Abstract:A chemical examination by the MS/MS based-molecular networking filtered with sorbyl fragment in association with HSQC-TOCSY fingerprinting led to the characterization of sorbicilinoid-type metabolites of a deep-sea derived fungus Penicillium rubens F54. Chromatographic fractionation allowed the isolation of 11 undescribed sorbicilinoid-type analogues namely sorbicillinolides A-K (1-11). Their structures were identified by extensive analyses of the spectroscopic data, including the calculated electronic circular dichroism and optical rotation for configurational assignments. Sorbicillinolides A-D represent newly rearranged sorbicilinoids featured a cyclopentenone core, and sorbicillinolides E-G as the stereoisomers are rare sorbicilinoids incorporated by a nitrogen atom. Sorbicillinolide A showed significant anti-neuroinflammatory effects in LPS-stimulated BV-2 macrophages due to the potent inhibition of NO and PGE2 production and the interruption of the RNA transcription of iNOS, COX-2 and IL6 of the NF-κB signaling pathway. Further investigation demonstrated that sorbicillinolide A stabilized and tightly bonded to COX-2, suggesting COX-2 as a potential target protein. This study provided sorbicillinolide A as an additional non-steroidal natural scaffold for the development of anti-inflammatory agent.
