Abstract:We have developed a new class of plasmonic vesicular nanostructures assembled from amphiphilic gold nanocrystals with mixed polymer brush coatings. One major finding is that the integration of gold nanocrystals (nanoparticles and nanorods) with two types of chemically distinct polymer grafts, which are analogous to block copolymers as a whole, creates a new type of hybrid building block inheriting the amphiphilicity-driven self-assembly of block copolymers to form vesicular structures and the plasmonic properties of the nanocrystals. In contrast to other vesicular structures, the disruption of the plasmonic vesicles can be triggered by stimulus mechanisms inherent to either the polymer or the nanocrystal. Recent advances in nanocrystal synthesis and controlled surface-initiated polymerization have opened a wealth of possibilities for expanding this concept to other types of nanocrystals and integrating different types of nanocrystals into multifunctional vesicles. The development of multifunctional vesicles containing stimuli-responsive polymers could enable their broader applications in biosensing, multimodality imaging, and theragnostic nanomedicine.

Abstract:In this study, we report the light-triggered disassembly of gold nanovesicles (GVs) from amphiphilic photo-responsive block copolymer (polystyrene-b-polyethylene glycol, PS-C4-PEG) tethered gold nanoparticles (Au NPs) for controlled drug release. Amphiphilic photo-responsive PS-C4-PEG, with the group of truxillic acid at the junction of the two blocks, is prepared and grafted on the surface of Au NPs. GVs with tunable size and structures can be generated by self-assembly of the PS-C4-PEG tethered Au NPs in a selective solvent. Drugs can be effectively encapsulated into the cavity of the GVs during the self-assembly process. Moreover, under UV light irradiation, the PS-C4-PEG on the Au NPs can be decomposed, resulting in the formation of defects on the surface of the GVs, and sustainable drug release can thus be achieved. More interestingly, the entire contour of the GVs remains after UV light irradiation due to the slow debonding of truxillic acid, which is important for combined chemo- and photothermal-therapies. With these features, the obtained GVs can be used as a multifunctional platform for cancer diagnosis, bioimaging, and combined therapies.

Abstract:Multifunctional triggered drug release systems have been one of the most intriguing and challenging topics. In this study, we synthesize amphiphilic copolymers composed of hydrophilic oligo ethylene glycol methyl ether methacrylate and hydrophobic azobenzene-containing methacrylate. These amphiphilic copolymers can easily self-assemble into micelles in aqueous media and show smart responses to temperatures, UV/vis lights and ions. Interestingly, the obtained copolymers can self-assemble onto hydrophobic upconversion nanoparticles in aqueous media via hydrophobic interaction to form hybrid colloidal clusters with the hydrophobic nanocrystal core and hydrophilic polymeric shell. The hybrid colloidal clusters exhibit a thermo-responsive and photosensitive performance. When served as a drug carrier, both the near-infrared light and temperature triggered drug release can be realized, which have potential applications in intelligent drug delivery.

Abstract:A high throughput way to generate gold nanoparticle-based microcapsules was demonstrated based on the interfacial assembly in a water and 2-ethyl-1-hexanol two phase system. The obtained gold microcapsules exhibit superior stability, good flexibility, ductility, and a significant enhancement (about 10-fold) in the biphasic catalytic reaction rate upon near-infrared light irradiation.

Abstract:The hierarchical assembly of multifunctional materials is challenging. In their Communication on page 15809 ff., Z. H. Nie, X. Y. Chen, and co-workers report the stepwise assembly of block-copolymer-tethered gold nanoparticles into strings that subsequently fold into vesicles with chain-like ordering of the nanoparticles in the membrane. The resulting vesicular assemblies can be used for photoacoustic imaging and drug delivery owing to their strong absorption in the near-infrared region.

Abstract:Using atom transfer radical polymerization (ATRP) and macromolecular azo coupling reaction, both o-nitrobenzyl (ONB) group and azobenzene group were efficiently incorporated into the center of the amphiphilic diblock copolymer chain. The prepared diblock copolymer was well characterized by UV-vis, H-1 NMR, and GPC methods. Self-assembly of the amphiphilic copolymer in selected solvents can result in uniform self-assembly aggregates. In the presence of external stimuli [upconversion nanoparticles (UCNPs)/NIR light or enzyme], the amphiphilic diblock copolymer chain could be broken by the cleavage of ONB or azobenzene group, which would lead to the disruption of the self-assembly aggregates. This photo- and enzyme-triggered disruption process was proved by using transmission electron microscopy (TEM) and GPC method. Fluorescence emission spectra measurements indicated that the release of Nile red, a hydrophobic dye, encapsulated by the self-assembly aggregates, could be successfully realized under the NIR light and enzyme stimuli. Abstract:Self-assembly of gold nanoparticles (AuNPs) into highly ordered superstructures provides a promising route toward fabricating materials with new functionalities or enhanced physical properties. Although self-assembly of AuNPs has garnered significant research attention recently, a highly ordered superlattice of AuNPs under a low concentration in a confined geometry formed by nonfunctionalized materials has not been reported. Herein, we investigate the self-assembly of a 2D AuNPs superlattice in a polymer vesicle layer using hydrophobic interactions, which exhibits centered rectangular lattice symmetry. To create the highly ordered AuNPs superlattice, the P(EGx-b-iPGEy) block copolymers that form the thickness of the hydrophobic vesicle layer comparable to the size of the AuNP are used as a template to control the AuNP degree of freedom. To the best of our knowledge, this study provides the first demonstration of a centered rectangular structure formation of AuNPs at the vesicle layer in 2D confined geometry.

Abstract:The stepwise self-assembly of hollow plasmonic vesicles with vesicular membranes containing strings of gold nanoparticles (NPs) is reported. The formation of chain vesicles can be controlled by tuning the density of the polymer ligands on the surface of the gold NPs. The strong absorption of the chain vesicles in the near-infrared (NIR) region leads to a much higher efficiency in photoacoustic (PA) imaging than for non-chain vesicles. The chain vesicles were further employed for the encapsulation of drugs and the NIR light triggered release of payloads. This work not only offers a new platform for controlling the hierarchical self-assembly of NPs, but also demonstrates that the physical properties of the materials can be tailored by controlling the spatial arrangement of NPs within assemblies to achieve a better performance in biomedical applications.

Abstract:In contrast to the conventional hierarchical self-assembly process, effective methods to enable reversible hierarchical self-assembly of block copolymers are comparatively few and limited in scope. Herein, we report, for the first time, a simple yet robust strategy for light-induced reversible hierarchical self-assembly of an amphiphilic diblock copolymer, poly(4-vinylpyridine)-block-poly[6-[4-(4-butyloxyphenylazo)phenoxy]hexyl methacrylate] (denoted P4VP-b-PAzoMA). The hierarchical structures are constructed via a two-step self-assembly process (first-level reverse micelles, second-level compound micelles, and rearrangement into micrometer-sized vesicles) driven by use of solvent. Intriguingly, because of reversible photoinduced trans-to-cis isomerization of azobenzene moieties in PAzoMA, the vesicles could disassemble into subunits upon UV light and then recover the nearly identical vesicular morphology upon visible light. Such a reversible hierarchical self-assembly process is accompanied by reversible fluorescence, encapsulation, and controlled release of dyes and can be used as a template for the synthesis of nanoparticles. Clearly, the ability to render the light-enabled reversible hierarchical self-assembly provides a unique platform for smart delivery vehicles and templates for nanomaterials.

Abstract:A new strategy for preparing luminescent and intelligent gold nanodots based on supramolecular self-assembly is described in this paper. The supramolecular self-assembly was initiated through electrostatic interactions and ion pairing between palmitic acid and hyperbranched poly(ethylenimine). The resulting structures not only have the dynamic reversible properties of supramolecules but also possess torispherical and highly branched architectures. Thus they can be regarded as a new kind of ideal nanoreactor for preparing intelligent Au nanodots. By preparing Au nanodots within this kind of supramolecular self-assembly, the environmental sensitivity of intelligent polymers and the optical, electrical properties of Au nanodots can be combined, endowing the Au nanodots with intelligence. In this paper, a supramolecular self-assembly process based on dendritic poly(ethylenimine) and palmitic acid was designed and then applied to prepare fluorescent and size-controlled Au nanodots. The pH response of Au nanodots embodied by phase transfer from oil phase to water phase was also investigated.

Abstract:The preparation of a new near-infrared (NIR) light-responsive nanocarrier for controlled drug release is demonstrated. Upconversion nanoparticles (UCNPs) were coated with an amphiphilic diblock copolymer through surface-initiated atom transfer radical polymerization, in which the inner block is hydrophobic, ultraviolet (UV)-sensitive poly(4,5-dimethoxy-2-nitrobenzyl methacrylate) (PNB), and the outer block is hydrophilic poly(methoxy polyethylene glycol monomethacrylate) (POEG). The resulting polymer/UCNP nanocarrier is thermally stable in water over a wide temperature range (5-70 degrees C) and is uniform in size (120 nm hydrodynamic diameter, polydispersity index <0.1). The diblock copolymer self-assembly on the surface of each UCNP occurs in aqueous solution, which allows encapsulation of antitumor drugs like doxorubicin (DOX) by the hydrophobic micelle-like core of PNB surrounding the NIR-sensitive UCNP. Under 980 nm laser exposure, the UV light emitted by the single UCNP is absorbed by the PNB inner layer, which results in cleavage of o-nitrobenzyl groups and formation of carboxylic acid groups. The increasing hydrophilicity of the diblock copolymer resulting from the NIR light-triggered photochemical reaction can thus disrupt the nanocarrier and leads to the release of DOX molecules. This diblock copolymer self-assembly-based approach to constructing NIR light-responsive nanocarriers of well-defined structures is general and offers possibilities for photocontrolled drug delivery.

Abstract:Plasmonic nanovesicles show broad applications in such as cancer theranostics and drug delivery, but the preparation of nanovesicles from shaped nanoparticles remains challenging. This article describes the vesicular self-assembly of shaped nanoparticles, such as gold nanocubes, grafted with amphiphilic block copolymers in selective solvents. The nanocubes assembled within the vesicular membranes exhibit two distinctive packing modes, namely square-like and hexagonal packing, depending on the relative dimension of the copolymer ligands and nanocubes. The corresponding optical properties of plasmonic nanovesicles could be tuned by varying the length of grafted copolymers and the size of nanocubes. This work provides guidance for the fabrication of functional plasmonic vesicles for applications in catalysis, nanomedicines and optical devices.

Abstract:Controllable assemblies of nanocrystals have attracted considerable interest because they often exhibit unique collective properties that differ from those displayed by individual nanocrystals and bulk samples. Reported approaches to prepare nanocrystal assemblies based on the molecular recognitions of small molecules or biomacromolecules are effective, but often require complicated and time-consuming modification processes of nanocrystals. In this paper, we demonstrate a simple and universal approach to assemble gold nanocrystals (AuNCs) into linear chains and complex networks in aqueous silver nitrate medium under irradiation with UV light without the involvement of any modification step. Due to the strong plasmon resonance coupling verified by finite difference time domain calculation, the assembled structures of AuNCs can be used as excellent surface-enhanced Raman scattering substrates and dark-field light-scattering bioimaging probes.

Abstract:Nanosized near infrared (NIR) responsive nanohybrids are synthesized by surface-initiated atom transfer radical polymerization (SI-ATRP) of N-isopropylacrylamide (NIPAAm) on gold nanorods (Au NRs). Transmission electron microscopy images demonstrate that the nanohybrids have a clear core/shell structure. Temperature-variable H-1 NMR spectroscopy, quasi-elastic light scattering (QELS), UV-vis spectroscopy, and laser irradiation prove that the nanohybrids are both temperature and NIR responsive. Norvancomycin (NVan) loading and release experiments show that the drug release rate can be modulated by NIR irradiation. Such an intelligent drug-delivery system might find applications in non-invasive controlled drug release in future.

Abstract:A two-step sequential gelation process is developed by C.-H. Chen and co-workers to produce agarose/alginate double network multi-compartmental hydrogel particles, using droplet assemblies induced by superhydrophobic surfaces as templates. The hydrogel particles can be formed with diverse hierarchical structures, showing combinational functionalities. On page 4886 it is shown how the synthesized hydrogel particles, when loaded with polypyrrole (PPy) nanoparticles that act as photothermal nanotransducers, function as NIR light-triggered and deformation-free hydrogel materials.

Abstract:Luminescent Au nanoparticles (AuNPs) and their organic/inorganic nanohybrids are of interest due to their favorable properties and promising biomedical applications. However, most existing AuNPs-based hybrid nanostructures cannot satisfy high efficiency in synthesis, deep tissue penetration, and long blood circulation simultaneously, thus cannot be employed in dynamic monitoring of biomedical applications. In this paper, using Pluronic F127 as a template, we report a robust approach for one-pot synthesis of AuNPs-based organic/inorganic nanohybrids (AuNHs) with bright luminescence in the second near-infrared (NIR-II) window, tunable shape, and controllable surface polyethylene glycol (PEG) density. The nanohybrids could be controlled from a necklace-like shape with dense brush PEG configuration to a spherical structure with brush PEG coating, which greatly impacts the in vivo biological behaviors. Compared to spherical AuNHs, the necklace-shaped AuNHs present a higher quantum yield and longer blood circulation, which are superior to most of individual AuNPs. With these outstanding features, the necklace-shaped AuNHs could achieve real-time, dynamic visualization of vascular dysfunction, capable of directing the precise administration of thrombolytics (a medicine for the breakdown of the blood clot). These findings could provide a powerful guide for designing novel NIR-II nanoprobes toward in vivo dynamic information visualization.

Abstract:A new theranostic system has been successfully fabricated by incorporating thermo-sensitive poly(N-isopropyl acrylamide) nanospheres with luminescent peptide-Au clusters (PNIPAM-peptide-Au nanospheres). With excellent stability, thermo-responsiveness, red fluorescence and targeting ability, these PNIPAM-peptide-Au nanospheres hold great potential as multifunctional platforms for controlled release, fluorescence imaging and targeted therapy.
