Abstract:A full-length cDNA, BcMF11, has been cloned from Chinese cabbage (Brassica campestris L. ssp. chinensis Makino) using rapid amplification of the cDNA ends (RACE) based on a pollen-specific cDNA fragment (DN237921). The BcMF11 cDNA has a total length of 828bp with poly (A) tail. Analysis of the sequence demonstrated that BcMF11 is a novel non-coding RNA which has no prominent open reading frame (ORF) or coding capacity. No significant similarities were observed between BcMF11 and previously published sequences in GenBank. Transcription analysis indicated that BcMF11 is a novel pollen-specific ncRNA and may be involved in the pollen development of Chinese cabbage.

Abstract:A transcript-derived fragment (GenBank accession number DN237920.1) accumulated in the wild-type flower buds of Chinese cabbage (Brassica campestris L. ssp. chinensis Makino, syn. B. rapa ssp. chinensis) was isolated and further investigated. The full length DNA and cDNA of the fragment were cloned by rapid amplification of cDNA ends. The gene, BcMF13, encodes a protein of 73 amino acids and is interrupted by an intron of 106 bp (GenBank accession number EF158459). Southern blot analysis revealed that BcMF13 could be a single-copy gene in the Chinese cabbage genome. Sequence blast analysis showed that BcMF13 was a new gene. In EST database, those sequences share 96-98% identity with BcMF13 cDNA all came from flower buds, microspores, anthers of Brassica, which proved BcMF13 homologs closely related to the development of male gametogenesis in Brassica. RT-PCR discovered that it is exclusively expressed in stage four and five flower buds of fertile line, strongly expressed in stamens. Successful suppression of BcMF13 gene expression by RNA antisense strategy greatly reduced the normal pollen grains, suggesting that BcMF13 was essential in pollen development in Brassica.

Abstract:The BcMF22 (Brassica campestris Male Fertility 22) gene was isolated from Chinese cabbage-pak-choi (B. campestris L. ssp. chinensis Makino, syn. B. rapa ssp. chinensis). The cDNA clone of BcMF22 was 1,870 bp in length and contained an open reading frame (OFF) of 1,260 bp, while the genomic sequence contained no introns. Sequence prediction indicated that BcMF22 might encode a methytransferase. Spatial and temporal expression analyses indicated BcMF22 was preferentially expressed in pollen. Transcripts of BcMF22 were first detected in pollen mother cells and continued to be present in mature pollen. In addition, transcripts were detected in pollinated pistils, approximately 4 h after pollination (4HAP). These results indicated that BcMF22 might be a pollen-preferential gene and was closely involved in pollination.
BcMF21 is important for pollen development and germination in Brassica campestris ssp. chinensis

Abstract:Brassica campestris Male Fertility 21 ( BcMF21 ) was previously isolated from the flower buds of Chinese cabbage ( Brassica campestris L. ssp. chinensis Makino, syn. B. rapa ssp. chinensis ) and expressed specifically in tapetum and microspores during the meiosis stage and the uninucleate stage of microspore development. Here, we used antisense RNA technology to knock down the expression level of BcMF21 in B. campestris and analyzed the phenotype of the transgenic plants. Alexander staining and scanning electron microscope revealed sterility and exine deformities in the mature pollen grains of BcMF21 antisense RNA transgenic plants. The germination furrow of the BcMF21 antisense RNA transgenic pollen was covered by lipid like materials. The pollen tubes burst and could not grow normally in vitro. Therefore, we presented here BcMF21 might be an important gene for pollen development and germination.
Characterisation of BcMF10, a Novel Gene Involved in Pollen Wall Development of Brassica Rapa Ssp. Chinensis

Abstract:Comparative expression profiling of flower buds in two male sterile lines [genic male sterile (GMS) and cytoplasmic male sterile (CMS)] with its male fertile maintainer line in Chinese cabbage pak-choi was performed using cDNA-AFLP technology to identify the genes implicated in male sterility. A novel gene BcMF10, sharing high sequence similarity to the function-unknown DUF1216 family in Arabidopsis was isolated, whose expression was absent in the flower buds of the GMS and CMS lines but present in the male fertile maintainer line. Temporal and spatial expression pattern analysis revealed that BcMF10 began to be expressed in tapetal cells and microspores during meiosis. Expression in tapetal cells was persistent until the degeneration of tapetum, and expression in microspores reached a peak during the tetrad stage but gradually declined as development proceeded. RNA interference technology was used to address the biological function of BcMF10. The RNAi transgenic Chinese cabbage pak-choi lines showed normal vegetative growth and reproductive development, but poor pollen germination. Scanning electron microscopy (SEM) showed that most of the transgenic pollen was deformed and exhibited an irregular shape with an abnormal number and distribution of germinal furrows. It is speculated that BcMF10 may encode a protein that plays a role in the formation of intine wall.
The Zinc-Finger Transcription Factor BcMF20 and Its Orthologs in Cruciferae Which Are Required for Pollen Development.

Abstract:Brassica campestris Male Fertility 20 (BcMF20) is a typical zinc-finger transcription factor that was previously isolated from flower buds of Chinese cabbage (Brassica campestris ssp. chinensis). By applying expression pattern analysis, it can be known that BcMF20 was specifically and strongly expressed in tapetum and pollen, beginning from the uninucleate stage, and was maintained during the mature-pollen stage. As BcMF20 was highly conserved in Cruciferae, it can be indicated that this zinc-finger transcription factor is important during the growth of Cruciferae. In this study, 12 C2H2-type zinc-finger TFs which shared high homology with BcMF20 were found from NCBI via BLAST. A new molecular phylogenetic tree was constructed by the comparison between BcMF20 and these 12 C2H2-type zinc-finger TFs with NJ method. By analyzing this phylogenetic tree, the evolution of BcMF20 was discussed. Then, antisense RNA technology was applied in the transgenesis of Arabidopsis thaliana to get the deletion mutants of BcMF20, so that its function during the pollen development can be identified. The results showed: BcMF20 are in the same clade with three genes from Arabidopsis. The inhibition of BcMF20 expression led to smaller amounts of and lower rate in germination of pollen and lower rate in fruit setting in certain transgenetic plants. This also led to the complete collapse of pollen grains. By SEM and TEM, pollen morphology and anther development processes were observed. In the middle uninucleate microspore stage, a relatively thin or even no primexine was formed in microspores. This may result in the malformation of the pollen wall and finally cause the deformity of pollens. Above all, it can be indicated that BcMF20 may act as a part of regulation mechanisms of TAZ1 and MS1. Together they play a role in a genetic pathway in the tapetum to act on proliferation of tapetal cells and keep the normal development of pollens.

Abstract:In this report a full length cDNA, Brassica campestris Male Fertile 21 (BcMF21) was successfully isolated from one of the cDNA-amplified fragment length polymorphism (cDNA-AFLP) transcript-derived fragments (TDFs), BBP10, that was found down-regulated in the flower buds of sterile plants in Brassica campestris L. ssp. chinensis Makino genic male sterile (GMS) AB line system (Bcajh 97-01A/B). BcMF21 protein structure analysis showed a signal peptide at the N-terminus; two protein kinase C phosphorylation sites, five N-myristoylation sites and one casein kinase II phosphorylation site. The promoter region of BcMF21, a 779 bp upstream of ATG was isolated by thermal asymmetric interlaced-PCR (TAIL-PCR). Bioinformatics analysis revealed that the promoter of BcMF21 contained several classical cis-acting elements and three pollen specific elements. Transient expression analysis showed that the promoter could drive green fluorescence protein (GFP) expression. Quantitative reverse transcript-PCR analysis revealed that BcMF21 was specifically expressed in flower buds. The transcript level of BcMF21 was much lower in the sterile flower buds than in the fertile flower buds in 'Bcajh 97-01A/B' system. In situ hybridization further showed that BcMF21 was only expressed in the tetrads and the microspores at the tetrad stage and the uninucleate stage. In addition, phylogenetic analysis demonstrated that the BcMF21 was relative conserved within family Crucifereae and might be originated from the ancestor diploid B. campestris within genus Brassica according to the Triangle of U theory.
Isolation and Characterization of a Novel BcMF14 Gene from Brassica Campestris Ssp. Chinensis

Abstract:A putative RALF (rapid alkalinization factor)-like gene (GenBank accession number EF523517), named BcMF14, was isolated from Chinese cabbage (Brassica campestris L. ssp. chinensis Makino, syn. B. rapa ssp. chinensis) by rapid amplification of cDNA ends (RACE) based on a cDNA-AFLP differential fragment exclusively expressed in fertile line. Semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) discovered that BcMF14 was prominently expressed in stage four and five flower buds of fertile line, no expression in vegetative structures or in sterility line. Detailed RT-PCR illuminated its strong expression in stamens. Successful suppression of BcMF14 gene expression greatly reduced the normal pollen grains. The frequency of abnormal pollen grains was 48.95% in the mutant with many shriveled pollen grains with irregular shape and some larger ones with deep hollows along the germination ditch. Pollen germination was stopped because of the severely twisted pollen tubes. These results demonstrate a potential role of the BcMF14 gene in the development of male gametogenesis in Chinese cabbage.
Characterization of a Novel Gene, BcMF7 , That is Expressed Preferentially in Pollen of Brassica Campestris L. Ssp. Chinensis Makino

Abstract:Pollen formation is important for plant sexual reproduction. To identify the genes that are involved in pollen formation, we performed the genome-wide transcriptional profiling in the flower buds of both male meiotic cytokinesis (mmc) mutant and its wild-type plants of Brassica campestris L. ssp. chinensis , syn. B. rapa L. ssp. chinensis . cDNA-amplified fragment length polymorphism (cDNA-AFLP) analysis showed that the mmc mutation resulted in changes in expression of a variety of genes. BcMF7 , a transcript-derived fragment (TDF) accumulated in the wild-type flower buds was further characterized. The BcMF7 gene has 1161 bp in length with two introns. The full-length BcMF7 cDNA has 609 bp in length and encodes a protein of 129 amino acids. The deduced amino acid sequence of BcMF7 protein shares no similarity to any function-known protein in Swiss-Prot database, but has 8 protein kinase C phosphorylation sites, 2 caselin kinase II phosphorylation sites, 2 tyrosine kinase phosphorylation sites, 2 N -glycosylation sites and 2 N -myristolyation sites. Spatial and temporal expression patterns analysis showed that BcMF7 was expressed exclusively in pollen. The expression signal of BcMF7 was first detected at the tetrad stage of microspore development, reached a peak level at the uninucleate stage, and decreased to a slightly low level at the mature pollen stage. All these results show that BcMF7 may play a certain role in the signal transduction during pollen development.

Abstract:The Arabidopsis transcription factor (At1g26610) was shown to be down-regulated in male sterile lines compared to their maintainer line in Chinese cabbage (Brassica campestris ssp. chinensis) in our previous study. The BcMF20 gene, homologue of the At1g26610 gene was isolated from Chinese cabbage. It encodes a putative C2H2 zinc finger protein. The spatial and temporal expression patterns examined by qRT-PCR and in situ hybridization, indicated BcMF20 is specifically expressed in the developing pollen grains and the tapetum from the uninucleate pollen stage to mature pollen stage.

Abstract:A novel membrane protein gene, BcMF12, was isolated from Chinese cabbage (Brassica campestris L. ssp. chinensis Makino) using rapid amplification of the cDNA ends based on a pollen-specific cDNA fragment (DN237936). The cDNA was 1,155 bp in length with an open reading frame of 894 bp capable of encoding a putative polypeptide of 297 amino acids with an estimated molecular mass of 34.6 kDa and a predicted isoelectric point of 9.6. Comparative and bioinformatics analyses revealed that BcMF12 showed high similarities with some membrane protein sequences previously published in the public database and contained six highly conserved transmembrane domains corresponding to six highly hydrophobic regions. This indicates that BcMF12 may be a putative membrane protein. RNA gel blot analysis indicated that the transcripts of BcMF12 were abundant in the flower bud, flower and anther, but not detected in the root, stem, leaf and pistil. Moreover, the BcMF12 transcripts were detectable at the late stages of pollen development. Morphological investigations of pollen from the BcMF12 antisense transgenic plants showed that most of pollen grains of transgenic plants were abnormal. These results strongly suggest that BcMF12 is a novel pollen-preferentially membrane protein which play an important role during the pollen development in Chinese cabbage.
Morphological and Functional Characterization of BcMF13 in the Antisense-Silenced Plants of Brassica Campestris Ssp. Chinensis Var. Parachinensis

Abstract:The gene Brassica campestris male fertility 13 (BcMF13, GenBank accession number EF158459) was isolated as a reproductive organ-specific gene from Chinese cabbage (Brassica campestris L. ssp. chinensis Makino, syn. B. rapa ssp. chinensis). It is exclusively expressed in stage four and five flower buds of fertile lines and is most strongly expressed in stamens. Here, we report a functional characterization of this BcMF13 gene in the antisense-silenced plants. The inflorescence of the BcMF13 mutant was compacted with anthers curved outside. The fertility of this mutant was greatly reduced with less than 5 seeds per silique. Under scanning electron microscopy, the mutant demonstrated numerous shriveled pollen grains with deep invaginations. The frequency of normal pollen grains was just 45.34%. The pollen mother cell, the tetrad, and the mature pollen of the BcMF13 mutant were abnormal resulting in the poor pollen vitality. Germination test in vivo suggested BcMF13 delayed the pollen tubes' extension in the style. All these indicated BcMF13 had a vital role in pollen development of Chinese cabbage.
Bcmf8, A Putative Arabinogalactan Protein-Encoding Gene, Contributes to Pollen Wall Development, Aperture Formation and Pollen Tube Growth in Brassica Campestris

Abstract:BACKGROUND AND AIMS:The arabinogalactan protein (AGP) gene family is involved in plant reproduction. However, little is known about the function of individual AGP genes in pollen development and pollen tube growth. In this study, Brassica campestris male fertility 8 (BcMF8), a putative AGP-encoding gene previously found to be pollen specific in Chinese cabbage (B. campestris ssp. chinensis), was investigated.METHODS:Real-time reverse transcription-PCR and in situ hybridization were used to analyse the expression pattern of BcMF8 in pistils. Prokaryotic expression and western blots were used to ensure that BcMF8 could encode a protein. Antisense RNA technology was applied to silence gene expression, and morphological and cytological approaches (e.g. scanning electron microscopy and transmission electron microscopy) were used to reveal abnormal phenotypes caused by gene silencing.KEY RESULTS:The BcMF8 gene encoded a putative AGP protein that was located in the cell wall, and was expressed in pollen grains and pollen tubes. The functional interruption of BcMF8 by antisense RNA technology resulted in slipper-shaped and bilaterally sunken pollen with abnormal intine development and aperture formation. The inhibition of BcMF8 led to a decrease in the percentage of in vitro pollen germination. In pollen that did germinate, the pollen tubes were unstable, abnormally shaped and burst more frequently relative to controls, which corresponded to an in vivo arrest of pollen germination at the stigma surface and retarded pollen tube growth in the stylar transmitting tissues.CONCLUSIONS:The phenotypic defects of antisense BcMF8 RNA lines (bcmf8) suggest a crucial function of BcMF8 in modulating the physical nature of the pollen wall and in helping in maintaining the integrity of the pollen tube wall matrix.

Abstract:The BcMF8 (Brassica campestris male fertility 8) gene, possessing the features of 'classical' arabinogalactan protein (AGP) was isolated from Brassica campestris L. ssp. chinensis, Makino syn. B. rapa L. ssp. chinensis. This gene was highly abundant in the fertile flower buds but silenced in the sterile ones of genic male sterile A/B line ('ZUBajh97-01A/B') in B. campestris. Expression patterns analysis suggested BcMF8 was a pollen-specific gene, whose transcript started to be expressed at the uninucleate stage and maintained throughout to the pollen at pollination stage. BcMF8 is highly homologous to the known pollen-specific AGP genes Sta 39-4 and Sta 39-3 from B. napus. Isolation and multiple alignment of the homologs of BcMF8 gene in the family Cruciferae indicated that BcMF8 was highly conserved in this family, which reflect the conservation in biological function and importance of this putative AGP gene in plant development. Similarity analysis also demonstrated Sta 39-4 and Sta 39-3 may originate from different genomes.
[Bcmf4 Gene, Encoding a Leucine-Rich Repeat Protein, Plays a Role in Male Fertility in Chinese Cabbage-Pak-choi].

Abstract:The BcMF4 (Brassica campestris Male Fertility 4) gene was previously isolated from the fertile B line of Chinese cabbage-pak-choi (Brassica campestris ssp. chinensis var. communis, syn. B. rapa ssp. chinensis var. communis). In the present paper, based on the cDNA sequence of BcMF4, primers were designed and used to amplify two fragments from the cDNA of flower buds of Chinese cabbage-pak-choi. Two produced fragments were introduced separately into binary vector pBI121 in antisense and sense orientations. The generated RNA interference (RNAi) vector was then mobilized into Agrobacterium tumefaciens strain LBA4404. The A. tumefaciens harboring the BcMF4 fragments was transformed to flowering Chinese cabbage (B. campestris ssp. chinensis var. parachinensis) via tissue culture. Approximately 45.8% of the pollen grains from 72.2% of RNAi plants exhibited abnormal in their shapes, and only 23.7% of the pollen grains from these plants germinated normally. Northern blotting demonstrated that the phenotypic change of pollen grains resulted from the inhibition of expression of the BcMF4 due to the insertion of the transgene. This indicates that functional interrupting of BcMF4 by RNAi resulted in partial pollen abortion in flowering Chinese cabbage, suggesting that the product of BcMF4 gene plays an important role during pollen development of Chinese cabbage such as Chinese cabbage-pak-choi and flowering Chinese cabbage.
Cloning and Characterization of a New Male Sterility-Related Gene BcMF1 in Brassica Campestris L. Ssp. Chinensis Makino

Abstract:【Objective】This paper aims to screen a novel gene related with the male sterility in Chinese cabbage for studying the molecular mechanism of plant male sterility. 【Method】The gene differential expression analysis was performed by cDNA-AFLP in the genic male sterile-fertile line ZUBajh97-01AB of Chinese cabbage, and a differentially expressed cDNA fragment, BcMF-A15T17, was found in fertile line. The full cDNA and DNA sequences of the gene related with BcMF-A15T17 were amplified by RACE and PCR, and the expression characteristics of the gene were proved by Northern blot analysis. 【Result】 The gene, designated as BcMF1, was 1 684 bp long in cDNA and 1 985 bp long in DNA, and specially expressed in middle and big floral buds of the fertile line. The hypothetical protein BcMF1 including 471 amino acids, which was predicted being a extracellular protein in secretory pathway, had higher similarity in the amino acids sequence to some members from a unknown protein family DUF1216 in Arabidopsis thaliana【.Conclusion】 All the results in this study suggest that BcMF1 is a new gene related with genic male sterility in Chinese cabbage.
Characterization of a male sterile related gene BcMF15 from Brassica campestris ssp. chinensis

Abstract:Data from cDNA-AFLP analysis based on the genome-wide transcriptional profiling on the flower buds of the male meiotic cytokinesis ( mmc ) mutant and its wild-type of Brassica campestris L. ssp. chinensis Makino, syn. B. rapa L. ssp. chinensis , indicated that mutation of the MMC gene resulted in changes in expression of a variety of genes. A transcript-derived fragment specifically accumulated in the wild-type flower buds was isolated, and the corresponding full-length cDNA and DNA was subsequently amplified. Bioinformatical analyses of this gene named BcMF15 (GenBank accession number EF600901) showed that it encoded a protein with 103 amino acids. The BcMF15 had a 88% nucleotide similarity to a lipid transfer protein-like gene. Moreover, sequence prediction indicated that BcMF15 might encode a membrane protein with a signal peptide at the N-terminus. Meanwhile, six domains were predicted in the deduced BcMF15 protein, such as the AAI domain existing in some crucial proteins of pollen development - preferential, signal peptide, transmembrane domain, vWF domain, ZnF_C4 domain, and Tryp_alpha_amyl domain. Spatial and temporal expression patterns analysis by RT-PCR indicated that BcMF15 was exclusively expressed in the fertile line, which indicated this gene is male sterile related. Phylogenetic analysis in Cruciferae revealed that the BcMF15 was relative conservative in evolution. We suppose BcMF15 may be a critical molecule in the transmembrane transportation and signal transduction during microspore development.

Abstract:A novel male-sterile mutant which lacks mature pollen, Brassisa campestris male sterile (bcms), was identified in Brassica campestris L. ssp. chinensis Makino (syn. B. rapa ssp. chinensis). Genetic analysis revealed that bcms was controlled by a single recessive mutation locus. Genome-wide transcriptional profiling was performed on the flower buds of both the bcms mutant and the wild-type from which it originated, and profiling analysis indicated that there were numerous changes in gene expression attributable to the gene mutation. This mutation resulted in down-regulation of a variety of genes and up-regulated expression of a few other genes. A total of 51 transcript-derived fragments (TDFs) were isolated: 32 specifically and 12 predominantly accumulated in wild-type flower buds, and two specifically and five predominantly accumulated in bcms flower buds. Sequence analysis showed that some of these TDFs share significant similarities with genes involved in different aspects of cellular development, such as signal transduction, cell wall biosynthesis and regulation. Most other TDFs showed no or very poor sequence similarities to entries in any database and might represent new candidate proteins involved in pollen development. Furthermore, spatial and temporal expression pattern analysis of 20 genes derived from cDNA-amplified fragment length polymorphism in different tissues of both the bcms and wild-type plants revealed their complex and dynamic expression patterns. The bcms mutant and the genes isolated in this paper provide excellent material for future studies on the molecular mechanism of male sterility.

Abstract:The polygalacturonase (PG) gene family has been found to be enriched in pollen of several species; however, little is currently known about the function of the PG gene in pollen development. To investigate the exact role that the PG gene has played in pollen development and about this family in general, one putative PG gene, Brassica campestris Male Fertility 9 (BcMF9), was isolated from Chinese cabbage (Brassica campestris ssp. chinensis, syn. B. rapa ssp. chinensis) and characterized.RT-PCR, northern blotting and in situ hybridization were used to analyse the expression pattern of BcMF9, and antisense RNA technology was applied to study the function of this gene.BcMF9 is expressed in particular in the tapetum and microspore during the late stages of pollen development. Antisense RNA transgenic plants that displayed decreased expression of BcMF9 showed pollen morphological defects that resulted in reduced pollen germination efficiency. Transmission electron microscopy revealed that the homogeneous pectic exintine layer of pollen facing the exterior was over-developed and predominantly occupied the intine, reversing the normal proportional distribution of the internal endintine layer and the external exintine in transgenic pollen. Inhibition of BcMF9 also resulted in break-up of the previously formed tectum and baculae from the beginning of the binucleate stage, as a result of premature degradation of tapetum.Several lines of evidence, including patterns of BcMF9 expression and phenotypic defects, suggest a sporophytic role in exine patterning, and a gametophytic mode of action of BcMF9 in intine formation. BcMF9 might act as a co-ordinator in the late stages of tapetum degeneration, and subsequently in the regulation of wall material secretion and, in turn, exine formation. BcMF9 might also play a role in intine formation, possibly via regulation of the dynamic metabolism of pectin.
Isolation and characterisation of the microspore-related gene BcMF4 in Chinese cabbage-pak-choi and its functional identification in Arabidopsis

Abstract:To provide information relevant to understanding the molecular mechanism of genic male-sterility in plants, differential expression of genes in flower buds at different developmental stages, and in other organs, between a sterile A line and a fertile B line of Chinese cabbage-pak-choi (Brassica rapa ssp. chinensis; syn. B. campestris ssp. chinensis) was analysed by cDNA-AFLP. Four cDNA fragments, including 13cMF-A17T14, were obtained from flower buds of the fertile B line. In this paper, on the basis of BcMF-A17T14, a spliced full-length cDNA designated BcMF4 was generated by rapid amplification of cDNA ends (RACE) from flower buds of the fertile B line of Chinese cabbage-pak-choi. The complete nucleotide sequence of the cDNA of BcMF4 was then amplified only from flower buds of the fertile B line. BcMF4 was expressed specifically in tetrads and uni-nucleate pollen of the fertile B line.,ne 1,102 bp DNA sequence of BcMF4 contains a single 95 bp intron and two exons. Its largest open reading frame (999 nt) encodes a polypeptide of 332 amino acids. The putative BcMF4 protein contains a signal peptide and displays five possible N-glycosylation sites, which are typical structures found in polygalacturonase inhibiting proteins (PGIPs). It also has eight Leucine-Rich Repeat (LRR) regions, and has 69 - 90% identity to PGIPs from other plants. Functional interruption of its ortholog in Arabidopsis thaliana, using an RNAi vector, resulted in aborted pollen grains and decreased pollen germination, and the abnormal pollen phenotype was stably inherited. This study suggests that the product of BcMF4 plays an important role in the development of microspores or pollen.
