Abstract:As a powerful technique for the investigation of interactions,surface plasmon resonance (SPR) is suitable for related researches,especially with the purpose of active ingredient screening due to its high analytical speed,low sample consumption and capability of real-time label-free analysis.However,conventional SPR instruments and methods could only provide limited and often insufficient structural information about the sample molecules,when facing unknown samples in complex matrix,the identification of active ingredient would become a problem.Thus,the combination of SPR with analytical techniques that could give abundant structural data like mass spectrometry(MS) has been highly desired.This review aimed to give an overview of the existing methods combining SPR and MS,which were mainly based on matrix assisted laser desorption ionization (MALDI),electrospray ionization(ESI),and ambient ionization methods including direct analysis in real time (DART),dielectric barrier discharge ionization(DBDI) and direct electrospray formed on the sensor chip.The typical methods combining SPR and MS using the mentioned ionization methods were discussed.MALDI based SPR-MS methods were the main way to combine SPR and MS with a straightforward principle,and was first realized,but it could hardly be operated online.ESI based SPR-MS methods could be used in online mode,but it had to face the incompatibility of the non-volatile salts in SPR samples mimicking the physiological conditions,and usually it necessitated additional instruments like high performance liquid chromatography(HPLC) systems.As other solutions,SPR-MS interfaces employing ambient ionization methods were reported in recent years,including DART,DBDI,and direct electrospray formed on the sensor chip,showing improved performances than those of the MALDI and ESI based interfaces.The development of SPR-MS interfaces would keep going on,making it possible and suitable for further SPR-MS studies concerning unknown samples.
