Abstract:We report a novel strategy for the synthesis of chiral 2-oxazolidinones via a dinuclear copper-catalyzed asymmetric propargylic amination-carboxylative cyclization sequence of propargylic esters with nucleophilic alkyl amines under ambient pressure of carbon dioxide. A variety of chiral 2-oxazolidinones featuring an exocyclic methylene moiety was obtained in good yields with high enantioselectivities via a one-pot operation.

Abstract:A gold-catalyzed asymmetric intermolecular [3 + 2]-cycloaddition of gamma-1-ethoxyethoxy-propiolate and aldehydes by the use of the chiral phosphoramidite (S,R,R)-5a is developed, which provides facile access to highly substituted 2,5-dihydrofurans in up to 84% yield with up to 97% ee. Control experiments support the result that this transformation indeed proceeds via an all-carbon gold 1,3-dipole with an open carbocation rather than a S(N)2 type reaction.

Abstract:AbstractHighly enantioselective gold‐catalyzed intramolecular cyclization of N‐allenamides was implemented by utilizing a designed chiral sulfinamide phosphine ligand (PC‐Phos). This represents the first example of highly enantioselective intramolecular cyclization of N‐allenamides. The practicality of this reaction was validated in the total synthesis of (R)‐desbromoarborescidine A and formal synthesis of (R)‐desbromoarborescidine C and (R)‐deplancheine. Moreover, the catalyst system PC‐Phos/AuNTf2 proved to be specifically efficient to promote the desymmetrization of N‐allenamides in excellent yields with satisfactory ee values.

Abstract:A regio- and stereoselective stepwise (4+2) annulation of N-propargylamides and α,β-unsaturated imines/ketones has been accomplished with synergetic catalysis by a combination of a gold-complex and a chiral quinine-derived squaramide (QN-SQA), leading to highly functionalized chiral tetrahydropyridines/dihydropyrans in good to high yields with generally excellent enantioselectivity. Mechanistic studies and DFT calculations indicate that the in-situ formed alkylgold species is the key intermediate in this transformation, and the amide group served as a traceless directing group in this highly selective transformation. This method complements the enantioselective (4+2) annulation of allene reagents, providing the formal internal C-C π-bond cycloaddition products, which is challenging and remains elusive.

Abstract:We report a tandem asymmetric Cu-catalyzed propargylic animation (ACPA)/Ag-catalyzed carboxylative cyclization (SCC) with CO 2 to 5-methylidene-2-oxazolidinones, even with tetrasubstituted stereocenters. By varying pyridine bisoxazoline (PYBOX) ligands, a general and highly enantioselective ACPA of unprecedentedly broad scope of secondary propargylic acetates and primary amines is achieved. Both α-aryl and α-aliphatic propargylic acetates could react with either aromatic or aliphatic primary amines to give the corresponding N -aryl or N -aliphatic ethynylamines with α-aliphatic or α-aryl groups in over 90% ee for use in the next step. The key to developing this one-pot sequence is to use a chelator triethylenetetramine (TETA) to mask the copper ion, to avoid its negative effect on Ag-catalyzed cyclization, whilst releasing PYBOX to activate the silver catalyst. With the methylidene moiety, these oxazolidinones can be readily elaborated. The value of the sequence is further shown by the catalytic enantioselective total synthesis of (−)-cytoxazone and the potent ezetimibe analogue.

Abstract:Novel catalytic systems consisting of cationic gold complexes, N-hydroxyphthalimide (NHPI), and transition-metal-based Lewis acids have been developed for the one-pot synthesis of functionalized oxazoles from N-propargylamides with excellent functional group tolerance. These transformations demonstrated the excellent compatibility of homogeneous gold catalysis with organocatalytic oxidative carbon-nitrogen bond formations using tert-butyl nitrite as the terminal oxidant. Moreover, oxazolecarbonitriles or carboxamides can be easily synthesized in a one-pot protocol according to the different synthetic requirements.

Abstract:A rhodium-catalyzed stepwise asymmetric 1,4- and 1,2-addition of arylboronic acids to α,β-unsaturated cyclic N-sulfonyl ketimines has been developed, providing a wide range of gem-diaryl-substituted chiral benzosulfamidates with high optical purities. C1-Symmetric chiral diene and branched chiral sulfur-olefin ligands were sequentially utilized in this double-arylation process for high stereocontrol. Further synthetic utility offers new opportunities for the facile construction of otherwise difficult to access polycyclic heterocycles.

Abstract:A simple, new type of chiral sulfinamide monophosphines, the so-called Ming-Phos ligands, is reported; these ligands could be easily prepared from inexpensive and commercially available starting materials. The Ming-Phos ligands performed well in the enantioselective gold-catalyzed cycloaddition reaction of 2-(1-alkynyl)-alk-2-en-1-ones with nitrones. Both enantiomers of the products could be obtained in good yields and with excellent diastereo- and enantioselectivity through transformations that were catalyzed by gold complexes derived from two diastereomers of Ming-Phos ligand M5 (Ar=1-naphthyl).

Abstract:Starting with vinyl sulfoxides and propargyl silane, the Au(I)-catalyzed asymmetric [3,3]-sigmatropic rearrangement of sulfonium intermediate furnished, after protodemetallation step and hydrolysis of thionium ion, the corresponding 4-oxo-2-aryl-2-alkyl-pentanal derivatives. From the fine analysis of crude mixtures representative of this transformation, formal α-arylated products of acetone (1-arylpropan-2-one derivatives) and aryl sulfanes were also isolated and characterized, depending on the substrates and reaction conditions used. Therefore, a tentative mechanistic explanation of the formation of these unexpected products was highlighted and DFT calculations have also streamlined the reactivity of these cationic gold(I)-catalyzed transformations.

Abstract:In the past decade, there have been many extraordinary advances in the development of gold-catalyzed enantioselective annulations, such as cycloadditions, cyclizations, cycloisomerizations, and tandem annulations, which are of particular interest owing to their potential for rapid construction of optically active hetero- and carbocyclic molecules. This Review summarizes the methods to construct chiral cyclic compounds by gold-catalyzed enantioselective annulations reported since 2005. The Review is organized according to the general annulation types catalyzed by chiral gold complexes or chiral gold salts, which have four main types (cycloadditions, cyclizations of C-C multiple bonds with tethered nucleophiles, cycloisomerization or cyclization of enynes, and tandem annulations), as well as some other strategies. The general reaction mechanisms of each subcategory, key intermediates for some unusual transformations, and the application of several novel ligands and chiral gold salts are also discussed.

Abstract:A novel gold(I)‐catalyzed intramolecular dehydrative amination method has been developed to synthesize diverse cyclic sulfamides efficiently. Reactions proceed smoothly at room temperature using a combination of (IPr)AuCl (5 mol%) and AgBF 4 (5 mol%). This method showcases a broad substrate scope, encompassing challenging seven‐membered ring structures, and exhibits functional group compatibility. Moreover, the reaction demonstrates high stereoselectivity, allowing the synthesis of enantiomerically enriched cyclic sulfamides from chiral secondary allylic alcohols. The synthesized cyclic sulfamides serve as valuable intermediates for generating 1,3‐diamines. This approach provides an improved route for cyclic sulfamide synthesis and a strategic entry point for obtaining essential 1,3‐diamine derivatives.

Abstract:An enantioselective cyclopropanation reaction of sulfoxonium ylides with β,γ-unsaturated ketoesters catalyzed by a chiral rhodium catalyst has been realized. A variety of optically pure 1,2,3-trisubstituted cyclopropanes was synthesized in 48-89% yields, with up to 99% ee, and with dr >20:1. Furthermore, research shows that a weak coordination between the chiral rhodium catalyst and β,γ-unsaturated ketoesters was responsible for the high diastereoselectivity and enantioselectivity of the corresponding products.

Abstract:The first intramolecular enantioselective cyclopropanation of indenes and trisubstituted alkenes was accomplished by using new chiral phosphine X5 derived gold(I) complexes. This reaction is a straightforward, efficient method for constructing [5-3-6] fused-ring compounds with two vicinal all-carbon quaternary stereogenic centers, a core structure shared by numerous pharmacological products, and bioactive compounds. The salient features of this transformation include high enantioselectivity (up to >98% ee), excellent yield (>97%), and nice functional group tolerance.

Abstract:Ordered mesoporous SBA-15 supported chiral phosphine gold (I) complexes were successfully developed. The gold(I) complexes were covalently immobilized on mercaptopropyl modified SiO(2)surface through radical reaction. In this way, the chiral carbon and chiral sulfur of the homogenous catalyst were perfectly remained in situ. And thus, the prepared catalysts demonstrated highly effective asymmetric cycloaddition activity, which was likely because the chiral induction effect of the Au complex was hardly influenced by this immobilization method. Moreover, they could be easily recovered by simple filtration and directly used for the subsequent catalytic runs. The prepared catalyst was highly stable and efficient without obvious activity loss after eight cycles.
