Abstract:A simple and distinctive method for the ultrasensitive detection of Cu2+ and Hg2+ based on surface-enhanced Raman scattering (SERS) using cysteine-functionalized silver nanoparticles (AgNPs) attached with Raman-labeling molecules was developed. The glycine residue in a silver nanoparticle-bound cysteine can selectively bind with Cu2+ and Hg2+ and form a stable inner complex. Silver nanoparticles co-functionalized with cysteine and 3,5-Dimethoxy-4-(6′-azobenzotriazolyl)phenol (AgNP conjugates) can be used to detect Cu2+ and Hg2+ based on aggregation-induced SERS of the Raman tags. The addition of SCN− to the analyte can successfully mask Hg2+ and allow for the selective detection of Cu2+. This SERS-based assay showed an unprecedented limit of detection (LOD) of 10pM for Cu2+ and 1pM for Hg2+; these LODs are a few orders of magnitude more sensitive than the typical colorimetric approach based on the aggregation of noble nanoparticles. The analysis of real water samples diluted with pure water was performed and verified this conclusion. We envisage that this SERS-based assay may provide a general and simple approach for the detection of other metal ions of interest, which can be adopted from their corresponding colorimetric assays that have already been developed with significantly improved sensitivity and thus have wide-range applications in many areas.

Abstract:Near infrared (NIR) emission Ag 2 S quantum dots (QDs) are of great value for biochemical sensing with strong anti-interference and low toxicity. Herein, NIR fluorescence Ag 2 S QDs were synthesized successfully. Combined with the excellent oxidase-like characteristics of manganese dioxide (MnO 2 ) nanosheets, a fluorescence and colorimetric dual-mode sensor for sulfide anion was developed. MnO 2 nanosheets could effectively catalyze the oxidation of TMB to produce blue TMB oxide (ox TMB), at the same time, the fluorescence of Ag 2 S QDs could be effectively quenched by fluorescence internal filtration effect (IFE) and dynamic quenching effect. The enzyme-like activity was weakened and the NIR fluorescence of Ag 2 S QDs was restored when sulfide anion (S 2− ) was added, due to the reduction of MnO 2 to Mn 2+ .The linear ranges for fluorescence and colorimetric analysis of S 2− were 2–250 μM and 0.3–50 μM, with detection limits of 0.6 and 0.215 μM, correspondingly. The dual-mode sensor had a wider detection range, higher sensitivity and shorter reaction time, which could be used for highly selective detection of S 2− in different concentration ranges. In addition, it had been successfully applied to the determination of sulfide in water samples with satisfactory accuracy and sensitivity.

Abstract:We present a high-throughput strategy for sensitive detection of H2S by using individual spherical Au-Ag core-shell plasmonic nanoparticles (PNPs) as molecular probes. This method is based on quantification of color variation of the single PNPs resulting from formation of Ag2S on the particle surface. The spectral response range of the 51 nm PNP was specifically designed to match the most sensitive region of color cameras. A high density of immobilized PNPs and rapid color RGB (red/green/blue) analysis allow a large number of individual PNPs to be monitored simultaneously, leading to reliable quantification of color change of the PNPs. A linear logarithmic dependence on sulfide concentrations from 50 nM to 100 μM was demonstrated by using this colorimetric assay. By designing PNPs with various surface chemistries, similar strategies could be developed to detect other chemically or biologically important molecules.

Abstract:Widespread occurrence of sulfides in domestic and industrial wastewater contributes to environmental pollution and poses risks to human health. Therefore, the development of highly selective, sensitive, and rapid sulfur ion (S 2− ) detection probes in aquatic ecosystems is of paramount importance. In this study, lignin-stabilized silver nanoprisms (EHL@AgNPRs) were prepared using the seed growth and self-assembly methods. Based on this, a novel, high-performance, and environmentally friendly S 2− colorimetric detection method was proposed. Lignin is believed to coat the surface of AgNPRs through cation-π and electrostatic interactions, acting as an excellent dispersant and stabilizer to prevent aggregation and shape deformation. This allows AgNPRs to maintain localized surface plasmon resonance (LSPR) characteristics and superior colorimetric sensing sensitivity towards S 2− even after 30 d. The EHL@AgNPRs exhibited remarkable selectivity towards S 2− with a minimum detection limit of 41.3 nM. The conjugation of lignin with AgNPRs offers a highly promising approach for the rapid detection of S 2− in natural aquatic environments and for the valorization of lignin.

Abstract:Hydrogen sulfide (H2S) is a gaseous signaling agent that has important regulatory roles in many biological systems but remains difficult to measure in living biological specimens. Herein, we report a novel near-infrared (NIR) activated surface-enhanced Raman scattering (SERS) nanosensor for in vitro and in vivo H2S assay utilizing Au/Ag alloy nanoboxes with local surface plasmon resonance (LSPR) at 741 nm. This detection is realized by SERS spectrum changes of the NIR probes, which are labeled on Au/Ag nanoboxes under NIR laser irradiation, resulting from the reaction of H2S with Ag in the substrate of Au/Ag nanoboxes. Owing to the well-matching between the incident laser light frequency and the LSPR of the substrates at the NIR region, this SERS nanosensor with a high selectivity and sensitivity can be utilized to monitor the distribution of H2S in living cells and zebrafish embryos. Noticeably, the NIR SERS signal is detectable in tissues at depths of up to 12 mm, demonstrating its huge potential in the in vivo studies of pathophysiological processes involving H2S.

Abstract:A new colorimetric assay for the detection of sulfide anions with high sensitivity and selectivity is reported, utilizing Au-Hg alloy nanorods (Au-HgNRs) as probe. Au-HgNRs were prepared by modifying gold nanorods (AuNRs) with reducing agent and mercury ions. In an aqueous solution with sulfide anions, the formation of mercuric sulfide due to redox reaction between the amalgams and sulfide anions greatly changed the surface chemistry and morphology of the Au-HgNRs, leading to a red shift of the localized surface plasmon resonance (LSPR) absorption peak, accompanied by a change in colorimetric response. A good linear relationship was obtained between the LSPR peak wavelength shift and concentration of sulfide anion in the range of 1 × 10 −5 −1 × 10 −4 mol/L. The selectivity of this method has been investigated by other anions. The colorimetric sensing system successfully detected sulfide in wastewater from leather industry.

Abstract:High selectivity to specific analyte is essential for chemical sensors but difficult to achieve. For most chemical sensors, although the response to the target analyte can be more significant than interference analytes, they still show obvious responses to the interference analytes. Here, highly selective chemical sensors are developed with negligible responses to other interference vapors. Instead of the widely investigated electronically conductive metal-organic frameworks (EC-MOFs), ionically conductive MOFs (IC-MOFs) are used as the sensing materials, and the unique interaction between the ion charge carrier and the analyte is utilized to achieve high sensing selectivity. Through the modulation of the metal nodes (Cu, Co, Ni, Zn, Mg) and organic ligands (H2TCPP, H2THPP, H4BTEC), sensor arrays based on a set of IC-MOFs are fabricated and achieve highly selective detection toward volatile sulfur compounds (VSCs). H2S and CH3SH can be selectively detected at concentrations down to 1 ppb and 1 ppm, respectively. The facile preparation and low cost endow the device with disposability. These results suggest new approaches for the development of highly selective chemical sensors.
Remarkable SERS Detection by Hybrid Cu2O/Ag Nanospheres

Abstract:Cu2O nanospheres (NSs) were synthesized by modifying the glucose reduction method. Based on this method, Cu2O/Au (Ag) NSs were further prepared by in situ reduction of HAuCl4 (via electron beam evaporation of Ag). With Rhodamine 6G (R6G) as probe, the surface-enhanced Raman scattering (SERS) characteristics of the three samples were systematically studied. The experiment results showed that the enhancement factor (EF) of Cu2O/Au (Ag) NSs as 1.25 × 108 (2.74 × 109) and the ultralow detection limit (LOD) as 8.07 × 10-12 (1.13 × 10-13) M for R6G. The excellent performance of SERS may be due to the charge transfer (CT) between metal-semiconductor (MS) molecules and the strong electromagnetic field (E-field) of each hot spot. In addition, discrete dipole approximation (DDA) simulations were performed to simulate the E-field enhancement of the Cu2O and Cu2O/Au (Ag) NSs in a three-dimensional (3D) configuration. These further supported that the high SERS performance for R6G is because of the powerful E-field coupling between neighboring Au (Ag) NPs and the surface plasmon resonance (SPR) effect. The Cu2O/Ag NSs have potential in applications such as biomedicine, food safety, and environmental monitoring because of their high sensitivity and good reproducibility.
Dual-Mode Scattering Nanoprobes for Imaging Hydrogen Sulfide in Living Cells

Abstract:Hydrogen sulfide (H2S), a gaseous signaling agent, was considered to play an essential role in the regulation of many biological systems. Herein, we design a surface-enhanced Raman scattering (SERS) and plasmon resonance Rayleigh scattering (PRRS) dual-mode nanoprobe for real-time imaging of H2S in living cells. To realize the dual-mode imaging, we fabricated a core–shell nanostructure with built-in electromagnetic hot spots by embedded Raman dyes between a gold nanostar (AuNS) core and a silver shell. When exposed to sulfide, the Ag shell will rapidly transform into Ag2S, resulting in the variation of PRRS caused by the change of the dielectric constant and the reduction of the SERS signal resulted from the blocking effect of Ag2S. There was well-defined linearity between the dual signals and the sulfide concentrations from 1 to 100 nM. The limit of detection (LOD) was 87 pM for the SERS mode and 53 pM for the PRRS mode, respectively. Complementary performances were proclaimed between the two proposed sensing modes, i.e., the PRRS mode sensing based on single nanoprobes can provide local concentration information of H2S in single cells, while the SERS signal can directly provide the average concentration information of H2S in single cells. Abstract:Hydrogen sulfide (H2S) is an appealing signaling molecule that plays fundamental roles in health and disease. However, H2S-mediated selective chemical transformations for the construction of imaging probes are limited, retarding the interrogation of H2S-related biological processes. Here, we present an alternative approach for engineering a new generation of efficient probes with a nonchemoselective moiety as a building block. To demonstrate our design concept, we developed a sulfoxide-functionalized BODIPY that exhibited a substantial redshift in its absorption and emission spectra upon reduction with H2S. However, such a probe also showed reactivity toward various competing biothiols under aqueous buffer conditions. To achieve high chemoselectivity, we used core-shell silica nanoparticles as an encapsulation matrix to confine the designed molecule probe within their interiors. The inherent molecular-size sieving character of the porous silica shell was capable of impeding competing biothiols from accessing the molecule probe within the core while allowing the specific reaction with the small target H2S. Thus, this strategy avoided disturbance from coexisting biothiols and achieved highly chemoselective detection in ratiometric and near-infrared (NIR) turn-on fluorescence modes. In light of these promising features, together with fast responsiveness and favorable cellular uptake, such a silica nanocomposite was successfully used to detect the endogenous production of H2S in estrogen-induced cardiomyocytes and living mouse model. To our knowledge, the approach reported here is the first to exploit the usefulness of common thiol-sensitive moieties for building chemoselective probes.
Detection of nitrite with a surface-enhanced Raman scattering sensor based on silver nanopyramid array

Abstract:Nutrient pollution is of worldwide environmental and health concerns due to extensive use of nitrogen fertilizers and release of livestock waste, which induces nitrite compounds in aquatic systems. Herein a surface-enhanced Raman scattering (SERS) sensor is developed for nitrite detection based on coupling between the plasmonic gold nanostars and the silver nanopyramid array. When nitrite is present in the assay, an azo group is formed between the 1-naphthylamine-functionalized silver nanopyramids and the 4-aminothiophenol-functionalized gold nanostars. This not only generates the SERS spectral fingerprint for selective detection, but also creates "hot spots" at the gap between the Au nanostars and the Ag nanopyramids where the azo group is located, amplifying SERS signals remarkably. Finite-difference time-domain (FDTD) simulation shows a SERS enhancement factor of 4 × 1010 at the "hot spots". As a result, the SERS sensor achieves a limit of detection of 0.6 pg/mL toward nitrite in water, and enables nitrite detection in real-world river water samples. In addition, this sensor eliminates the use of any Raman reporter and any expensive molecular recognition probe such as antibody and aptamer. This highly sensitive, selective and inexpensive SERS sensor has unique advantages over colorimetric, electrochemical and fluorescent devices for small molecule detection.
One-step method to prepare coccinellaseptempunctate-like silver nanoparticles for high sensitivity SERS detection

Abstract:The surface morphology of metal particles can be adjusted by the annealing process to enhance the activity of Raman substrate and achieve an ultra-low concentration detection limitation. In this study, a surface-enhanced Raman scattering (SERS) substrate of coccinellaseptempunctate-like silver nanospheres coated with a silver nanoisland structure (Ag NI@Ag NSs) was prepared using a simple annealing process. Numerous silver nanospheres were dispersed on the surface of the silver nanoislands, which exhibited a single crystal plane (111) orientation. The SERS substrate had an enhancement factor of 108 for Rhodamine B (Rh B) and was highly sensitive to phosphoethanolamine (PETA). The SERS response toward PETA showed excellent correlation during the range of 5 × 10−7 - 1 × 10−5 mol/L with the detection limitation as low as 5 × 10−7 mol/L, and the correlation coefficient is 0.9918. The simulated high-pressure test showed that the SERS response could be still observed at 1100 m sub ocean pressure even with a PETA concentration of 1 mM. The test results of selectivity, reproducibility, and accuracy indicated that the Ag NI@Ag NS sensor possessed potential applications in the deep-sea analytical and other related fields.

Abstract:As a highly toxic gas pollutant and also an endogenous gaseous signaling molecule existing in a variety of physiological processes, the rapid and accurate in-field detection of hydrogen sulfide is of great concern. Nevertheless, two drawbacks as for the optical probes for H 2 S detection, taking about a long time to reach the optical signal balance or the low selectivity, always exist. Herein, by using a highly photoluminescent and H 2 S-stimuli responsive silver-based metal-organic frameworks (MOFs): Ag-BDC (BDC = 1, 4-benzene dicarboxylate), we demonstrated that the luminescence intensity of Ag-BDC MOFs was inversely proportional to the concentration of H 2 S due to the Ag–S coordination and the obstruction of ligand-to-metal charge transfer (LMCT) transition process, and there was a quick response time of below 3.0 min. Combined with a simple customized device to separate H 2 S from the sample, the selectivity of the method for H 2 S detection could be greatly improved, and no interference would be caused even if the other sulfur-containing species coexisted. The luminescence probe presented a favorable sensitivity within a linear range of 0.1–1000 μM along with a detection limit of 23.7 nM. When employed to assay the endogenous sulfide level in the human serum and mouse brain tissue, the approach showed recoveries from 96.3% to 102% with relative standard derivation ( RSD ) less than 2.0%. By the integration of the responsive luminescent silver-based MOFs with a simple self-assembled headspace separation device, obviously the present strategy could be beneficial to the development and design of the in-field fast H 2 S measurement, possessing particular advantages in biological systems to eliminate the potential interferences.

Abstract:Hydrogen sulfide (H 2 S) in natural water plays an important role in carbon and sulfur cycles in biosphere. Current detection protocol is complicated, which need to "bring back water" to lab followed by gas chromatograph analysis. In situ , field detection is still challenging. Herein, a portable, sensitive surface enhanced Raman scattering (SERS) chip was proposed for in situ H 2 S sampling and SERS signal stabilizing, enabling a "bring back chip" manner for lab analysis. The SERS chip was composed of single core-shell gold nanorod-ZIF-8 framework (Au NR@ZIF-8) nanoparticle. Relying on headspace adsorption, evaporated H 2 S was enriched in the ZIF-8 shell and then reacted with Au NR, resulting in the weakening of the Au-Br bond Raman peak (175 cm −1 ) and the appearance of the Au-S bond Raman peak (273 cm −1 ). The SERS signal reached equilibrium in 10 min. The detection range of H 2 S was 0.1–2000 μg/L and limit of detection was 0.098 μg/L. SERS signal was not interfered by normal volatile gases. Moreover, SERS signal of a reacted chip was stable at an ambient condition, allowing for in situ sampling and bring-back detection. The applicability of the chip was verified by dynamic H 2 S monitoring during artificial black-odor water evolution, and in-field quantitative analysis of H 2 S content in river water and sediment. Finally, the chip was sealed in a waterproof breathable membrane device, which realized the detection of vertical profiles of H 2 S in the river. This work provided a promising tool for field analysis of H 2 S in natural environments.

Abstract:The widespread use of silver raises concerns about environmental and health risks, necessitating highly sensitive detection methods for trace silver ions (Ag+). Surface plasmon resonance (SPR) sensors offer benefits like label-free detection and rapid response, but their sensitivity for Ag+ detection is limited due to weak ion adsorption. Here, we developed an SPR sensor with MoS42−-intercalated NiAl-layered double hydroxide (LDH) as the adsorption layer of Ag+ to enhance detection sensitivity. Our sensor achieves a sensitivity of 254.75 nm/μg/L and detects Ag+ at a low concentration of 2.8 pM, outperforming various existing sensors. It also shows excellent repeatability, long-term stability, and selectivity, proving effective in real-world environmental samples. This work advances high-performance SPR sensors for heavy metal ion detection.

Abstract:Metabolic disorders involving endogenous H 2 S have been linked to a variety of serious human diseases, particularly cancer. In this study, we employed nanofibers with surface-enhanced Raman scattering (SERS) activity for the detection of H 2 S within live cells. These nanofibers were chosen for their minimal invasiveness, high spatial resolution, and enhanced SERS sensitivity. To improve the performance of SERS, highly sensitive core-shell multibranched-Au NPs (MBAuNP)@Ag NPs were decorated on the nanofibers as SERS tags for H 2 S detection. A SERS probe named MBN, embedded between the Au core and Ag shell, was utilized for quantitative detection. These nanofibers exhibited excellent reproducibility (relative standard deviation (RSD) within 5.7 %) and demonstrated a strong linear relationship with sulfide concentrations ranging from 50 nM to 1 μM, with an estimated detection limit of 0.12 nM. As a proof of concept, the aforementioned nanofibers were successfully applied to detect endogenous H 2 S in living cells, offering a potential analytical method in the related research of detection.

Abstract:Surface-enhanced Raman spectroscopy (SERS) is a promising analytical technique for fast and accurate disease detection due to its attractive features. However, realizing label-free direct detection is still challenging as most probes have extremely low Raman cross sections and little affinity to SERS substrates. Disappointingly, SERS spectra of most bacteria and other biological samples look similar as the differences in their molecular compositions are subtle and not detectable. Hence, the fabrication of highly enhancing plasmonic nanoparticles with excellent uniformity is demanded. Moreover, as SERS substrates are not reusable, cost-effective and simplistic fabrication methods are critical. Here, we report a facile approach to synthesizing Ag nanoparticle array on Cu-foil in less than 3 min, using only Cu-foil, silver nitrate, and hydroquinone. We employed the idea of galvanic replacement in combination with a seed-mediated particle-growth approach. The label-free bacterial detection has shown that our Cu/Ag nanoparticle substrate is superior to highly acclaimed Ag nanocubes. Creating strong second and third-generation SERS hot-spots through cooperative interaction of homo- (Ag–Ag) and heterogeneous (Ag–Cu) surfaces contributes mainly to the observed excellent enhancement. Interestingly, direct liquid bacteria sample analysis showed a 6-fold higher detection sensitivity than completely dried samples. We believe that our approach will offer remarkable advantages and change how SERS substrates are prepared and conducted in SERS-based bacterial detection.

Abstract:Colorimetric and chemoresistive gas sensing methods have aroused great interest in H2S monitoring due to their unique merits of naked-eye readout, and highly sensitive and rapid detection. However, combining these two methods for gas detection, especially utilizing one material as their common sensing material is a grand challenge because they are inconsistent in sensing mechanism. Taking advantage of the strong chemical affinity of Cu2O for H2S and the excellent performance of localized surface plasmon resonance (LSPR) of Au nanoparticles (NPs) in the visible regions and its ability as a noble metal to enhance gas sensing property, the Cu2O-Au nanochains (NCs) were prepared for dual-mode detection of H2S gas. The Cu2O-Au chemoresistive gas sensor shows a 5-fold higher response than Cu2O sensor at room temperature with a low detection limit of 10 ppb. Such good performance is attributed to the spillover effect and catalytic activity of Au NPs, and the enhanced H2S adsorption after Au loading as revealed by density functional theory calculation. Test strips containing Cu2O-Au produced for gaseous H2S detection show superior color gradient changes (blue, yellow, and brown). Finally, the practicability of the method was validated by real-time monitoring H2S released from cell culture.
