cancer model is similar to human epithelial ovarian cancer stageⅢ and Ⅳ.The immune or drug therapy efficacy is easier observed in subcutaneous tumor model. Establishing the ID-8cells ovarian cancer tumor model in immunocompetent C57BL/6 mice is helpful for clinical study of the treatment of ovarian cancer and immunological study of molecular models.
Establishment of highly metastatic ovarian cancer model with omental tropism via in vivo selection

Abstract:Epithelial ovarian cancer (OC) is often diagnosed at an advanced stage with peritoneal metastasis, and preclinical models mimicking the natural course of OC peritoneal metastasis are essential to improve treatment. We implanted ES2 and ID8 cells in the ovaries of mice and obtained highly metastatic (HM) sublines from their omental metastases after three cycles in vivo selection. Orthotopic xenografts derived from the HM sublines showed enhanced omental tropism and more extensive metastasis with earlier onset. The HM cells exhibited increased in vitro migration and invasion properties, and RNA sequencing revealed that the genes related to epithelial-mesenchymal transition and extracellular matrix regulation were significantly altered in the HM cells. Among them, the upregulated genes were significantly associated with poorer survival in OC patients. In conclusion, these HM sublines can be leveraged to establish spontaneous metastatic OC mouse models, which may serve as ideal preclinical models for anti-metastasis therapy for OC patients.

Abstract:Immunodeficient murine models are usually used as the preclinical models of osteosarcoma. Such models do not effectively simulate the process of tumorigenesis and metastasis. Establishing a suitable animal model for understanding the mechanism of osteosarcoma and the clinical translation is indispensable. The UMR-106 cell suspension was injected into the marrow cavity of Balb/C nude mice. Tumor masses were harvested from nude mice and sectioned. The tumor fragments were transplanted into the marrow cavities of SD rats immunosuppressed with cyclosporine A. Through muti-rounds selection in SD rats, we constructed orthotopic osteosarcoma animal models using rats with intact immune systems. The primary tumor cells were cultured in-vitro to obtain the immune-tolerant cell line. VX2 tumor fragments were transplanted into the distal femur and parosteal radius of New Zealand white rabbit to construct orthotopic osteosarcoma animal models in rabbits. The rate of tumor formation in SD rats (P1 generation) was 30%. After four rounds of selection and six rounds of acclimatization in SD rats with intact immune systems, we obtained immune-tolerant cell lines and established the orthotopic osteosarcoma model of the distal femur in SD rats. Micro-CT images confirmed tumor-driven osteolysis and the bone destruction process. Moreover, the orthotopic model was also established in New Zealand white rabbits by implanting VX2 tumor fragments into rabbit radii and femurs. We constructed orthotopic osteosarcoma animal models in rats with intact immune systems through muti-rounds in-vivo selection and the rabbit osteosarcoma model.

Abstract:Objective To establish a highly metastatic model of colorectal cancer in mice by using organ selection of metastasis. Methods Histologically intact C26 colorectal cancer tissues were subcu- taneously implanted into the NOD-SCID mice which is immunodeficiency. In order to gain visible meta- static focus, the tumor bearing mice were undertaken tumor resection in first selection when tumor reached 1.5 cm in diameter. Screening was conducted according to the method of subcutaneous implantation→lung metastasis→subcutaneous inoculation→lung metastasis. Then the same steps were duplicated in BALB/c mice. Tumorgenicity, invasion, metastasis and morphological characteristics of the implanted tumors were studied by light microscopy, electron microscopy and flow cytometry. Results Highly metastatic models of mice colorectal cancer were obtained after organ screening. After 5 generations in vivo, the carcinoma metastasis rate accounted to 100 percent in NOD-SCID mice, and the metastases of lungs and lymph nodes were noted in all of animals. While only a lower metastasis rate was found when the tumor was subcutaneously implanted in natural mice at the first time. But a 100 percent lung metastatic rate was observed again after being passaged in vivo for 4 generations in natural mice. Conclusions We had established a highly metastasis colorectal cancer model of natural mice which was high, steady, and intuitionistic metastasis and had same biological characteristics of mice colorectal carcinoma. The result provided a useful tool for the study of metastatic mechanism and treatment of human colorectal carcinoma.
Development of Intraperitoneally Transplantated Human Ovarian Carcinoma Model with Immune Reconstruction in Severe Combined Immunodeficient Mice

Abstract:OBJECTIVE:To develop an intraperitoneally transplanted human ovarian carcinoma model in humanized severe combined immunodeficient (SCID) mice.METHODS:Twelve CB17SCID mice were randomly divided into 4 groups: group A intraperineally injected with phosphate-buffered saline, group B injected with human ovarian carcinoma cells SKOV3, group C injected with human peripheral blood lymphocyte (PBL) for immune reconstruction, and group D injected with PBL and SKOV3 cells. The behaviors of mice, tumor growth and morphology, human IgG in peripheral blood, cancer antigen 125 (CA125) immunohistochemical staining, tumor infiltrating lymphocyte (TIL), and status of graft versus host disease (GVHD) were detected.RESULTS:The survival period was > 28 days in all groups. The ratio of successful tumor transplantation was 3/3 in groups B and D. The tumor was widespread in peritoneal cavity, mainly in diaphragm, liver, and mesentery, with bloody ascites. The number and size of tumor were less in the humanized group. CA125 expression was positive in primary transplanted tumor cells and SKOV3 cells. IgG was detected in humanized groups (C and D). The level of human IgG was significantly higher in group D than in group C (P < 0.05). TIL infiltration was remarkable in group injected with PBL and SKOV3 cells and failed to be found in group injected with SKOV3 cells. No GVHD was found in all groups.CONCLUSION:An intraperitoneal transplanted human ovarian carcinoma model has been established in humanized SCID mice that simulates the biological behavior of human ovarian carcinoma disseminating intraperitoneally in patients with immune function and may function as an ideal animal model for preclinical research of ovarian carcinoma treatment.
The establishment and evaluation of the animal disease model of human endometrial carcinoma in nude rat

Abstract:Objective:To establish the subcutaneous transplantable tumor and orthotopic implanted tumor model of the human endometrial carcinoma in nude rat,and observe its biological features.Methods:Ishikawa cell line was cultured in-vitro and inoculated subcutaneously into eight female nude rats.Two weeks later,two nude rats were sacrificed and the subcutaneous tumors were excised.The subcutaneous tumors were cut into carcinoma mass with volume of 2mm3 and then transplanted into the left uterine cavity of 10 nude rats.All ten nude rats were sacrificed 4 weeks after the transplantion.The left uterus were excised and the carcinoma mass were studied through histopathology,immunohistochemistry and flow cytometer.Results:The subcutaneous tumors of the human endometrial carcinoma were generated in all eight nude rats.The transplanted carcinoma was generated in the left uterine cavity in seven rats.The biological features of transplanted tumors were similar with the human endometrial carcinoma.The flow cytometer indicated the S-phase tumor cells accounted for 37.83 % in the human endometrial carcinoma of early stage in nude rats.Conclusion:The subcutaneous transplantable tumor model and orthotopic implanted tumor model using nude rat can serve as an ideal disease model in vivo study of human endometrial carcinoma and contribute to developing the new therapies of human endometrial carcinoma.
Characterization of CC-531 as a Rat Model of Colorectal Liver Metastases

Abstract:Purpose: Surgical resection of colorectal liver metastases is not achievable in more than 70% of the cases. Although the liver directed therapies have become a part of the stand of care, lack of a preclinical model impedes the assessment of toxicity and therapeutic benefits attributed several candidate drugs or treatment regimens that can be designed. In the present study we aim develop and characterize a rat colorectal liver metastasis model. Materials and methods: Growth characteristics of CC-531 cells were determined in vitro followed by subcapsular liver implantation in syngeneic WAG/Rij rats. Tumor growth progression was followed over 3 weeks by ultrasound (US) and magnetic resonance imaging (MRI). Growth characteristics were also assessed by histopathology and immunohistochemistry in harvested tumor tissues. Results: The doubling time of CC-531 cells was found be under 24hrs and all the implanted rats grew tumors. US imaging showed hypoechoic masses and MRI showed contrast enhancement representing complex tumor microenvironments. Hematoxylin and Eosin staining confirmed tumor growth and uniform CD31 staining in tumor confirmed even vessel density. Conclusion: CC-531 can be used as a metastatic rat tumor colorectal liver metastases model with well-defined characteristics that can be readily followed by imaging whilst having a therapeutic window for interventions.

Abstract:Abstract Background Γ-Ionizing radiation (IR) therapy is one of major therapeutic tools in cancer treatment. Nevertheless, γ-IR therapy failed due to occurrence of metastasis, which constitutes a significant obstacle in cancer treatment. The main aim of this investigation was to construct animal model which present metastasis during radiotherapy in a mouse system in vivo and establishes the molecular mechanisms involved. Materials and methods The C6L transfectant cell line expressing firefly luciferase (fLuc) was treated with γ-IR, followed by immunoblotting, zymography and invasion assay in vitro. We additionally employed the C6L transfectant cell line to construct xenografts in nude mice, which were irradiated with γ-IR. Irradiated xenograft-containing mice were analyzed via survival curves, measurement of tumor size, and bioluminescence imaging in vivo and ex vivo . Metastatic lesions in organs of mice were further assessed using RT-PCR, H & E staining and immunohistochemistry. Results γ-IR treatment of C6L cells induced epithelial-mesenchymal transition (EMT) and increased cell invasion. In irradiated xenograft-containing mice, tumor sizes were decreased dramatically and survival rates extended. Almost all non-irradiated xenograft-containing control mice had died within 4 weeks. However, we also observed luminescence signals in about 22.5% of γ-IR-treated mice. Intestines or lungs of mice displaying luminescence signals contained several lesions, which expressed the fLuc gene and presented histological features of cancer tissues as well as expression of EMT markers. Conclusions These findings collectively indicate that occurrences of metastases during γ-IR treatment accompanied induction of EMT markers, including increased MMP activity. Establishment of a murine metastasis model during γ-IR treatment should aid in drug development against cancer metastasis and increase our understanding of the mechanisms underlying the metastatic process.

Abstract:Lung cancer is the leading cause of malignancy-related death worldwide, and metastasis always results in a poor prognosis. However, therapeutic progress is hampered by a deficiency of appropriate pre-clinical metastatic models. To bridge this experimental gap, we developed an in vivo metastatic model via subcutaneous (s.c.) injection. The original cell line (XL-2) adopted in this model was newly isolated from the ascites of a patient with extensive metastases of lung adenocarcinoma, thereby avoiding any alteration of its initial molecular biology features from artificial serial cultivation. After comprehensive phenotypical and histological analysis, it was identified as a lung adenocarcinoma cell line. Additionally, the drug test showed that XL-2 cell line was sensitive to docetaxel, and resistant to doxorubicin, indicating it might serve as a cell line model of drug resistance for identifying mechanisms of tumors resistant to doxorubicin. Through this s.c. model, we further obtained a highly metastatic cell line (designated XL-2sci). The metastatic rate of mice in XL-2 group was 3/10, in contrast to the rate of 9/10 in XL-2sci group. Optical imaging, micro-computed tomography (micro-CT) scanning and Transwell assays were further applied to identify the enhanced metastatic capacity of XI-2sci cells both in vivo and in vitro. Compared with XL-2 cells, ITRAQ labeled proteomics profiling study showed that some tumor metastasis-associated proteins were upregulated in XL-2sci cells, which also indicated the reliability of our model. Proliferation ability of XL-2 and XL-2sci were also evaluated. Results showed that highly metastatic XL-2sci possessed a decreased proliferation capacity versus XL-2, which demonstrated that its increased metastatic activity was not facilitated by a faster growth rate. In conclusion, we successfully developed an in vivo metastatic model using a newly established lung adenocarcinoma cell line, which will be beneficial to further investigations of lung cancer metastasis and to the development of anti-metastasis drugs.
Development of an immunosuppressed orthotopic hepatocellular carcinoma rat model for the evaluation of chemo- and radioembolization therapies

Abstract:Hepatocellular carcinoma (HCC) is widely known to be chemo-resistant and presents with significant liver disease resulting in low tolerability to systemic chemotherapy. As a counter measure, more targeted therapies such as trans -arterial chemoembolization (TACE) and trans -arterial radioembolization (TARE) have been developed. To further optimize these therapies, animal models are critical in elucidating the molecular events in disease progression and test new treatment options. The present study focuses on the development of a hepatoma bearing rat model. N1S1 rat hepatoma cells were transfected by a lentiviral method and injected into the liver of Sprague Dawley (SD) and Rowett Nude (RNU) rats. Longitudinal tumor growth was observed by bioluminescence imaging (BLI) and liver/tumor histology. In both models, tumors were visible within 4 days post cell inoculation. Tumor take rates were 52 % and 73 % for male and female SD rats, respectively, and 100 % for male RNU rats. By day 12 and 15 post inoculation, we recorded complete tumor regression in male and female SD rats. Liver histology showed advanced fibrosis in the tumor regressed SD rats, whilst RNU rats exhibited the characteristic sheet pattern of Novikoff tumor with mild liver fibrosis . Increased CD3 and TUNEL staining observed in SD rat livers may be key factors for tumor regression. Our data reveal that the immunocompetent SD rats are not recommended as a model for therapeutic investigations. The immunosuppressed RNU rats, however, are characterized by consistent and reliable tumor growth and thus a desirable model for future therapeutic investigations.

Abstract:Objective To establish the athrmic inouse model of breast cancer in normal position and imitated metastatic breast cancer. Methods Breast cancer cells MDA-MB-231-luc carrying luciferase gene was injected into the athymie mice.The optieal imaging in vivo system was used to observe the establislament of the model. Reseults The breast tumor emerged after we planted the MDA-MB-231-lue cells in the mammary gland fatpads,the volume and photon of the tumor increased during the second weekto the fifth week.After injection by the tail vein,the tumors mainly located in the lungs While after infusion in the left alrtrium.the tumolrs metastate to all over the body. Conclusions We succeeded in the establishment of the athymic mice model of breast cancer.in situ and imitated metastatic breast cancer by iniection into the vena caudalis and the left alttrium.The optical imaging in vivo system could distinctly show the formation of the tumors.
Establishment of Novel Rat Models for Premalignant Breast Disease.

Abstract:BACKGROUND:Breast cancer has become one of the most common malignant tumors among females over the past several years. Breast carcinogenesis is a continuous process, which is featured by the normal epithelium progressing to premalignant lesions and then to invasive breast cancer (IBC). Targeting premalignant lesions is an effective strategy to prevent breast cancer. The establishment of animal models is critical to study the mechanisms of breast carcinogenesis, which will facilitate research on breast cancer prevention and drug behaviors. In this study, we established a feasible chemically-induced rat model of premalignant breast cancer.METHODS:Following the administration of the drugs (carcinogen, estrogen, and progestogen) to Sprague-Dawley (SD) rats, tumors or suspicious tumors were identified by palpation or ultrasound imaging, and were surgically excised for pathological evaluation. A series of four consecutive steps were carried out in order to determine the carcinogen: 7,12-dimethylbenzaanthracene (DMBA) or 1-methyl-1-nitrosourea, the route of carcinogen administration, the administration period of estrogen and progestogen, and the DMBA dosage.RESULTS:Stable premalignant lesions can be induced in SD rats on administration of DMBA (15 mg/kg, administered three times) followed by administration of female hormones 5-day cycle.RESULTS:were confirmed by ultrasound and palpation.CONCLUSION:Under the premise of drug dose and cycle, DMBA combined with estrogen and progestogen can be used as a SD rat model for breast premalignant lesions.
Enhanced Efficacy and Specificity of Epithelial Ovarian Carcinogenesis by Embedding A Dmba-Coated Cloth Strip in the Ovary of Rat

Abstract:Abstract Background Ovarian cancer is predominant of epithelial cell origin and often present at an advanced stage with poor prognosis. Most animal models of ovarian carcinoma yield thecal/granulose cell tumors, rather than adenocarcinomas. The best reported induction rate of adenocarcinoma in rats is 10-45% by an ovarian implantation of 7, 12-dimethylbenz[a]anthracene (DMBA) coated silk suture. We provided an improved procedure to construct the model by the ovarian implantation of DMBA-coated cloth strip. Methods A sterile suture (as S group) or a piece of cloth strip (as CS group) was soaked in DMBA before ovarian implantation in Wistar rats. Tumor size, incidence rate and pathological type were analyzed. Results Ovarian tumors in rats of CS group were first noted at 16 wk post implantation and reached a cumulative incidence of 75% (96/128) at 32 wk, while the tumor incidence rate in S group at 32 wk was only 46.25% (37/80). The tumor size in CS group (3.63 ± 0.89 cm) was larger than that of S group (2.44 ± 1.89 cm) (P < 0.05). In CS group, there were only two types of tumor formed: adenocarcinoma (90/96) and sarcoma (6/96). While in S group, there were different types, including adenocarcinoma (21/37), squamous carcinoma (3/37), granulosa cell tumor (3/37), sarcoma (4/37), undifferentiated carcinoma with no adeno character (2/37), benign ovarian tumor (2/37), and malignant teratoma (1/37). Conclusion The model in our study yields much higher incidence and specificity of epithelial derived tumors and showed histological similarities to human ovarian cancers, which would be more suitable for therapeutic research.
Establishment of a stable hepatic metastasis mouse model of murine colorectal cancer by microsurgical orthotopic implantation

Abstract:Background: Liver metastasis is a common cause of death from colorectal cancer (CRC). In this paper we developed a liver metastasis mouse model by microsurgical orthotopic implantation (MSOI) to illuminate the CRC progression with an eye toward developing effective drug treatment. Methods: Murine colon carcinoma CT-26 cells were cultured and then injected to male BALB/c athymic nude mice right flank to generate subcutaneous implantation tumor with 2×107 CT-26 cell suspension in DMEM. Tumor tissue at an average size of 1 cm3 was injected into another nude mice right flank with 20-gauge inoculating needle. Between fourth and sixth generations, tumor tissue sewn into the cecal surface establishes orthotopic transplanted CRC model by MSOI. Then on the 7th, 14th, 21st and 35th day, body weight, abdomen circumference, volume of ascites and local tumor weight were observed and weighed. On the 21st day and 35th day, local tumor rate was calculated, and metastatic tumors of other organs were observed. Tumor tissue was stained by HE for pathologic analysis. Results: On the 35th day, body weight and abdomen circumference of the model group were significantly higher than the control group (P<0.01). Local tumor weight increased rapidly from the 21st d to the 35th d (P<0.01), and take rate was high (100%). Metastatic tumor appeared only in liver on the 21st day and then invaded to liver, stomach, retroperitoneal lymph node and abdominal wall on the 35th day. The metastatic rate of liver tumor respectively was 83.3% and 100% on the 21st day and 35th day, but liver function remained normal. Pathologic analysis showed that colorectal tumor invaded the normal tissue of liver, abdominal wall and stomach. Conclusions: A stable hepatic metastasis mouse model of murine CRC was established by MSOI.
Establishment of Rat C6 Glioma Model

Abstract:Objective To establish an easy, economical, practical rat C6 glioma model.Method After fixation by self-made frame, C6 gliomaous cells were implanted into the specific plot of brain. The behavior and survival time of the rats were observed.Result After inoculation, diminished food intake, weight loss, intraocular hemorrhage, ocular proptosis, per se whirl, epileptic attack and hemiplegia ocurred in model rats. After 2-3weeks, pathological dissection showed oncogenesis in cranium.Conclusion The rat model of brain tumor we made is similar to the human's. The tumor is quick to form, and relatively unchangeable during the survival, so it can be used for studing the tumor's foundational mechanism, experimental cure and pharmacodynamics.
Liver Tumor Model with Implanted Rhabdomyosarcoma in Rats: MR Imaging, Microangiography, and Histopathologic Analysis.

Abstract:In compliance with institutional regulations for care and use of laboratory animals, the aim of this study was to establish and characterize a rodent liver tumor model to provide a platform for preclinical assessment of new diagnostic and therapeutic strategies. A rhabdomyosarcoma tumor was implanted in the right and left liver lobes of 20 rats, for a total of 40 tumors. T1- and T2-weighted magnetic resonance (MR) images, diffusion-weighted images, and dynamic susceptibility contrast agent-enhanced perfusion-weighted images were obtained up to 16 days after tumor implantation and were compared with postmortem three-dimensional computed tomographic (CT) images, digital microangiograms, and histopathologic findings. Fifteen tumors were examined with proton (H-1) MR spectroscopy. All tumors grew, with a mean volume doubling time of 2.2 days +/- 0.9 ( standard deviation) and a final size of 591 mm(3) +/- 124. The rhabdomyosarcoma tumor showed hypervascularity at MR imaging, three-dimensional CT, microangiography, and histologic analysis. On dynamic susceptibility contrast-enhanced perfusion- weighted images, the maximum signal intensity decrease differed in time and extent between the tumor and the liver, with a significantly (P <.001) higher relative blood volume, relative blood flow, and permeability value in the tumor than in the liver. With H-1 MR spectroscopy, the rhabdomyosarcoma tumor and the liver featured significant (P <.001) choline and lipid peaks, respectively. Implantation of a rhabdomyosarcoma tumor in the livers of rats is feasible and reproducible, and this animal model seems promising for future testing of new diagnostic and therapeutic strategies. Abstract:Background The formation of metastasis is the most common cause of death in patients with lung cancer. A major implement to understand the molecular mechanisms involved in lung cancer metastasis has been the lack of suitable models to address it. In this study, we aimed at establishing a highly metastatic model of human lung cancer and characterizing its metastatic properties and underlying mechanisms. Methods The human lung adeno-carcinoma SPC-A-1 cell line was used as parental cells for developing of highly metastatic cells by in vivo selection in NOD/SCID mice. After three rounds of selection, a new SPC-A-1sci cell line was established from pulmonary metastatic lesions. Subsequently, the metastatic properties of this cell line were analyzed, including optical imaging of in vivo metastasis, immunofluorescence and immunohistochemical analysis of several epithelial mesenchymal transition (EMT) makers and trans-well migration and invasion assays. Finally, the functional roles of fibronectin in the invasive and metastatic potentials of SPC-A-1sci cells were determined by shRNA analysis. Results A spontaneously pulmonary metastatic model of human lung adeno-carcinoma was established in NOD/SCID mice, from which a new lung cancer cell line, designated SPC-A-1sci, was isolated. Initially, the highly metastatic behavior of this cell line was validated by optical imaging in mice models. Further analyses showed that this cell line exhibit phenotypic and molecular alterations consistent with EMT. Compared with its parent cell line SPC-A-1, SPC-A-1sci was more aggressive in vitro , including increased potentials for cell spreading, migration and invasion. Importantly, fibronectin, a mesenchymal maker of EMT, was found to be highly expressed in SPC-A-1sci cells and down-regulation of it can decrease the in vitro and in vivo metastatic abilities of this cell line. Conclusions We have successfully established a new human lung cancer cell line with highly metastatic potentials, which is subject to EMT and possibly mediated by increased fibronectin expression. This cell line and its reproducible s.c . mouse model can further be used to identify underlying mechanisms of lung cancer metastasis.
