Abstract:A sugar functionalized naphthalimide derivative (H1) self-assembles into supramolecular nanotubes (25 nm pore diameter) by the reaction of 4-N-ethylaminenaphthalimide-N-propinyl and delta-gluconolactone in refluxed ethanol. The suspension of the tube assembly in water can directly form hydrogels when triggered by sonication, without change in morphology or molecular aggregates in the pH range of 5-8. Modified with aminocarproic acid, H2 with more hydrogen bonding sites can form pH tolerant hydrogels in the widest range of pH values from 1-14 accelerated by sonication. The gelation mechanism was studied in detail. To the best of our knowledge, this is the first paradigm wherein hydrogels were constructed from naphthalimide derivatives. Finally, the potential of the hydrogel as a drug delivery and release system for hydrophilic medicine was explored.
Encapsulation of 10-Hydroxy Camptothecin in Supramolecular Hydrogel As an Injectable Drug Delivery System

Abstract:10-Hydroxy camptothecin (HCPT) has been proven to be a cell cycle-specific chemotherapeutic agent, which is a necessary choice to inhibit tumor residue growth and prevent tumor metastasis after surgery. But it suffers from light decomposition, poor solubility, relatively low bioavailability, and some side effects, which are the major obstacles toward its clinical use. Integration of hydrophobic HCPT with hydrophilic hydrogel is a facile approach to change the disadvantageous situation of HCPT. In this study, a novel supramolecular hydrogelator with improved synthetic strategy was triggered by chemical hydrolysis, and then self-assembled to hydrogel. Taking advantage of the high-equilibrium solubility of HCPT in hydrogelator solution, this hydrogel was utilized to load HCPT via encapsulation as an effective carrier. HCPT hydrogels were characterized by several techniques including transmission electronic microscopy, rheology, and UV spectroscopy. In vitro release experiment indicated HCPT hydrogel could maintain long term and sustained release of HCPT at high accumulated rate. 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide assay showed that HCPT hydrogel had an optimized anticancer efficacy. Besides, with prominent physical properties of carrier, HCPT hydrogel possessed satisfactory stability, syringeability, and recoverability, demonstrating itself as a potential localized injectable drug delivery system. © 2015 Wiley Periodicals, Inc. and the American Pharmacists Association J Pharm Sci 104:2266–2275, 2015

Abstract:Hydrogels derived from fluorenylmethoxycarbonyl (Fmoc)-conjugated amino acids and peptides demonstrate remarkable potential in biomedical applications, including drug delivery, tissue regeneration, and tissue engineering. These hydrogels can be injectable, offering a minimally invasive approach to hydrogel implantation. Given their potential for prolonged application, there is a need for non-destructive evaluation of their properties over extended periods. Thus, we introduce a hydrogel characterization platform employing single-walled carbon nanotubes (SWCNTs) as near-infrared (NIR) fluorescent probes. Our approach involves generating supramolecular self-assembling hydrogels from aromatic Fmoc-amino acids. Integrating SWCNTs into the hydrogels maintains their structural and mechanical properties, establishing SWCNTs as optical probes for hydrogels. We demonstrate that the SWCNT NIR-fluorescence changes during the gelation process correlate to rheological changes within the hydrogels. Additionally, single particle tracking of SWCNTs incorporated in the hydrogels provides insights into differences in hydrogel morphologies. Furthermore, the disassembly process of the hydrogels can be monitored through the SWCNT fluorescence modulation. The unique attribute of SWCNTs as non-photobleaching fluorescent sensors, emitting at the biologically transparent window, offers a non-destructive method for studying hydrogel dynamics over extended periods. This platform could be applied to a wide range of self-assembling hydrogels to advance our understanding and applications of supramolecular assembly technologies.

Abstract:Hydrogels loaded with anticancer drugs can be regarded as depots for intratumoral chemotherapy to downsize tumors. However, the low drug loading content, the rapid drug release, and the subsequently undegraded blank hydrogel matrix may limit their applications. In this work, we prepared an injectable camptothecin (CPT) conjugated hydrogel with simultaneous drug release and degradation to resolve these problems. Poly[oligo(ethylene glycol) maleate] (POEGM) with two terminal hydroxyl groups was first synthesized by the direct polycondensation of oligo(ethylene glycol) (OEG) with maleic acid (MA). CPT, a highly hydrophobic anticancer drug, was conjugated to the chain end of POEGM via carbonic ester bond. The resultant water-soluble CPT-polymer conjugate could be in situ crosslinked by poly [oligo(ethylene glycol) mercaptosuccinate] (POEGMS) rapidly through thiol-ene "click" reaction under physiological conditions, which can be used as injectable CPT conjugated hydrogels. Bright blue fluorescence emitted from CPT-conjugated hydrogel under UV lamp demonstrated the homogeneity of CPT loading. The CPT release by the breakage of carbonic ester bond and the degradation of hydrogels by the breakage of ester bonds were observed simultaneously within one week to 16 days, depending on the solid content of hydrogels. The CPT-conjugated hydrogels exhibited significant cytotoxicity to HepG2 cells based on in vitro cell viability assays. As a contrast, the blank hydrogels without CPT conjugation showed excellent biocompatibility under the same conditions. This kind of CPT conjugated hydrogels could be a potential candidate for intratumor drug delivery.

Abstract:An amphiphilic gelatin macromolecule capable of self-assembling to form micelle-like nanospheres for entrapping hydrophobic therapeutic molecules was successfully synthesized by grafting hydrophobic hexanoyl anhydrides to the amino groups of primitive gelatin. The substitution and grafting efficiency of the modified gelatin displayed a concentration-dependent manner toward the hexanoyl anhydride. The particle size and stability of the resulting self-assembled nanoparticles strongly depend on the degree of substitution of the gelatin. These gelatin nanoparticles exhibited an outstanding cytocompatibility and highly efficient drug encapsulation efficiency. The drug release profile of the anticancer agent (camptothecin) from the amphiphilic gelatin nanoparticles shows that the drug release mechanism is dominated by a combined effect of diffusion and degradation. Furthermore, a tunable sustained release and degradation behavior can be controlled by changing the degree of substitution of the gelatin. This new type of amphiphilic gelatin nanoparticles with controlled biodegradability and rapid cellular internalization may serve in the intracellular delivery of specific proteins or genes, permitting a novel design for intracellularly-based nanotherapy.

Abstract:Poor nuclear delivery and accumulation are the main reasons for the reduced drug efficacy of many anticancer drugs that target DNA or enzymes in the nucleus, and it is a major obstacle to successful cancer therapy. To address this problem, developing practical drug delivery systems for nuclear delivery is urgently needed. Here we develop a supramolecular hydrogel by conjugating the anticancer agent 10-hydroxycamptothecine (HCPT) and macrocyclic polyamine cyclen to a self-assembling peptide. The cyclen fragment possesses nuclear localization and ATP hydrolysis properties, which can provide a synergistic therapeutic effect for cancer treatment. The HCPT-FFFK-cyclen nanofibers showed improved nuclear accumulation and inhibition capacity in cancer cells including drug-resistant cancer cells in vitro. The nanofibers also exhibited favorable ATP consuming ability in vitro. Moreover, the obtained nanomedicine showed enhanced anticancer efficiency and favorable biocompatibility in vivo when administered to mice via tail vein injection. This constructed self-delivery drug system significantly improved the delivery efficiency of the small molecule agents into the nucleus and showed favorable ATP consuming ability, offering new strategies for developing nanomedicines for cancer combination therapy.

Abstract:We reported a facile method to prepare camptothecin (CPT) loaded ultrathin hydrogel films based on a polymeric prodrug via layer by layer (LbL) thiol-ene "click" chemistry. Free camptothecin could be sustainedly released from the ultrathin hydrogel film within 24 hours through the hydrolysis of ester bonds between the hydrogel backbone and the camptothecin moieties.

Abstract:Hydrogels based on beta-D-glucopyranoside-substituent thiosemicarbazide derivatives were synthesized and used as template to fabricate netlike CdS nanofibers. The self-assembling properties of the hydrogels and their effect on the synthesis of CdS nanostructure were studied. Fourier transform infrared spectra, UV-visible absorption spectra and X-ray diffraction revealed that pi-pi stacking, hydrogen bonds and interdigitated interactions between hydrophobic chains had influence on the formation of self-assembled lamellar hydrogel. Transmission electron microscopic and electron diffraction observation showed the formation of hexagonal stacking nanofiber-based CdS semiconductor materials. The transmission electron microscopic pictures of the CdS nanostructure in difference growth stages revealed the growth mechanism of CdS nanofibers templated by hydrogel.

Abstract:Natural compounds as carriers for hydrophobic drugs have been increasingly used in drug delivery systems. In this study, disodium glycyrrhizin (Na2GA), tannic acid (TA) and camptothecin (CPT) were firstly used to prepare the camptothecin solid dispersion (CPT SD). When dissolved in a solution medium, Na2GA self-assembled to form micelles and CPT was encapsulated in micelles, meanwhile, TA connected with Na2GA through hydrogen bonds to form a contract shell. The average diameter of the CPT-loaded micelles is 80 nm with the critical micellar concentration of 0.303 mg/mL, the zeta potential of −33 mV, the PDI of 0.25 and drug loading 6.22%. In vitro experiments confirmed that the drug-loaded micelles exhibited excellent stability and permeability in the intestinal environment. Furthermore, the formulation showed excellent anti-tumor activity in vitro and in vivo. These findings imply that this nano-micelles provide a more potential and efficacious oral drug formulation for chemotherapy.

Abstract:A multiwalled carbon nanotube (MWNT)-based drug delivery system was developed by covalently combining carbon nanotubes with the antitumor agent 10-hydroxycamptothecin (HCPT) using hydrophilic diaminotriethylene glycol as the spacer between nanotube and drug moieties. The surface functionalizations of the nanotube were carried out by enrichment of carboxylic groups with optimized oxidization treatment, followed by covalently linking hydrophilic diaminotriethylene glycol via amidation reaction, and then HCPT was chemically attached to carbon nanotubes through a cleavable ester linkage. It is demonstrated that the obtained MWNT-HCPT conjugates are superior in antitumor activity both in vitro and in vivo to clinical HCPT formulation. In vivo single photon emission computed tomography (SPECT) imaging and ex vivo gamma scintillation counting analyses reveal that MWNT-HCPT conjugates have relatively long blood circulation and high drug accumulation in the tumor site. These properties together with the enhanced cell uptake and multivalent presentation of HCPT on a single nanotube benefit substantially the antitumor effects and would boost significantly the applications of carbon nanotubes in the biomedicine field.

Abstract:AbstractMost chemotherapeutics are hydrophobic molecules and need to be converted into hydrophilic form before administration. Based on the excellent hydrophilicity and programmability of DNA, now, a general strategy to construct a precise drug‐containing DNA framework for cancer treatment is reported. In this novel drug delivery system, carbonethyl bromide‐modified camptothecin (CPT) is employed to directly react with phosphorothioate (PS) modified DNAs, resulting in the formation of chemotherapeutics‐grafted DNAs with a responsive disulfide linkage. By tuning the number and site of PS modifications on DNA strands, hydrophilicity of the obtained DNA‐drug conjugates (DDCs) can be regulated to retain their aqueous solubility and capability of molecular recognition. Subsequently, programmable DNA nanotechnology enables the self‐assembly of a precise drug‐containing tetrahedral framework with stimuli‐responsive feature and enhanced antitumor efficacy both in vitro and in vivo.
Injectable hydrogel imbibed with camptothecin-loaded mesoporous silica nanoparticles as an implantable sustained delivery depot for cancer therapy

Abstract:In recent years, injectable stimuli-sensitive hydrogels are employed as suitable drug delivery carriers for the release of various anti-cancer drugs. However, large pore size of the microporous hydrogel trigger release of small molecular anticancer drug that limits hydrogel application in cancer therapy. Therefore, introducing reinforcing fillers such as mesoporous silica nanoparticles (MSNs) can not only load different type of anticancer drugs but also prevent the premature release of drugs due to the strengthening of the networks. Furthermore, high specific surface area, suitable size, large pore volume, and stable physicochemical properties of MSNs can improve the therapeutic efficacy. In this study, to sustain the release of hydrophobic anticancer drug, camptothecin (CPT) was loaded into MSNs, and then imbibed into the physiological stimuli-sensitive poly(ethylene glycol)-poly(β-aminoester urethane) (PAEU) hydrogels. MSN-imbibed PAEU hydrogels exhibited prolonged release of CPT than MSNs and PAEU hydrogel alone. Furthermore, MSN-imbibed PAEU copolymers form stable viscoelastic gel depot into the subcutaneous layers of Sprague-Dawley rats and found to be safe and not induced toxicity to healthy organs, implying biodegradability and safety of the hydrogels. Interestingly, CPT-loaded hydrogels shown dose-dependent toxicity to A549 and B16F10 cells. These results demonstrated that MSN-imbibed PAEU hydrogel with biocompatible, biodegradable, and in situ gel forming property could be a useful drug delivery depot for sustained release of anticancer drugs.

Abstract:Biotemplated nanomaterials offer versatile functionality for multimodal imaging, biosensing, and drug delivery. There remains an unmet need for traceable and biocompatible nanomaterials that can be synthesized in a precisely controllable manner. Here, we report self-assembled quantum dot DNA hydrogels that exhibit both size and spectral tunability. We successfully incorporate DNA-templated quantum dots with high quantum yield, long-term photostability, and low cytotoxicity into a hydrogel network in a single step. By leveraging DNA-guided interactions, we introduce multifunctionality for a variety of applications, including enzyme-responsive drug delivery and cell-specific targeting. We report that quantum dot DNA hydrogels can be used for delivery of doxorubicin, an anticancer drug, to increase potency 9-fold against cancer cells. This approach also demonstrated high biocompatibility, trackability, and in vivo therapeutic efficacy in mice bearing xenografted breast cancer tumors. This work paves the way for the development of new tunable biotemplated nanomaterials with multiple synergistic functionalities for biomedical applications.

Abstract:Free-standing, micropatterned silica nanotube membranes are in situ fabricated using a micropatterned silica-coated collagen hybrid hydrogel as template. They are substrate-free, and not only maintained their micropatterned microstructure well, but also exhibited strong cell contact guidance ability to direct cell alignment and differentiation, indicating their good potential for biomedical applications.

Abstract:Here, we report the synthesis and characterization of size-controllable and stimuli-responsive DNA nanohydrogels as effective targeted gene delivery vectors. DNA nanohydrogels were created through a self-assembly process using three kinds of building units, respectively termed Y-shaped monomer A with three sticky ends (YMA), Y-shaped monomer B with one sticky end (YMB), and DNA linker (LK) with two sticky ends. Hybridization at the sticky ends of monomers and LK leads to nanohydrogel formation. DNA nanohydrogels are size-controllable by varying the ratio of YMA to YMB. By incorporating different functional elements, such as aptamers, disulfide linkages, and therapeutic genes into different building units, the synthesized aptamer-based nanohydrogels (Y-gel-Apt) can be used for targeted and stimuli-responsive gene therapy. Y-gel-Apt strongly inhibited cell proliferation and migration in target A549 cells, but not in control cells. By taking advantage of facile modular design and assembly, efficient cellular uptake, and superior biocompatibility, this Y-gel-Apt holds great promise as a candidate for targeted gene or drug delivery and cancer therapy.

Abstract:PEGylated drugs and their self-assembled nanoparticles have been used as a first-line technology in nanomedicines. However, premature disassembly before reaching the lesions under dilute conditions limits their bioavailability. This challenge can be overcome by covalent crosslinking. However, undefined molecular structures of the prodrugs are introduced, which hinder large-scale production and clinical translation. Herein, a supramolecular PEGylated camptothecin nanodrug with a crosslinked architecture and defined molecular drug structure was developed. PEGylated mono-tetraethylenepentamine β-cyclodextrin (mPEG-TEPA-CD) and adamantane modified camptothecin (AD-SS-CPT) were first synthesized. The host-guest coupling of β-cyclodextrin (β-CD) and adamantine (AD) moieties between mPEG-TEPA-CD and AD-SS-CPT induced the formation of supramolecular PEGylated camptothecin, which could self-assemble into micelles in the aqueous medium. They were further stabilized by the addition of a diacrylate crosslinker for reaction with the amine groups on mPEG-TEPA-CD, maintaining the unaltered AD-SS-CPT. The nanodrugs can internalize into tumor cells quickly and release free camptothecin in response to glutathione. They also have prolonged blood circulation time and can accumulate efficiently at tumor sites, thus showing effective antitumor activity. This supramolecular drug delivery system provides a new strategy for the design of nanodrugs.
