Abstract:Two macrocyclic polyamine ligands and their dinuclear copper(II) (CuII) complexes were synthesized and characterized. In the complexes, two 1,4,7,10-tetraazacyclododecane (cyclen) moieties were bridged by a dipeptide spacer, and CuII as metal ion was coordinated to the ligands to form the novel complexes. These complexes were characterized by UV/VIS, IR, EPR, and ESI-MS analyses. The catalyzed DNA cleavage and DNA binding properties of these complexes were studied under physiological conditions. The effects of complex concentrations, cleavage time, and temperature on the DNA cleavage reactions were systematically investigated. The results strongly suggested that the plasmid DNA (pUC 19) can be cleaved by the dinuclear CuII complexes. At pH 7.0 and 37 degrees, incubating DNA with the complexes for 24 h, all Form I was selectively converted into Form II. At 55 degrees, pH 7.0, the time was shortened to less than 30 min. According to the mechanism experiment, these complexes cleave plasmid pUC19 DNA by oxidative mechanism. The synergistic effect of the CuII ions is important to improve the DNA cleavage.

Abstract:A hairpin polyamide-chlorobenzenesulfonyl conjugate was synthesized in solution by a haloform reaction and the dicyclohexylcarbodiimide/1-hydroxybenzotriazole (DCC/HOBT) coupling reaction. Its ability to cleave DNA was investigated with supercoiled DNA (pBluescript SK), and the DNA cleaving efficiency of chlorobenzenesulfonyl group was enhanced by the hairpin polyamide under UV irradiation (365 nm).

Abstract:In this paper, three [Formula: see text],[Formula: see text]-conjugated cationic porphyrin compounds were designed and synthesized. The structure of the intermediates and desired porphyrins were confirmed by UV, IR, 1H NMR, MS and elemental analysis. The interaction modes between these porphyrins and ct-DNA were studied by UV-vis spectroscopy and fluorescence emission spectroscopy. The results showed that PCP 1 had an external binding mode with DNA at low DNA concentration and could intercalate DNA with the increase of concentration. PCP 2 interacted with DNA through an external binding mode, and PCP 3 could insert into DNA. The binding constants ([Formula: see text] between PCP1[Formula: see text]PCP3 and ct-DNA were calculated to be 8.41 × 104, 7.33 × 104 and 4.14 × 104 M[Formula: see text], respectively. The singlet oxygen (1O[Formula: see text] generation of PCP1[Formula: see text]PCP3 was determined by the 1,3-diphenylisobenzofuran (DPBF) method using tetrapyridylporphyrin (H2TMPyP) as a reference. The 1O2 generation rate of PCP1[Formula: see text]PCP3 followed the order of PCP2 >PCP1>H2TMPyP >PCP3. Subsequently, the photocleavage effect of porphyrins on pBR322 plasmid DNA was studied by gel electrophoresis. At 10.0 [Formula: see text]M, PCP1 and PCP2 could cleave DNA completely. At 2.0 [Formula: see text]M, the cleavage rate of DNA by PCP3 was 57.5%, which was significantly higher than that of H2TMPyP (38.8%). These results verified that the amount of cationic ions in the porphyrin structure could affect the binding modes of porphyrins with DNA and their cleavage ability of DNA.

Abstract:The DNA binding and cleavage activity of a synthetic polyamide containing dipeptide Ser-His has been investigated by spectroscopic techniques, such as electronic absorbance and circular dichroism (CD) spectropolarimetry, as well as gel electrophoresis. The results show that the molecule has a strong interaction with DNA and can improve DNA-cleavage ability about 100 fold as compared with single dipeptide Ser-His.

Abstract:We report the design and synthesis of a novel cationic AIE-gen, i.e., (E)-TPEDEPy-DBz, which can be derived from a facile synthetic route. (E)-TPEDEPy-DBz is a stereo-pure isomer and shows evident AIE properties with yellow-greenish emission in the solid state (ΦF = 43.7%). Distinct from most of the ionic AIE-gens reported in the literature, which are usually exemplified by their water solubility and the capacity for interaction with charged biological species, (E)-TPEDEPy-DBz demonstrated moderate solvatochromism and evident mechanoluminochromism behaviours. The emission of (E)-TPEDEPy-DBz can be switched between yellow-greenish in the crystalline state to red-orange in the amorphous state by grinding and fuming treatments. Due to the AIE-characteristics, cationic modification and the thus-acquired water solubility, (E)-TPEDEPy-DBz performed as a sensitive fluorescent probe to detect DNA in Tris-HCl buffer solution. More importantly, (E)-TPEDEPy-DBz showed good membrane permeability and cell viability, and the potentiality to be used as a mitochondria-specific probe working in a fluorescence turn-on mode.

Abstract:A water-soluble porphyrin dimer (Por Dimer) containing eight positive charges, bridged by 4,4′-dicarboxy-2,2′-bipyridine, has been synthesized. With Meso-tetrakis(N-methyl-pyridium-4-yl)porphyrin (H2TMPyP) as the reference compound, the water-soluble porphyrin dimer was investigated for its interaction with DNA by absorption, fluorescence, and circular dichroism (CD) spectroscopy. The apparent affinity binding constant (K app = 1.2 × 106) of Por Dimer binding to CT DNA was measured by a competition method with ethidium bromide (EB) (that of H2TMPyP was 6.9 × 106). The cleavage ability of Por Dimer to pBR322 plasmid DNA was studied by gel electrophoresis. The results suggest that the binding modes of Por Dimer were complex and involve both intercalation and outside binding.
Potent DNA photocleavage by zinc(II) complexes of cationic bis-porphyrins linked with aliphatic diamine

Abstract:We have prepared zinc(II) complexes of cationic bis-porphyrins, as one of the attempts to improve less DNA photocleavage activities of the metal-free bis-porphyrins composed of two H(2)TMPyP-like chromophores, linked with a series of aliphatic diamines. The less activities seemed to be derived from their intermolecular self-aggregation properties in aqueous solution. The zinc(II) insertion into the metal-free cationic bis-porphyrins completely removed their self-aggregation properties, most probably due to steric hindrance between axial ligands of zinc(II) chromophores of the cationic bis-porphyrins. The DNA photocleavage activities of the zinc(II) complexes were fully enhanced, which were three times larger than that of the lead compound H(2)TMPyP. Quantitative analysis of singlet oxygen production by photosensitization of cationic bis-porphyrins was performed using 1,3-diphenylisobenzofuran, and the singlet oxygen productivities of them were found to be related to their solution properties. There is a good relationship between the activities and the productivities, which will provide insights into the further development of more effective DNA photocleavage agents.

Abstract:A highly water soluble 3:2 complex of copper(II) and 2,4,6-tris(di-2-pyridylamine)-1,3,5-triazine (TDAT) has been synthesized and structurally characterized. The complex crystallized in a triclinic P1 space group with a molecular formula of [Cu-3(TDAT)(2)Cl-3]Cl-3 center dot 2H(2)O (1), where each copper ion is coordinated by four pyridine nitrogen atoms and an apical chloride. The trinuclear complex is stable at physiological relevant conditions. It can bind to DNA through electrostatic attraction and cleave efficiently the supercoiled pBR322 DNA into its nicked and linear forms at micromolar concentrations. Active oxygen intermediates such as hydroxyl radicals and singlet oxygen generated in the presence of 1 may act as active species for the DNA scission.

Abstract:Copper(II) complexes of ethyl 2-[bis(2-pyridylmethyl)amino]propionate ligand (ETDPA) have been synthesized and characterized by elemental analyses, IR spectra, UV spectra and ES-MS. These complexes are stable in air with the formula of [(ETDPA)CuCl2] and [(ETDPA)Cu(phen)](ClO4)2. The DNA-binding properties of the Cu(II) complexes have been investigated by ultraviolet spectroscopy and fluorescence spectroscopy, which showed that the binding mode of the two complexes with DNA was different. The binding mode of [(ETDPA)CuCl2] is not the classical intercalation binding, and the binding mode of [(ETDPA)Cu(phen)](ClO4)2 with DNA is intercalation. The cytotoxic assay shows that the [(ETDPA)Cu(phen)](ClO4)2 is more active on the proliferation of cancer cells than the [(ETDPA)CuCl2].

Abstract:The interaction between salmon testes DNA (st-DNA) and a series of Cu(II) polypyridyl complexes, i.e. [Cu(dmbpy)(NO3)2] (1) (dmbpy = 4,4'-dimethyl-2,2'-bipyridine), [Cu(bpy)(NO3)2] (2) (bpy = 2,2'-bipyridine), [Cu(phen)(NO3)2] (3) (phen = phenanthroline), [Cu(terpy)(NO3)2]·H2O (4) (terpy = 2,2':6',2″-terpyridine), [Cu(dpq)(NO3)2] (5) (dpq = dipyrido-[3,2-d:2',3'-f]-quinoxaline) and [Cu(dppz)(NO3)2] (6) (dppz = dipyrido[3,2-a:2',3'-c]phenazine) was studied by UV/Vis absorption, Circular Dichroism, Linear Dichroism, EPR, Raman and (UV and vis) resonance Raman spectroscopies and viscometry. These complexes catalyse enantioselective C-C bond forming reactions in water with DNA as the source of chirality. Complex 1 crystallizes as an inorganic polymer with nitrate ligands bridging the copper ions, which adopt essentially a distorted square pyramidal structure with a fifth bridging nitrate ligand at the axial position. Raman spectroscopy indicates that in solution the nitrate ligands in 1, 2, 3 and 4 are displaced by solvent (H2O). For complex 1, multiple supramolecular species are observed in the presence of st-DNA in contrast to the other complexes, which appear to interact relatively uniformly as a single species predominantly, when st-DNA is present. Overall the data suggest that complexes 1 and 2 engage primarily through groove binding with st-DNA while 5 and 6 undergo intercalation. For complexes 3 and 4 the data indicates that both groove binding and intercalation takes place, albeit primarily intercalation. Although it is tempting to conclude that the groove binders give highest ee and rate acceleration, it is proposed that the flexibility and dynamics in binding of Cu(II) complexes to DNA are key parameters that determine the outcome of the reaction. These findings provide insight into the complex supramolecular structure of these DNA-based catalysts.

Abstract:Macroayclic complexes of Ni(II), Cu(II) and Co(II) were synthesized via the reaction of 1,4-bis(o-aminobenzyl)-1,4-diazacycloheptane (L) with the respective metal salts. The polyamine ligand and the corresponding complexes were characterized by mass spectrometry, elemental analysis and FT-IR and for [CuL](ClO4)2 (2), by a single crystal X-ray structural analysis which revealed the copper was in a square planar environment. The interaction of the ligand and its complexes with calf thymus DNA (ct-DNA) at pH 7.4 was investigated by UV–Vis, competitive fluorescence spectroscopic methods and dynamic viscosity measurements. These results of these experiments supported intercalative mode of binding. The competitive fluorescence studies in the presence of methylene blue (MB) as an intercalator probe showed that the ligand and its complexes were able to release MB from DNA helix. Viscosity measurements were in good agreement with the results of the competitive fluorescence, indicating that all compounds are intercalating species.

Abstract:A novel oxalato-bridged copper(II) complex has been prepared and structurally characterized: [Cu(bpa)(mu-C2O4)]center dot H2O (bpa?=?bis(2-pyridylmethyl) amine). In the complex, the copper ion is linked in an unusual mu 1,2,3-C2O42- bridging mode, generating one-dimensional zigzag chain disposition. Variable-temperature magnetic susceptibility studies (2300 K) reveal a weak ferromagnetic coupling, J?=?0.63 cm-1, between the copper ions. The interaction of the complex with CT-DNA has been studied using UVvisible absorption and emission spectral methods, and the binding constant of the complex with CT-DNA is Kapp?=?9?X?104 m-1, which indicates that the interaction of the complex with DNA is a moderate intercalative mode. Furthermore, the complex cleaves supercoiled plasmid DNA efficiently in the presence of hydrogen peroxide. The mechanistic investigations suggest that the hydroxyl radical and singlet oxygen are involved in the DNA degradation. Copyright (c) 2012 John Wiley & Sons, Ltd.

Abstract:Here, we describe the unexpected discovery of a Cu-catalyzed condensation polymerization reaction of propargylic electrophiles (CPPE) that transforms simple C3 building blocks into polydiynes of C6 repeating units. This reaction was achieved by a simple system composed of a copper acetylide initiator and an electron-rich phosphine ligand. Alkyne polymers (up to 33.8 kg/mol) were produced in good yields and exclusive regioselectivity with high functional group compatibility. Hydrogenation of the product afforded a new polyolefin-type backbone, while base-mediated isomerization led to a new type of dienyne-based electron-deficient conjugated polymer. Mechanistic studies revealed a new α-α selective Cu-catalyzed dimerization pathway of the C3 unit, followed by <i>in situ</i> organocopper-mediated chain-growth propagation. These insights not only provide an important understanding of the Cu-catalyzed CPPE of C3, C4, and C6 monomers in general but also lead to a significantly improved synthesis of polydiynes from simpler starting materials with handles for the incorporation of an α-end functional group.

Oxidative DNA Cleavage Promoted by Polynuclear Copper Complexes Bearing Iminodiacetate Chelator

Abstract:The rigid-spaced polycopper(II) complexes of different benzyl polyiminodiacetic acid (IDA) ligands, Cu2L1 (complex 1, meta-dicopper complex), Cu2L2 (complex 2, para-dicopper complex) and Cu3L3 (complex 3) (H4L1, N,N,N′,N′-m-xylylenediamine tetraacetic acid; H4L2, N,N,N′,N′-p-xylylenediamine tetraacetic acid; H6L3, N,N,N′,N′,N″,N″-mesitylenetriamine hexaacetic acid) were prepared to explore the chelator’s role in determining the DNA cleavage behavior of polycopper complexes by comparing these complexes of IDA chelator with their analogues of bis(2-pyridylmethyl)amine (BPA) chelator. PolyCu(IDA) complexes 1–3 show the synergic effect between copper(II) centers. Their DNA cleavage activity follows an order of 3>1>2, which is consistent with their DNA binding abilities. However, this consistence was not observed in the case of polyCu(BPA) complexes according to our previous study. In addition, both the DNA binding ability and cleavage efficiency of polyCu(IDA) complexes are distinctly lower than those of polyCu(BPA) complexes. Furthermore, both the meta- and para-dicopper complexes are able to mediate linear DNA from supercoiled DNA at high complex concentration for polyCu(IDA) complexes, while for their analogues with BPA chelators, only the meta-dicopper analogue could induce the linear DNA from supercoiled DNA. These results suggested that the replacement of BPA chelator with the anionic IDA chelator alters distinctly the DNA cleavage behavior of rigid-spaced polycopper complexes.
Double-strand cleavage of DNA by a polyamide-phenazine-di-N-oxide conjugate

Abstract:Phenazine and its derivatives have been widely applied as nucleic acid cleavage agents due to active oxygen activating the C-H bond of the substrate. However, diffusion of oxygen radicals limits their potential applications in the DNA-targeted metal-free drug. Introduction of groove binder moiety such as polyamide enhanced the regional stability of radical molecules and reduced cytotoxicity of the drugs. In this work, we described the design and synthesis of a polyamide-modified phenazine-di-N-oxide as a DNA double-strand cleavage agent. The gel assays showed the hybrid conjugates can effectively break DNA double strands in a non-random manner under physiological conditions. The probable binding mode to DNA was investigated by sufficient spectral experiments, revealing weak interaction between hybrid ligand and nucleic acid molecules. The results of our study have implications on the design of groove-binding hybrid molecules as new artificial nucleases and may provide a strategy for developing efficient and safe DNA cleavage reagents.

Abstract:An asymmetric aza-BODIPY analogue bearing quinoxaline moiety was synthesized via a titanium tetrachloride-mediated Schiff-base-forming reaction of 6,7-dimethyl-1,4-dihydroquinoxaline-2,3-dione and benzo[d]thiazol-2-amine. This novel aza-BODIPY analogue forms a complementary hydrogen-bonded dimer due to the quinoxaline moiety in the crystal structure. It also shows intense absorption and fluorescence, with fluorescence quantum yields close to unity. The electrochemical measurements and the DFT calculations revealed the presence of the low-lying HOMO, which benefits their potential applications as an electron-transporting material.
