Abstract:The application of small molecules as synthetic blocks is a field of great meaning and full of challenges, especially the use of inert ether molecules, such as THF as starting materials. A versatile and easily handled cross-dehydrogenative coupling between ethers and quinaldine is reported here. Compared to the developed reaction, Selectfluor is used as a mild oxidant, and a variety of functional groups are tolerated. Nitrogen protection, anhydrous systems, and external base are not needed, making this method a convenient route to functional heterocycles.

Abstract:2-Perfluoroalkylated quinolines were efficiently synthesized via the Michael addition of perfluoroalk-2-ynoates with isatins, followed by the reaction with alcohols through an intramolecular rearrangement cyclization in the presence of Na2CO3. Under the mild and metal-free reaction conditions, a broad scope of quinolines was synthesized and the mechanism was proposed.

Abstract:An efficient method for the synthesis of substituted quinolines is described. By FeCl3-promoted C–C bond formation, 2,3-disubstituted and 2,3,4-trisubstituted quinolines were successfully synthesized from the reaction between imines and electron-deficient alkynes in good yields. The reaction can also be performed in a sequential three-component manner by using aromatic amines and aromatic aldehydes instead of the corresponding imines.

Abstract:We here developed an effective approach for construction of 2- or 4-(1,4-dioxan-2yl) substituted quinazolines under mild conditions. The silver-K2S2O8 catalyzed direct CDC reaction between quinazolines and 1,4-dioxane for the synthesis of a series of 2- or 4-(1,4-dioxan-2yl) substituted quinazolines hybrids, which conglomeration of quinazoline and cyclic ether moieties in one molecular, will have the potential applications for the development of new drugs or drug candidates. The reaction proceeded well in water under mild conditions and showed broad substrate scope and good functional group compatibility. The catalytic reaction mechanism had been studied through EPR strategy and simultaneously HRMS analysis.

Abstract:An efficient and rapid synthesis of multiply substituted quinolines is described. This method is enabled by a three-component cascade annulation of readily available aryl diazonium salts, nitriles, and alkynes. This reaction is catalyst- and additive-free. Various aryl diazonium salts, nitriles, and alkynes can participate in this transformation, and the yields are up to 83%.

Abstract:An efficient silver‐catalyzed δ‐regioselective unactivated C(sp3 )–H of free alcohols dehydrogen‐coupling with quinazolines for the synthesis of δ‐hydroxylalkyl substituted quinazolines in aqueous media has been developed. Various alcohols including 1o and 2o alcohols, reacted with quinazolines through 1,5‐hydrogen‐atom transfer strategies to give the corresponding 2‐ or 4‐ δ‐hydroxylalkyl substituted quinazolines with good to excellent yields under mild reaction conditions. Notably, several substituted quinazolines and quinazolinones could be selectively dehydrogenative‐coupled with divergent free alcohols. This protocol provides a platform to access divergent functionalizations of quinazolines and quinazolinones to meet the growing needs for screening.

Abstract:A metal-free (NH4)2S2O8-mediated decarboxylative trifluoromethylation reaction of alkenes with 3,3,3-trifluoro-2,2-dimethylpropionic acid has been proposed. This method offers a novel route for the direct synthesis of a series of CMe2CF3-containing quinazolinones from basic chemical raw materials. The reaction mechanism was studied by radical trapping test and DFT methods, verifying an oxidative-triggered cascade process promoted by the CMe2CF3 radicals. This strategy provides advantages such as high yield, wide substrate compatibility, and high atom economy.

Abstract:An atom and step economy cascade trifluoromethylation/cyclization of unactivated alkene with Langlois reagent as a CF3 source is described. A variety of polycyclic quinazolinones were successfully synthesized in 52–81% yields under transition metal‐ and oxidant‐free conditions. The Langlois reagent used in this strategy as a CF3 reagent possesses the advantages of bench‐stablity, cost‐effectivity and high‐efficiency. Additionally, gram‐scale reaction, broad substrate scope and good functional group tolerance demonstrated the synthetic usefulness of this protocol.

Abstract:A diastereoselective synthesis of 2-methyl-4-amino-1,2,3,4-tetrahydro-quinoline derivatives was achieved through the reaction of aromatic amines and tert-enamides in water under reflux conditions. Abstract:Quinolines and their derivatives bearing fluorine-containing substituents are essential structural motifs found in numerous bioactive compounds and advanced functional materials. Therefore, there is an urgent need to develop new synthetic methods for fluoroalkylated quinoline derivatives. In this study, we present a facile and efficient approach to access fluoroalkylated quinoline derivatives by using Eaton's reagent as a cost-effective, and commercially available reagent. This Combes cyclization reaction demonstrated compatibility with various arylamines bearing diverse functional groups, resulting in the synthesis of 2,4-bis(trifluoromethyl)quinolines and 2,4-bis(difluoromethyl)quinolines in moderate to high yields. Furthermore, this reaction can be conveniently carried out in a solvent-free one-pot protocol.

Abstract:A silver-catalyzed cascade cyclization strategy has been developed for the synthesis of 4-aminotetrahydrocarbazole, a common core structure found in various alkaloids. This target molecule can be synthesized through a one-step tandem cyclization reaction, thereby eliminating the need for a prior synthesis of tetrahydrocarbazole. Furthermore, the use of chiral tert-butylsulfinamide facilitates in situ chiral resolution of the resulting product.

Abstract:An effective and straightforward Ag(I)-mediated annulation of 2-(2-enynyl)quinolines and N'-(2-alkynylbenzylidene)hydrazides was developed, forging various synthetically challenging 17bH-isoquinolino[2'',1'':1',6']pyridazino[4',5':3,4]pyrrolo[1,2-a]quinolines, including different nitrogen-containing fused rings, in moderate to excellent yields. This one-pot cycloaddition strategy features exclusive regioselectivity, high atom economy, and broad substrate scope under mild conditions. The practicality and reliability of this cycloaddition reaction was demonstrated by a successful scale-up synthesis.
