Abstract:An ultra-high pressure liquid chromatography-tandem mass spectrometry method has been developed and validated for identification and quantification of five major bioactive components in rat plasma after oral administration of Qihuotongqiao tablets. The analysis was performed on an Acquity, UPLC HSS T3 column (100 mm x 2.1 mm, 1.8 mu m; Waters, USA) utilizing a gradient elution profile and a mobile phase consisting of(A) water containing 0.5 mM ammonium chloride and (B) acetonitrile. Electrospray ionization (ESI) tandem interface was employed prior to mass spectrometric detection. The calibration curve was linear over the range of 4.2-416.0 ng/mL for notoginsenoside RI, 38.4-3840.0 ng/mL for ginsenoside Rg(1), 3.7-368.0 ng/mL for ginsenoside Re, 37.6-5640.0 ng/mL ginsenoside Rb-1 and 4.5-448.0ng/mL for icariin. respectively. The average accuracies ranged from 87.2 to 109.3% with RSD <= 13.7%. The results indicated that ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS) provided improved chromatographic parameters resulting in significantly increased sample throughput including lower solvent consumption and lower limits of quantitation (LLOQ) for most of target analytes compared to previous method employing conventional high-performance liquid chromatography (HPLC) separation. Abstract:A sensitive and specific method was developed and validated for the quantitation of one major metabolite of genipin in rats plasma. The major metabolite was isolated from rat bile via semi-preparative HPLC technology and its chemical structure was identified as genipin-1-o-glucuronic acid (GNP-GLU), which was for the first time used as a standard compound for quantitative analysis in rat plasma after administration of genipin. The application of high-performance liquid chromatography-tandem mass spectrometry in negative mode in multiple reaction monitoring mode was investigated. Chromatographic separation was achieved on an Eclipse XDB-C-18 column using a mobile phase consisting of water with 0.1% formic acid (A)-acetonitrile (B). The limit of detecation was 0.214ng/mL and the lower limit of quantification was 0.706ng/mL. The calibration curve was linear from 1.27 to 3810ng/mL for plasma samples, with a correlation coefficient of 0.9924. The intra- and inter-day precisions and accuracy were all within 15%. The recoveries of GNP-GLU and puerarin were above 90.0 and 76.2%, respectively. The highly sensitive method was successfully applied to estimate pharmacokinetic parameters of GNP-GLU following oral and intravenous administration of genipin to rats. Determination of Imatinib in Rat Plasma by HPLC and Study of Its Pharmacokinetics

Abstract:OBJECTIVE To develop a high performance liquid chromatography method for the determination of imatinib in rat plasma and study its pharmacokinetics.METHODS Plasma was deposited by perchloric acid.The analytical column was ZORBAX SB-C18(4.6 mm×150 mm,5 μm).The mobile phase was acetonitrile-water-0.1% trifluoroacetic acid(19∶61∶20) and the flow rate was 1.0 mL·min-1.The UV detection wavelength was 282 nm.Six male SD rats were given a single dose of 50 mg·kg-1 imatinib by garage.Blood samples were collected from tail vein at different time points.The concentrations of imatinib in plasma were determined by the established HPLC method.The pharmacokinetic parameters were analyzed by DAS program.RESULTS Excellent liner relationship was obtained in the range of 0.10-20.00 mg·L-1(r=0.999 8),and the lower limit of qauntification was 0.05 mg·L-1.The recoveries were(98.86±2.77)%,(100.35±2.31)% and(100.14±1.66)% respectively at three concentrations(0.25,5.00,15.00 mg·L-1),the intra-day RSDs were 2.64%,2.50% and 1.62% with REs of-1.60%,-1.00% and 0.10%,and the inter-day RSDs were 3.51%,2.77% and 1.34% with REs of-0.80%,0.72% and 0.21%,respectively.After oral administration in rats,the pharmacokinetic profile of imatinib was fitted with a two-compartment model,the half-lives of α phase and β phase were(2.40±0.84) h and(7.82±0.87) h,respectively.CONCLUSION The method is accurate,simple,rapid and can be used to determine imatinib concentration in rat plasma and study its pharmacokinetics.
Determination of Ginsenosides Rb1, Rb2, and Rb3 in Rat Plasma by a Rapid and Sensitive Liquid Chromatography Tandem Mass Spectrometry Method: Application in a Pharmacokinetic Study.

Abstract:A sensitive rapid resolution liquid chromatography–tandem mass spectrometry method was developed to determine the pharmacokinetics of ginsenoside Rb1, Rb2, and Rb3 in rats, after oral administration (50mg/kg) and intravenous administration (10mg/kg) of Rb1, Rb2, and Rb3, respectively. The plasma samples were extracted by saturated N-butanol with Rg2 as internal standard. Chromatographic separation was performed on a Zorbax SB-C18 column (50mm×4.6mm, 1.8μm) with a mobile phase consisting of methanol and 1mM ammonium formate (74:26, v/v). Multiple reaction monitoring mode was performed using the fragmentation transitions of m/z 1107.7→m/z 178.9, m/z 1077.7→m/z 148.6, and m/z 1077.7→m/z 783.4 for Rb1, Rb2, and Rb3, respectively. Calibration curves were recovered over a concentration range of 20–1000ng/ml for Rb1 and Rb2, and 50–2500ng/ml for Rb3. The limits of detection were 3.0ng/ml, 4.0ng/ml, and 6.5ng/ml. Both intra-day and inter-day variances were less than 15% and the accuracy was within 86–114% for the three ginsenosides. All three ginsenosides had poor oral bioavailability (0.78%, 0.08%, and 0.52% for Rb1, Rb2, and Rb3, respectively). The value of Rb1 is higher than that of Rb2 or Rb3, indicating that ginsenosides with hexose and hydroxyl groups (Rb1) could present better pharmacokinetic behaviors than those with pentose groups in the same glycosylation site by oral administration.
Determination of GL-V9, a Derivative of Wogonin, in Rat Plasma by UPLC-MS/MS and Its Application to a Pharmacokinetic Study after Oral and Pulmonary Administration.

Abstract:GL-V9, a derivative of wogonin, shows much more potent anticancer properties than wogonin. In this study, a selective, sensitive and rapid ultra-high-performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS) method was developed and validated for the determination of GL-V9 in rat plasma. Plasma samples were processed using methanol to precipitate protein. Chromatographic separation of analytes was achieved on a C-18 column using gradient elution within 4.5 min. The mobile phase consisted of acetonitrile and water including 0.1% (v/v) formic acid and 5 mm ammonium acetate. GL-V9 and caffeine (internal standard) were monitored by positive electrospray triple quadrupole mass spectrometer and quantified using multiple reaction monitoring (MRM) mode with the transitions of m/z 410.20 -> 126.10 (GL-V9) and 195.10 -> 138.00 (IS: caffeine), respectively. Good linearity was obtained over the range of 2-1000 ng/mL (R-2 > 0.99) and the extraction recovery was 101.91 +/- 11.34%. The intra- and inter-day precision variations were small (RSD 1.35-6.96%) and the relative error (RE) of accuracy was -7.35-6.27%. The established and validated UPLC-MS/MS method was successfully applied to study the pharmacokinetic behavior of GL-V9 after administration through different delivery routes. The results demonstrated that pulmonary delivery exhibited a greater advantage in terms of improving bioavailability compared with oral administration.
Determination and pharmacokinetic study of nodakenin in rat plasma by RP-HPLC method.

Abstract:A simple, sensitive and selective RP-HPLC method has been developed for quantification of nodakenin in rat plasma. Nodakenin in rat plasma was extracted with acetonitrile, which also acted as a deproteinization agent. Chromatographic separation of nodakenin was performed on an analytical Diamonsil (TM) ODS C-18 column, with a mobile phase of MeOH-H2O (1:1, v/v) at a flow-rate of 1.0 mL/min, and UV detection was set at 330 nm. The calibration curve was linear over the range 0.2-12.0 mu g/mL (R-2 = 0.9995) in rat plasma. The lower limit of detection and quantification were 0.01 and 0.1 mu g/mL, respectively, using the rat plasma sample. The extraction recoveries were 77.36 +/- 4.56, 82.89 +/- 1.84 and 81.66 +/- 2.49% at concentrations of 1.0, 5.0 and 10.0 mu g/mL, respectively. The intra- and inter-day precision and accuracy were validated by relative standard deviation and relative error, which were in the ranges 5.07-5.83 and 3.95-6.29%, respectively. After i.v. administration to rats at a single dose of 40 mg/ kg, the plasma concentration-time curve of nodakenin was best conformed to a two-compartment open model. Abstract:Deltonin is a naturally occurring spirostanol glycoside from Dioscorea zingiberensis C.H. Wright, which is used in traditional Chinese medicine. It exerts strong cytotoxic effect on C26 cells, inhibits C26 derived-tumor growth, and prolongs the survival of tumor-bearing mice after its oral administration, indicating its potential for use as an anti-tumor drug. To investigate the pharmacokinetic profiles of deltonin, a rapid, sensitive, and simplified high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) assay was developed and validated for the determination of deltonin in rat plasma. After acetonitrile-mediated plasma protein precipitation, chromatographic separation of deltonin was achieved using a reversed phase Hypersil Gold column (150mm×2.1mm, 5μm), with gradient elution using 0.1% formic acid and acetonitrile. Thereafter, deltonin was quantified using MS/MS with electrospray ionization (ESI) in positive multiple reaction monitoring (MRM) mode. The flow rate of the mobile phase was 200μL/min, and the retention time was 9.03min for deltonin and 6.31min for the internal standard (IS: 20(S)-ginsenoside Rb1). The linear range of the calibration curve was 2-5000ng/mL (r(2)>0.99), and the limit of detection (LOD) was 0.46ng/mL. The intra- and inter-day accuracies ranged from -2.8% to 11.1% and precisions (RSD) were within 13.1%. Deltonin was found to be stable under short-term temperature conditions, post-preparative temperature conditions, and after 3 freeze-thaw cycles conditions. The validated method was successfully applied to a pharmacokinetic study in rats after oral administration of deltonin (50 and 100mg/kg). The pharmacokinetics is characterized by high apparent clearance (CL/F) and apparent volume of distribution (Vd/F).
Determination of Sennoside A in Rat Plasma by Liquid Chromatography/Electrospray Ionization Tandem Mass Spectrometry and Its Application to a Rat Pharmacokinetic Study

Abstract:To characterize pharmacokinetic profile of sennoside A in rats after intravenous and oral administration, a simple and sensitive liquid chromatography tandem mass spectrometry method was established and validated for quantitative determination of sennoside A in rat plasma. After prepared by protein precipitation with acetonitrile, sennoside A and internal standard were separated on a Waters ACQUITY HSS T3 (2.1 × 100 mm, 1.8 μm) column using acetonitrile and 5‐mM ammonium acetate in water as mobile phase by gradient elution. The method showed excellent linearity over the range of 0.5–1000 ng/mL with acceptable intra‐ and inter‐day precision, accuracy, matrix effect, and recovery. The stability assay indicated that sennoside A was stable in plasma during the sample collection, preparation, and analysis. Next, the method was applied to pharmacokinetic study of sennoside A in rats. After intravenous and intragastric administrated to rats, the concentrations of sennoside A in plasma at different time points were quantitated and the pharmacokinetic parameters were calculated by software of DAS 2.0. Pharmacokinetic parameters suggested that after oral administration, sennoside A was reached to the peak at 2.9–3.6 h with a Cmax value of 13.2–31.7 ng/mL. Sennoside A was eliminated slowly from the plasma with T1/2 value between 15.4 and 18.3 h. The oral absolute bioavailability was among 0.9%–1.3%, which indicated low blood exposure level.

Abstract:A rapid and high selective ultra-performance liquid chromatography（UPLC） with tandem mass spectrometry method for simultaneous determination of six compounds including albiflorin, paeoniflorin, picroside I, picroside II, saikosaponin A, and saikosaponin D in rat plasma was developed and validated using butyl p-hydroxybenzoate as an internal standard. One-step direct protein precipitation with acetonitrile was used to extract the compounds from the rat plasma samples. Chromatographic separation was achieved using an ACQUITY UPLC BEH C 18 column（100 mm × 2.1 mm, 1.7 μm） at a flow rate of 0.4 m L/min, using gradient mode containing 0.1% formic acid in water and acetonitrile were used as the Mobile phase A and B. Electrospray ionization in negative ion mode and multiple reaction monitoring were used to identify and quantify active components. Calibration curves showed good linearity（R~2 ＞ 0.9908） over a wide concentration range for all compounds. The intra-and interday precision（relative standard deviation） ranged 2.4%–7.0% and 2.6%–8.0%, respectively. The accuracy（relative error） was from-13.0% to 13.2% at all quality control levels. The recovery ranged from 81.1% to 92.5%. The validated method was successfully applied to pharmacokinetic study in rats after oral administration of Qing Gan-Shu Yu-Fang. The results show that one can draw a conclusion that these six active ingredients can be quickly absorbed and play a pharmacodynamic role rapidly in vivo.
Determination of Sparstolonin B by Ultra-High Performance Liquid Chromatography Coupled with Triple Quadrupole Mass Spectrometry: Application to Pharmacokinetic Study of Sparstolonin B in Rat Plasma

Abstract:Sparstolonin B (SsnB), a spontaneous isocoumarin compound isolated from the tuber of Scirpus yagara Ohwi. (Cyperaceae), possesses potent anti-inflammatory and antitumor activity. In the present study, a rapid and simple UHPLC/MS/MS method for determination of SsnB in rat plasma was developed and validated. Plasma samples were pretreated by liquid-liquid extraction with ethyl acetate containing rhein as an internal standard and separated on a C-18 column at 35 degrees C, with a gradient mobile phase consisting of acetonitrile and water containing 0.2% (v/v) formic acid within 2.1min. MS/MS detection was accomplished in multiple reaction monitoring mode with negative electrospray ionization. The precursor-product ion transitions were m/z 266.9 [M-H](-)m/z 211.0 for SsnB and m/z 283.2 [M-H](-)m/z 239.0 for IS. The intra- and inter-day precision (RSD) was <8.98% and the accuracy (RE) ranged from -7.40 to 4.50%. The extraction recoveries ranged from 96.28 to 97.30%. The pharmacokinetic parameters were calculated using Win Nonlin53 software. The absolute bioavailability of SsnB was estimated to be 6.98%. The proposed method was successfully applied to a pharmacokinetic study of SsnB in rats after intravenous administration with a dose of 0.5mg/kg and oral administration at a dose of 5mg/kg. Wogonin Pharmacokinetic Behavior in Rats after Intravenous Injection

Abstract:Objective: To establish a method for determining plasma level of wogonin and study its pharmacokinetic behavior in rats after intravenous injection.Method: The concentration of wogonin in plasma was determinated by HPLC on a Lichrospher ODS(150 mm×6.0 mm,5 μm) column with the mobile phase of methanol-water-acetic acid(60:40:0.5);UV detector was set at 275 nm,the temperature of column was 35 ℃,flow rate was set at 1 mL·min-1.Wogonin was injected into rat through tail vein after 16h fasting,and its plasma concentrations at different times were determined.The data were processed with the software 3p97.Results:The linear range was 0.02-5.0 μg·mL-1,the lowest limit of quantification was 20 ng·mL-1.The intra-day and inter-day variations of RSD were less than 10%.The primary pharmacokinetic parameters were : t1/2,α0.0656 h,t1/2,β4.447 h,AUC 883 h·ng·mL-1.Conclution:This assay method is simple,precise and sensitive enough for intravenous pharmacokinetic study of wogonin and the pharmacokinetic behavior of wogonin in rats fitted to the two compartment model. The wogonin distributed quickly but eliminated slowly.

Abstract:A simple, rapid and sensitive LC-MS/MS analysis method was developed and validated for the determination of Raddeanin A (RA) in rat plasma. Protein precipitation with three volumes of methanol as the precipitation reagent was used as the sample preparation method. The analysis process was performed on a Thermo Syncronis C18 column with the mobile phase of methanol-water (containing 5mM ammonium formate, pH 2.2) (85:15, v/v). RA and glycyrrhetinic acid (internal standard) were monitored under negative electrospray ionization in multiple reaction monitoring (MRM) mode. Retention time of RA and IS were 2.1 min and 3.5 min, respectively. The limit of detection was 5 ng/mL and the linear range was 50-50,000 ng/mL. The intra-day and inter-day precision was 1.87-2.94% and 3.25-5.36%, and the intra-day and inter-day accuracy ranged from 5.9% to 10.5% and 5.6% to 11.1%, respectively. The absolute recovery was above 90.3%. The method has been successfully translated to the pharmacokinetic study of RA in rats after intravenous and intraperitoneal administration (0.75 mg/kg).
Determination of astragaloside III in rat plasma by liquid chromatography-tandem mass spectrometry and its application to a rat pharmacokinetic study

Abstract:Astragaloside III (AST III), a naturally occurring saponin compound isolated from Radix Astragali, has been demonstrated to have anti-gastric ulcer, immunomodulatory and antitumor effects. To evaluate its pharmacokinetics in rats, a rapid, sensitive and specific high-performance liquid chromatography-tandem mass spectrometric (HPLC-MS/MS) method has been developed and validated for the quantification of astragaloside III in rat plasma. Samples were pretreated using a simple protein precipitation with methanol-acetonitrile (50:50, v/v) and the chromatographic separation was performed on a C18 column by a gradient elution using a mobile phase consisting of water containing 0.1% formic acid and acetonitrile containing 0.1% formic acid. Astragaloside III and the internal standard (buspirone) were detected using a tandem mass spectrometer in positive multiple reaction monitoring mode. Method validation revealed excellent linearity over the range of 5.00-5000 ng/mL together with satisfactory intra- and inter-day precision, accuracy and recovery. Stability testing showed that astragaloside III spiked into rat plasma was stable for 24 h at 20°C temperature, for up to 30 days at -80°C, and during three freeze-thaw cycles. The method was successfully used to investigate the pharmacokinetic profile of AST III after oral (10 mg/kg) and intravenous (1.0 mg/kg) administration in rats. The oral absolute bioavailability of AST III was calculated to be 4.15 ± 0.67% with an elimination half-life value of 2.13 ± 0.11 h, suggesting its poor absorption and/or strong metabolism in vivo.

Abstract:The purpose of this study was to simultaneously investigate the pharmacokinetics of five bioactive compounds in rat plasma after oral administration of Buyang Huanwu decoction (BYHWD) using high-performance liquid chromatography coupled with mass spectrometry (HPLC-MS). The separations were performed on a Thermo Hypersil Gold C-18 analytical column (50 x 2.1 mm, 3 mu m) with the column temperature kept at 30 degrees C. The quantitative analysis was performed using a quadrupole mass spectrometer detector operated under selected ion monitoring mode. A linear gradient elution of A (0.1% formic acid solution) and B (100% acetonitrile) was used at a flow rate of 0.2 mL/min. The method was validated within the concentration ranges 1.8-450, 6.0-1500, 2.0-500, 1.2-300 and 1.2-150 ng/mL for paeoniflorin, calycosin-7-O-beta-D-glucoside, ononin, calycosin and formononetin, respectively. Simultaneous determination by UPLC-MS/MS of seven bioactive compounds in rat plasma after oral administration of Ginkgo biloba tablets: application to a pharmacokinetic study

Abstract:A rapid, reliable, and sensitive method was developed using ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) with an electrospray ionization (ESI) source for determination of seven bioactive compounds in rat plasma after oral administration of Ginkgo biloba tablets (GBTs). The method simultaneously detects bilobalide (BB), ginkgolide A (GA), ginkgolide B (GB), ginkgolide C (GC), quercetin (QCT), kaempferol (KMF), and isorhamnetin (ISR) for pharmacokinetic study. The analytes and internal standard (IS) were extracted from rat plasma by acetidin. An MS/MS detection was conducted using multiple reaction monitoring (MRM) and operating in the negative ionization mode. The calibration curve ranges were 5-500, 5-500, 2.5-250, 1-100, 1-100, 1-100, and 1-100 ng/ml for BB, GA, GB, GC, QCT, KMF, and ISR, respectively. The mean recovery of the analytes ranged from 68.11% to 84.42%. The intra- and inter-day precisions were in the range of 2.33%-9.86% and the accuracies were between 87.67% and 108.37%. The method was used successfully in a pharmacokinetic study of GBTs. The pharmacokinetic parameters of seven compounds were analyzed using a non-compartment model. Plasma concentrations of the seven compounds were determined up to 48 h after administration, and their pharmacokinetic parameters were in agreement with previous studies.
A Rapid and Sensitive Ultra-High-pressure Liquid Chromatography-Tandem Mass Spectrometry Method for the Determination of Notoginsenoside Ft1 in Rat Plasma with Application to Pharmacokinetic Study

Abstract:Notoginsenoside Ft1 (NGFt1), a dammarane triterpene glycoside isolated from Panax notoginseng, showed potent effective in stimulating platelet aggregation in our previous assay, yet its pharmacokinetic behavior is still unclear. This study describes a rapid and sensitive ultra-high-pressure LC-tandem mass spectrometry assay for determining of NGFt1 in rat plasma. Methanol-mediated precipitation was used for sample pre-treatment. Chromatographic separation was achieved on a C-18 column with gradient elution using water and acetonitrile as mobile phase. Determination was obtained using an electrospray ionization source in negative selected reaction monitoring (SRM) mode at the transitions of m/z 915.9 -> m/z 783.8 and m/z 799.8 -> m/z 637.8 for NGFt1 and internal standard, respectively. The assay was linear over the concentration range 0.25-2500 ng/mL (r > 0.995) with the lower limit of quantification of 0.25 ng/mL. The intra- and inter-day precisions (relative standard deviation, %) ranged 1.65%-9.84% and 2.46%-13.49%, respectively, whereas accuracy (relative recovery, %) ranged from 96.21% to 99.45%, respectively. The recovery ranged from 95.09% to 102.22% and the matrix effect from 98.29% to 100.13%. The analyte was stable under tested storage conditions. The method has been successfully applied to a preclinical pharmacokinetic study in rats after a single intravenous (2 mg/kg) and oral (50 mg/kg) administration.

Abstract:A simple, rapid and sensitive method for the determination of geniposidic acid (GSA) in rat plasma was developed using liquid chromatography tandem mass spectrometry (LC-MS/MS). Geniposide (GS) was used as the internal standard. Rat plasma pretreated by solid-phase extraction (SPE) was analyzed by LC-MS/MS with negative ion mode electrospray ionization (ESI) in the multiple reaction monitoring (MRM) mode. The analytical column was C₈ column and the mobile phase was methanol (A) and water (B). The flow rate was set at 0.8 mL/min with split ratio of 1:3, the total run time was 15 min. The MS/MS ion transitions monitored were m/z 373.3-211.1 for GSA and m/z 387.3-225.3 for GS. The quantification limit was 5 ng/mL within a linear range of 10-4000 ng/mL. The inter-day and intra-day accuracy and precision were within ±10%. The method was fully validated for its sensitivity, selectivity, matrix effect, stability study and recovery. The data indicate that our LC-MS/MS assay is an effective method for the pharmacokinetics study of GSA in rat plasma.

Abstract:Toosendanin (TSN) is a major triterpenoid existing in Melia toosendan, which has been used as a digestive tract parasiticide and insecticide but with serious hepatotoxicity. An ultra-performance liquid chromatographyelectrospray ionizationmass spectrometry method was developed for determination of TSN in rat plasma. Plasma samples were separated on Acquity UPLCTM BEH C18 column with acetonitrile and water as flow phase by gradient elution and determined by quadrupole mass spectrometer in negative selective ion monitoring mode. Usolic acid was used as internal standard. The calibration curves were linear over 0.023.0?mu g/mL for TSN with a lower limit of quantification (LLOQ) of 20?ng/mL in rat plasma. The extraction recoveries of TSN were within 74.380.7% with an accuracy of 94.5108.9%. The intra- and inter-day precision values of the assay at three quality control levels were 8.813.8% and <13.9% at LLOQ level, respectively. The method was successfully applied to a pharmacokinetic study of TSN in rats after a single intravenous and oral administration of 2 and 60?mg/kg. The shorter Tmax, higher Vd and Cl of TSN after oral administration indicated that TSN could be absorbed, distributed and eliminated quickly in rats in vivo. Abstract:To evaluate the pharmacokinetics of a novel analogue of ginkgolide B, 10- O -dimethylaminoethylginkgolide B (XQ-1) in rat plasma in pre-clinical studies, a sensitive and specific liquid chromatographic method with electrospray ionization mass spectrometry detection (LC–ESI–MS) was developed and validated. After a simple extraction with ethyl acetate, XQ-1 was analyzed on a Shim-pack C18 column with a mobile phase of a mixture of 1 μmol L −1 ammonium acetate containing 0.02% formic acid and methanol (55:45, v / v ) at a flowrate of 0.3 mL min −1 . Detection was performed in selected ion monitoring (SIM) mode using target ions at [M + H] + m / z 496.05 for XQ-1 and m / z 432.10 for the internal standard (lafutidine). Linearity was established for the concentration range from 2 to 1,000 ng mL −1 . The extraction recoveries ranged from 86.0 to 89.9% in plasma at concentrations of 5, 50, and 500 ng mL −1 . The lower limit of quantification was 2 ng mL −1 with 100 μL plasma. The validated method was successfully applied to a pharmacokinetic study after intragastic administration of XQ-1 mesylate in rats at a dose of 20 mg kg −1 .

Abstract:A simple and sensitive UPLC-MS/MS assay was developed and validated for rapid determination of BZG in rat plasma and tissues. All biological samples were prepared by protein precipitation method using Imatinib as an internal standard (IS). The analyte and IS were separated on a C18 reverse phase analytical column with 4.5 min of analytical run, at flow rate of 0.3 mL/min. The detection was performed on a triple quadrupole tandem mass spectrometer equipped with electrospray ionization (ESI) by multiple reactions monitoring (MRM) of the transitions at m/z 451.0→254.0 for BZG and m/z 494.3→394.1 for IS, respectively. The linearity of this method was found to be within the concentration range of 0.5-2500 ng/mL with a lower limit of quantification of 0.5 ng/mL. All validation parameter results were within the acceptable range described in guideline for bioanalytical method validation. The method was successfully applied to a pharmacokinetic and tissue distribution study of BZG in rats. With the preliminary knowledge of in vivo pharmacokinetics and disposition properties, this study will be beneficial for further development of BZG.
