Abstract:Selenoprotein W (SelW) is expressed in various tissues of many animals and acts as an oxidoreductase in mammals. However, little is known about the role of the SelW in birds. To investigate the role of the chicken SelW on H(2)O(2)-induced apoptosis in CHO-K1 cells, overexpression of a chicken SelW cell lines (CHO-K1/SelW) were constructed. Using acridine orange/ethidium bromide (AO/EB) double staining and terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling (TUNEL) assays, as well as WST-1 cell viability assay, we monitored the extent of the H(2)O(2)-induced apoptosis and detected the abundance of the caspase-3, caspase-8, and fas mRNA by real-time quantitative reverse transcription PCR (qPCR). We here found that overexpression of SelW cells, compared with the wild-type cells, resulted in a markedly decrease in sensitivity to H(2)O(2)-induced oxidative stress and had a lower apoptotic cell death in AO/EB and TUNEL assays. Cell viability revealed that overexpression of SelW cells had higher cell viability than wild-type cells. qPCR results found that overexpression of SelW cells had a lower levels of caspase-3, caspase-8, and fas mRNA than wild-type cells. Taken together, our findings suggested that SelW could reduce the oxidative damage induced by H(2)O(2) and had an important protective function in against oxidative damage.
Selw Protects Against H2o2-Induced Liver Injury In Chickens Via Inhibiting Inflammation And Apoptosis

Abstract:Selenium (Se) is recognized as a necessary trace mineral in animal diets. Se deficiency induces a number of diseases and injuries in chickens including liver damage, which is related to oxidative stress. Selenoprotein W (SelW) plays a crucial role in antioxidant defense mechanisms in mammals; however, little is known about the role of SelW and cytokines in H2O2-mediated chicken liver damage since chicken SelW has no cysteine (Cys) at residue 37 (Cys37) that is required for its presumed antioxidant function in mammals. The aim of this study was to evaluate whether chicken SelW possessed the same function as mammal SelW and to investigate the protective role of SelW against H2O2 by influencing the expression of inflammation and apoptosis via building a model of overexpressed or/and knocked down SelW in cultured chicken hepatic cells. The results showed that after the cells were exposed to H2O2, the levels of P53, caspase-3, Bax, Bak, COX-2, PTGEs, TNF-alpha, iNOS and NF-kappa B were lower in the SelW overexpressed cells but higher in the SelW knockdown cells compared to the H2O2-only treatment group. However, the result for Bcl-2 was opposite to that of the other apoptosis-promoting genes. Moreover, chickens were fed with a Se-deficient diet containing 0.032 mg kg(-1) Se for 15, 25, 35, 45, and 55 days, and their liver tissues were collected and examined for apoptosis and inflammation. The results showed that the levels of P53, Bax, Bak, COX-2, PTGEs, TNF-alpha, iNOS and NF-kappa B in the Se-deficient chicken livers were dramatically higher than the levels in the normal control group. In conclusion, our findings suggested that chicken SelW possess an antioxidant function similar to the mammalian homologs despite the lack of Cys37 in the peptide, and SelW regulated related-inflammation and apoptosis during H2O2-dependent chicken liver damage.

Abstract:The aim of the present study was to examine the role of Selenoprotein W (Sepw1) in modulating the expression of other selenoproteins. In the present study, we silenced and overexpressed the expression of Sepw1 in chicken myoblasts and subsequently treated the myoblasts with a reactive oxygen species (ROS) scavenger, N-acetyl-L-cysteine (NAC), and H2O2. Thereafter, the levels of expression of 25 selenoproteins and the activities of certain antioxidative enzymes, glutathione peroxidase (Gpx), superoxide dismutase (SOD), and catalase (CAT) were analyzed. In addition, principal component analysis (PCA) was used to define the most important parameters that could be used as key factors. The results indicated that as a highly expressed selenoprotein (only lower than Gpx1, Selk, Sels and Sep15), Sepw1 could interact with H2O2 (P < 0.05) and influence the expression of some selenoproteins (Gpx3, Gpx4, Txnrd1, Selt, Selh, Sepp1, Sels and Sep15, P < 0.05) and the sensitivity of the cells to H2O2. Both the overexpression and silencing of Sepw1 influenced the mRNA levels of selenoproteins. However, the responses of selenoproteins to altered Sepw1 expression were different. The results indicated that Sepw1 played a special role in H2O2 metabolism and may modulate the expression of certain selenoproteins through the redox pathway. Therefore, these results indicate that Sepw1 is an essential antioxidative selenoprotein in chicken myoblasts.

Abstract:In this study, a novel avian selenoprotein W ( SelW ) gene was cloned from chicken cerebral tissue. The complete nucleotide sequence of the gene contained a 258 bp open reading frame encoding 85 amino acids. Bioinformatics approaches identified the chicken SelW protein is characterized by a β-α-β-β-β-α secondary structure pattern, wherein β 1 and β 2 are parallel strands forming a classical β 1 -α 1 -β 2 motif, which is also observed in thioredoxin-like fold proteins. The protein has a candidate CXXU redox motif which is located in the loop (residues 10–13) between β 1 and α 1 . The 3D structural similarity between mouse and chicken SelW protein suggests that the proteins may exhibit similar functions. Additionally, a selenocysteine insertion sequence (SECIS) element was found in the 3′-untranslated region of the SelW mRNA. The SECIS element was classified as form II. Moreover, we analyzed the mRNA expression of SelW genes in 36 different tissues of 60-day-old chickens; the expression of SelW was detected in all tissues, indicating that SelW is expressed widely in chicken tissue. Hence, we suggest that SelW might play an important role in the biochemical functions of Se in birds.
Selenomethionine Attenuated H2O2-Induced Oxidative Stress and Apoptosis by Nrf2 in Chicken Liver Cells

Abstract:Earlier studies have shown that selenomethionine (SM) supplements in broiler breeders had higher deposition in eggs, further reduced the mortality of chicken embryos, and exerted a stronger antioxidant ability in offspring than sodium selenite (SS). Since previous studies also confirmed that Se deposition in eggs was positively correlated with maternal supplementation, this study aimed to directly investigate the antioxidant activities and underlying mechanisms of SS and SM on the chicken hepatocellular carcinoma cell line (LMH). The cytotoxicity results showed that the safe concentration of SM was up to 1000 ng/mL, while SS was 100 ng/mL. In Se treatments, both SS and SM significantly elevated mRNA stability and the protein synthesis rate of glutathione peroxidase (GPx) and thioredoxin reductase (TrxR), two Se-containing antioxidant enzymes. Furthermore, SM exerted protective effects in the H2O2-induced oxidant stress model by reducing free radicals (including ROS, MDA, and NO) and elevating the activities of antioxidative enzymes, which performed better than SS. Furthermore, the results showed that cotreatment with SM significantly induced apoptosis induced by H2O2 on elevating the content of Bcl-2 and decreasing caspase-3. Moreover, investigations of the mechanism revealed that SM might exert antioxidant effects on H2O2-induced LMHs by activating the Nrf2 pathway and enhancing the activities of major antioxidant selenoenzymes downstream. These findings provide evidence for the effectiveness of SM on ameliorating H2O2-induced oxidative impairment and suggest SM has the potential to be used in the prevention or adjuvant treatment of oxidative-related impairment in poultry feeds.

Abstract:Selenium (Se) mainly performs its function through Se-containing proteins. Selenoprotein W (SelW), one member of the selenoprotein family, plays important roles in the normal function of the heart. To investigate the possible relationship between Se and SelW for the regulation of oxidative damage in chicken embryo myocardial cells, we treated myocardial cells with Se and H 2 O 2 . Then, the levels of lactate dehydrogenase (LDH) and 3,4-methylenedioxyamphetamine in the culture media, levels of SelW, inflammatory genes NF-κB, tumor necrosis factor (TNF)-α, p53, and the cell cycle were analyzed. Furthermore, the correlation between SelW and the levels of these factors was determined. The results indicated that Se treatment increased the expression of SelW ( P < 0.05) and caused a downregulation of p53, NF-κB, and TNF-α ( P < 0.05). In contrast, H 2 O 2 increased the expression of p53, NF-κB, TNF-α, and LDH ( P < 0.05) and induced early cell apoptosis, which was alleviated by treatment with Se. In addition, SelW had a positive correlation with the levels of inflammatory genes investigated. Taken together, our findings suggested that SelW is sensitive to Se levels and oxidative stress, and may play a role in the protective function of Se against oxidative damage and inflammation in chicken myocardial cells.
Antioxidative Role of Selenoprotein W in Oxidant-Induced Chicken Splenic Lymphocyte Death

Abstract:To verify the antioxidative role of SelW in oxidant-induced chicken splenic lymphocyte, in this report, the influence of selenite supplementation and SelW gene silence on H 2 O 2 -mediated cell viability and cell apoptosis in cultured splenic lymphocyte derived from spleen of chicken were examined. The cultured cells were treated with sodium selenite and H 2 O 2 , or knocked down SelW with small interfering RNAs (siRNAs). The lymphocytes were examined for cell viability, cell apoptosis and mRNA expression levels of SelW and apoptosis-related genes (Bcl-2, Bax, Bak-1, caspase-3 and p53). The results show that the mRNA expression of SelW were effectively increased after treatment with sodium selenite, and H 2 O 2 -induced cell apoptosis was significantly decreased and cell viability was significantly increased. 20 μM H 2 O 2 was found to induce cell apoptosis and decrease cell viability, which was alleviated obviously when cells were pretreated with sodium selenite before exposure to 20 μM H 2 O 2 . Meanwhile, H 2 O 2 induced a significantly up-regulation of the Bax/Bcl-2 ratio, Bax, Bak-1, caspase-3 and p53 and down-regulation of Bcl-2 ( P < 0.05). When lymphocytes were pretreated with Se before treated with H 2 O 2 , the Bax/Bcl-2 ratio and mRNA expression of those genes were significantly decreased, and Bcl-2 was increased ( P < 0.05). SelW siRNA-transfected cells were more sensitive to the oxidative stress induced by treatment of H 2 O 2 than control cells. Silencing of the lymphocyte SelW gene decreased their cell viability, and increased their apoptosis rate and susceptibility to H 2 O 2 . Silencing of SelW significantly up-regulated the Bax/Bcl-2 ratio, Bax, Bak-1, caspase-3 and p53 and down-regulated Bcl-2 ( P < 0.05). The present study demonstrates that SelW plays an important role in protection of splenic lymphocyte of birds from oxidative stress.
Selenoprotein W Depletion in Vitro Might Indicate That Its Main Function is Not As an Antioxidative Enzyme

Abstract:Examination of the antioxidative homeostasis in skeletal muscle cells in the presence or absence of selenoprotein W (SelW) is necessary to understand the importance of SelW in the antioxidative system. Depletion of SelW by RNA interference was achieved by introducing a synthetic small interfering RNA into the mouse skeletal muscle cell line C2C12 (C3H). Transfectant screening was performed by real-time reverse transcription-PCR, Western blotting, flow cytometry, fluorescence staining, cell viability, and glutathione assays. SelW expression and mRNA levels were downregulated by 62.1 and 72.4%, respectively. In addition, acute cytotoxicity and an apoptosis rate of ∼36% in SelW-depleted cells demonstrated that RNA interference was successful. As compared with non-SelW-depleted cells, the enzyme activities of glutathione peroxidase, superoxide dismutase, and catalase and total antioxidative capability and glutathione level increased by 47.6, 103.0, 31.0, 205.6, and 30.0%, respectively (P < 0.05). Thus, SelW is important for the antioxidative system of muscle cells. Depletion of SelW, however, could be compensated by other intracellular antioxidative enzymes because oxidative stress was not the causative factor for apoptosis in SelW-depleted cells. Thus, the main function of SelW in muscle cells is not in the antioxidative system.

Abstract:The 15-kDa selenoprotein (Sep15) is a thioredoxin-like protein. The expression of Sep15 is regulated by dietary selenium (Se) and plays important roles in mammals. However, the structure and function of chicken Sep15 and its response to Se are still unclear. In the present study, we replicated the chicken Se deficiency models and Sep15 deficiency models in splenocytes. Then, the homology, structure analysis, and levels of Sep15 were analyzed. In addition, the oxidative stress levels were examined in Sep15 deficiency splenocytes. The results indicated that chicken Sep15 preserved high similarity with that of other 14 animals in the coding nucleotide sequences (CDS) and deduced amino acid sequence, which suggested that chicken Sep15 may be derived from the same ancestor with other animals. The predicted structure and function showed that chicken Sep15 preserved the conserved thioredoxin-like fold CxU, which suggested an antioxidative function. Chicken Sep15 was also decreased by Se deficiency in immune organs (P < 0.05). In addition, Sep15 deficiency induced the occurrence of higher oxidative stress and enhanced the sensitivity of cells to H2O2 (P < 0.05). So the in vitro study further verified its antioxidative function. Thus, similar to its mammal homolog, chicken Sep15 preserves the typical characteristic of selenoprotein and may play some roles in the redox regulation.

Abstract:Selenoprotein N (SEPN1) is critical to the normal muscular physiology. Mutation of SEPN1 can raise congenital muscular disorder in human. It is also central to maturation and structure of skeletal muscle in chicken. However, human SEPN1 contained an EF-hand motif, which was not found in chicken. And the biochemical and molecular characterization of chicken SEPN1 remains unclear. Hence, protein domains, transcription factors, and interactions of Ca2+ in SEPN1 were analyzed in silico to provide the divergence and homology between chicken and human in this work. The results showed that vertebrates' SEPN1 evolved from a common ancestor. Human and chicken's SEPN1 shared a conserved CUGS-helix domain with function in antioxidant protection. SEPN1 might be a downstream target of JNK pathway, and it could respond to multiple stresses. Human's SEPN1 might not combine with Ca2+ with a single EF-hand motif in calcium homeostasis, and chicken SEPN1 did not have the EF-hand motif in the prediction, indicating the EF-hand motif malfunctioned in chicken SEPN1.

Abstract:Selenoprotein W (SelW) has been found to be ubiquitously expressed in tissues in vivo and was purified more than 18 years ago. However, little in vitro research has been performed on SelW from birds. To detect the mRNA levels of chicken SelW in cultured cell lines, chicken SelW cDNA was cloned into an expression vector. The chicken SelW expression construct was then transfected into CHO-K1 cells. Using RT-PCR and real-time quantitative reverse transcription PCR, we detected the expression of the chicken SelW mRNA. Moreover, the selenocysteine-synthase (SecS) and selenophosphate synthetase-1 (SPS-1) mRNA levels were analyzed. The expression of SelW was detected in SelW-transfected cells; no expression was observed in control cells. Significant increases in the SelW mRNA levels were obtained in chicken SelW-transfected cells relative to control cells. SecS mRNA levels were significantly increased in chicken SelW transfected cells. No significant difference in the SPS-1 level was observed. Our findings show that chicken SelW could be studied in vitro and that SecS and SPS-1 may have potential roles in SelW biosynthesis.
Characterization of Selenoprotein M and Its Response to Selenium Deficiency in Chicken Brain

Abstract:Selenoprotein M (SelM) may function as thiol disulfide oxidoreductase that participates in the formation of disulfide bonds and can be implicated in calcium responses. SelM may have a functional role in catalyzing free radicals and has been associated with Alzheimer's disease (AD). However, studies of SelM in chicken remain very limited. In this study, two groups of day-old broiler chicks (n = 40/group) were fed a corn-soy basal diet (BD, 13 μg Se/kg) and BD supplemented with Se (as sodium selenite) at 0.3 mg/kg. The brain was collected at 14, 21, 28, and 42 days of age. We performed a sequence analysis and predicted the structure and function of SelM. We also investigated the effects of Se deficiency on the expression of Selt, Selw, and Selm and the Se status in the chicken brain. The results show that Se deficiency induced the lower (P < 0.05) Se content, glutathione peroxidase (GPx), and catalase (CAT) activities; increased (P < 0.05) malondialdehyde (MDA) content; and reduced (P < 0.05) the expression of Selm messenger RNA (mRNA) and protein abundance of SelM in the brain. However, there were no significant brain Selt and Selw mRNA levels by dietary Se deficiency in chicks. The different regulations of these three redox (Rdx) protein expressions by Se deficiency represent a novel finding of the present study. Our results demonstrated that SelM may have an important role in protecting against oxidative damage in the brain of chicken, which might shed light on the role of SelM in human neurodegenerative disease. More studies are needed to confirm our conclusion.

Abstract:Selenoprotein W (SelW) is abundantly expressed in skeletal muscles of mammals and necessary for the metabolism of skeletal muscles. However, its expression pattern in skeletal muscle system of birds is still uncovered. Herein, to investigate the distribution of SelW mRNA in chicken skeletal muscle system and its response to different selenium (Se) status, 1-day-old chickens were exposed to various concentrations of Se as sodium selenite in the feed for 35 days. In addition, myoblasts were treated with different concentrations of Se in the medium for 72 h. Then the levels of SelW mRNA in skeletal muscles (wing muscle, pectoral muscle, thigh muscle) and myoblasts were determined on days 1, 15, 25, and 35 and at 0, 24, 48, and 72 h, respectively. The results showed that SelW was detected in all these muscle components and it increased both along with the growth of organism and the differentiation process of myoblasts. The thigh muscle is more responsive to Se intake than the other two skeletal muscle tissues while the optimal Se supplementation for SelW mRNA expression in chicken myoblasts was 10 −7 M. In summary, Se plays important roles in the development of chicken skeletal muscles. To effect optimal SelW gene expression, Se must be provided in the diet and the media in adequate amounts and neither at excessive nor deficient levels.

Abstract:Selenoprotein W (SelW) is mainly understood in terms of its antioxidant effects in the cellular defense system. Inflammation is an important indicator of animal tissue injury, and the inflammatory cells may trigger a sophisticated and well-orchestrated inflammatory cascade, resulting in exaggerated oxidative stress. To investigate the role of SelW in inflammatory injury in chicken immune tissues and cultured splenic lymphocyte, in this report, the effects of selenium (Se) on mRNA expressions of SelW and inflammatory factors (iNOS, COX-2, NF-κB, PTGEs, and TNF-α) in the chicken immune organs (spleen, thymus and bursa of Fabricius) and cultured splenic lymphocyte treated with sodium selenite and H 2 O 2 , or knocked down SelW with small interfering RNAs (siRNAs) were examined. The results showed that Se-deficient diets effectively decreased the mRNA expression of SelW ( P < 0.05), and induced a significantly up-regulation of COX-2, iNOS, NF-κB, PTGEs and TNF-α mRNA levels ( P < 0.05). The histopathological analysis showed that immune tissues were obviously injured in the low-Se groups. In vitro, H 2 O 2 induced a significantly up-regulation of the mRNA levels of inflammation-related genes (iNOS, COX-2, NF-κB, PTGEs, and TNF-α) in cultured splenic lymphocyte ( P < 0.05). When lymphocytes were pretreated with Se before treated with H 2 O 2 , the inflammation-related genes were significantly decreased ( P < 0.05). Silencing of SelW significantly up-regulated the inflammation-related genes (iNOS, COX-2, NF-κB, PTGEs, and TNF-α) in cultured splenic lymphocyte ( P < 0.05). The results suggested that the expression levels of inflammatory factors (iNOS, COX-2, NF-κB, PTGEs, and TNF-α) and SelW can be influenced by Se in birds. SelW commonly played an important role in the protection of immune organs of birds from inflammatory injury by the regulations of inflammation-related genes.
Possible Correlation of Selenoprotein W with Inflammation Factors in Chicken Skeletal Muscles

Abstract:The aim of the present study was to investigate the possible correlation of selenoprotein W (SelW) with inflammatory injury induced by dietary selenium (Se) deficiency in chicken. One-day-old male chickens were fed either a commercial diet or a Se-deficient diet for 55 days. Then, the expression levels of SelW messenger RNA (mRNA) and inflammation-related genes (NF-κB, TNF-α, iNOS, COX-2, and PTGES) in chicken skeletal muscles (wing muscle, pectoral muscle, and thigh muscle) were determined at 15, 25, 35, 45, and 55 days old, respectively. In addition, the correlation between SelW mRNA expression and inflammation-related genes were assessed. The results showed that dietary Se deficiency reduced the mRNA expression of SelW in chicken wing, pectorals, and thigh muscles. In contrast, Se deficiency increased the mRNA expression levels of inflammation-related genes in chicken skeletal muscle tissues at different time points. The Pearson’s correlation coefficients showed that the mRNA expression levels of inflammation-related genes were significantly negative related to SelW ( p < 0.05). These data showed that Se deficiency induced the inflammatory response in chicken skeletal muscle. As one important selenoprotein gene in skeletal muscles, SelW may play a role in the regulation of inflammation reaction in Se-deficiency myopathy.

Abstract:Selenoprotein U (SelU) may regulate a myriad of biological processes through its redox function. In chicks, neither the nucleotide sequence nor the amino acid sequence is known. The main objectives of this study were to clone and characterize the chicken Selu gene and investigate Selu messenger RNA (mRNA) and protein expression in chicken tissues. The coding sequence (CDS) of Selu contained 387 bases with a typical mammalian selenocysteine insertion sequence (SECIS) located in the 3′-untranslated region. The deduced amino acid sequence of chicken SelU contains 224 amino acids with UAA as the stop codon. Like all SelU genes identified in different species, chicken SelU contains one well-conserved selenocysteine (Sec) at the 85th position encoded by the UGA codon. The SECIS element was with the conserved denosine (--AAA--) rather than the motif cytidine (--CC--) motif. Moreover, the expression pattern of Selu mRNA in muscle, liver, kidney, heart, spleen, lung, testis, and brain was analyzed with real-time quantitative PCR in young male chickens fed a Se-deficient corn-soybean meal basal diet supplemented with 0.0 and 0.3 mg Se/kg in the form of sodium selenite. We found that the abundance of Selu mRNA in muscle, liver, kidney, heart, spleen, and lung was downregulated (P < 0.05) by Se deficiency. However, it was not affected by dietary Se concentrations in testis and brain. Furthermore, protein abundance of SelU in these seven tissues was consistent with the mRNA abundance. Hence, we suggest that Selu might play an important role in the biochemical function of Se in birds.
Probing Selenium-Deficient Chicken Spleen Th1/Th17 Differentiation Based on Selenoprotein W Targeting of PKM2/HIF1α

Abstract:Selenium regulates the differentiation and function of immune cells mainly through selenoproteins. Selenoprotein W (SelW) has been shown to mitigate inflammatory bowel disease in mice by modulating the differentiation of helper T (CD4+ T) cell. Previous studies by our team have underscored SelW's critical role in safeguarding chicken spleens and splenic lymphocytes against inflammatory injury. However, research examining SelW's involvement in regulating CD4+ T cell differentiation in avian spleens remains scarce. Therefore, the selenium-deficient chicken model was constructed in this study. It was found that the spleen of selenium-deficient chickens showed significant inflammatory damage, accompanied by decreased SelW expression, diminished antioxidant capacity, heightened glycolysis, and an elevated count of Th1/Th17 cells. To elucidate the specific mechanism of SelW regulating Th1/Th17 cell differentiation, this study used molecular docking technology, fluorescence colocalization, and co-immunoprecipitation and initially confirmed the targeting relationship between SelW and pyruvate kinase M2 (PKM2). Subsequently, an in vitro model of SelW overexpression, knockdown, and TEPP-46 (PKM2 tetramer activator) cotreatment of chicken primary splenic lymphocytes was replicated. Our findings revealed that selenium deficiency triggers oxidative stress and promotes PKM2 nuclear translocation via SelW downregulation, which stabilizes HIF1α transcription in the nucleus, enhancing glycolysis and skewing chicken splenic CD4+ T cells toward the Th1/Th17 phenotype. Our study, for the first time, demonstrates the existence of an interaction between SelW and PKM2 in poultry, emphasizing SelW's paramount significance in CD4+ T cell differentiation, providing fresh perspectives on the contributions of selenoproteins to T cell biology and immune processes.

Abstract:Selenoprotein S (SelS) is a new endoplasmic reticulum (ER) and plasma membrane resident selenoprotein. Previous studies reveal that SelS protects cells from oxidative damage and endoplasmic reticulum stress-induced apoptosis. It participates in the retro-translocation of misfolded proteins from the ER lumen to cytosol for degradation (ER-associated protein degradation). SelS is involved in spermatogenesis, lipoprotein metabolism and the regulation of inflammatory response. Polymorphism of SelS is closely associated with chronic inflammatory diseases such as diabetes, coronary heart disease and preeclampsia. This paper reviews the recent research progress on SelS, especially its biological functions, with authors's work about the role of SelS in inflammatory response. Finally, some issues to be addressed and potential orientations to be further studied are also put forward and prospected.

Abstract:Selenoprotein W (SelW) is a selenocysteine containing protein with redox activity involved in the antioxidant response. In this study, a selenoprotein W was cloned from pearl mussel Cristaria plicata (designated as CpSelW), and the expression patterns were characterized in tissues after Aeromonas hydrophila challenged. The full-length cDNA of cpSelW was of 858bp, containing a 5′ untranslated region (UTR) of 145bp, a 3′ UTR of 455bp with a poly (A) tail, and an open reading frame (ORF) of 258bp encoding a polypeptide of 85 amino acids with the predicted molecular mass of 9.277kDa, which shared 61% identity with SelW from Gallus gallus. A tertiary structure model generated for the CpSelW displayed a β-α-β-β-β-α secondary structure pattern, which was similar to mouse SelW protein 3D structure. The mRNA of CpSelW was constitutively expressed in tested tissues of healthy mussel, including mantle, gill, hemocytes, muscle, and hepatopancreas, and it was highly expressed in hepatopancreas. After mussels were stimulated by A. hydrophila, the mRNA expression of CpSelW in hemocytes at 6, 12 and 24h, in gill at 12h and in hepatopancreas at 24h was significantly down-regulated.
