Abstract:Quantitative analysis of up-regulated biomarkers in pathological tissues is helpful to tumor surgery yet the loss of biomarker extraction and time-consuming operation limited the accurate and quick judgement in preoperative or intraoperative diagnosis. Herein, an immobilization-free electrochemical sensing platform is developed by constraint coupling of electron transfer cascade on electrode-nanosensor interface. Specifically, electrochemical indicator (Ri)-labeled single-stranded DNA on electroactive nanodonor (polydopamine, PDA) can be responsively detached by formation of DNA complex through the recognition and binding with targets. By applying the oxidation potential of Ri, nanosensor collisions on electrode surface trigger a cascade redox cycling of PDA and Ri through synchronous electron transfer, which boost the amplification of current signal output. The developed nanosensor exhibit excellent linear response toward up-regulated biomarkers (miRNA-21, ATP, and VEGF) with low detection limits (32 fM, 386 pM, and 2.8 pM). Moreover, background influence from physiological interferent is greatly reduced by restricted electron transfer coupling on electrode. The practical applicability is illustrated in sensitive and highly repeatable profiling of miRNA-21 in lysate of tumor cells and tumor tissue, beneficial for more reliable diagnosis. This electrochemical platform by employing electron transfer cascades at heterogeneous interfaces offers a route to anti-interference detection of biomarkers in tumor tissues. An immobilization-free nanosensor is fabricated through the constraint of cascade redox and electron transfer synchronous on electrode-nanosensor interface for repeatable sensing of biomarkers in tumor tissues. Abstract:Herein, we develop a novel method for designing electrochemical biosensors with both current and potential signal outputs for the simultaneous determination of two species in a living system. Oxygen (O 2 ) and pH, simple and very important species, are employed as model molecules. By designing and synthesizing a new molecule, Hemin‐aminoferrocene (Hemin‐Fc), we create a single electrochemical biosensor for simultaneous detection and ratiometric quantification of O 2 and pH in the brain. The reduction peak current of the hemin group increases with the concentration of O 2 from 1.3 to 200.6 μ m . Meanwhile, the peak potential positively shifts with decreasing pH from 8.0 to 5.5, resulting in the simultaneous determination of O 2 and pH. The Fc group can serve as an internal reference for ratiometric biosensing because its current and potential signals remain almost constant with variations of O 2 and pH. The developed biosensor has high temporal and spatial resolutions, as well as remarkable selectivity and accuracy, and is successfully applied in the real‐time quantification of O 2 and pH in the brain upon ischemia, as well as in tumor during cancer therapy.

Abstract:CEA and AFP standard grade antigens, anti-CEA and anti-AFP antibodies were purchased from Linc-Bio Science Co. (Shanghai, China). Carboxyl graphene sheet (CGS) dispersion (the content of carboxyl >5 wt%) was obtained from Nanjing XFNANO Materials Tech Co., Ltd., (Nanjing, China). N-(3-dimethylaminopropyl)-N’-ethyl-carbodiimide hydrochloride (EDC), N-hydroxysuccinimide (NHS), bovine serum albumin (BSA) were purchased from Sigma (St. Louis, USA). MB, potassium ferricyanide (K3Fe(CN)6), ferric chloride (FeCl3·6H2O) came from Shanghai Chemical Reagent Company (Shanghai, China). ITO-coated (thickness: 100 nm; resistance: 10 Ω sqr) aluminosilicate glass slides were purchased from CSG (Shenzhen, China). Serum specimens provided by Gulou Hospital (Nanjing, China) were stored at 4 °C. All other chemicals were of analytical grade. Ultrapure water (18.2 MΩcm) was obtained from a Milli-Q water purification system and used in all experiments. Electrochemical Microfluidic Paper-Based Aptasensor Platform Based on a Biotin-Streptavidin System for Label-Free Detection of Biomarkers

Abstract:Timely and rapid detection of biomarkers is extremely important for the diagnosis and treatment of diseases. However, going to the hospital to test biomarkers is the most common way. People need to spend a lot of money and time on various tests for potential disease detection. To make the detection more convenient and affordable, we propose a paper-based aptasensor platform in this work. This device is based on a cellulose paper, on which a three-electrode system and microfluidic channels are fabricated. Meanwhile, novel nanomaterials consisting of amino redox graphene/thionine/streptavidin-modified gold nanoparticles/chitosan are synthesized and modified on the working electrode of the device. Through the biotin-streptavidin system, the aptamer whose 5'end is modified with biotin can be firmly immobilized on the electrode. The detection principle is that the current generated by the nanomaterials decreases proportionally to the concentration of targets owing to the combination of the biomarker and its aptamer. 17β-Estradiol (17β-E2), as one of the widely used diagnostic biomarkers of various clinical conditions, is adopted for verifying the performance of the platform. The experimental results demonstrated that this device enables the determination of 17β-E2 in a wide linear range of concentrations of 10 pg mL-1 to 100 ng mL-1 and the limit of detection is 10 pg mL-1 (S/N = 3). Moreover, it enables the detection of targets in clinical serum samples, demonstrating its potential to be a disposable and convenient integrated platform for detecting various biomarkers.

Abstract:We present a highly efficient multichannel microfluidic electrochemical sensor integrated with an electroactive nanocarbon microelectrode for sensitive and selective detection of multiple biomarkers in different biological samples. Our results have shown that ionic liquid-assisted wet spinning followed by tailored growth of metal-organic frameworks and pyrolysis treatment led to structural and molecular engineering of mechanically robust all-carbon microfibers for excellent electrochemical activities. The flexible bottlebrush-like nanocarbon microelectrode features a "stem" of freestanding N, B-codoped graphene fiber and high-density "bristles" of Co, N-codoped carbon nanotube arrays, leading to promoted electrocatalytic mechanism that has been substantiated by density functional theory calculations. The structural characteristics, high catalytic activities, and favorable biocompatibility of the bottlebrush nanocarbon electrodes provide opportunities for multichannel, microfluidic detection of redox-active biomolecules, including hydrogen sulfide (H<sub>2</sub>S), dopamine (DA), uric acid (UA), and ascorbic acid (AA), and have been applied to on-chip monitoring of H<sub>2</sub>S and DA released from live cancer cells or neuroblastoma cells and DA, UA, and AA in trace amounts of body fluids such as sweat, finger blood, tears, saliva, and urine, which is of great significance for clinical diagnosis and prognosis in point-of-care testing.

Abstract:An electrochemical method for the simultaneous detection of two different tumor markers, carcinoembryonic antigen (CEA) and α-fetoprotein (AFP), in one-pot, using CdS/DNA and PbS/DNA nanochains as labels was developed. Herein, magnetic beads (MBs) as bimolecule immobilizing carriers, were used for co-immobilization of primary anti-CEA and anti-AFP antibodies. The distinguishable signal labels were synthesized by in situ growth of CdS and PbS nanoparticles on DNA chains, respectively, which were further employed to label the corresponding secondary antibodies. A sandwich-type immunoassay format was formed by the biorecognition of the antigens and corresponding antibodies. The assay was based on the peak currents of Cd2+ and Pb2+ dissolved from CdS and PbS nanoparticles by HNO3 using square wave stripping voltammetry. Experimental results show that the multiplexed electrochemical immunoassay has enabled the simultaneous monitoring of CEA and AFP in a single run with wide working ranges of 0.1–100ngmL−1 for CEA and 0.5–200ngmL−1 for AFP. The detection limits reach to 3.3pgmL−1 for CEA and 7.8pgmL−1 for AFP.

Abstract:Here a novel electrochemical microarray platform towards high-throughput detection is presented. This microarray owns 8×5 well-organized oval cells and 8×5 pairs of ITO discs (two ITO discs for each pair in one cell). 8 mobile and aligned Pt–Ag/AgCl coaxial electrodes with compact configuration, where Pt column is used for counter electrode and Ag/AgCl for reference electrode, are inserted in 8 cells separately and made up 8 groups of three-electrode detection system together with 8 pairs of ITO discs. To demonstrate the applicability of such design, two kinds of bioconjugates, carboxyl graphene nanosheets–methylene blue (CGS–MB) and carboxyl graphene nanosheets–Prussian blue (CGS–PB) nanocomposites selected as model redox indicators and crosslinked with two kinds of antibodies were coated on the pair of ITO discs to realize a high-throughput detection of two tumor markers based on the increasing spatial blocking and impedance from the formed immunocomplex.

Abstract:This paper describes a novel electrochemiluminescence (ECL) imaging platform for simultaneous detection of cancer biomarkers based on a closed bipolar electrode (BPE) array. It consists of two separated channel arrays: detection channel array and sensing channel array, which are connected by a group of parallel ITO BPEs on a glass substrate. Besides, two parallel ITO strips are fabricated at the two sides of BPE array and employed as driving electrodes. After Au films are electrochemically deposited on the cathodes of the BPE array, nanobioprobes including biorecognition elements (aptamer or antibody) and a novel electrochemical tag, which is synthesized by doping thionine in silica nanoparticles (Th@SiO2 NPs), are introduced into the cathodes by immunoreaction or DNA hybridization. The Th@SiO2 coupled nanobioprobes as both recognition probes and signal amplification indicators could mediate the ECL signals of Ru(bpy)3(2+)/tripropylamine (TPA) on the anodes of BPE array through faradaic reaction due to the charge neutrality of BPE. Thus, multiplex detection of cancer biomarkers (adenosine triphosphate (ATP), prostate-specific antigen (PSA), α-fetoprotein (AFP) and thrombin) is realized by forming specific sensing interfaces onto the cathodic poles of BPEs in different sensing channels and reported by the ECL images of the Ru(bpy)3(2+)/TPA system on the anodic poles of BPEs in detection channels. The results demonstrate that this visual ECL platform enables sensitive detection with excellent reproducibility, which may open a new door toward the development of simple, sensitive, cost-effective, and high throughput detection methods on biochips.

Abstract:Recently, we have developed a series of robust, nanobiotechnology-based electrochemical biosensing approaches with high sensitivity, and reproducibility toward the simultaneous multiplex detection and classification of both acute myeloid leukemia and acute lymphocytic leukemia cells, quantitative evaluation of DR4/DR5 expression on leukemia cell surface, dual-channel detection of matrixmetalloproteinase-2 (MMP-2) in human serum as well as ultrasensitive circulating tumor cells (CTCs) electrochemical cytosensing using Fe3O4@AuPd nanocages as the nonenzymatic nanoelectrocatalysts . Self-powered biosensor based on enzyme biofuel cell (EBFC) was another kind of super-sensitive self-contained sensing system which was powered by EBFC, meanwhile, the output of EBFC was directly related to the analyte concentration. The system was capable of detecting cancer cells, quantifying two types of cancer-related biomarkers (miR-21 and miR-141). Furthermore, we have developed a self-powered cascading integrated system for tumor diagnosis, therapy, and prognosis supervision. These versatile electrochemical biosensing platforms hold great promise as new point-of-care diagnostic tools for early detections and are believed to be of great clinical value for the evaluation of therapeutic effects on patients after radiation therapy or drug treatment, moreover, provide potential opportunity for developing economical preclinical theranostic platform.

Abstract:Electrically addressable covalent immobilization of probes on a multiplexed electrode for the simultaneous detection of multiple targets within the same sample is often regarded as a difficult milestone to be achieved. Herein, we demonstrated a reagentless disposable multiplexed electrochemical DNA and aptamer-based sensing platform for the simultaneous determination of various targets. The electrochemically triggered "click" chemistry was developed, and three biomarkers, including p53, thrombin, and VEGF165 were used as model analytes. The proposed sensor consisted of three independent screen-printed carbon electrodes (SPCE), with an alkyne-azide cycloaddition reaction that was activated selectively by means of electrical triggering, so that different DNA probes can be modified on the desired electrode units in sequence. In terms of simultaneous detection, the sensor was able to quantify the DNA target of p53 with a detection limit of 0.35 nM, whereas the limits of detection for protein quantification of thrombin and VEGF165 were 0.22 nM and 0.014 nM, respectively. The proposed sensor not only showed encouraging reproducibility and stability, but also performed well even in 50% serum samples. Therefore, the work described here offers a general strategy for developing a multiplexed sensor with promising potential to achieve rapid, simple and cost-effective analysis of biological samples.

Abstract:Herein we reported the construction of dual-mode electrochemiluminescence (ECL) and electrochemical (EC) sensing platform using the bipolar silica nanochannel array (bp-SNA) confined with the bifunctional probe (tris(2,2′-bipyridyl) ruthenium, Ru(bpy)32+) inside the nanochannels (designated as Ru@bp-SNA). With the asymmetric surface charges, bp-SNA on the indium tin oxide electrode can effectively function as electrostatic nanocage for the stable confinement of Ru(bpy)32+ via dual electrostatic force, namely electrostatic attraction of inner negative surface and electrostatic repulsion of outer positive surface. Ru(bpy)32+ immobilized into the nanospace acts as both highly efficient ECL luminophore and well-defined EC redox molecule to generate strong signal. Since the modification of bio-recognition units on the top of Ru@bp-SNA is able to confer excellent recognitive capacity and obtain the formation of complex in the presence of specific analyte, the decrease of ECL and EC signals enables the quantitative detection of this analyte. As proof-of-concept experiments, two tumor related biomarkers, CEA and CA15–3, have been successfully determined in aqueous solution and human serum by ECL and EC modes, respectively.

Abstract:It is a great challenge to fabricate multiplex and convenient photoelectrochemical biosensors for ultrasensitive determination of biomarkers. Herein, a fascinating potentiometric addressable photoelectrochemical biosensor was reported for double biomarkers' detection by varying the applied bias in the detection process. In this biosensor, the nanocomposite of cube anatase TiO2 mesocrystals and polyamidoamine dendrimers modified a dual disk electrode as an excellent photoelectrochemical sensing matrix. Subsequently, two important biomarkers in serum for prostate cancer, prostate-specific antigen and human interleukin-6, were immobilized onto the different disks of modified electrode via glutaraldehyde bridges. Then another two photosensitizers, graphitic-carbon-nitride-labeled and CS-AgI-labeled different antibodies, were self-assembled onto the electrode surface by a corresponding competitive immune recognition reaction. The change in photocurrent with the target antigen concentration at different critical voltages enables us to selectively and quantitatively determine targets. The results demonstrated that this potentiometric addressable photoelectrochemical biosensing strategy not only has great promise as a new point-of-care diagnostic tool for early detection of prostate cancer but also can be conveniently expanded to multiplex biosensing by simply change biomarkers. More importantly, this work provides an unambiguous operating guideline of multiplex photoelectrochemical immunoassay.

Abstract:Some of the cancer biomarkers often lack specificity and sensitivity; thus, simultaneous detection of multiple biomarkers can make the diagnosis more accurate. Also, simple sensing system without utilization of extra reagents like mediator or substrate during detection event is desirable for point-of-care testing. To address this, mediator and substrate-free amperometric biosensor for simultaneous detection of cancer biomarkers carcinoembryonic antigen (CEA) and alpha-fetoprotein (AFP) have been demonstrated by designing two different redox-labelled detection probes. Colloidal nanoparticles of polyaniline-pectin conjugated with AFP antibody along with ferrocene and silver nanoparticles conjugated with CEA antibody along with anthraquinone were used as redox probes to bind with AFP and CEA during the detection event. Sensor constructed using carboxylic acid tethered polyaniline as immobilization matrix displayed 5 times wider linear range than conventional polyaniline for AFP and CEA detection by sandwich electrochemical assay. The detection limit was 30 pg mL −1 for AFP and 80 pg mL −1 for CEA. The biosensor displayed appropriate sensitivity, good specificity, and negligible cross-reactivity between the two targets. The proposed sensor was used to determine APF and CEA in human blood serum. The strategy demonstrated can be further extended for detection of panel of cancer biomarkers by designing appropriate redox probes.
Application of Multiplex Microfluidic Electrochemical Sensors in Monitoring Hematological Tumor Biomarkers

Abstract:Rapid yet accurate detection of disease-related biomarkers is key for point of care testing, where there is an increasing demand for multi-index analysis. Here, we present a versatile device for multianalyte quantification based on the microfluidic technique and electrochemical sensor array. The analytes were shunted through inner-built channels to screen-printed electrodes installed at different positions of the chip. These electrodes were modified with different nanomaterials and sensing agents to afford specific responses to the corresponding indicators. To prove the applicability of the platform for multifunction, we chose leukemia as the model disease and determined four relevant markers: methotrexate (MTX), lactate dehydrogenase (LDH), uric acid (UA), and urea. They are indicative as/for the therapeutic drug (MTX), prognosis (LDH), and side effect (UA and urea). The sensing chip exhibited low detection limits of 35 nM, 25 U/L, 450 nM, and 20 μM toward the four analytes, which are much lower than their minimum contents in human serum. Furthermore, practical application of the chip was demonstrated by simultaneous detection of the four analytes in the blood plasma of rabbit. By simply replacing the modification agents, the sensing platform is expected to serve the detection of a wide range of chem/biosubstances in various fields.
