Abstract:A first robust synthetic method for the construction of quinoline fused triazolo-azepine derivatives via intramolecular 1,3-dipolar cycloaddition reaction from quinoline containing azide and alkyne functionality has been described. This novel approach has significant potential to create five- and seven-membered ring in a one-pot operation without use of any metal-catalyst or additives. The in-situ cyclization reactions afforded interesting and biological significant triazolo-azepines derivatives in high yields (65-87%). Moreover, this interesting chemistry has several advantages like metal-free, additives-free, high atom economy, diversified library of compounds, high yields of products, in-situ formation of two ring and pharmaceutical importance of synthesized prototypes.
