Abstract:Single-nanoparticle detection has received tremendous interest due to its significance in fundamental physics and biological applications. Here, we demonstrate an optical nanofibre-enabled microfluidic sensor for the detection and sizing of nanoparticles. Benefitting from the strong evanescent field outside the nanofibre, a nanoparticle close to the nanofibre can scatter a portion of the field energy to the environment, resulting in a decrease in the transmitted intensity of the nanofibre. On the other hand, the narrow and shallow microfluidic channel provides a femtoliter-scale detection region, making nanoparticles flow through the detection region one by one. By real-time monitoring of the transmitted intensity of the nanofibre, the detection of a single polystyrene (PS) nanoparticle as small as 100 nm in diameter and exosomes in solution is realised. Based on a statistical analysis, the mean scattering signal is related to the size of the nanoparticle. Experimentally, a mixture of nanoparticles of different diameters (200, 500, and 1000 nm) in solution is identified. To demonstrate its potential in biological applications, high-throughput counting of yeasts using a pair of microchannels and dual-wavelength detection of fluorescently labelled nanoparticles are realised. We believe that the developed nanoparticle sensor holds great potential for the multiplexed and rapid sensing of diverse viruses. Progress on Optical Measurements in Single-Molecule Analysis with Nanopores

Abstract:Nanopore technology is widely used for single molecule/single particle detection due to its advantages of label-free sensing, low cost, portability and ultra-high sensitivity. The ionic current based electrical measurement is the common method for collecting nanopore signals. However, the electrical measurements suffer from several deficiencies such as the non-ideal noise components, low throughput and insufficient temporal or spatial resolution. Therefore, many strategies have been proposed to overcome these limitations. In this review, we summarize the progress of optical detection techniques with high bandwidth and high throughput that can be used to supplement or/and replace electrical measurements. It has been demonstrated that single-molecule optical detection using fluorescently labeled analytes (such as DNA) in nanopores is feasible. Moreover, methods to further improve the signal-to-noise ratio (SNR) of optical measurements in nanopores, including background noise suppression and the use of spectrally resolvable fluorescent groups, were also assessed. To avoid the fluorescence labeling of analytes, Ca2+ fluorescent probe-based ion flux imaging can achieve the optical measurement of unlabeled analytes directly. In addition to fluorescence imaging, plasmonic nanopores also provide solutions for optical nanopore sensing with the advantages of tunable optical/electrical sensitivity and scalable parallel detection. Plasmonic structures can enhance electromagnetic fields and focus them to a "hot spot" region located only at the nanopore depending on the structure of the pore, and the characteristic optical signals of the analyte are affected as it translocates through the pore. The SNR of optical signals can be improved by designing pores with reasonable size, shape, material and dielectric environment for enhancing the confinement of the electromagnetic field. The highly constrained electromagnetic field of plasmons have been used to help resolve single fluorescent molecules. Appropriate modifications of the surface of the plasmonic nanostructure can effectively suppress the fluorescence background and further enhance the fluorescence signal. In addition to enhancing labeled fluorescence emission, the unlabeled resonance shift and Raman scattering spectroscopy can also be performed directly for measuring single molecules. The principle of detecting the plasmonic resonance shift is that when the analytes enter the plasmonic nanopore, the local refractive index changes, which affects the intensity and frequency of the light scattered. Single DNA molecules have already been shown to transport through nanopores which can be efficiently detected by monitoring the intensity of scattered light. In addition, unlabeled surface-enhanced Raman scatting (SERS) can not only provide rich fingerprint information of molecular fine structure characteristics, but also be used to identify and measure a variety of complex samples, with chemical contrast. SERS with single molecule sensitivity is very important for the potential application of nanopores in chemical identification of different types of molecules. To achieve accurate and real-time measurement of the reaction of local targets, nanopipettes and SERS technology can be further combined. The highly sensitive plasmonic nanopipettes enable the measurement of single particles, single biomolecules and the environmental stimulus. However, controlling the molecular translocation through the pore is the main challenge for the enhanced SERS sensing at present. The typical sub-millisecond translocation time is too short to obtain the full spectral information with SERS. To address this limitation, several techniques based on the plasmonic trapping effect have been proposed to regulate the translocation speed. To conclude, nanopore technology based on single-molecule optical sensing has made great progress, but there are still several major challenges that needs to be solved. We believe that optical nanopore technology with high temporal and spatial resolution may open up a new horizon for nanopore analysis.

Abstract:Single nanoparticle sensing (SNS) is an emerging research field which utilizes single nanoparticles as individual nano-sensors to acquire the qualitative and quantitative information of the analytes in a localized and microscopic sample environment. Both the molecular recognition and signal transduction take place at the surface of a single nanoparticle. Versatile kinds of optical microscopy, such as dark-field microscopy and fluorescence microscopy, are often applied to locating the nano-sensor, and to accessing and analyzing the optical signal it reports. Compared to traditional sensing mechanisms that rely on ensemble nanomaterials, throughput and multiplex analysis. More importantly, single nanoparticles can be easily introduced to microscopic and dynamic systems such as living cells to probe specific analytes with high temporal and spatial resolution while maintaining the excellent sensitivity. In this review, we begin with a brief introduction on the history and development of SNS, which is followed by its major features. We subsequently survey the recent progresses in this field in the past five years, focusing on the different sensing principles, single nanoparticle counting and single nanoparticle tracking. We finally provide our perspectives that further developments on nano-probes, optical imaging techniques and data analysis are critical to the growth and applications of SNS in broad fields.

Abstract:A new sensor made of a sub-wavelength optical-fiber array is shown. On page 7462, Q. Gong, Y.-F. Xiao, and co-workers report that such a tiny optical device can detect and size single nanoscale particles, owing to its high sensitivity and real-time response ability. This simple and promising design may facilitate important applications, such as early-stage biomedical diagnostics, process control of semiconductor manufacturing, and explosives and environmental monitoring.

Abstract:Optical nanopore methods are widely believed to be practical approaches to address the flicker noise and the throughput limitations of nanopore detection. However, most optical nanopore methods in the literature are based on fluorescent labeling, which perplexes the detection operation and limits their applications. In this paper, we demonstrate a brand new optical nanopore technique named "scattering nanopore method". This method achieves single nanopore readouts without the introduction of dyes by detecting scattering light through a single subwavelength aperture on Au film. The single particle induced optical fluctuations could be detected simultaneously with the nanopore ionic current readouts. The method we demonstrated here opens a new direction for nanopore sensing of single particles, even single molecules in the near future.

Abstract:A photonic sensor based on a random slot surface-emitting semiconductor laser is theoretically demonstrated by utilizing multi-order diffraction. The results show that the variation of far-field local speckles at single wavelength lasing can be utilized to confirm the refractive index of the substances and the flow velocity of erythrocytes in capillaries. The similarity of the intensity contrast is the other protocol to measure the flow velocity of erythrocytes. Additionally, the similarity of the spectral contrast at multiple wavelengths lasing can be used to determine the concentration of erythrocytes in capillaries. The random slot semiconductor laser may provide a new and flexible platform for versatile active sensing.

Abstract:<section class="article-section__content"><p>Electrical detection of single molecule based on solid‐state nanopores is recently combined with optical methods. The synergistic effect of the electrical and optical measurements makes the combination powerful. It is promising for the next‐generation single‐molecule detection approach. Conventional solid‐state nanopore sensors monitor the electrical pulses caused by molecules translocating through the nanopore. However, the electrical signal is limited by insufficient resolutions, low throughput, lack of specificity, and molecule manipulation. Optical detection of single molecules has been extensively studied since the past decades. Optical signals, such as surface‐enhanced Raman spectroscopy (SERS) and fluorescence, have unique sensitivity and specificity of single molecules. Recently, optical methods are developed to complement the limitations of electrical detection of solid‐state nanopores. This review describes the main approaches used for electro‐optical detection of single molecules based on solid‐state nanopores, including total internal reflection fluorescence (TIRF), confocal microscopy, and plasmonic nanostructures. Laser‐induced heating, optical trapping, and strategies to reduce background fluorescence are also discussed. Moreover, recent experiments on electro‐optical detection of DNA and other analytes are introduced.</p></section>
Random Lasers with Controlled Emission Modes by Colloidal Semiconductor Quantum Dots Coupled to Plasmonic Nanorod Arrays

Abstract:Random lasers based on scattering for light amplification have gained extensive attention due to their simple structures. Colloidal semiconductor quantum dots (CSQDs) are excellent candidates as active media for random lasing. However, achieving CSQDs-based random lasers is challenging because the non-radiative Auger recombination produces a remarkable carrier loss. Here, we report random lasers with low thresholds based on the CSQDs coupled to the plasmonic nanorod arrays. The random lasers result from the enhancing excitation and emission rates of the CSQDs through plasmon resonance energy transfer induced by the spectral overlap between the resonant cavity modes of the plasmonic nanorod arrays and the excitons of the CSQDs, which reduces the carrier loss and establishes the optical gain in the CSQDs. Moreover, the incoherent and coherent emission modes can be controlled by varying the filling configurations (i.e., full-filling and partial-filling) of the plasmonic nanorods embedded in anodic aluminum oxide templates. Consequently, the incoherent random laser with a threshold of 19.3 11J/mm2 and coherent random lasers with a threshold of 13.5 11J/mm2 are performed, respectively. The proposed system provides a feasible method for low-threshold CSQDs-based random lasers with controlled emission modes, showing a promising avenue for potential applications.

Abstract:Colloidal quantum wells (CQWs) are a new type of atomic flat semiconductor nanocrystals, which present excellent opportunities as optical gain materials owing to their ultranarrow emission linewidth, tunable emission wavelength, suppressed Auger effect, and large gain cross-section. Here we synthesized monodispersed silica nanospheres and CdSe(4ML)/CdZnS core/shell CQWs. We observed random lasing by optically pumping the composites of CdSe(4ML)/CdZnS core/shell CQWs and silica nanoparticle scattering layer. The random lasing ranges from 615 nm to 625 nm with a threshold of 0.55 mJ cm −2 . Fourier analysis reveals that sufficient Rayleigh scattering is the primary reason for the low threshold in the spatial frequency spectrum of the random laser. Our findings suggest that CQWs can serve as an efficient optical gain medium for random lasing, enabling various applications such as speckle-free laser imaging, random number generation for secure communication, chemical and biological sensing, high brightness and energy-efficient lighting, and label-free bioimaging.

Abstract:Due to the stimulated emission amplification, lasers with excellent characteristics, including the high energy density, ultra-narrow spectral linewidth, and high directionality, are extremely favorable for sensing, detection, and imaging. Bringing these merits into the micro/nano scale, micro/nano lasers with miniaturized device sizes further enable outstanding spatial and temporal confinement, greatly boosting the light-matter interaction and bridging the size mismatch between light and biomolecules. Thanks to these advantages, micro/nano lasers have drawn widespread attention and opened new opportunities for a variety of biomedical and biochemical applications. In this paper, we review recent developments in biomolecular sensing and cellular analysis based on micro/nano lasers. We first describe the fundamental building blocks of micro/nano lasers, with discussions on gain material considerations, cavity structures, and pumping. We then review recent applications using micro/nano lasers as biosensors and bioprobes, including biomolecule (mainly proteins and DNAs) sensing, wavelength-multiplexed cell labeling/tracking/probing, and high-resolution cellular/tissue bioimaging. Finally, an outlook of the challenges and potential developments of micro/nano lasers for biological sensing and clinical applications is provided.

An Anderson-localized Random Nanolaser

Abstract:Precision is a virtue throughout science in general and in optics in particular where carefully fabricated nanometer-scale devices hold great promise for both classical and quantum photonics [1-6]. In such nanostructures, unavoidable imperfections often impose severe performance limits but, in certain cases, disorder may enable new functionalities [7]. Here we demonstrate on-chip random nanolasers where the cavity feedback is provided by the intrinsic disorder in a semiconductor photonic-crystal waveguide, leading to Anderson localization of light [8]. This enables highly efficient and broadband tunable lasers with very small mode volumes. We observe an intriguing interplay between gain, dispersion-controlled slow light, and disorder, which determines the cross-over from ballistic transport to Anderson localization. Such a behavior is a unique feature of non-conservative random media that enables the demonstration of all-optical control of random lasing. Our statistical analysis shows a way towards ultimate thresholdless random nanolasers.
Far-Field Single Nanoparticle Detection and Sizing

Abstract:Whispering gallery mode based optical microcavities are important for highly sensitive optical sensing. However, the current experimental realizations are strongly dependent on high-resolution tunable lasers and evanescent coupling, which are too cumbersome and too expensive for portable devices. Herein we experimentally demonstrate a cost-effective and robust approach to detect and size a single nanoparticle with far-field laser emissions. By placing a limacon microdisk close to a spiral microdisk, chiral resonances have been successfully generated. In contrast to previous research, here the internal chirality is strongly correlated with the far-field patterns (FFPs) and thus can be transduced to far-field emissions. Once a nanoparticle is attached to the limacon microdisk, the asymmetrical backscattering at the notch of the spiral can be averaged by the symmetrical scattering of the nanoparticle. Consequently, the internal chirality and the corresponding FFPs are changed. By measuring a far-field directional laser emission, nanoparticles with a radius of similar to 50 nm have been successfully detected and sized without employing any spectral information. As a narrow-linewidth tunable laser is not used in our experiment and microdisks lasers may be electrically driven, this research will provide a new path to cost-effective, portable, highly sensitive optical sensors. Abstract:Tremendous progress in nanotechnology has enabled advances in the use of luminescent nanomaterials in imaging, sensing and photonic devices. This translational process relies on controlling the photophysical properties of the building block, that is, single luminescent nanoparticles. In this Review, we highlight the importance of single-particle spectroscopy in revealing the diverse optical properties and functionalities of nanomaterials, and compare it with ensemble fluorescence spectroscopy. The information provided by this technique has guided materials science in tailoring the synthesis of nanomaterials to achieve optical uniformity and to develop novel applications. We discuss the opportunities and challenges that arise from pushing the resolution limit, integrating measurement and manipulation modalities, and establishing the relationship between the structure and functionality of single nanoparticles.

Explaining the mechanism of random lasing based sensing

Abstract:Here we report a random lasing based sensor which shows pH sensitivity exceeding by 2-orders of magnitude that of a conventional fluorescence sensor. We explain the sensing mechanism as related to gain modifications and lasing threshold nonlinearities. A dispersive diffusive lasing theory matches well the experimental results, and allow us to predict the optimal sensing conditions and a maximal sensitivity as large as 200 times that of an identical fluorescence-based sensor. The simplicity of operation and high sensitivity make it promising for future biosensing applications.

Abstract:Single nanoparticle detection is of critical importance in many applications. Optical microcavities featuring high-Q factors and small mode volumes have been widely investigated in sensing applications. In general, the microcavity sensing depends mainly on reactive (i.e., dispersive) interactions, resulting in a resonance wavelength shift or mode splitting, which essentially responds to the real part of the polarizability of the targets. In the first part of this talk, we report the experimental demonstration of single nanoparticle detection using either resonance mode broadening or microcavity Raman laser splitting. In the second part, a dissipative sensing method is demonstrated to detect single lossy nanoparticles. This dissipative sensing method holds great potential in detecting nanoparticles of high absorption or ultralow polarizabilities, such as carbon nanotubes and metal nanoparticles, and in characterizing nanoparticle properties in combination with the reactive sensing method.

Abstract:Single-molecule detection is a rapidly developing area within the analytical chemistry field that requires ultrasensitive technologies to detect a range of molecules. Over the past few decades, various optically-, mechanically-, and electrically-based strategies have been employed for single-molecule detection to uncover information in biological processes. These strategies enable real-time monitoring with single-molecule/single-event sensitivity. In addition, their high temporal resolution enables investigation of the underlying mechanisms of biological functions from static to dynamic, from qualitative to quantitative, and from one to multiple disciplines. In this review, we provide a brief overview of the prominent, real-time single-molecule detection nanotechnologies and their potential applications within the life science fields.

Abstract:Detection of nanoscale objects is highly desirable in various fields such as early-stage disease diagnosis, environmental monitoring and homeland security. Optical microcavity sensors are renowned for ultrahigh sensitivities due to strongly enhanced light-matter interaction. This review focuses on single nanoparticle detection using optical whispering gallery microcavities and photonic crystal microcavities, both of which have been developing rapidly over the past few years. The reactive and dissipative sensing methods, characterized by light-analyte interactions, are explained explicitly. The sensitivity and the detection limit are essentially determined by the cavity properties, and are limited by the various noise sources in the measurements. On the one hand, recent advances include significant sensitivity enhancement using techniques to construct novel microcavity structures with reduced mode volumes, to localize the mode field, or to introduce optical gain. On the other hand, researchers attempt to lower the detection limit by improving the spectral resolution, which can be implemented by suppressing the experimental noises. We also review the methods of achieving a better temporal resolution by employing mode locking techniques or cavity ring up spectroscopy. In conclusion, outlooks on the possible ways to implement microcavity-based sensing devices and potential applications are provided.
