Abstract:This study examined the effects of organic manure and chemical fertilizer on soil microflora, soil respiration, number of inorganic phosphate solubilizing bacteria, and organic phosphorus (P) mineralizing bacteria. The inorganic phosphate solubilizing rate, organic P mineralizing rate, and selected enzyme activities in a blue purple paddy soil were also studied. The results showed that organic manure significantly increased the total number of fungi, actinomyces, bacteria, P solubilizing bacteria, organic P mineralizing bacteria, P solubilizing rate and organic P mineralizing rate, soil respiration rate, and selected enzyme activities, whereas chemical fertilizer resulted in a smaller effect, and bacteria were affected more than fungi and actinomyces by organic manure. The enhancement of biological activities caused by organic manure might be due to the introduction of a large amount of living microorganisms and readily-utilizable carbon source on which microorganisms live. This study showed that the augment of inorganic phosphate solubilizing bacteria and organic P mineralizing bacteria was one of the reasons that organic manure increased the avilability of P in a blue purple paddy soil.
Unveiling the Role of Dissolved Organic Matter on Phosphorus Sorption and Availability in a 5-Year Manure Amended Paddy Soil.

Abstract:Dissolved organic matter (DOM) is an active component of organic manure that is widely used in agroecosystems to increase nutrient availability and consequently enhance crop yields. However, the ways in which soil DOM characteristics are influenced by organic manure and how it contributes to crop yield and soil P availability remains unclear. Here, we conducted a 5-year field experiment and demonstrated that partial replacement of chemical P fertilizer with swine manure could maintain high rice yield and soil available P levels and increase P fertilizer use efficiency (PUE) in comparison to chemical fertilization, even when the total P input was reduced. This suggests that organic manure application can significantly mobilize soil P and increase P availability. Structural equation modeling analysis indicated that the soil pH and humification degree of DOM, rather than DOM content, directly decreased maximum P adsorption capacity. The combined results of the optical spectroscopy and ultrahigh-resolution mass spectroscopy obtained from the laboratory validation experiment based on the DOM-removed soil demonstrated that manure-derived DOM competing with P for adsorption was one of the main reasons for the increase in soil P availability and that the effective DOM components were N-containing lignins, tannins, and condensed polycyclic aromatics with higher O/C and lower H/C ratios. Overall, our results provide solid evidence that soil DOM characteristics are influenced by manure application and facilitate soil P availability, which could help guide the sustainable P management and manure application in agroecosystems.
Effects of Long-Term Organic Material Applications on Soil Carbon and Nitrogen Fractions in Paddy Fields

Abstract:Soil organic carbon (C) and nitrogen (N) pools are the most important soil components, and their characteristics directly or indirectly determine the stability of the soil structure and the soil fertility. Using a rice fields experiment in the coastal areas of Southeast China, how a long-term (10 years) continuous application of organic materials consisting of green manure (Chinese milk vetch), pig manure, and rice straw affected the soil C and N fractions were studied. The results showed that 10-year continuous applications of organic materials to the paddy fields significantly increased the C fraction content in terms of parameters such as the soil organic C (SOC), microbial biomass C (MBC), and active organic C (AOC). It also increased the soil N fraction content, including the soil total N (TN), dissolved total N (DTN), microbial biomass N (MBN), and active N. The rice straw return had the most significant effect in relation to increasing the SOC and dissolved organic C (DOC), with 23.9% and 42.3% increases, respectively, compared to applying chemical fertilizer alone. Organic pig manure had the most significant effect on the soil MBN and DTN, with 49.3% and 50.7% increases, respectively, compared to the application of chemical fertilizer alone. MBC and MBN are the primary components of soil AOC and active N, respectively. The application of organic materials increased the ratio of soil MBC and AOC to SOC, as well as the ratio of MBN, DTN, and active N to the soil TN. Our findings indicated that long-term applications of organic materials could Improve and enhance paddy soil fertility and promote increased rice yields.
High-molecular-weight Dissolved Organic Matter Enhanced Phosphorus Availability in Paddy Soils: Evidence from Field and Microcosm Experiments

Abstract:Dissolved organic matter (DOM) derived from organic fertilizers may increase soil phosphorus (P) availability. However, the frequently observed correlation between soil P availability and dissolved organic carbon (DOC) content has led to an excessive focus on DOC content at the expense of DOM properties. The present study investigated the influence of DOM characteristics on soil P availability by using a 6-year field experiment and microcosms of P sorption in paddy soil. Our results showed that partial replacement of chemical P fertilizer with manure or crop straw increased P fertilizer-use efficiency, even when decreasing chemical P input by 34%, compared to normal chemical fertilization. The microcosm experiment demonstrated that DOM properties, rather than total DOC content, determine soil P sorption capacity, despite the significant correlation between DOC content and P availability observed in the field experiment. Manure-DOM exerted stronger inhibitory effects on P sorption than straw-DOM, and high molecular weight (HMW)-fractionated DOM exerted stronger inhibitory effects on P sorption than low-molecular-weight-fractionated DOM by 16–20%. The mechanism is primarily attributed to high molecular weight (HMW) DOM, which possesses unique characteristics such as strong aromaticity. These attributes lead to competition for soil P sorption sites and a decrease in soil surface charge, collectively suppressing P sorption in paddy soil. Our study points out a promising avenue for regulating organic matter properties with organic fertilization to improve P use efficiency in agricultural soils.
Organic Matter Accelerated Microbial Iron Reduction and Available Phosphorus Release in Reflooded Paddy Soils

Abstract:The cycling of soil phosphorus (P) is inherently linked with soil organic carbon-iron (C-Fe) cycling, yet empirical integration of these processes within paddy soils remains scarce. In this study, we conducted a microcosm experiment using paddy soils subjected to six distinct fertilization regimes involving varying P inputs for five years. In addition to evaluating P activation under reflooding conditions, we assessed the Fe reduction process and characterized the properties of dissolved organic matter (DOM) at the molecular level using Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR MS), alongside profiling the composition of soil microbial communities with high-throughput sequencing. Our findings revealed that after 25 days of reflooding, soil Olsen-P content increased by an average of 73% compared to its initial state, showing a strong correlation with the Fe reduction process. Specifically, treatments involving pig manure application exhibited higher Fe reduction rates and enhanced P activation, highlighting the role of organic matter in facilitating Fe reduction. Investigations on the relative abundance of typical iron-reducing microbes further supported their importance in P activation, but the rate of iron reduction is limited by soil organic matter content. Delving deeper into DOM properties, soil DOM composition profiling and network analysis suggested that high-molecular-weight DOM, particularly lignins, served as the primary resources driving Fe reduction by iron-reducing microbes, consequently promoting Fe reduction and P release. Taken together, our study assembled the C-Fe-P cycling dynamics in paddy soils, emphasizing the pivotal role of microbial-driven Fe reduction facilitated by soil DOM in P availability and subsequently sustainable agricultural practices.
Differences in Carbon and Nitrogen Mineralization in Soils of Differing Initial Ph Induced by Electrokinesis and Receiving Crop Residue Amendments

Abstract:Initial soil pH has been reported to have a great impact on the decomposition of added organic materials and hence to determine the direction and magnitude of subsequent soil pH changes. However, most previous investigations have been conducted on different soil types differing in initial pH and other soil properties. Here, we investigated the effects of initial soil pH on the subsequent soil pH and N mineralization changes caused by addition of crop residues to two soils (a Paleudalf and a Plinthudult) with different pHs induced by treatment with direct electric current. This produced pH gradients of 6.50-3.20 and 6.74-3.81, respectively. Three typical field crop residues differing in C/N ratio, i.e. rice straw C/N = 42, canola residue C/N = 36 and Chinese milk vetch C/N = 14 (vetch), were incubated with the soils for 102 days. With both soils, total CO2 fluxes differed between the type of added crop residues, with vetch > canola residue > rice straw, and decreased with decreasing initial soil pH. The incorporation of crop residues into the two soils at all pHs increased soil pH except for the Paleudalf at pH 3.81 and 4.25 amended with rice straw and canola residue, where pH slightly decreased. As expected, vetch incorporation caused a greater pH increase than the two non-legume residues (rice straw and canola residue), but this effect was transient in soils of higher initial pH. The pH declined rapidly with time in soils of initial pH >= 4.40 treated with vetch, while addition of rice straw and canola residue maintained stable pHs. Irrespective of crop residue addition, soil pH continuously increased or remained steady over the 102-day incubation period in the highly acidic soils (pH <= 4.25). While NH4+ accumulated in highly acidic soils, NO3- accumulated in higher pH soils. Contrasting effects on nitrification and subsequent pH changes occurred between soil pH <= 4.25 and pH >= 4.40. However, no clear relationship between net N mineralization and soil pH was found. This study demonstrated that low soil pH greatly inhibited nitrification, while net N mineralization was generally less affected. Effect of Organic Manure on Phosphorus Adsorption-Desorption and Availability in Paddy Soil Derived from Red Earth

Abstract:A field test with the traditional rotation of paddy rice/upland crop (wheat)was carried out on a paddy soil derived from red earth to elucidate the effect of organic manure on the phoshporus adsorption-desorption by soil and its P availability.Soil samples were taken from different treatments at rice havesting stage and analysed.The isothermal adsorption of P by the samples fitted very well with Langmuir equation,and hence,the parameters in the equation,i.e.,maximum adsorption(qm),constant related to bonding energy(k) and their product(k×qm) could be used as a comprehensive index to characterize the potential P adsorptivity of the soil.Organo-inorganic fertilization and organic manuring could decrease qm and k,while mineral P application had little effect on them.The isothermal desorption of P was significantly correlated with initially added and isothermally adsorbed P.Part of P added was fixed,which represented the P fixation capacty of soil,and organic manuring could obviously lower the P fixation,The content of soil available P had a significant negative correlation with qm,k and fixed P.It is conclued that organic manure could increase the P availability of paddy soil dervied from red earth by decreasing qm.k,maximum buffering capacity(MBC=k×qm)and fixation capacity.
Soil Microbial Biomass and pH as Affected by the Addition of Plant Residues

Abstract:The soil microbial biomass is involved in the decomposition of organic materials and thus, the cycling of nutrients in soils. Reductions in the size and activity of the microbial biomass are frequently used as an early indicator of changes in soil chemical and physical properties resulting from management and environmental stresses in agricultural ecosystems. In a laboratory-incubated soil, we found a strong relationship between microbial biomass C and microbial biomass N. Irrespective of the type of plant residues added, soil pH was significantly correlated with microbial biomass C and microbial biomass N. Different C/N ratio of the residues was the main characteristic that affected soil microbial biomass C, N and soil pH. Microbes played a main role in plant residues decomposition and indirectly influenced of soil pH.
The role of plant residues in pH change of acid soils differing in initial pH

Abstract:Reports on the effect of plant residues on soil pH have been contradictory. The conflicting accounts have been suggested to result from differences in compositions and types of plant residues and characteristics of soils. This incubation study examined the effect of plant residues differing in concentrations of N (3–49gkg−1) and of alkalinity (excess cations) (220–1560mmolkg−1) on pH change of three soils differing in initial pH (3.9–5.1 in 0.01M CaCl2). The addition of plant residues at a rate of 15gkg−1 soil weight increased the pH of all soils by up to 3.4 units and the pH reached the maximum at day 42 after incubation for Wodjil (initial pH 3.87) and Bodallin (pH 4.54) soils and day 14 for Lancelin soil (pH 5.1). The amount of pH buffering was decreased by residue addition in Wodjil soil, increased in Bodallin soil and remained unchanged in Lancelin soil, which closely related to changes of soil pH. Residue addition increased NH4+ concentration and the increase in NH4+ concentration generally correlated positively with the concentration of residue N. The NH4+ concentration increased with time, reached the peak at Days 42–105 for Wodjil soil, Days 14–105 for Bodallin soil and Days 14–42 for Lancelin soil, and then decreased only in Lancelin soil. The concentration of NO3− was kept minimal in Wodjil and Bodallin soils. In Lancelin soil, NO3− concentrations increased with incubation time from days 14–28. Irrespective of plant residue and incubation time, the amounts of alkalinity produced due to residue addition correlated highly with the sum of the alkalinity added as plant residues (excess cations) and those resulting from mineralization of residue N, with the slope of regression lines decreasing with increase of the initial soil pH. Direct shaking of soil with the residues at the same rate of alkalinity (excess cations) under sterile conditions increased the pH of the Wodjil soil but decreased it in the Lancelin soil. It is suggested that the decarboxylation of organic anions (as indicated by excess cations) of added plant residues and ammonification of the residue N causes soil pH increase whereas nitrification of mineralised residue nitrogen causes soil pH decrease, and that the association/dissociation of organic compounds also plays a role in soil pH change, depending initial pH of the soil. The overall effect on soil pH after addition of plant residues would therefore depend on the extent of each of these processes under given conditions.

Abstract:Purpose This study aimed to investigate the effect of initial soil pH and organic anion-to-acid ratio on changes in soil pH. Materials and methods Two soils (Podosol and Tenosol) along with two carboxylic acids (malic and citric acid) and their anions (sodium malate and citrate), commonly found in plant residues, were used in this study. Stock solutions of either malic acid and disodium malate or citric acid and trisodium citrate were added to pre-incubated soils at anion-to-acid ratios of 0:100, 10:90, 25:75, 50:50, 75:25, 90:10, 100:0 and at 0.25 g C kg −1 soil. Soils were adjusted to 80 % field capacity and mixed thoroughly, and three replicates of 50 g of each soil were transferred into individual plastic cores and incubated at 25 °C in the dark for 30 days. Soil pH, respiration, NH 4 + , and NO 3 − were determined. Results and discussion Soil pH increased linearly with increasing organic anion-to-acid ratio. The addition of organic anions to soil resulted in net alkalinisation. However, the addition of organic acids immediately decreased soil pH. During subsequent incubation, soil pH increased when the organic anions were decomposed. Alkalinity generation was lower in the Podosol (initial pH 4.5) than in the Tenosol (initial pH 6.2), and was proportional to anion-to-acid ratio across all the treatments. Cumulative CO 2 -C release was approximately three times lower in the Podosol than the Tenosol at day 2 due to lower microbial activity in the low-pH Podosol. Conclusions Increasing anion-to-acid ratio of organic compounds increased soil pH. Increases in soil pH were mainly attributed to direct chemical reactions and decomposition of organic anions. Low pH decreased the amount of alkalinity generated by addition of organic compounds due to incomplete decomposition of the added compounds. This study implies that organic anion-to-acid ratio in plant residues plays an important role in soil pH change.
Carbon Compounds Differ in Their Effects on Soil pH and Microbial Respiration

Abstract:The mechanisms of soil pH change after the addition of organic matter to soil are not fully understood. The aim of this study was to investigate changes in pH after addition of carbon compounds over a 60-d incubation period. Seven organic compounds commonly found in plant residues (acetic acid, malic acid, citric acid, benzoic acid, ferulic acid, glucosamine hydrochloride and glucose) were selected based on the number and type of functional groups, and added at 0.5 mg C.g(-1) soil to two soils differing in initial pH. Addition of organic acids (R-COOH) immediately decreased pH. The magnitude of the pH decrease depended on dissociation constant of the acid and the initial soil pH. In subsequent incubation, pH was slowly returned to original levels as organic anions were mineralized, consuming H+ ions. Glucose which contains hydroxyl (R-OH) group did not alter soil pH. However, carboxyl (R-COOH) and amine (R-NH2) groups changed pH significantly. Soil respiration was also increased by the addition of C compounds. Cumulative respiration was higher in soil with malic acid, citric acid, ferulic acid and glucose than with other compounds. The addition of glucose, citric acid and malic acid resulted in priming as cumulative respiration was greater than the actual amount of C added.
[Study on mechanism of SOM stabilization of paddy soils under long-term fertilizations]

Abstract:Fourier transform infrared spectroscopy (FTIR) was applied to study the structure of soil organic matter (SOM) of paddy soils under long-term different fertilization treatments. The aim was to clarify the different distribution of SOM between different fertilization methods and between topsoil and subsoil, and to explore the stability mechanism of SOM under different fertilization treatments. The results showed that the content of topsoil organic carbon (SOC) was the highest under organic-inorganic fertilizations, with the increment of SOC by 18.5%, 12.9% and 18.4% under high organic manure (HOM), low organic manure (LOM) and straw returning (STW) respectively compared with no fertilization treatment (CK). The long-term fertilizations also changed the chemical structure of SOM. As compared with CK, different fertilization treatments increased the functional group absorbing intensity of chemical resistance compounds (aliphatic, aromaticity), carbohydrate and organo-silicon compounds, which was the most distinctive under treatments of HOM, LOM and STW. For example, the absorbing intensity of alkyl was 0.30, 0.25 and 0.29 under HOM, LOM and STW, respectively. These values were increased by 87% , 56% and 81% as compared with that under CK treatment. The functional group absorbing intensity of SOM in the topsoil was stronger than that in the subsoil, with the most distinctive difference under HOM, LOM and STW treatments. The present research indicated that the enhanced chemical resistance of functional group of SOM may contribute to the high contents of SOC in the paddy soils under long-term organic-inorganic fertilizations, which also suggested a chemical stabilization mechanism of SOM in the paddy soils.
Model organic compounds differ in priming effects on alkalinity release in soils through carbon and nitrogen mineralisation

Abstract:The influence of organic matter and its cycling on soil pH change is still unclear. This study investigated the effect of organic compounds on carbon and nitrogen dynamics and their relationship with pH changes in two soils differing in initial soil pH (Podosol of pH 4.5 and Tenosol of pH 6.2). Seven organic compounds representing common compounds in decomposing plant residues or root exudates were added to the soils and incubated for 60 d. The largest cumulative soil respiration occurred when glucose, malic acid and citric acid were added. In addition, the Tenosol had the greater respiration compared to the Podosol. The addition of organic acids (acetic, malic, citric, ferulic and benzoic acid) instantly decreased soil pH due to the dissociation of H+ from the acids. The pH of both soils was then restored over time, which was positively correlated with decomposition % of these compounds. The pH of the Tenosol amended with all the organic acids and of the Podosol with malic acid exceeded that of the control, and net alkalization occurred, with the degree of alkalization being greater with malic and citric acid. Adding organic acids to the Tenosol generally increased NH4 concentrations but decreased NO3 concentrations. The addition of glucose decreased pH in Podosol but slightly increased it in the Tenosol. The addition of glucosamine hydrochloride decreased pH due to significant nitrification. The results suggest that the addition of organic acids stimulates microbial NO3 uptake, and ammonification and decomposition of indigenous soil organic matter, resulting in a priming effect on alkalinity release, and that the degree of the priming effect is influenced by the type of organic acid and initial soil pH.
Dynamics, Biodegradability, and Microbial Community Shift of Water-Extractable Organic Matter in Rice-Wheat Cropping Soil under Different Fertilization Treatments

Abstract:Although fertilization plays an important role in determining the contents of soil dissolved organic matters or water-extractable organic matter (DOM, WEOM), knowledge regarding the dynamics, biodegradability, and microbial community shifts of WEOM in response to different fertilization treatments is very limited, particularly in rice-wheat cropping soil. Thus, in the present study, we performed biodegradation experiments using WEOM extracted from samples of soil that had been subjected to four different fertilization treatments: unfertilized control (CK), chemical fertilizer (CF), 50% chemical fertilizer plus pig manure (PMCF), and 100% chemical fertilizer plus rice straw (SRCF). UV spectrum and fluorescence 3D excitation-emission matrix analyses applied to investigate the chemical composition of WEOM revealed that all examined WEOMs were derived from microbial activity and the dominant portion comprised humic acid-like compounds. After the incubation, 31.17, 31.63, 43.47, and 33.01% of soil WEOM from CK, CF, PMCF, and SRCF treatments, respectively, were biodegraded. PMCF- derived WEOM had the highest biodegradation rate. High-throughput sequencing analyses performed to determine the microbial community before and after the incubation indicated that Sphingomonas, Bacillus, and Flavisolibacter were the predominant bacterial genera in the original inoculum derived from the four fertilization treatments. Following biodegradation, we observed that the dominant bacteria differed according to fertilization treatments: Curvibacter (43.25%) and Sphingobium (10.47%) for CK, Curvibacter (29.68%) and Caulobacter (20.00%) for CF, Azospirillum (23.68%) and Caulobacter (13.29%) for PMCF, and Ralstonia (51.75%) for SRCF. Canonical correspondence analysis revealed that, shifts in the microbial community were closely correlated with pH and specific UV absorbance at 254 nm. We speculated that the inherent traits of different WEOM and the properties of soil solutions under different fertilization treatments shaped the soil microbial community structure, thereby influencing the biodegradation of WEOM. Effects of Different Organic Fertilizers on Improving Soil from Newly Reclaimed Land to Crop Soil

Abstract:The rapid development of cities in the recent 10 years caused a reduction in the cultivated land area, which only accounts for 14% of the total land area in China. Land development and reclamation have been regarded as an effective way to compensate farmland occupation. However, most of the newly reclaimed land has poor soil fertility and suitability; in some cases, the production capacity is only 10–30% of the occupied farmland. In order to ameliorate the soil quality of the newly reclaimed land, this study evaluated the effects of commercial organic fertilizer (0.75, 1.50, and 2.25 kg/m2), mushroom residue (1.50, 2.25, and 3.00 kg/m2), biogas slurry (150, 225, and 300 kg/m2), vegetable cake (0.30 and 0.60 kg/m2), and chemical compound fertilizer (37.50 g/m2) on the pH, moisture content and organic matter content (OMC), available phosphate, total nitrogen, alkaline hydrolysis nitrogen, microbial biomass carbon and nitrogen, and number of total bacteria and phosphate-solubilizing bacteria, as well as the growth of maize seedlings. The results from this study indicate that the soil quality (OMC is an indicator) was ameliorated by chemical and organic fertilizers, in particular commercial organic fertilizers, which caused a 9.35–16.35% increase in moisture content, a 11.56–18.72% increase in pH, a 1.73–2.15 fold increase in OMC, a 338.44–491.41% increase in available P, a 36.80–48.14% increase in total N, a 95.32–128.34% increase in alkaline hydrolysis N, a 92.57–178.38% increase in total bacterial numbers, and a 7.57–20.87 fold increase in microbial biomass carbon compared with the control. The pot experiment further indicated that soil amended with commercial organic fertilizers caused a 20.35–30.55% increase in the height and a 12.50–16.67% increase in the total dry weight of maize seedlings. In addition, representative strains with the ability to dissolve phosphorus and fix nitrogen were successfully isolated using the culture method, and were then identified based on colony morphological observation and 16S rDNA sequence analysis, which help us to not only understand why organic fertilizer has great effect on soil improvement, but also provides beneficial microbial resources for further study.
Short-term Response of Organic and Inorganic Fertilizers on Dissolved Nitrogen and pH in Soil

Abstract:The short-term changes of soil dissolved nitrogen and soil pH after fertilizations were studied by pot experiment in greenhouse.The study aimed to discuss the conversion between organic and mineral nitrogen,so as to evaluate the interrelationships between dissolved nitrogen and pH after fertilization in soil.During the five weeks after Brassica chinensis were transplanted into soil amended with different nitrogen fertilizers,soil pH of cake manure,pig compost and inorganic fertilizer treatments followed sharp decreases with slow rebound;sludge compost and control treatments almost didn't alter pH.Total dissolved nitrogen(TDN),dissolved organic nitrogen(DON),NH+4-N and NO-3-N in soil under different fertilizations followed the sequence: Cake manure,inorganic fertilizer >pig compost >sludge compost >control.NH+4-N declined on the whole,while the rise trends of NO-3-N differed among four fertilization treatments.The results represent transformations of different organic fertilizers varied obviously in soil.Soil pH of cake manure and inorganic fertilizer treatments were significantly positive correlation with NH+4-N,while negative with NO-3-N;except that DON of pig compost treatment was correlated with both NH+4-N and NO-3-N negatively,the other four treatments all had significant postive correlation with TDN.
Effect of Long-term Fertilizer Application on the Stability of Organic Carbon in particle Size Fractions of a paddy Soil in Zhejiang province, China

Abstract:Effects of chemical fertilizers and organic manure on the soil organic carbon ( SOC) content in particle size fractions of paddy soil were investigated in a 17-year long-term fertilization field experiment in Zhejiang Province, China. The inherent chemical composition of silt- and clay-associated SOC was evaluated with solid-state 13C-NMR spectroscopy. Compared to CK ( no fertilizer treatment), NPKRS (NPK fertilizers plus rice straw), NPKOM (NPK fertilizers plus organic manure), NPK (NPK fertilizers) and OM (organic manure alone) treatments significantly (P ﹤ 0. 05) increased the SOC content of sand- (2-0. 02 mm), silt- (0. 02-0. 002 mm) and clay-sized ( ﹤ 0. 002 mm) fractions. However, no significant difference was observed in the accumulation of silt- and clay-associated SOC between CK and rice straw (RS) treatments. Besides, in comparison with plots applied with NPK fertilizers alone, combined application of organic amendments and NPK fertilizers facilitated the storage of newly sequestered SOC in silt- and clay-sized fractions, which could be more conducive to the stability of SOC. Based on 13C-NMR spectra, both silt and clay fractions were composed of Alkyl-C, O-alkyl-C, Aromatic-C and carbonyl-C. Changes in the relative proportion of different C species were observed between silt and clay fractions: the clay fraction had relatively more Alkyl-C, carbonyl-C and less O-alkyl-C, Aromatic-C than those in the silt fraction. This might be ascribed to the fact that the organic matter complexed with clay was dominated by microbial products, whereas the silt appeared to be rich in aromatic residues derived from plants. The spectra also showed that the relative proportion of different C species was modified by fertilization practices. In comparison with organic amendments alone, the relative proportion of Alkyl-C was decreased by 9. 1% - 11. 9% and 13. 7% - 19. 9% under combined application of organic amendments and chemical fertilizers, for silt and clay, respectively, and that of O-alkyl-C was increased by 2. 9% -6. 3% and 13. 4% -22. 1% , respectively. These results indicated that NPKOM and NPKRS treatments reduced the decomposition rate of SOC. The aromaticity, hydrophobicity and, hence, chemical recalcitrance of silt- and clay-associated SOC in the NPK fertilizer treatments were lower than those of the organically amended plots and unfertilized treatments, indicating decreased recalcitrance of SOC against decomposition. We concluded that long-term application of organic manure combined with chemical fertilizers, either through increased accumulation of both recalcitrant compounds and carbohydrates or reduced decomposition of organic matter, was a sustainable strategy for facilitating carbon accumulation of the paddy soil investigated in this study.
The Influence of Soil Properties on the Size and Structure of Bacterial and Fungal Communities along A Paddy Soil Chronosequence

Abstract:In agroecosystems, soil bacterial and fungal communities are crucial for soil and plant health because of their diverse metabolic functions. However, little is known about the effect of long term paddy management on microbial communities. This study was conducted to assess the responses of the soil bacterial and fungal communities to cultivation history along a paddy soil chronosequence by using phospholipid fatty acid (PLFA) profiling and Illumina HiSeq sequencing. Soil samples were collected from the paddy fields (50, 100, 300, 1000, and 2000 years of paddy use; P50-P2000-), the adjacent arable chronosequence (50, 100 and 300 years of land use; NP50-NP300) and mudflat (estuarine sediment, representing the parent material for paddy and arable soil reclamation). The results showed that soil microbial biomass PLFAs increased significantly with increasing soil organic C which accumulated more under paddy than arable management. Bacterial taxonomic groups assigned to Proteobacteria and Acidobacteria changed relatively in the transition from tidal wetland to agricultural land. The relative abundances of dominant bacterial phyla in paddy soil orderly changes with cultivation time. The dominant fungal phyla in all samples were Ascomycota, Chytridiomycota, Basidiomycota, Glomeromycota and Zygomycota, representing 18.50%, 18.46%, 10.02%, 7.34%, and 7.19%, respectively. The succession of fungal community structure was mainly associated with changes in Ascomycota. Correlation analysis showed that higher soil total carbon and nitrogen related to long-term cultivation were associated with lower Proteobacteria and Ascomycota, but higher Verrucomicrobia. Furthermore, different land use type differed significantly in their fungal composition, but likely had similar effects on the succession of bacterial composition, mainly the Proteobacteria and Acidobacteria. Our results indicate that orderly succession of soil bacterial and fungal communities occurred along the long-term development of paddy soil, which in turn was associated with changes in soil physicochemical properties over time.
