Abstract:Objective:To investigate the mechanismsof Zusanli(ST36) in intracerebral hemorrhage.Methods:96 rats were randomly divided into the sham operation group,the sham acupuncture group,the intracerebral hemorrhage group and the acupuncture group with 24 in each group. The intracerebral hemorrhage model was established by injected 0. 5U collagenase type Ⅶ stereotaxically into globus pallidus in rats. The acupuncture group was needled Zusanli(ST36) . The sham acupuncture group was stimulated non-acupoints skins. The positional variations of the expression of vascular endothelial growth factor(VEGF) and the VEGF mRNA were observed at the 3th,7th and 14th day after model establishment. Results:There were no obvious VEGF expressions in different time points of the sham operation group. In the 3th day,a plenty of the microvessel with positive VEGF around the hematoma area were observed in the intracerebral hemorrhage group,the sham operation group and the acupuncture group. In the 7th day,these microvessels with positive VEGF were digging into the hematoma area. And then,they were appeared in this area. After detected by the RT-PCR,there were no obvious expressions of VEGF mRNA in the sham operation group. Its expression was more remarkable in the 3 days after intracerebral hemorrhage. This condition was lasted to the 14th day. In every time point,the VEGF mRNA expressions of the acupuncture group were significantly higher than those of the intracerebral hemorrhage group. Nevertheless,this expression in the sham acupuncture group was approximate to the intracerebral hemorrhage group. Conclusion:Needing Zusanli(ST36) do not promote the VEGF expression in the intracerebral hemorrhage rats,but also repair the injury of the microvessel of the hemorrhage damage area. This could be realized by the reconstruction of the vessel system.
Electroacupuncture improves recovery after hemorrhagic brain injury by inducing the expression of angiopoietin-1 and -2 in rats

Abstract:Background Angiopoietin (Ang) is one of the major effectors of angiogenesis, playing a critical role in neurovascular remodeling after stroke. Acupuncture has been widely used for treating stroke in China for a long time. Recently, we have demonstrated that electroacupuncture (EA) can accelerate intracerebral hemorrhage (ICH)-induced angiogenesis in rats. In the present study, we investigated the effect of EA on the expression of Ang-1 and Ang-2 in the brain after ICH. Methods ICH was induced by stereotactic injection of collagenase type VII into the right globus pallidus. Adult male Sprague–Dawley rats were randomized into the following four groups: sham-operation (SHAM), stroke-no electroacupuncture (SNE), stroke-EA at the Zusanli acupoint (SEZ), and stroke-EA at a nonacupoint (SEN). EA was applied to the bilateral Zusanli (ST36) acupoint in the SEZ group and a nonacupoint in the SEN group. The expression of Ang-1 and Ang-2 was evaluated by immunohistochemistry and quantitative real-time reverse transcription-polymerase chain reaction (RT-PCR). Results Some Ang-1 and Ang-2 immunoreactive microvessels with a dilated outline were detected in the perihematomal tissues after ICH, and the vessels extended into the clot from the surrounding area since day 7. The expression of Ang-1 increased notably as long as 2 weeks after ICH, while Ang-2 immunoreactivity declined at about 7 days following a striking upregulation at 3 days. EA at the Zusanli (ST36) acupoint upregulated the expression of Ang-1 and Ang-2 at both the protein and mRNA levels. However, EA at a nonacupoint had little effect on the expression of Ang-1 and Ang-2. Conclusions Our data suggest that EA at the Zusanli (ST36) acupoint exerts neuroprotective effects on hemorrhagic stroke by upregulation of Ang-1 and Ang-2.
Time-dependent effects of electroacupuncture at the Ren channel on extracellular signal-regulated kinases 1/2 expression in focal cerebral ischemia rats

Abstract:BACKGROUND: The onset of focal cerebral ischemia activates extracellular signal-regulated kinases 1 and 2, regulates cell cycle, promotes cell proliferation and differentiation, and affects the normal stage and function of brain cells. OBJECTIVE: To observe the effects of electroacupuncture at the Ren channel on extracellular signal-regulated kinases 1/2 expression in the lateral cerebral ventricle wall of rats with focal cerebral ischemia. The effects were analyzed at different time points after intervention. DESIGN: Randomized controlled study. SETTING: Department of Anatomy, Sun Yat-Sen University. MATERIALS: A total of 60 healthy adult male Wistar rats weighing (250±10) g were provided by the Experimental Animal Center, Medical College of Sun Yat-Sen University. The animal experiment was conducted with confirmed consent by the local ethics committee. The GB6805-Ⅱ electric acupuncture apparatus was provided by Shanghai Medical Equipment High-techno Company. METHODS: The experiment was performed at the Laboratory of Anatomy, Sun Yat-Sen University, from February to July 2007. All experimental animals were randomly divided into the following groups: normal group (n = 6), sham operation group (n = 18), model group (n = 18), and electroacupuncture group (n = 18). Middle cerebral artery occlusion (MCAO) was performed in the model group and electroacupuncture group. Zea Longa's grading standard was used to assess neurological impairment after reperfusion; animals whose grades were between 1 and 4 were included in this study. The normal control group was not exposed to MCAO. In sham operation animals, the right common carotid artery (CCA) was isolated, and the external carotid artery (ECA) was damaged, but no embolism was induced. The electroacupuncture group was given acupuncture on the second day after surgery. The acupoint locations were chosen according to Experimental Acupuncture (People's Publishing House; 1997; First Edition). The Chengjiang, Qihai, and Guanyuan acupoints were labeled and connected to a G6805 electroacupuncture apparatus with sparse-dense waves (sparse waves were 30 Hz, dense waves were 100 Hz), with a frequency of 6–15 V. The duration was 20 minutes. Two days after surgery, the model and sham operation groups were placed with their backs on the operating table, but they received no acupuncture. However, the normal group received acupuncture. The experimental animals under anesthesia were sacrificed on days 7, 14, and 28 post-surgery. Western blot analysis was used to measure expression of extracellular signal-regulated kinases 1/2 in the inferior region of the lateral cerebral ventricle wall. Expression was measured in the normal group at time points corresponding to the sham operation group. MAIN OUTCOME MEASURES: Expression of extracellular signal-regulated kinases 1/2 in the inferior region of the lateral cerebral ventricle wall at different time points after intervention. RESULTS: All 60 rats were included in the final analysis, without any loss. Seven days after MCAO, there was no significant difference in extracellular signal-regulated kinases 1/2 expression in the electroacupuncture group compared to the model group (P > 0.05). However, extracellular signal-regulated kinases 1/2 expression significantly increased in the model group at 14 and 28 days after treatment (P < 0.05). CONCLUSION: Electroacupuncture at the Ren channel can enhance extracellular signal-regulated kinases1/2 expression in the inferior region of the lateral cerebral ventricle wall of rats with focal cerebral ischemia. However, this effect is not apparent until 14 days after electroacupuncture intervention.
The Poly-Adp Ribose Polymerase-1/Apoptosis-inducing Factor Pathway May Help Mediate the Protective Effect of Electroacupuncture on Early Brain Injury after Subarachnoid Hemorrhage.

Abstract:Subarachnoid hemorrhage (SAH) is a clinically common, acute, critical cerebrovascular disease associated with high mortality. Here, we investigated the effects of electroacupuncture on early brain injury after SAH. We successfully established a Sprague–Dawley rat model of the SAH model, and randomly divided the rats into four groups: sham-operated group, SAH group, positive control group, and electroacupuncture group. Electroacupuncture effectively decreased the number of transferase UTP nick end labeling-positive cells and extent of DNA fragmentation compared with the control, indicating a decrease in apoptosis. Moreover, electroacupuncture decreased the expression of proteins involved in the poly-ADP ribose polymerase-1/apoptosis-inducing factor (PARP-1/AIF) pathway in vivo, and the difference was statistically significant (P < 0.05). Treatment with electroacupuncture resulted in a significant improvement in neurological function. It inhibited the increase in blood–brain barrier permeability by regulating the protein expression of matrix metalloproteinase-9, occludin, and claudin-5. Additionally, electroacupuncture limited the development of cerebral edema and microglial activation in early brain injury after SAH. In conclusion, electroacupuncture can ameliorate early brain injury after SAH, and this may occur via inhibition of the PARP-1/AIF pathway.
Electroacupuncture enhances cerebral blood perfusion by inhibiting HIF-1α in rat subarachnoid hemorrhage

Abstract:Objective Cerebral blood perfusion (CBP) reduction is a prevalent complication following subarachnoid hemorrhage (SAH) in clinical practice, often associated with long-term cognitive impairment and prognosis. Electroacupuncture (EA), a widely utilized traditional Chinese therapy for central nervous system disorders, has demonstrated promising therapeutic effects. This study aims to investigate the therapeutic potential of EA in restoring CBP in SAH rats and to explore the mechanisms involving HIF-1α in this process. Methods Rats were randomly assigned to one of five groups, including Sham, SAH, EA, EA + Saline, and EA + dimethyloxallyl glycine (DMOG) groups. EA treatment was administered for 10 min daily, while DMOG were intraperitoneally injected. Behavioral tests, cerebral blood flow monitoring, vascular thickness measurement, western blotting, and immunofluorescence staining were conducted to assess the therapeutic effects of EA on cerebral blood flow. Results SAH resulted in elevated levels of HIF-1α, endothelin (ET), ICAM-1, P-SELECTIN, E-SELECTIN, and decreased level of eNOS in the brain. This led to cerebral vasospasm, decreased CBF, and cognitive deficits in the rat SAH model. EA intervention downregulated the expression of HIF-1α, ET, ICAM-1, P-SELECTIN, and E-SELECTIN, while increasing eNOS expression. This alleviated cerebral vasospasm, restored CBF, and improved cognitive function. However, the administration of the HIF-1α stabilizer (DMOG) counteracted the therapeutic effects of EA. Conclusion EA promotes the recovery of cerebral blood flow after SAH injury, attenuates cerebral vasospasm, and accelerates the recovery of cognitive dysfunction, and its mechanism of action may be related to the inhibition of the HIF-1α signaling pathway.

Abstract:Electroacupuncture (EA) at ST36 can improve the survival rate in rats after hemorrhagic shock (HS). The current study investigated rats with 60% blood loss. 144 rats were divided into four groups: hemorrhage without fluid resuscitation (HS), EA after hemorrhage without fluid resuscitation (EA), hemorrhage with delayed resuscitation (DFR), and EA after hemorrhage with delayed resuscitation (EA + DFR). The survival rate and biological parameters 0, 3, 12, and 24 h after HS were investigated. The 24 h survival rate of EA + DFR was significantly higher than that of DFR. 12 h after hemorrhage, the level of mean arterial blood pressure of EA + DFR was significantly higher than that of DFR, and the levels of renal blood flow, intestinal mucosal blood flow, and hepatic blood flow of EA + DFR were also significantly higher than those of DFR. Three hours after hemorrhage, the levels of lactate, PaCO2, alanine aminotransferase, and creatinine of groups receiving EA were significantly lower than those of non-EA groups, and the levels of pH, PaO2, and diamine oxidase of groups receiving EA were significantly higher. EA at ST36 can improve the 24 h survival rate and produce the experimental antishock effects on tissue perfusion and organ protection from fatal HS.
Effects of electroacupuncture on angiogenesis after ischemia and reperfusion

Abstract:Objective To explore the effect of eletroacupuncture (EA) on angiogenesis after cerebral ischemia and reperfusion through observing the number of peripheral blood endothelial progenitor cells (EPCs) and the expression of vascular endothelial growth factor (VEGF) in the hippocampus.Methods Among 50 male Sprague-Dawley (SD) rats,10 were randomly selected as a control group (CG).Qi deficiency and blood stasis were induced in the other 40 rats by intragastric administration of dahuang daily for 7 days.A model of focal cerebral ischemia and reperfusion injury was then created by thread-occlusion of the middle cerebral artery,except in 10 rats selected for the sham operation group (SOG) who received the same operation without tightening the thread.The successfully modeled rats were divided into a model group (MG,n =10) and an electroacupuncture group (EAG,n =10).The rats in EAG started acupuncture treatment the next day after modeling,while no treatment was given to those in the other groups.On day 1,3 and 7 after the modeling,the neurological functioning of the 4 groups was assessed using Basso-Beattie-Bresnahan locomotion scoring and the number of EPCs in the peripheral circulation was detected by flow cytometry.On the 8th day,all the rats were decapitated and the VEGF expression was detected immunohistochemically.Results The average BBB locomotion score of the EAG rose significantly from (4.50 ± 1.05) on day 1,to (13.67 ± 1.21) on day 3,and then further to (20.00 ±0.89) by day 7,significantly higher than that of MG at the same time points,but still significantly lower than that of the control group.The average number of EPCs in the peripheral circulation in the MG also increased from (26.83 ± 6.05) on day 1 to (33.67 ± 5.39) on day 3,and decreased to (32.83 ± 5.04) on day 7 after modeling,significantly lower than that of the CG [(45.50 ±9.40),(42.17 ±4.62) and (41.33 ±5.50)],the SOG [(58.00 ±8.05),(59.67±4.84) and (53.83 ±5.38)[and the EAG [(66.17 ±4.36),(127.50 ±73.75) and (55.00 ±35.15)] at each time point.After occlusion,the average expression of VEGF-positive cells in the hippocampus of the EAG [(27.80 ± 2.39)％] was significantly higher than that of the MG,and both were significantly higher than that of the CG.Conclusions Eletroacupuncture can up-regulate VEGF expression and EPCs in the peripheral blood in rats after cerebral ischemia-reperfusion,which can promote angiogenesis and neural functional recovery.
Effect of electroacupuncture on cerebral blood flow and secretion of ET-1 in rats with ischemic stroke sequelae

Abstract:[Objective] To observe the effect of electroacupuncture on cerebral blood flow and secretion of ET-1in rats with ischemic stroke sequelae.[Methods] Thirty healthy adult male Wistar rats were randomly divided into electroacupuncture group, model group and sham operation group with 10 rats in each group. Referencing the ZeaLonga line tied method the model of middle cerebral artery occlusion(MCAO) was reproduced. Five weeks later the model of ischemic stroke sequelae was considered to be formed. Rats in electroacupuncture groups were punctured on Baihui,Shuigou. Two weeks later, the Laser Doppler flow-meter was used to monitor the cerebral blood flow of rats and immunohistochemical method was used for determination of the level of endothelin-1(ET-1) in brain tissue. [Results] Compared with the sham operation group, the cerebral blood flow was reduced in the model group(P0.05). Compared with model group, the cerebral blood flow was significantly increased in the EA group(P0.05).In the model group the total area with ET-1 protein expression-positive substance was higher than that in sham group(P0.05). The total area of the positive cells of ET-1 protein expression in the brain tissue of EA group had a lower trend than that in the model group. But there was no significant difference between these two groups(P 0.05).[Conclusion] After cerebral ischemia, cerebral tissue ET-1 levels has increased significantly. EA can inhibit the upregulation of ET-1 and improve the blood flow of the rats with ischemic stroke sequelae.
Effect of electro-acupuncture at Zusanli acupoint on blood coagulation during intestinal ischemia-reperfusion in rats

Abstract:Objective To evaluate the effect of electro-acupuncture (EA) at Zusanli (ST36) acupoint on blood coagulation during intestinal ischemia-reperfusion (I/R) in rats.Methods Forty healthy male Sprague-Dawley rats,aged 6-8 months,weighing 250-300 g,were divided into 5 groups (n =8 each) using a random number table:sham operation group (group S),intestinal I/R group (group I/R),EA at Zusanli acupoint group (group EA),EA at non-acupoint group (group NE) and α7 nicotinic acetylcholine receptor antagonist α-bungarotoxin (α-BGT) group (group α-BGT).Intestinal I/R was induced by clamping the superior mesenteric artery for 4-5 min followed by 120 min of reperfusion.Bilateral Zusanli acupoints were stimulated with an electric stimulator (frequency 3 Hz,voltage 2-4 V,wave length 2 ms) for 30 min starting from the time point immediately after beginning of ischemia in group EA,while EA was performed at the points 5 mm lateral to the bilateral Zusanli instead in group NE.In group α-BGT,α-BGT 1 μg/kg was intraperitoneally injected at 45 min before ischemia,and the other treatments were similar to those previously described in group EA.Blood samples were collected from the abdominal aorta at 120 min of reperfusion for determination of the concentrations of tumor necrosis factor alpha (TNFα),tissue factor (TF),antithrombin (AT),tissue plasminogen activator (tPA),fiber plasminogen activator inhibitor-1 (PAl-l) and D-dimer in plasma (by enzyme-linked immunosorbent assay) and platelet count (PLT).The animals were sacrificed after blood sampling,the distal ileum specimens were removed for examination of the pathological changes with a light microscope,and the damage to the intestinal mucous membrane was assessed and scored according to Chin.Results Compared with group S,the concentrations of plasma TNFα,TF,tPA,PAI-1 and D-dimer were significantly increased,and the plasma AT concentration and PLT were decreased in I/R,NE and α-BGT groups,the concentrations of plasma TNFα and TF were significantly increased,and the plasma AT concentration was decreased in group EA,and Chiu's scores were significantly increased in I/R,EA,NE and α-BGT groups (P＜ 0.05).Compared with group I/R,the concentrations of plasma TNFα,TF,tPA,PAI-1 and D-dimer were significantly decreased,the plasma AT concentration and PLT were increased,and Chiu's scores were decreased in group EA (P＜0.05),and no significant change was found in the variables mentioned above in NE and α-BGT groups (P＞0.05).Compared with group EA,the concentrations of plasma TNFα,TF,tPA,PAI-1 and D-dimer were significantly increased,the plasma AT concentration and PLT were decreased,and Chiu's scores were increased in group NE (P＜0.05).Conclusion EA at Zusanli acupoint can improve blood coagulation during intestinal I/R in rats,and the mechanism is related to activating the cholinergic anti-inflammatory pathway.
Electroacupuncture improves neurovascular unit reconstruction by promoting collateral circulation and angiogenesis

Abstract:Acupuncture at Shuigou (GV26) shows good clinical efficacy for treating stroke, but its mechanism remains poorly understood. In this study, a cerebral infarction model of ischemia/reperfusion injury received electroacupuncture at GV26 (15 Hz and 1 mA, continuous wave [biphasic pulses], for 5 minutes). Electroacupuncture effectively promoted regional cerebral blood flow on the infarct and non-infarct sides, increased infarct lesions, lectin, and number of blood vessels, upregulated von Willebrand factor and cell proliferation marker Ki67 expression, and diminished neurological severity score. These findings confirm that electroacupuncture at GV26 promotes establishment of collateral circulation and angiogenesis, and improves neurological function.
The Effect of Electro-acupuncture on Focal Cerebral Angiogenesis in Cerebral Ischemia-reperfusion Rats

Abstract:Objective:To observe the effects of electro-acupuncture on the quantity of endothelial progenitor cells(EPCs) in peripheral blood and on the expression of Vessel Endothelium Growth Factor Receptor-2(VEGFR-2) and Platelet Endothelial Cell Adhesion Molecule-1(PECAM-1) in ischemic cerebral cortex of cerebral ischemia-reperfusion rats,discussing the mechanism of angiogenesis after cerebral ischemia with the effect of electro-acupuncture.Methods:Male rats were randomly and evenly assigned to normal group,sham-operation group,model group and EA group."quchi" and "housanli" were selected.EPCs quantity was displayed using flow cytometry.VEGFR-2 and PECAM-1 expression are observed with immunohistochemistry.Results:EPCs quantity in blood achieved peak in model group at 24hrs,and began to decreased after 48hrs;while in EA group EPCs quantity in blood started to increase at 24hrs,and reached peak in 48hrs,then decreased at 72hrs.VEGFR-2 and PECAM-1 expression began to increase at 24hrs in model group and EA group,and the expression increased with the time prolonged.The expression of electro-acupuncture group was more than that in model group.Conclusion:The electro-acupuncture could increase EPCs quantity in peripheral blood,and raise VEGFR-2 and PECAM-1 expression in ischemic cerebral cortex.Those can contribute to promote the formation of blood vessels in cerebral ischemia region.
Long-Term Stimulation with Electroacupuncture at DU20 and ST36 Rescues Hippocampal Neuron through Attenuating Cerebral Blood Flow in Spontaneously Hypertensive Rats

Abstract:This study was designed to investigate the effect of long-term electroacupuncture at Baihui (DU20) and Zusanli (ST36) on cerebral microvessels and neurons in CA1 region of hippocampus in spontaneously hypertensive rats (SHR). A total of 45 male Wistar rats and 45 SHR were randomly grouped, with or without electroacupuncture (EA) at DU20 and ST36, once every other day for a period of 8 weeks. The mean arterial pressure (MAP) was measured once every 2 weeks. Cerebral blood flow (CBF) and the number of open microvessels in hippocampal CA1 region were detected by Laser Doppler and immunohistochemistry, respectively. Nissl staining and Western blotting were performed, respectively, to determine hippocampus morphology and proteins that were implicated in the concerning signaling pathways. The results showed that the MAP in SHR increased linearly over the observation period and was significantly reduced following electroacupuncture as compared with sham control SHR rats, while no difference was observed in Wistar rats between EA and sham control. The CBF, learning and memory capacity, and capillary rarefaction of SHR were improved by EA. The upregulation of angiotensin II type I receptor (AT1R), endothelin receptor (ETAR), and endothelin-1 (ET-1) in SHR rats was attenuated by electroacupuncture, suggesting an implication of AT1R, ETAR, and ET-1 pathway in the effect of EA.

Abstract:Objective. Electroacupuncture (Ea) is a useful complementary and alternative therapy for intracerebral hemorrhage (ICH). However, the neurobiological basis for the Ea treatment of ICH is still unclear. The primary aim of the present study was to explore whether Ea prevents brain edema, apoptosis, excitotoxicity, and neuroinflammation in rats after hemorrhagic stroke. Methods. Rats were randomly divided into Sham, Control, and Ea groups. We used modified neurological severity score (mNSS) and gait analysis to estimate neurological function in rats, and PET/CT to assess glucose uptake and the hemorrhagic focus volume. Measurement of the brain water content and TUNEL staining were used to evaluate brain edema and cell apoptosis, respectively. The serum myelin basic protein (MBP), neuron-specific enolase (NSE), calcium-binding protein B (S100B), and tumor necrosis factor-α (TNF-α) concentrations were examined with ELISA. The expression levels of the CD68, GALC, Arg-1, iNOS, NR2A, Glu2R, AQP4, MAP2, GFAP, AQP9, Bcl-2, Bax, and Glu proteins around the hematoma were detected via immunohistochemistry staining. Western blot was used to analyze the levels of the AQP4, AQP9, Bax, Bcl-2, iNOS, and Arg-1 proteins. Results. Ea treatment improved neurological function and reduced the hemorrhagic area and brain water content in rats after ICH. The serum concentrations of MBP, NSE, S100B, and TNF-α all decreased significantly in the Ea group compared with the Control group. Expression levels of the Glu, NR2A, AQP4, AQP9, Bax, GFAP, iNOS, and CD68 proteins in brain tissue surrounding the hematoma were obviously suppressed in ICH rats following Ea treatment. Moreover, Ea stimulation increased the levels of the MAP2, GALC, Glu2R, Arg-1, and Bcl-2 proteins, but reduced the number of TUNEL-positive cells in rats after ICH. Conclusion. The results of this study suggest that Ea may exert neuroprotective effects by suppressing brain edema, apoptosis, excitotoxicity, and neuroinflammation.

Abstract:Intracerebral hemorrhage (ICH) is the most fatal form of acute stroke, with 17.9 million patients worldwide, accounting for 10–15% of all stroke patients [ 1 ]. Despite treatments such as surgical hematoma evacuation and blood pressure management, mortality from ICH remains as high as 68% [ 1 ]. ICH-induced injuries are classified into primary injuries and secondary injuries. The primary injury was related to the mass effect caused by initial hematoma, hematoma expansion and hydrocephalus. Secondary brain injury is caused by microglia activation, which drives inflammatory, oxidative and cytotoxic reactions that further lead to cell death and dysfunction [ 2 ]. After ICH, reactive oxygen species (ROS) act as mediators of demyelination and blood-brain barrier disruption, causing mitochondrial dysfunction, and may be involved in the development of early and late brain edema. A recent multicenter clinical trial confirmed that early hematoma removal after ICH does not effectively improve patient prognosis [ 3 ]. In addition to limited progress in the management of ICH patients, effective treatment options are yet to be developed for the secondary brain injury of ICH.

Electric Acupuncture Treatment Promotes Angiogenesis in Rats with Middle Cerebral Artery Occlusion Through EphB4/EphrinB2 Mediated Src/PI3K Signal Pathway

Abstract:Background: Cerebral infarction is one of the most common causes of disability and death worldwide. It is reported that electric acupuncture was able to improve the prognosis of cerebral infarction by promoting angiogenesis. However, the corresponding signal pathways of angiogenesis promotes by electric acupuncture treatment needs to be further explored. Methods: MCAO rat was employed as the animal model, and clopidogrel hydrogen sulfate treatment was set as the positive control. Behaviors of rats, H&E staining, and TTC-staining was used to evaluate the recovery of infarcted brain tissue and nervous function. After that, immunocytochemical and immunofluorescence staining was used to quantify the angiogenesis and compensatory circulation, which including the analysis of microvessel density, field/ microvessel area ratio, and microvessel diameter. Western blot and RT-PCR for the detection of the related signal molecule, PI3K, Src, and EphB4/ephrinB2. Results: The neurologic impairment scores were decreased, and the brain tissue damage that showed with H&E and TTC-staining was relieved by the treatment of electric acupuncture in MCAO rat. The quantification of microvessel density and field/ microvessel area ratio was improved obviously, and the microvessel diameter was decreased which represent the angiogenesis of capillary in day 3 and 7 by the electric acupuncture treatment. We also found that the level of Src and PI3K was increased markedly followed by the up-regulation of EphB4 and EphrinB2 mRNA during the electric acupuncture treatment, and the pre-treatment of Src and/or PI3K inhibitor was able to disturb the angiogenesis of capillary. Conclusions: We proved that electric acupuncture was able to accelerate the recovery of infarcted brain tissue and nervous function in MCAO rat by the promotion of angiogenesis, which was regulated by EphB4/EphrinB2 mediated Src/PI3K signal pathway. Our study provides a potential therapy and theoretical basis for the clinical treatment of cerebral infarction by the use of electric acupuncture.
Neuroprotective Effect of Electroacupuncture and Upregulation of Hypoxia-Inducible Factor-1α during Acute Ischaemic Stroke in Rats

Abstract:Background Acupuncture is a traditional method that has been widely used in various fields of medicine with therapeutic effect. However, evidence of effectiveness to support the application of electroacupuncture (EA) during the process of ischaemia is scarce. Objectives To investigate dynamic changes in hypoxia-inducible factor (HIF)-1α expression as well as its association with neurological status in rats subjected to acute ischaemic stroke and EA intervention. Methods Forty adult male rats were randomly divided into three groups that received sham surgery (Control group, n=10) or underwent middle cerebral artery occlusion and EA (MCAO+EA group, n=15) or minimal acupuncture as a control treatment (MCAO+MA group, n=15). The rats in the MCAO+EA and MCAO+MA groups received EA or acupuncture without any electrical current, respectively, during 90 min of ischaemia. Rats in the Control group received the same surgical procedure but without MCAO. EA involved electrical stimulation of needles inserted into the quadriceps at 50 Hz frequency and 3 mA current intensity. Neurological status was evaluated on postoperative day 1, and cerebral infarction volume (IV) and HIF-1α expression 24 hours later. Results Neurological scores were improved and cerebral IV was decreased in the MCAO+EA group compared to the MCAO+MA group (both p<0.05). Moreover, HIF-1α expression was higher in the MCAO+EA group versus the MCAO+MA group (p<0.05). Conclusions EA enhanced recovery of neurological function, decreased cerebral IV and increased HIF-1α expression in ischaemic rats. Further research is needed to determine whether EA is effective for stroke treatment through the stimulation of muscle contraction.

Abstract:Objective: To observe the effect of electroacupuncture on the expression of voltage-gated sodium channel Na(v)1.6 after cerebral ischemia in rats. Methods: A total of 105 healthy adult male Sprague-Dawley rats were randomly allocated into sham operation (n = 15) , ischemic (n = 45) and electroacupuncture (n = 45) groups. A model of permanent right middle cerebral artery occlusion (MCAO) was induced by the intraluminal filament method. After wakening, the rats in the electroacupuncture group were treated with electroacupuncture by choosing the following acupoints; Neiguan, Waiguan, Zusanli, and Sanyinjiao, the needle retaining time was 30 minutes, once a day. Neurological deficit, striatum Na (v) 1.6 expression (immunofluorescence staining and real-time fluorescence quantitative PCR method), and infarct volume changes in rats were observed at 6 hour, days 1, 2, 3, and 7 after ischemia. Results: Circled digit oneNeurological deficit was most severe at day 2 after ischemia in the ischemic group and electroacupuncture group; at the same time point, the neurological deficit in the ischemic group was more severe than that in the electroacupuncture group (P < 0.05). Circled digit twoImmunofluorescence staining showed that the expression of Na (v) 1.6 reached the peak at day 1 in both the ischemic and electroacupuncture groups, and then it began to be down-regulated. Real-time quantitative PCR showed that the expression of Na (v) 1.6 was up-regulated from 6 hour to day 1, it was highest at day 1, and the expression showed a downward trend from day 2 to day 7; the expression of Na (v) 1.6 was down-regulated from 6 hour to day 7 in the electroacupuncture group. At the same time point, the above two methods showed that the expression of Na (v) 1.6 was down-regulated in the electroacupuncture group compared to the ischemic group (P < 0.05). Circled digit threeThe infarct volume became the largest at day 3 after ischemia in both ischemic and electroacupuncture groups. At the same time point, the infarct volume in electroacupuncture group was smaller than that in the ischemic group (P<0.05). Conclusion: Na (v) 1.6 may be involved in the pathogenic process of ischemic brain injury. Electroacupuncture may exert protective effect for ischemic cerebral injury through the down-regulation of the expression of Na (v) 1.6.

Abstract:Objective: To observe the expression of Na(v)1.1 and Na(v)1.6 after the electroacupuncture therapy (ET) on acute cerebral ischemia, and discuss the mechanism of function of ET.Methods: Focal acute cerebral ischemia model was established by the occlusion of right middle cerebral artery. Rats were randomly divided into sham operation control (SC), ischemia control (IC) and ET groups. Four acupoints, 'NEIGUAN', 'WAIGUAN', 'SANYINJIAO', and 'ZUSANLI', were selected to be acupunctured. Immunofluorescence and real-time PCR methods were used to detect Na(v)1.1 and Na(v)1.6 expression, and 2,3,5-triphenyl tetra-zolium chloride staining was used to detect infarct volume at 6 hours, 1, 2, 3, and 7 days after ischemia.Results: There is no change in the expression of Na(v)1.1 and Na(v)1.6 in SC group. After ischemia the expression of Na(v)1.1 and Na(v)1.6 was up-regulated compared with that of SC group. The Na(v)1.1 expression was down-regulated from 6 hours to 2 days, then up-regulated from 3 to 7 days. The Na(v)1.6 expression was up-regulated from 6 hours to 1 day, then down-regulated from 2 to 7 days after ischemia. In ET group the neurological deficit behavior, the change in Na(v)1.1 and Na(v)1.6 expression, and the infarct volume were more dramatic than those in IC group at the same time point, and the difference had a statistic value (P < 0.05).Conclusion: Na(v)1.1 and Na(v)1.6 play a role in the injury of cerebral ischemia, ET could regulate the expression of Na(v)1.1 and Na(v)1.6 after ischemia, reduce the infarction volume and decrease cerebral ischemic damage. ET had a cerebral protective function, and one of its important mechanism may be it could regulate the expression of Na(v)1.1 and Na(v)1.6 after ischemia.

Abstract:Objective: To investigate whether electro-acupuncture can improve neurological function and up-regulation the expression of Angiopoietin/Tie pathway after focal brain ischemia. Method: Eighteen male adult Sprague-Dawley rats were subjected to 60min right middle cerebral artery occlusion(MCAO). All rats were randomly assigned to one of the three groups: electro-acupuncture group, 2 week control group and sham group. Electro-acupuncture group undergo 30min electro-acupuncture treatment per day for 2 weeks. Result: After 2 weeks, the neurological score of electro-acupuncture group were higher when compared with spontaneous recovery(P0.05). Angiopoietin-1、angiopoietin-2 and its recepter Tie-2 were up-regulated in electro-acupuncture group(P0.05). Conclusion: Electro-acupuncture can improve neurological function after focal brain ischemia, which may associated with the up-regulation of Angiopoietin/Tie pathway.
Electro‐nape‐acupuncture regulates the differentiation of microglia through PD‐1/PD‐L1 reducing secondary brain injury in acute phase intracerebral hemorrhage rats

Abstract:Introduction This study aimed to investigate the effect of electro‐nape‐acupuncture (ENA) on the differentiation of microglia and the secondary brain injury in rats with acute‐phase intracerebral hemorrhage (ICH) through the programmed cell death protein‐1/ligand‐1 (PD‐1/PD‐L1) pathway. Methods A total of 27 male Sprague‐Dawley rats were randomly divided into three groups: sham group, ICH group, and ENA group. The autologous blood infusion intracerebral hemorrhage model was used to study the effects of ENA by administering electroacupuncture at GB20 (Fengchi) and Jiaji (EX‐B2) acupoints on 24 h after the modeling, once per day for 3 days. The neurological function damage, hematoma lesion, and inflammatory cell infiltration were measured by the beam walking test and hematoxylin‐eosin staining. The expression of PD‐1, PD‐L1, CD86, CD206, and related cytokines around the hematoma was measured by western blot, quantitative reverse transcription polymerase chain reaction, and immunofluorescence. Results The ICH group had significant neurological deficits (p < .001), hematoma lesions, and inflammatory cell infiltration. The levels of CD86 protein, inflammatory factors tumor necrosis factors (TNF)‐α, interleukin (IL)‐1β, and IL‐6 were increased (p < .001), while CD206 protein was reduced (p < .01), and the number of CD86+/CD11b+ cells was also increased (p < .001) compared to the sham group. However, after ENA intervention, there was a significant reduction in neurological function damage (p < .05), infiltration of inflammatory cells, and the expression levels of CD86+/CD11b+ cells (p < .05), resulting in the increased expression of PD‐1 protein and differentiation of M2 phenotype significantly (p < .001). Conclusion The study concludes that ENA could reduce neurological function damage, inhibit the expression of pro‐inflammatory cytokines, and improve the infiltration of inflammatory cells to improve secondary brain injury in acute‐phase intracerebral hemorrhage rats. These effects could be related to the increased expression of PD‐1 around the lesion, promoting the differentiation of microglia from M1 to M2 phenotype.
