Abstract:In this work, we synthesized a series of boronate ester fluorescence probes (E)-4,4,5,5-tetramethyl-2-(4-styrylphenyl)-1,3,2-dioxaborolane (STBPin), (E)-N,N-dimethyl-4-(4-(4,4,5,5-tetramethyl-1,3,2-dioxaborolan-2-yl)styryl)aniline (DSTBPin), (E)-4-(4-(4,4,5,5-tetramethyl-1,3,2-dioxaborolan-2-yl)styryl)benzonitrile (CSTBPin), (E)-2-(4-(4-methoxystyryl)phenyl)-4,4,5,5-tetramethyl-1,3,2-dioxaborolane (MSTBPin), (E)-N,N-dimethyl-4-(4-(4,4,5,5-tetramethyl-1,3,2-dioxaborolan-2-yl)styryl)naphthalen-1-amine (NDSTBPin), and N,N-dimethyl-4-(2-(4-(4,4,5,5-tetramethyl-1,3,2-dioxaborolan-2-yl)phenyl)oxazol-5-yl)aniline (DAPOX-BPin) for the detection of hydrogen peroxide (H2O2). DSTBPin and MSTBPin displayed an "Off-On" fluorescence response towards H2O2, owing to the loss of the intramolecular charge transfer (ICT) excited state. Whereas, CSTBPin displayed a decrease in fluorescence intensity in the presence of H2O2 owing to the introduction of an ICT excited state. STBPin, on the other hand, produced a small fluorescence decrease, indicating the importance of an electron-withdrawing or electron-donating group in these systems. Unfortunately, the longer wavelength probe, NDSTBPin, displayed a decrease in fluorescence intensity. Oxazole-based probe DAPOX-BPin produced a "turn-on" response. Regrettably, DAPOX-BPin required large concentrations of H2O2 (>3mm) to produce noticeable changes in fluorescence intensity and, therefore, no change in fluorescence was observed in the cell imaging experiments.

Abstract:In current work, one novel benzothiazole-based fluorescent chemosensor (TZ-BO) with ESIPT luminescence mechanism was designed for the detection of hydrogen peroxide (H2O2) as typical reactive oxygen species (ROS). The boronate catenary in our designed probe molecule TZ-BO is both the blocking group of ESIPT and the reactive group of H2O2. The synthesized probe TZ-BO has the advantages of large Stokes shift, high sensitivity, good selectivity, and fast reaction speed to H2O2. The ability of the probe to detect H2O2 in aqueous media and exogenous H2O2 in cells by fluorescence signal was investigated. The experimental results manifested that probe TZ-BO is able to sensitively detect H2O2 using a fluorescence method in an excellent linear range of H2O2 concentration from 0 to 60 μM (R2 = 0.994). The detection limit was determined as 1.0 × 10−6 M. Most importantly, the probe TZ-BO with highly low cytotoxicity can effectively detect exogenous H2O2 in HeLa cells. This probe provides a promising tool for monitoring the real concentration level of H2O2 in related physiological and pathological researches.

Abstract:A water-soluble boronate-based fluorescent probe was evaluated for the detection of peroxynitrite (ONOO-) in the presence of a monosaccharide. The enhanced fluorescence of the probe when bound with o-fructose was switched off in the presence of peroxynitrite. In contrast, other reactive oxygen/nitrogen species (ROS/RNS) led to only slight fluorescence decreases due to protection by an internal N-B interaction. The interaction of the probe with o-fructose not only strengthens the fluorescence signal, but also protects the boronic acid from oxidation by other ROS/RNS. Therefore, under conditions generating various ROS/RNS, the boronate-based saccharide complex preferentially reacts with peroxynitrite. The probe was used in cell imaging experiments for the detection of endogenous and exogenous peroxynitrite. The sensor displays good "on-off" responses towards peroxynitrite, both in RAW 264.7 cells and HeLa cells.

Abstract:The synthesis, properties and applications of a water-soluble boronate-functioned hemicyanine-naphthol hybrid as a novel ratiometric fluorescent sensor for hydrogen peroxide are presented. The dye displayed remarkable a colour change from pale orange ((em)=590nm) to pink ((em)=690nm) in the presence of H2O2, which could be rationalized by the chemoselective H2O2-mediated transformation of arylboronate to phenolate with high selectivity and a fast response (within 2min). A good linear relationship (R-2=0.9951) was obtained with the H2O2 concentration ranging from 0 to 25M, with a limit of detection of 0.09M according to the signal-to-noise ratio (S/N=3). The advantages of this fluorophore include easy modification, excellent aqueous solubility and superior photostability, and it has been applied to the detection of trace amounts of hydrogen peroxide in water samples. Alizarin-based Molecular Probes for the Detection of Hydrogen Peroxide and Peroxynitrite.

Abstract:Phenol fluorophores are a large family of fluorophores, which have attracted more and more attention in the design of probes. Using the self-assembly of aromatic boronic acid with Alizarin Red S (ARS) and Gallein (GAL), the novel chemosensors ARS-CBA and GAL-CBA were provided for hydrogen peroxide (H2O2), which demonstrated their ability to detect H2O2 with indicator displacement assay (IDA) by colorimetric and electrochemical measurements. After ARS-CBA and GAL-CBA reacted with H2O2, the systems displayed a red-shifted visible color change in aqueous media and off-on electrochemical signals showing generation of phenol. The chemosensor ARS-CBA also had good performance in fluorometric measurements and turn-off fluorescent response indicated removal of aromatic boronic acid. In addition, a designed near-infrared (NIR) dual-modal fluorescent probe alizarin blue S (ABS) was used for peroxynitrite (ONOO-) with a visible colorimetric change in dimethyl sulfoxide (DMSO) and "on-off" fluorescent response indicating the oxidation of hydroxyl. The flexible Phenol fluorophores are allowed to prepare multiple fluorescent probes towards H2O2 or ONOO- for environmental and physiological applications.
Monitoring The Fluctuation Of Hydrogen Peroxide In Diabetes And Its Complications With A Novel Near-Infrared Fluorescent Probe

Abstract:Diabetes is one of the metabolic diseases marked by hyperglycemia and is often accompanied by the occurrence of some complications. As a biomarker of oxidative stress, hydrogen peroxide (H2O2) has close association with the occurrence and development of diabetes and its complications. Unfortunately, there is no fluorescent probe reported for imaging H2O2 in diabetic mice. Here, a novel near-infrared (NIR) fluorescent probe named QX-B was designed and synthesized to detect H2O2. For the probe, the quinolinium-xanthene dye is used as the fluorophore and borate ester is chosen as the response group. After the addition of H2O2, a strong MR fluorescence signal at 772 nm is observed. The probe not only shows high sensitivity with 10-fold enhancement but also displays excellent selectivity to H2O2 over other possible interfering species. In the meantime, the possible response mechanism of QX-B toward H2O2 was proposed and verified by the high-performance liquid chromatography (HPLC) experiment, mass spectra (MS) experiment, and density functional theory (DFT) calculation. Furthermore, based on the low cell cytotoxicity of QX-B, it has been applied in imaging exogenous and endogenous H2O2 in HeLa cells, HCT116 cells, 4T1 cells, and zebrafish successfully. More importantly, inspired by the performance of NIR fluorescence, QX-B has been used in monitoring H2O2 in diabetic mice for the first time. This provides very important information for the diagnosis and treatment of diabetes and its complications.

Abstract:A complicated relationship exists between viscosity and hydrogen peroxide (H2O2) in biological systems, whose changes are important indication of many diseases. To provide efficient tools for exploring this, we devise and synthesize three boronate-based fluorescence probes (L1–L3) by incorporating different N-hydroxyethyl substituted aminobenzaldehydes with boronated pyridinium group respectively. L3 with good properties and water solubility has been chosen for further study. L3 exhibits good response to viscosity and H2O2 with excellent sensitivity and selectivity. The reaction mechanism has been proposed as the restriction of twisted intramolecular charge transfer (TICT) to high viscosity resulting in strong red fluorescence, while boronic acid moiety is cleaved to release by H2O2 causing green fluorescence due to intramolecular charge transfer (ICT). Moreover, L3 has been successfully applied to monitor viscosity and H2O2 in living cells. This work demonstrates that N-hydroxyethyl substitution does not alter the electron donating property of the aminobenzene group, which provides an ideal example to develop promising tools for monitoring bioactive processes/species in living systems.
Dual Mechanism of an Intramolecular Charge Transfer (Ict)-Fret-based Fluorescent Probe for the Selective Detection of Hydrogen Peroxide

Abstract:A dual-mechanism intramolecular charge transfer (ICT)-FRET fluorescent probe for the selective detection of H2O2 in living cells has been designed and synthesized. This probe used a coumarin-naphthalimide hybrid as the FRET platform and a boronate moiety as the recognition group. Upon the addition of H2O2, the probe exhibited a redshifted (73nm) fluorescence emission, and the ratio of fluorescence intensities at =558 and 485nm (F-558/F-485) shifted notably (up to 100-fold). Moreover, there was a good linearity (R-2=0.9911) between the ratio and concentration of H2O2 in the range of 0 to 60m, with a limit of detection of 0.28m (signal to noise ratio (S/N)=3). This probe could also detect enzymatically generated H2O2. Importantly, it could be used to visualize endogenous H2O2 produced by stimulation from epidermal growth factor.

Abstract:Intracellular H2O2 plays an important role in regulating a variety of cellular functions. Fluorescent probes that can make response to intracellular levels of H2O2 would provide valuable tools for revealing the functions of H2O2 in living organisms. However, traditional pH-insensitive probes and lysosome-targetable probes can only provide spatially nonspecific visualization of intracellular H2O2 and specific sensing of lysosomal H2O2, respectively. In this work, we developed a H2O2-responsive and pH-switchable fluorescent probe (HP-L1) which can make response sequentially to intracellular H2O2 and lysosomal pH. The fluorescent probe is comprised of a H2O2-responsive boronate moiety and a pH-switchable spirobenzopyran fluorophore. When the probe was applied for intracellular H2O2 sensing, only fluorescent emission from lysosomes is visible, and the fluorescence from other regions is not able to be obviously detected, which is due to the pH-switchable property of the spirobenzopyran fluorophore. Thus, the developed fluorescence probe enables the spatially confined (i.e., lysosome-specific) visualization of the intracellular H2O2. We envisioned that this kind of fluorescent probe (or the proposed sensing strategy) would allow the visualization of the overall levels of intracellular H2O2 without interferences of possible fluorescent signals from other sources (e.g., dyes for cellular staining and multiplex analysis).

Abstract:As one of the important components of active oxygen species, H2O2 (Hydrogen peroxide) plays an important role in life activities. Many studies have shown that high concentration of H2O2 can cause cell necrosis, resulting in oxidative stress and oxidative damage in the body, and then induce various diseases. The application of organic small molecule fluorescent probes in life science has attracted the attention of many researchers because of its advantages of easy to obtain, high spatial and temporal resolution, little damage to biological samples and simple operation. In this study, a new hydrogen peroxide near infrared fluorescence probe JH was constructed using oxanthracene analogues as molecular framework. The fluorescence emission wavelength of the probe is located in the near infrared region and has a large Stokes shift, which makes the probe less damaging to biological samples and lower background interference, which is conducive to improving the detection resolution. The probe can complete the response to H2O2 within 95 min with a low detection limit (4.72 μM), and the color of the solution changes from dark blue to light blue after the response, and H2O2 can be recognized by the naked eye without any tools. In addition, probe JH has been successfully used for the detection of H2O2 in cells, which is expected to become a tool for early diagnosis of diseases.

Abstract:Hydrogen peroxide (H2O2) is one of the important reactive oxygen species (ROS), which is closely related to many pathological and physiological processes in living organisms. Excessive H2O2 can lead to cancer, diabetes, cardiovascular diseases, and other diseases, so it is necessary to detect H2O2 in living cells. Since this work designed a novel fluorescent probe to detect the concentration of H2O2, the H2O2 reaction group arylboric acid was attached to the fluorescein 3-Acetyl-7-hydroxycoumarin as a specific recognition group for the selective detection of hydrogen peroxide. The experimental results show that the probe can effectively detect H2O2 with high selectivity and measure cellular ROS levels. Therefore, this novel fluorescent probe provides a potential monitoring tool for a variety of diseases caused by H2O2 excess.

Abstract:Hydrogen peroxide(H2O2), as a reliable signaling biomolecule for oxidative stress, its accurate detection during agent-stimulated oxidative stress plays a vital role in pathological and physiological mechanism exploration for disease theranostics. It's necessary to develop an efficient method for their detection. In view of the advantages of fluorescent probes, we rationally constructed a novel fluorescent probe Compound 2 based on 4-(Bromomethyl)benzeneboronic acid pinacol ester_Herein, a small molecule fluorescent probe was fabricated using isoflore nitrile as fluorescent group, phenylboronic acid pinacol ester as the response group, to detect H2O2. The probe Compound 2 has a strong fluorescence intensity at 575 nm, indicating that the structure of the probe molecule is reasonably designed, and the Stokes shift is up to 172 nm. While the detection time is as low as 30 s and the LOD of the probe for H2O2 is as low as 3.7 μmol/L,the quantum yield is Φ = 40.31 %. It has been successfully used for imaging detection of H2O2 in HepG2 cells and zebrafish for its low toxicity. It can be found that this small molecule fluorescent probe can identify H2O2 in tumor cells significantly and efficiently, which would realize the early diagnosis of tumor.
Near-infrared fluorescence probe for hydrogen peroxide detection: design, synthesis, and application in living systems

Abstract:Using fluorescent probes to detect endogenous hydrogen peroxide, which is associated with many diseases in the human body, remains an essential technique. Cyanine fluorochromes are a class of dyes that have attracted much attention and are widely used in the synthesis of fluorescent probes. In this article, a novel near-infrared (NIR) fluorescence probe for the detection of hydrogen peroxide was constructed and successfully applied to imaging endogenous hydrogen peroxide in vivo. Notably, probe 1 was designed by connecting 4-(bromomethyl)benzeneboronic acid pinacol ester as the sensing unit to the IR-780 hemicyanine skeleton, which exhibits excellent properties like NIR fluorescence emission over 700 nm. Probe 1 has satisfactory sensitivity to hydrogen peroxide with a low detection limit of 0.14 μM (S/N = 3), attributed to a responding mechanism that leads to the oxidation of phenylboronic acid pinacol ester and thereby releases fluorophore 2. Moreover, probe 1 displays excellent selectivity towards hydrogen peroxide over other substances. Taking advantage of these properties, the probe proved to be cell-permeable. Based on the results of N-acetylcysteine and rotenone together, probe 1 is capable of clearly visualizing endogenously produced hydrogen peroxide in living HepG2 cells and mice. The superior performance of the probe, as a reliable chemical tool, makes it of great potential application for exploring the role played by hydrogen peroxide in biological systems.

Abstract:Abnormal intracellular peroxynitrite (ONOO-) concentration is related to oxidative damage, which is correlated with many pathological consequences, such as local inflammation and other diseases. In this work, a series of resorufin benzyl ether-based fluorescent probes were designed using boronate as a recognizing moiety installed on a phenyl moiety for ONOO- detection via a self-immolation mechanism. The location of the boronate as well as the substitution patterns on the phenyl moiety were investigated and the responding behaviors of the designed probes to ONOO-, other reactive oxygen species, and biothiols were examined. It was found that all the immolative probes were inevitably dominated by ONOO-. Compared with other probes, p-Borate possessed favorable selectivity and high sensitivity to ONOO-. Moreover, p-Borate was successfully used to detect ONOO- in cells and inflamed mice.

Abstract:Hydrogen peroxide (H2O2) plays an important role in the initiation and progression of many diseases. Therefore, the detection of H2O2 in vivo is very important to elucidate its biological functions in various diseases. At present, the research of fluorescent probes has attracted wide attention. Fluorescent probes are becoming more and more useful as tools for detecting various substances. In this paper, we present a novel fluorescent probe capable of selectively detecting H2O2 in organisms. This probe is based on the design strategy of the recognition of H2O2 enhancing the water-solubility, as a result, producing the turn-on fluorescence intensity. With the increase of hydrogen peroxide concentration, the fluorescence intensity of the newly as-prepared probe is enhanced. There was a good linear relationship when the concentration range of hydrogen peroxide was from 1 μM to 100 μM. In terms of cell imaging work, the detection of endogenous and exogenous H2O2 in RAW 264.7 macrophage cells showed obvious fluorescence enhancement under confocal microscopy. So this new probe has been shown to be a reliable chemical tool for detecting the generation and development of H2O2-related diseases in living systems.

Abstract:The multifunctional hemin@carbon dot hybrid nanozymes (hemin@CD) with simultaneous peroxidase-like activity and fluorescence signalling property was prepared for the first time. Based on these properties, hemin@CD was applied to develop a dual-channel fluorescent probe for H2O2 and H2O2-based biocatalytic systems. By virtue of the peroxidase-like activity, hemin@CD can catalyze the oxidative coupling of 4-aminoantipyrine with phenol in the presence of H2O2 to form a pink-red quinoneimine dye with a maximum absorbance at 505 nm. Under the excitation wavelength of 480 nm, the green fluorescence of hemin@CD peaks at 540 nm and is quenched by the generated quinoneimine dye due to an inner filter effect, and also by H2O2 because of dynamic quenching. Thus, a colorimetric and fluorimetric dual-channel optical probe for H2O2 is obtained. Due to the glucose/xanthine transformations under formation of H2O2 by the relevant oxidase catalysis, the probe can be applied for detection of glucose and xanthine. The colorimetric detection limits for H2O2, glucose and xanthine are 0.11, 0.15, 0.11 μM, and the and fluorimetric detection limits are 0.15, 0.15, 0.12 μM, respectively.
A Fast-Response, Highly Sensitive and Selective Fluorescent Probe for the Ratiometric Imaging of Hydrogen Peroxide with a 100 Nm Red-Shifted Emission

Abstract:Recently, growing attention has been paid to the accurate determination of hydrogen peroxide (H2O2) for elucidating its detailed biological function in physiology and pathology. A fluorescence method with the help of a fluorescent probe is the preferred technique for in situ visualization of biologically important species in vivo, even in single living cells. In the present manuscript, we developed a simple, fast response and highly selective fluorescent probe (1) with a receptor of the boronate moiety for the ratiometric imaging of H2O2 in living cells. Probe 1 could quantifiably detect H2O2 in the range of 18540 mM by a ratiometric fluorescence spectroscopy method with a detection limit of 4 mM. Importantly, probe 1 exhibited 81 nm red-shifted absorption spectra accompanied by the color changes from colorless to yellow, and 100 nm red-shifted emission spectra upon addition of H2O2. Thus, 1 can serve as a ''naked-eye'' probe for H2O2. Preliminary bioimaging application and low cytotoxicity investigations further demonstrated that the proposed probe would be of great benefit to biomedical researchers for investigating the detailed biological function of H2O2 in biological systems.

Abstract:A benzothiazole-based derivative aggregation-induced emission (AIE) fluorescent 'turn-on' probe named 2-(2-((4-(4,4,5,5-tetramethyl-1,3,2-dioxaborolan-2-yl)benzyl)oxy)phenyl)benzo[d]thiazole (probe BT-BO) was developed and synthesized successfully for detecting hydrogen peroxide (H2O2) in living cells. The synthesis method of probe BT-BO is facile. Probe BT-BO demonstrates a well-resolved emission peak at 604 nm and the ability to prevent the interference of reactive oxygen species (ROS), various metal ions and anion ions, and good sensitivity. Additionally, the probe boasts impressive pH range versatility, a fast response time to H2O2 and low cytotoxicity. Finally, probe BT-BO was applied successfully to image A549 and Hep G2 cells to monitor both exogenous and endogenous H2O2.

Abstract:The authors describe a fluorometric and colorimetric nanoprobe for H2O2. The detection scheme is based on the in-situ formation of silver(I) ions from a composite consisting of nitrogen-doped carbon quantum dots (N-CQDs) and silver nanoparticles (AgNPs). A drastic change occurs both in fluorescence and color of the solution of the N-CQD/AgNPs composite. The fluorescence of composite (with excitation/emission peaking at 320/384 nm) is enhanced on increasing the concentration of H2O2 due to the oxidation of silver metal in the N-CQD/AgNPs to form Ag(I) ions. The latter undergo strong coordination with the nitrogen atoms of the N-CQDs. In-situ formation of Ag(I) ions further results in a change in color of the solution from pale yellow (with a peak at 408 nm) to colorless. Under optimized conditions, the probe gives a fluorometric and colorimetric response in the 10 to 50 μM H2O2 concentration range with a 4.7 μM limit of detection. The probe is highly selective over several potentially interfering ions and agents. It was successfully applied to the determination of H2O2 in spiked samples without prior treatment.
