Abstract:Four petroleum-contaminated soil samples at different degree were collected and used for isolation of petroleum-degrading bacterium. 28 petroleum-degrading strains were screened and identified as Brevibacterium, Pseudomonas, Micrococcus and Plesiomonas, respectively. There are more petroleum-degrading bacteria in heavy petroleum-contaminated soil than that in light contaminated soil, and Brevibacterium and Pseudomonas are dominant petroleum-degrading bacteria. A culture-independent molecular biological method was applied to analyze the genetic diversity of microbial communities of the four soil samples. Application of Denaturing Gradient Gel Electrophoresis (DGGE) in this study had provided new ideas for study of the microbial community in petroleum- contaminated soil. The genomic DNA of microbial community were extracted from the four soil samples in chemical method, which is based on lysis with a high salt extract ion buffer (1.5mol/L NaCl) and extend heating (2 to 3 h) of the soil suspension in the presence of sodium dodecyl sulfate (SDS), CTAB and proteinase K. After purification of the genomic DNA with DNA gel recovery kits, the V3 variable fragments of genes coding for 16S rRNA were amplified by using specific primers. The result of agarose gel(1.5%) electrophoresis showed that the PCR products were about 230 bp. The amplified DNA fragments were then separated by paralleled DGGE with the denaturant (urea and formamide) from 30% to 60% to generate DNA fingerprints, nine major bands from DGGE profiles of the four samples were eluted from gel, then reamplified and sequenced. The results suggested that they belonged to 2 OTUs at 97% homo logy cutoff, 4 OTUs shared 97%-100% homology with known sequences in the database, while 3 OTUs had 91%-95% homology with known sequences. The isolates from 8 OTUs grouped together with some uncultured eubacteria in the database. A phylogenetic analysis of 9 OTUs classified them into three major groups: the OTU2 lineage and the other 8 OTUs lineage, the latter was classified further into 6 subgroups. The results indicated the diversity of soil microbes was significantly correlated with the degree of petroleum contamination. Owing to the selective pressure of petroleum pollutants, there was less diversity of microbes in heavy contaminated soil than that in light contaminated soil. Sequences alignment results showed Mycobacterium and Bacillus bacteria are dominant population in light contaminated soil and Flavobacterium and Azospira are dominant population in heavy contaminated soil. The results of GC-MS indicated the degree of contamination, especially the content of aromatic hydrocarbons affect the diversity of soil microbes. The petroleum-degrading strains isolated in this study will be good candidates for bioremediation of petroleum-contaminated soils, and the analysis of the microbe diversity in petroleum-contaminated soils will offer a valid scientific basis for better bioremediation of petroleum-contaminated soil.
