Abstract:Transition metal-catalysed C-H bond functionalisations have been extensively developed in organic and medicinal chemistry. Among these catalytic approaches, the selective activation of C(sp(3))-H and C(sp(2))-H bonds is particularly appealing for its remarkable synthetic versatility, yet it remains highly challenging. Herein, we demonstrate the first example of temperature-dependent selective C-H functionalisation of unactivated C(sp(3))-H or C(sp(2))-H bonds at remote positions through palladium catalysis using 7-pyridyl-pyrazolo[1,5-a]pyrimidine as a new directing group. At 120 degrees C, C(sp(3))-H arylation was triggered by the chelation of a rare [6,5]-fused palladacycle, whereas at 140 degrees C, C(sp(2))-H arylation proceeded instead through the formation of a 16-membered tetramer containing four 7-pyridyl-pyrazolo[1,5-a]pyrimidine-palladium chelation units. The subsequent mechanistic study revealed that both C-H activations shared a common 6-membered palladacycle intermediate, which was then directly transformed to either the [6,5]-fused palladacycle for C(sp(3))-H activation at 120 degrees C or the tetramer for C(sp(2))-H arylation at 140 degrees C with catalytic amounts of Pd(OAc)(2) and AcOH. Raising the temperature from 120 degrees C to 140 degrees C can also convert the [6,5]-fused palladacycle to the tetramer with the above-mentioned catalysts, hence completing the C(sp(2))-H arylation ultimately.

Abstract:A C-H arylation of thiopyran derivatives with aryl halides has been developed. Under the catalysis of Pd(OAc)2/Ag2CO3, the C-H arylation takes place at the α-position of the thiopyran ring. When dibromo-substituted compounds are used as reactants, double C-H arylations may occur on the same thiopyran ring at its α- and β-positions.

Abstract:A novel palladium catalyzed direct ortho-arylation of N-phenacylpyridinium bromide was developed. The amazing N-phenacyl group regioselectively activates the C-H bond of pyridine and automatically departs from the arylated products. A kinetic isotope effect study proved that the reaction went through a C-H-bond activation pathway and 2,6-diphenylpyridine was produced stepwise from 2-phenylpyridine.

Abstract:Palladium-catalyzed C(sp2)-H arylation of ortho C-H bonds involving 2-(1-methylhydrazinyl)pyridine (MHP) as the directing group has been investigated. The reaction proceeds smoothly under an air atmosphere to generate biaryl derivatives in an environmentally friendly manner while tolerating a wide range of functional groups. Notably, the directing group present in the product could be easily removed under mild reductive conditions.

Abstract:Palladium-catalyzed C-H activation reactions directed by a removable 2-pyridylsulfinyl group were developed. Aromatic olefination products were formed in good yields on treatment of 2-(phenylsulfinyl)pyridines with alkenes in the presence of a Pd catalyst. The reaction tolerates a wide range of substituted alkenes, including various acrylates and styrenes. The controlled experiments indicated that the 2-pyridyl moiety, rather than the sulfinyl, played the role of ligand. The final reductive desulfonylation affords the stilbenes, sulfides, and disulfides with different reductive conditions, respectively. More importantly, this transformation could also be applied in arylation through dual C-H activation.

Abstract:Palladium-catalyzed [4+2] annulation of aryl and alkenyl carboxamides with 1,3-dienes via C–H functionalization is developed using air as the terminal oxidant. The method demonstrates good functional group tolerance and high stereoselectivity, affording a series of 3,4-dihydroisoquinolones and 5,6-dihydropyridinones in yields of up to 99%.

The reaction employs aryltrifluoroborates as the arylation reagent, leading to the corresponding products in moderate to good yield with broad substrate scope. Abstract:prehalogenation of the substrates is required, and the formation of undesired halide by-products is unavoidable. In addition to the conventional Heck reaction, the use of organometallic reagents is also an attractive strategy. These reactions usually require a catalytic amount of Pd in the presence of an appropriate oxidant (Scheme 1, path B). Despite the wide application of this “oxidative Heck reaction”, preactivation of the aromatic carbon fragments is also required. Catalytic functionalization of arenes via C H bond activation provides a potentially economic and clean method for making many chemicals directly from hydrocarbons. Fujiwara et al. developed an efficient coupling of arenes and activated alkenes through an oxidative palladium(II)-catalyzed process, which is known as the Fujiwara– Moritani oxidative Heck reaction (Scheme 1, path C). Since then, substantial progress has been made to improve the efficiency and practicality of this reaction, such as the improvement of the regioselectivity, and the use of molecular dioxygen as a reoxidant. Abstract:An enantioselective palladium-catalyzed C-H arylation of functionalized pyrazoles/triazoles/imidazoles is developed, affording a variety of axially chiral ortho-nitro/formyl-substituted heterobiaryls with excellent enantioselectivities and good yields. The method features a deuterated P-chiral phosphorus ligand CD(3)-AntPhos, a broad substrate scope of functionalized heterobiaryls, mild reaction conditions, and low palladium loadings.

Abstract:Directed C–H arylations have proven to be some of the most advantageous strategies for the synthesis of biaryls. However, their sensitivity toward steric hindrance is a key limitation. Couplings of 2,6-disubstituted aryl halides with arenes have so far been elusive. This weakness is overcome by a ruthenium 3,4,7,8-tetramethyl-1,10-phenanthroline catalyst. It allows the selective ortho-C–H arylation of widely available (hetero)aromatic acids with bulky aryl halides in up to 95% isolated yield. 46 examples of tri-substituted (hetero)biaryls, all outside the scope of established catalyst systems, demonstrate the efficiency of the protocol. Computational and experimental studies illustrate how this unique combination of carboxylate directing group and chelating N-ligand facilitates the selectivity determining C–H activation step. The preference for oxidative addition of the aryl halide over competing benzoic acid coordination is decisive to suppress unwanted dehydrogenative homocoupling.

Abstract:The direct transformation of unactivated C(sp(3))-H bonds into C-halogen bonds was achieved by palladium catalysis at room temperature with good functional group tolerance. Some drugs and natural products were readily modified by this method. Merged with substitution reaction, newly formed C-X bonds can be transformed into diverse C-O, C-S, C-C and C-N bonds. A preliminary mechanism study demonstrates that solvent is crucial for C-H activation and the C-H activation step is involved in the rate-limiting step. An isolated Pd(ii) intermediate can be transformed into a halogenated product with the retention of conformation which suggests that concerted reductive elimination from Pd(iv) to form a C-X bond was favored.

Abstract:Palladium-catalyzed C-H direct arylation generally occurs on the ortho-position of directing groups. By comparing meta-arylated products of 2-naphthyl urea to ortho-arylated products of phenyl urea, the ortho- and meta-regioselectivity of aryl ureas were found to depend on the aromaticity of the corresponding aryl substituents. Thus, aromaticity is a new factor which can affect the regioselectivity in C-H direct arylation. The finding was further confirmed by regioselective direct arylation of indole and pyrrole derivatives.

Abstract:Selective functionalizations of aryl C-H bonds in 2-arylpyridines have been developed via sequential borylation and aerobic oxidative copper catalysis, and the corresponding aryl halides, sulfones, azides and arylamines were obtained in good yields. The protocol uses cheap and readily available boron tribromide (BBr3) as the borylating reagent, and in-organic salts (potassium iodide, ammonium bromide, sodium alkylsulfinates, sodium azide) as the functional group sources. This method makes functionalizations of aryl C-H bonds easy.
