Abstract:A gold-catalyzed regioselective [3 + 2] cycloaddition of alkynyl triazenes with 1,2,4-dioxazoles was developed. The triazene group in the products could be replaced to obtain iodo-oxazoles, providing potential transformations to diverse oxazole structures. This protocol features readily available starting materials, mild reaction conditions and scalability. A plausible mechanism involving a nitrene transfer process was proposed.

Abstract:The C-O bond cleavage for activation of alcohols is synthetically useful and practically challenging. This work describes carbazate as a new type of electrochemically activated alkylating agent derived from ubiquitous alcohols for direct functionalization of heteroarenes under mild electrolytic conditions. The simple undivided cell at low oxidative potentials with carbon/platinum electrode set-ups offers excellent substrate tolerance, affording a variety of primary, secondary and tertiary alkyl-decorated heterocycles in good chemical yields. Furthermore, the mechanism for this electrochemical deoxyalkylation reaction has been investigated.

Abstract:The copper-catalyzed cyclization of activated alkenes with cyclobutanone O-acyl oximes under redox-neutral conditions has been reported. This facile protocol provided an efficient approach to a variety of cyanoalkylated oxindoles and dihydroquinolin-2(1H)-ones with a broad substrate scope and excellent functional group tolerance. In this reaction, sequential C-C bond cleavage, radical addition, and cyclization processes were involved, wherein multiple bonds were constructed in a one-pot reaction. Mechanistic studies suggest that the reaction probably proceeded via a radical pathway.

Abstract:A rhodium-catalyzed coupling-cyclization reaction of alkenyldiazoacetates with o-alkenyl arylisocyanides has been developed. In this reaction, the highly reactive alkenylketenimine intermediates generated by coupling reaction of alkenyldiazoacetates with o-alkenyl arylisocyanides undergo intramolecular [4 + 2] cycloaddition and provide a novel, highly efficient method for the one-step synthesis of carbazoles by formation of three new bonds and two rings from readily available acyclic starting materials.

Abstract:An array of redox-neutral alkylation/cyclization cascade reactions of N-functionalized acrylamides with cycloalkyl hydroperoxides were achieved via the alkoxyl radical-triggered C-C bond cleavage. Through adjusting the radical acceptors on the N atom, a variety of keto-alkylated chain-containing azaheterocycles, including indolo[2,1-a]isoquinolin-6(5H)-ones, quinoline-2,4-diones, and pyrido[4,3,2-gh]phenanthridines were constructed by a one-pot procedure with good yields and excellent functional group tolerance.

Abstract:An efficient method for the synthesis of isoquinolinone derivatives via photopromoted carboamination of alkynes is developed. Starting from the readily available propargyl alcohol derivatives, the polycyclic isoquinolinone derivatives could be obtained with good aryl and heterocycle tolerance. Both terminal and alkyl substituted alkynes could be employed. This protocol is operationally easy, and easily conducted on a gram-scale. A possible mechanism involving radical addition and cyclization following aromatization was proposed.

Abstract:A silver oxidant/Lewis acid catalyst cooperatively promoted three-component carbo-heterofunctionalization of alkenes is reported. A broad range of nucleophiles, including H2O, alcohols, and anilines, can combine with a variety of 1,3-dicarbonyl compounds or analogues as radical precursors to realize difunctionalization of various styrene derivatives. Interestingly, mechanistic investigations demonstrate that the Ag(I) complex of bidentate phosphine ligand quickly oxidizes the radical intermediate generated from the addition of initial radical across alkene double bonds to form carbocation intermediates, which may be the reason behind the excellent yields and high selectivity of such reactions.

Abstract:An electrochemical oxidation-induced cyclization of ortho-alkyl-substituted azobenzenes has been developed. The direct electrochemical benzylic C-H functionalization with respect to azobenzenes could proceed in the absence of any catalyst or external chemical oxidant to afford a number of 2H-indazole derivatives in moderate to good yields. This protocol enables the reuse of the byproduct to the same 2H-indazoles, thus significantly reducing pollution discharge in synthetic chemistry.

Abstract:A strategy for synthesis of azo‐derived compounds by electrochemical oxidative ring‐opening reactions of 2H‐indazoles with alcohols is reported. The reactions are carried out under galvanostatic electrolysis conditions in an undivided cell without the use of transition metal catalysts. On the basis of cyclic voltam¬metry and control experiments, a plausible mechanism for this ring‐opening reaction involving the radical pathway is proposed. Furthermore, various 2H‐indazoles and alcohols are suitable in this system, giving the desired ortho‐alkoxycarbonylated azobenzenes in 9‐84% yields.

Abstract:An effective and straightforward Ag(I)-mediated annulation of 2-(2-enynyl)quinolines and N'-(2-alkynylbenzylidene)hydrazides was developed, forging various synthetically challenging 17bH-isoquinolino[2'',1'':1',6']pyridazino[4',5':3,4]pyrrolo[1,2-a]quinolines, including different nitrogen-containing fused rings, in moderate to excellent yields. This one-pot cycloaddition strategy features exclusive regioselectivity, high atom economy, and broad substrate scope under mild conditions. The practicality and reliability of this cycloaddition reaction was demonstrated by a successful scale-up synthesis.

Abstract:A practical copper-catalysed oxidative radical alkylarylation of activated alkenes using AIBN (azobisisobutyronitrile) and related reagents has been developed. This has allowed the general incorporation of nitrile moieties into oxindoles by a cascade addition/C(sp2)–H cyclization process. This protocol demonstrates for the first time that DIAD (diisopropyl azodicarboxylate) can act as a new activator for the C(sp3)–H functionalization of acetonitrile instead of the usual Ag salts. The use of an inexpensive copper salt as the catalyst, the general access to primary and tertiary nitriles, as well as the air- and moisture-tolerant reaction conditions make this protocol a highly attractive approach to cyano-containing oxindoles.

Abstract:A new method of cobalt-catalyzed synthesis of pyrrolidinones from aliphatic amides and terminal alkynes was discovered through a C-H bond functionalization process on unactivated sp(3) carbons with the silver cocatalyst using a bidentate auxiliary. For the first time, a broad range of easily accessible alkynes are exploited as the reaction partner in C(sp(3))-H bond activation to give the important 5-ethylidene-pyrrolidin-2-ones in a site-selective fashion. The reaction tolerates a wide variety of functional groups including -F, -Cl, -Br, -CF3, ether, cyclopropane, and thiophene. Both pyridine ligand and aromatic solvent play the important role for the promotion of reactivity. This cobalt-catalyzed cyclization reaction can be successfully extended to a variety of aromatic amides to afford a variety of isoindolinones. Attractive features of this catalytic system include its low cost, easy operation, and convenient access to a wide range of pyrrolidinones and isoindolinones.
