Abstract:We characterize vesicle adhesion onto homogeneous substrates by means of a perturbative expansion around the infinite adhesion limit, where curvature elasticity effects are absent. At first order in curvature elasticity, we determine analytically various global physical quantities associated with adhering vesicles: height, adhesion radius, etc. Our results are valid for adhesion energies above a certain threshold, that we determine numerically. We discuss the haptotactic force acting on a vesicle in the limit of weak adhesion gradients. We also propose a few methods for measuring adhesion energies and we suggest a possible way of determining the size of suboptical vesicles using controlled adhesion gradients.

Abstract:A theoretical model for describing the adhesion of lipid vesicle with free edges is developed. For adhesion in contact potential or in finite-range potential, the total energy functional is defined as the sum of elastic free energy, the surface energy, the line tension energy and the contact potential or the long-ranged potential. The equilibrium differential equation and boundary conditions for opening-up lipid vesicles are derived through minimizing the total energy functional. Numerical solutions to these equations are obtained under the axial symmetric condition. These numerical solutions can be used to qualitatively explain the influence of the substrate on the open-up lipid vesicles.

Abstract:We develop a two dimensional model of a vesicle adhered on a curved substrate via long-range molecular interactions while subjected to a detachment force. The relationship between the force and displacement of the vesicle is investigated as a function of the substrate shape. It is shown that both the force– displacement relationship and the maximum force at pull-off are significantly dependent on the substrate shape. The results suggest that probes with different tip shapes may be designed for cell manipulation. For example, we demonstrate that a vesicle can be pulled off a flat surface using a probe with a curved tip.

Abstract:In this paper, elastic behaviors of single polymer chains adsorbed on the rough surfaces with a substrate and some periodically tactic pillars are investigated by the pruned-enriched-Rosenbluth method (PERM). In our simulation, a single polymer chain is firstly adsorbed on the substrate and then pulled along the z-axis direction, which is vertical to the substrate. We investigate the chain size and shape of polymer chains, such as mean-square radii of gyration per bond 〈S2〉xy/N, 〈S2〉z/N and shape factor 〈δ∗〉 in order to show how the size and shape of adsorbed polymer chains change during the desorption process. Due to the occurrences of separation of the chains from the substrate, farther adsorption on the upper surfaces of pillars and complete separation from the whole rough surfaces in the elastic process, the changes of 〈S2〉xy/N, 〈S2〉z/N and 〈δ∗〉 during the process are complicated. On the other hand, some thermodynamic properties such as average energy per bond, average Helmholtz free energy per bond, elastic force f are investigated, and our aim is to study the elastic behaviors of polymer chains adsorbed on the rough surface during the elasticity process. Elastic force f has some plateaus during the desorption process for strong adsorption interaction. If there is no adsorption interaction, the chains can get away from the rough surfaces spontaneously. These investigations can provide some insights into the elastic behaviors of polymer chains adsorbed on the rough surface.

Abstract:Elastic behaviors of single polymer chains adsorbed on the attractive surface are first investigated using Monte Carlo simulation method based on the bond fluctuation model. We investigate the chain size and shape of adsorbed chains, such as mean-square radius of gyration S2, mean-square bond length b2, shape factors sf(i) and delta*, and the orientation of chain segments P2, to illuminate how the shape of polymer chains changes during the process of tensile elongation. There are some special behaviors of the chain size and shape at the beginning of elongation, especially for strong attraction interaction. For example, mean fraction of adsorbed segments decreases abruptly in the region of small elongation ratio and then decreases slowly with increasing elongation ratio. In fact, the chain size and shape also changes abruptly for small elongation ratio with strong attraction interaction. Some thermodynamics properties are also investigated here. Average Helmholtz free energy increases fast for elongation ratio lambda<1.15, especially with strong attraction, and increases slowly for lambda>1.15. Similar behaviors are obtained for average energy per bond. Elastic force (f ) and energy contribution to force (f(U)) are also studied, and we find that elastic force decreases abruptly for lambda<1.15, and there is a minimum of elastic force for strong attraction interaction, then increases very slowly with increasing elongation ratio. However, there are different behaviors for weak attraction interaction. For energy contribution to force (f(U)), there is a maximum value for strong attraction interaction in the region of lambda<1.15. Some comparisons with the atomic force microscopy experiments are also made. These investigations may provide some insights into the elastic behaviors of adsorbed polymer chains.

Abstract:The morphology and motion behavior of a cell are highly influenced by its external biological, chemical, and physical stimuli, and geometric confinement. In this paper, it is revealed that the mean curvature of the substrate significantly influences the adhesion of vesicles. By employing the variational method and investigating the Helfrich free energy, the configuration of axisymmetric vesicles adhered to curved spherical substrates is obtained theoretically. Moreover, numerical simulations based on the finite element method are also carried out to investigate the adhesion of vesicles on curved substrates with complex shapes. It is found that for a fixed area of a vesicle, its total free energy depends mainly on the mean curvature of the adhesion region but is insensitive to the specific shape of the substrate, and the total free energy monotonically decreases with the increase in the mean curvature. In addition, possible biological significances of the curvature-dependent adhesion, such as the shape of the cell and antibiofouling, are discussed. This study may deepen our understanding of the underlying mechanisms of adhesion in cellular activities.

Abstract:A continuum model was introduced for the adhesion of vesicles to substrate surfaces. In the model, the vesicle membrane was assumed to be a closed shell with hyperelasticity. The vesicle cavity is filled with a liquid of fixed volume. The receptors on the membrane are mobile and initially uniformly distributed while the ligands on the substrate surface are fixed and also uniformly distributed. The formation of localized regions of tight binding between receptors and ligands, results in vesicle adhesion to the substrate surface. An adhesive model was introduced to describe the adhesive interaction between the receptors and the ligands. The growth of the adhesion area occurs via recruiting receptors from the non-adhered region through diffusion. Finite-element methods were used to solve the governing equations for the deformation of the vesicle and the receptor diffusion on the membrane surface. Effects of the membrane stiffness, the cohesive parameters and the receptor density on the adhesion kinetics of the vesicle were studied. In addition, the instability of the advancing front of the adhesion was also analyzed.

Abstract:A theoretic model is proposed to study the adhesion behavior of a vesicle adhering inside another vesicle in 2-D case. The equilibrium shape equations and boundary conditions are investigated. With zero pressure, the integral case of the shape equations with dimensionless parameters is derived. When a soft vesicle adheres inside a rigid ring, the bifurcation between two phases is revealed, the critical adhesion condition and phase diagram are obtained. The finite element method (FEM) is used to study the deformation processes induced by the change of work of adhesion and bending rigidity. The relationships between the energy, adhesion length, cell volume, bending rigidity and work of adhesion are obtained.

Abstract:On the basis of Helfrich's bending energy model, we show that the adsorption process of a small spherical particle to a closed vesicle can be analytically studied by retaining the leading terms in an expansion of the shape equation. Our general derivation predicts the optimal binding sites on a vesicle, where the local membrane shape of the binding site could be non-axisymmetric before the continuous adhesion transition takes place. Our derivation avoids directly solving the shape equation and depends on an integration of the contact-line condition. The results are verified by several examples of independent numerical solutions.

Abstract:Adhesion plays an integral role in diverse biological functions ranging from cellular transport to tissue development. Estimation of adhesion strength, therefore, becomes important to gain biophysical insight into these phenomena. In this Letter, we use curvature elasticity to present non-intuitive, yet remarkably simple, universal relationships that capture vesicle-substrate interactions. Our study reveals that the inverse of the height, exponential of the contact area, and the force required to detach the vesicle from the substrate vary linearly with the square root of the adhesion energy. These relationships not only provide efficient strategies to tease out adhesion energy of biological molecules but can also be used to characterize the physical properties of elastic biomimetic nanoparticles. We validate the modeling predictions with experimental data from two previous studies.

Abstract:In this work, we study the adhesion of multi-component vesicle membrane to both flat and curved substrates, based on the conventional Helfrich bending energy for multi-component vesicles and adhesion potentials of different forms. A phase field formulation is used to describe the different components of the vesicle. For the axisymmetric case, a number of representative equilibrium vesicle shapes are computed and some energy diagrams are presented which reveal the dependence of the calculated shapes and solution branches on various parameters including both bending moduli and spontaneous curvatures as well as the adhesion potential constants. Our computation also confirms a recent experimental observation that the adhesion effect may promote phase separation in two-component vesicle membranes.

Abstract:Focal adhesions are clusters of specific receptor-ligand bonds that link an animal cell to an extracellular matrix. A capability to control focal adhesions, for which a quantitative description of the collective behavior of multiple molecular bonds is a critical step, is essential for tissue and cellular engineering. While the behavior of single molecular bonds is governed by statistical mechanics at small scale, continuum mechanics should be valid at large scale. How can this transition be modeled and can this tell us something about the mechanics of cell adhesion? Here we develop a stochastic-elasticity model of a periodic array of adhesion clusters between two dissimilar elastic media subjected to an inclined tensile stress, in which stochastic descriptions of molecular bonds and elastic descriptions of interfacial traction are unified in a single modeling framework. A fundamental scaling law of interfacial traction distribution is established to govern the transition between uniform and cracklike singular distributions of the interfacial traction within molecular bonds. Guided by this scaling law, we perform Monte Carlo simulations to investigate the effects of cluster size, cell/matrix modulus and loading direction on lifetime and strength of the adhesion clusters. The results show that intermediate adhesion size, stiff substrate, cytoskeleton stiffening, and low-angle pulling are factors that contribute to the stability of focal adhesions. The predictions of our model provide feasible explanations for a wide range of experimental observations and suggest possible mechanisms by which cells can modulate adhesion and deadhesion via cytoskeletal contractile machinery.

Abstract:When pulling a vesicle adhered on a substrate, both the force-displacement profile and the maximum force at pull-off are sensitively dependent upon the substrate shape. Here we consider the adhesion between a two-dimensional vesicle and a rigid substrate via long-range molecular interactions. For a given contact area, the theoretical pull-off force of the vesicle is obtained by multiplying the theoretical strength of adhesion and the contact area. It is shown that one may design an optimal substrate shape to achieve the theoretical pull-off force.

Abstract:A two-dimensionally adhesive contact model is established, in which a gas-filled elastic membrane adheres on a stretched substrate. The free energy of the system is achieved, minimization of which leads to the relationship between the contact width and the global substrate strain. The contact solution exhibits three distinct regimes characterized by two threshold strains: (i) the contact size is hardly affected by the external loading acted on the substrate when the global substrate strain is below the first threshold value; (ii) the size of the contact is reduced quickly as the force is between the two threshold levels; (iii) the contact size tends to vanish when the global substrate strain exceeds the second threshold level. All the results share a number of common features with the experimental observation of cell orientation on a stretched substrate. Effects of the internal pressure, the tensile stiffness of the membrane and the interface work of adhesion on the two threshold levels are further discussed. The finding in the present paper should be helpful for deep understanding of adhesion mediated deformation sensing mechanism by which cells can detect mechanical signals in extracellular matrices and the design of adhesion mediated deformation sensors. Abstract:Abstract Adhesion between a solid sphere and a thin film is a common but crucial issue in the study of biological membranes and two-dimensional materials. To supplement quantitative knowledge of membrane adhesion, this work addresses the axisymmetric adhesive contact between a rigid sphere and a circular freestanding elastic membrane with initial radial tension. For the membranes following linear stretching elasticity, both the JKR- and DMT-type adhesion as well as the transition regime in-between are considered. The dependences of external force with respect to the contact radius and displacement are derived analytically. In essence, the general solution is governed by three dimensionless parameters, reflecting the effects of membrane stretching elasticity, the range of adhesion force and the membrane size, respectively. It is interestingly found that the membrane size does not affect the contact radius and displacement at zero-external force at all and has minor influence on the value of pull-off force. The presented closed form solutions might be useful for the understanding of adhesion behaviors of sphere-membrane systems.

Abstract:A fundamental understanding of cell-nanomaterial interaction is of essential importance to nanomedicine and safe applications of nanotechnology. Here we investigate the adhesive wrapping of a soft elastic vesicle by a lipid membrane. We show that there exist a maximum of five distinct wrapping phases based on the stability of full wrapping, partial wrapping, and no wrapping states. The wrapping phases depend on the vesicle size, adhesion energy, surface tension of membrane, and bending rigidity ratio between vesicle and membrane. These results are of immediate interest to the study of vesicular transport and endocytosis or phagocytosis of elastic particles into cells.

Abstract:This paper aims at constructing a general mathematical model for the equilibrium theory of adhering lipid vesicles from a geometrical point of view. Based on the generalized potential functional, a few differential operators and their integral theorems on curved surfaces, the general normal and tangential equilibrium differential equations and boundary conditions are given at the first time for inhomogeneous lipid vesicles. A general boundary condition ψ˙=2(w−γ/R)/kc is first put forward including line tension. No assumptions are made either on the symmetry of the vesicle or on that of the substrate. The physical and biological meaning of the equilibrium differential equations and the boundary conditions are discussed. Numerical simulation results based on the Helfrich energy for adhering lipid vesicles under the axial symmetric condition show the effectiveness and convenience of the present theory.

Abstract:This paper aims at developing a stochastic-elastic model of a soft elastic body adhering on a wavy surface via a patch of molecular bonds. The elastic deformation of the system is modeled by using continuum contact mechanics, while the stochastic behavior of adhesive bonds is modeled by using Bell's type of exponential bond association/dissociation rates. It is found that for sufficiently small adhesion patch size or stress concentration index, the adhesion strength is insensitive to the wavelength but decreases with the amplitude of surface undulation, and that for large adhesion patch size or stress concentration index, there exist optimal values of the surface wavelength and amplitude for maximum adhesion strength.

Abstract:The dynamics of an adhesive two-dimensional vesicle doublet under various flow conditions is investigated numerically using a high-order, adaptive-in-time boundary integral method. In a quiescent flow, two nearby vesicles move slowly towards each other under the adhesive potential, pushing out fluid between them to form a vesicle doublet at equilibrium. A lubrication analysis on such draining of a thin film gives the dependencies of draining time on adhesion strength and separation distance that are in good agreement with numerical results. In a planar extensional flow we find a stable vesicle doublet forms only when two vesicles collide head-on around the stagnation point. In a microfluid trap where the stagnation of an extensional flow is dynamically placed in the middle of a vesicle doublet through an active control loop, novel dynamics of a vesicle doublet are observed. Numerical simulations show that there exists a critical extensional flow rate above which adhesive interaction is overcome by the diverging stream, thus providing a simple method to measure the adhesion strength between two vesicle membranes. In a planar shear flow, numerical simulations reveal that a vesicle doublet may form provided that the adhesion strength is sufficiently large at a given vesicle reduced area. Once a doublet is formed, its oscillatory dynamics is found to depend on the adhesion strength and their reduced area. Furthermore the effective shear viscosity of a dilute suspension of vesicle doublets is found to be a function of the reduced area. Results from these numerical studies and analysis shed light on the hydrodynamic and rheological consequences of adhesive interactions between vesicles in a viscous fluid.

Abstract:A theoretical model is developed to describe the influence of external loads on the equilibrium shapes of adhering tubular lipid vesicles. In the case of nonspecific adhesion, the equilibrium shape equations and boundary conditions are derived through the method analogous to previous research. For a range of applied forces, locally stable bound shapes are simulated. Along with the increase or decrease the external force, the process for adhering vesicle disengaging from or sprawling to the substrate is predicted and analyzed.
