Effect of cadmium stress on ammonium assimilation enzymes and polyamine in the roots of rice (Oryza sativa) with different cadmium resistance

Abstract:The characteristics of ammonium assimilation enzymes and polyamine (Spm) in roots of rice under Cd stress are not clear, meanwhile, the difference between rice cultivars is also worth to explore. In order to investigate the effects of ammonium assimilation enzymes and polyamine on the stress of cadmium, a nutrient solution hydroponic experiment was conducted to investigate the effect of ammonium assimilation enzymes and polyamine (PAs)of seedling roots to cadmium(Cd) stress with two different rice varieties (D38B and Chuannongyou527). The results showed that the more Cd was accumulated in chuannonyyou527 than D83B with the increased Cd treatment; Cd stress was effective in increasing glutamate dehydrogenase (GDH) activity, decreasing glutamine synthetase (GS) and glutamate synthase (GOGAT) activity. This change is significantly larger in D83B than Chuannongyou527 under high-Cd (25μM) condition; Although Cd stress induce high arginine (Arg) content and ADC activity in Chuannongyou527, but the putrescine (Put) accumulation is not significantly different in two cultivars. At the same time, a concomitant decrease in spermidine (Spd) and spermine (Spm) was observed. However, Chuannongyou527 maintained a relatively high level of (fSpd+fSpm)/fPut compared with D83B under Cd stress. These results suggest that, in general, treatment with Cd affects nitrogen assimilation and polyamine synthesis to a greater extent in D83B than in Chuannongyou527. © Research India Publications.
