Abstract:The effects of hot water treatment in alleviating chilling injury and reducing ultrastructural damage of mature-green cherry tomatoes （Lycopersicun esculentum cv. cerasiform Alef） were investigated. Mature-green cherry tomato fruits were treated in water at 40°C or 45°C for 5 min or 15 min, and then stored at 5°C for 19 days followed by ripening at 20°C. Water treatment at 40°C for 15 min increased tolerance of cherry tomato fruits to chilling stress, indicating as low outbreak of skin lesion, high color a* value, and low electrolyte leakage. Treated fruits showed typical climacteric respiration and developed normal red color with chlorophyll degradation and lycopene accumulation during ripening, while fruits without treatment failed to develop red color and suffered skin lesion. After 19 days of chilling, heated fruits showed the conversion of chloroplast to chromoplast with the disappearance of thylakoids. Mitochondria and other cell organelles were not adversely affected in treated fruits. However, ultrastructures in pericarp cells in control fruits severely damaged with extensive disorganization of cytoplasm, swelled chloroplasts, distorted and unstacked thylakoids. Chloroplast was the first and most severely impacted organelle by chilling stress. Hot water treatment （40°C for 15 min） before storage alleviated chilling injury in cherry tomato fruits. The results suggest that chilling injury is related with the damage of cell structure under chilling stress.
Brassinosteroid Biosynthetic Gene SlCYP90B3 Alleviates Chilling Injury of Tomato (solanum Lycopersicum) Fruits During Cold Storage.

Abstract:Tomato is susceptible to chilling injury during cold storage. In this study, we found that low temperature promoted the expression of brassinosteroid (BR) biosynthetic genes in tomato fruits. The overexpression of SlCYP90B3 (SlCYP90B3-OE), a key BR biosynthetic gene, alleviated the chilling injury with decreased electrical conductivity and malondialdehyde. In SlCYP90B3-OE tomato fruits, the activities of antioxidant enzymes, including ascorbate peroxidase (APX), catalase (CAT), peroxidase (POD), and superoxide dismutase (SOD), were markedly increased, while the activity of membranous lipolytic enzymes, lipoxygenase (LOX), and phospholipase D (PLD), were significantly decreased when compared with the wild-type in response to cold storage. Furthermore, the expression level of the cold-response-system component, SlCBF1, was higher in SlCYP90B3-OE fruits than in the wild-type fruits. These results indicated that SlCYP90B3 might be involved in the chilling tolerance of tomato fruits during cold storage, possibly by regulating the antioxidant enzyme system and SlCBF1 expression.
Effect of UV-C Treatment on Modulating Antioxidative System and Proline Metabolism of Bamboo Shoots Subjected to Chilling Stress

Abstract:Bamboo shoots are highly nutritious but difficult to preserve even under low temperature since they are sensitive to chilling. In the present research, bamboo shoots were exposed to UV-C light (4.1 kJ/m2) prior to 56 days storage at 1 °C to evaluate the potential of UV-C treatment on alleviating chilling injury (CI) of bamboo shoots under cold storage. CI incidence and physiological and biochemical characteristics associated with CI were determined during storage. The results showed that UV-C treatment apparently inhibited the development of CI and resulted lower EI and MDA content than that of the control. Further investigation demonstrated that UV-C treatment increased activities of CAT, APX, GR and DPPH scavenging activity of bamboo shoots, which greatly contribute to alleviating CI. The increases in both O2 − production rate and H2O2 content of UV-C-treated bamboo shoots were significantly delayed, PAL activity was increased which contributed to enhancing accumulation of TP, and endogenous proline synthesis was promoted, which resulted from the increased activity of P5CS and decreased PDH activity. The results above suggest that the improved antioxidative defense system and enhanced proline accumulation is the underlying mechanism that UV-C treatment enhanced chilling tolerance in bamboo shoots.

Abstract:As an environmentally friendly approach for fruit quality improvement, the effect of preharvest UV-C on the physiology of strawberry fruit during postharvest storage remains to be assessed. Strawberry fruit developed with supplementary UV-C were stored at room temperature for 2 weeks. Preharvest UV-C attenuated fruit postharvest senescence and altered phytochemicals composition. Higher ester titer was found in the treated fruit at harvest, whereas higher terpene and furanone contents were detected after 72 h of storage. At harvest, polyphenolics accumulated to a higher level in UV-C group, but the difference disappeared after 24 h of storage. Meanwhile, the intrinsic level of abscisic acid and the expressions of FaPYR1, SnRK2, and FaASR in the UV-C-treated fruit was enhanced at harvest but returned to a lower level as storage proceeded. This study highlights the time-dependent effect of preharvest UV-C on strawberry fruit postharvest biochemical indexes and the possible involvement of abscisic acid signaling factors.

Abstract:In this study, the mechanism recruited by exogenous melatonin application at 100 μM for alleviating chilling injury in tomato fruits during cold storage was investigated. Alleviating chilling injury in tomato fruits in response to exogenous melatonin application at 100 μM may ascribe to providing sufficient intracellular ATP occur by higher H-ATPase, Ca-ATPase, cytochrome c oxidase (CCO), and succinate dehydrogenase (SDH) enzymes activity during cold storage. Also, higher unsaturated/saturated fatty acids (unSFA/SFA) ratio owing to higher linoleic and linolenic acids accumulation coincides with lower palmitic, stearic and oleic acids accumulation may be responsible for alleviating chilling injury in tomato fruits in response to exogenous melatonin application at 100 μM, which may occur by higher fatty acid desaturase 3 and 7 (FAD3 and FAD7) genes expression accompanying by lower phospholipase D (PLD) and lipoxygenase (LOX) genes expression and enzymes activity, in addition to providing sufficient intracellular ATP. Therefore, exogenous melatonin application may be a beneficial postharvest procedure for alleviating chilling injury in tomato fruits during cold storage.

Abstract:Recent studies presented preharvest ultraviolet C (UV-C) as an environmentally friendly approach for the management of horticultural crop diseases. The effect of this approach on quality preservation during postharvest storage has not yet been investigated. Strawberry fruit harvested from plants grown with supplemental UV-C were stored at room temperature for 72 h, and their postharvest shelf-life biochemical indicators were evaluated. The involvement of microRNAs (miRNAs) in the activation of UV-C-induced antioxidant systems was investigated. Preharvest UV-C contributed to the preservation of sugar and organic acid and reduced overall lipid peroxidation in strawberry fruit during storage. We found that miR159 and miR398 were downregulated by preharvest UV-C and that their respective targets were upregulated at the early stage of storage with enhancement of the activity of antioxidant enzymes. The initial burst of H2O2 and O2• - suggested that preharvest UV-C primed the fruit in an antioxidative activated state via reactive-oxygen-species-mediated feedback control with post-transcriptional involvement of miRNAs.

Abstract:Preharvest ultraviolet-C (UV-C) treatment of strawberry is a very new approach, and little information is available on the effect of this treatment on plant growth regulators. In this study, the effect of preharvest UV-C irradiations at three different doses on strawberry yield, fruit quality parameters and endogenous phytohormones was investigated simultaneously. The overall marketable yield of strawberry was not affected by the preharvest UV-C treatments, although more aborted and misshapen fruits were found in UV-C treated groups than in the untreated control. The fruits in the high dose group were firmer and had approximately 20% higher sucrose content and 15% higher ascorbic acid content than the control, while fruits from the middle and low dose groups showed no significant changes in these parameters. The lower abscisic acid (ABA) content found in the fruits in the high UV-C group may be associated with those quality changes. The citric acid content decreased only in the low dose group (reduction of 5.8%), with a concomitant 37% reduction in jasmonic acid (JA) content, compared to the control. The antioxidant status of fruits that received preharvest UV-C treatment was considered enhanced based on their oxygen radical absorbance capacity (ORAC) and malondialdehyde (MDA) content. In terms of aroma, three volatile alcohols differed significantly among the various treatments with obvious activation of alcohol acyltransferase (AAT) activity. The observed synchronous influence on physiological indexes and related phytohormones suggests that preharvest UV-C might affect fruit quality via the action of plant hormones.
Effect of 1-Methylcyclopropene on Chilling Injury of Tomato Fruit

Abstract:In order to determine the effectiveness of 1-methylcyclopropene(1-MCP)on chilling injury and disease resistance of tomato fruit after cold storage.Green mature 'Zheza 205' tomato fruit was treated by 1 μL · L-1 1-MCP before being stored at 3 ℃ for 14 days then ripen at 20 ℃ for 16 days.The quality and physiology of tomato fruit were investigated.1-MCP treatment not only promoted chilling injury and disease index,but also increased the activities of polyphenol oxidase and peroxidase.1-MCP treatment retarded the peak of ethylene and the increase of lycopene,maintained higher content of titratable acidity and firmness.The activities of chitinase and β-1,3-glucanase were also inhibited by 1-MCP treatment.This study indicated that 1 μL · L-1 1-MCP delayed tomato fruit ripen,but promoted chilling injury and disease incidence,and showed that it could not be considered for commercial use in tomato fruit after cold storage.

Abstract:The effects of 4 kJ/m2 UV-C irradiation on main qualities and antioxidants of tomato during storage were investigated.The experimental results indicated that 4 kJ/m2 UV-C irradiation maintained a high level of firmness and delayed the color development,significantly promoted the accumulation of phenolics,flavonoids and lycopene,increased the antioxidant enzyme activities of CAT,SOD,APX and GR,consequently showed higher antioxidant ability during storage.The results suggested that UV-C irradiation had the potential to apply on extending the shelf life and enhancing antioxidants of tomato fruit.

Abstract:To obtain an overall view on gene expression during the early stage (24 h) of tomato fruit in response to postharvest UV-C irradiation (4 kJ/m(2)), we performed a microarray analysis by using Affymetrix Tomato Genechip. The results showed that 274 and 403 genes were up- or down-regulated, respectively, more than two folds in postharvest tomato fruit irradiated with UV-C as compared with that in control fruit. The up-regulated genes mainly involve in signal transduction, defense response and metabolism. Conversely, genes related to cell wall disassembly, photosynthesis and lipid metabolism were generally down-regulated. These results opened ways to probe into the molecular mechanisms of the effects of postharvest UV-C irradiation on increased disease resistance, delayed softening, better quality maintenance and prolonged postharvest life in tomato fruit.
UV-C Treatment Affects Browning and Starch Metabolism of Minimally Processed Lily Bulb

Abstract:The effects of 4.5 kJ m (UV)-U-2-C radiation on the quality of minimally processed lily bulb, such as browning, growth of microorganism, total phenolic content, starch, total soluble sugar, reducing sugar content, the activities of phenylalanine ammonia lyase (PAL), polyphenol oxidase (PPO), peroxidase (POD), alpha-amylase, beta-amylase, starch phosphorylase and the antioxidant activity were studied. Results showed that UV-C treatment inhibited browning index and microbial growth by approximately 37% and 32% respectively. Compared with the control, PPO and POD activities of UV-C treated minimally processed lily bulb were reduced by about 26% and 18% respectively, while total phenolic content was increased by 13%. UV-C treatment down-regulated the activities of alpha-amylase, beta-amylase and starch phosphorylase, which resulted in higher content of starch, total soluble sugar content and lower content of reducing sugar. The antioxidant activity and PAL activity were higher than the control during storage. These results indicated UV-C treatment could effectively maintain quality of minimally processed lily bulb stored at 4 +/- 1 degrees C. Therefore, UV-C radiation might be an effective and safe alternative technology for minimally processed lily bulb. (C) 2017 Elsevier B.V. All rights reserved.

Abstract:Mature green cherry tomato fruit were harvested and treated with ultraviolet-C (UV-C) irradiation at a predetermined dose of 4.2 kJ m(-2), and stored at 18 C for 35 days. The effects of UV-C treatment on color change, pigment contents, and the expression of major genes involved in carotenoid metabolism, including Psy 1, Pds, Lcy-beta, and Lcy-epsilon, encoding phytoene synthase, phytoene desaturase, lycopene beta-cyclase and lycopene epsilon-cyclase, respectively, were examined. The UV-C treated fruit developed a pink red color in contrast to the normal orange red color of control fruit. Lycopene accumulation during ripening in UV-C treated fruit was significantly inhibited but its final content was not affected. However, both accumulation and final content of beta-carotene were significantly suppressed in UV-C treated fruit. The lower content of beta-carotene, leading to a higher lycopene to beta-carotene ratio, is probably responsible for the altered color phenotype in UV-C treated fruit. Psy 1, a major gene involved in lycopene synthesis was inhibited by UV-C irradiation. Significantly suppressed expression of Lcy-beta gene was also observed in UVC treated fruit. Thus it is possible that the lower transformation from lycopene to carotenes contributed to the relatively stable content of lycopene. (C) 2013 Elsevier B.V. All rights reserved.

Abstract:Low-temperature storage is widely used as a postharvest treatment applied for delaying senescence in vegetables and ornamentals and ripening in fruits, upholding their postharvest quality. But the refrigerated storage of tropical and subtropical crop plant species provokes a set of physiological alterations known as chilling injury (CI) that negatively affect their quality and frequently renders the product not saleable. The increasing demand for consumption of fresh fruits and vegetables, along with restriction on the use of synthetic chemicals to reduce CI, has encouraged scientific research on the use of heat treatments as an environment-friendly technology for CI mitigation. Membrane damage and reactive oxygen species production are multifarious adverse effects of chilling as oxidative stress in sensitive fruits and vegetables. Chilling mitigation in heat-treated fruits and vegetables could be attributed to (1) enhancement of membrane integrity by the increase of unsaturated fatty acids/saturated fatty acids (unSFA/SFA) ratio; (2) enhancement of heat shock protein gene expression and accumulation; (3) enhancement of the antioxidant system activity; (4) enhancement of the arginine pathways which lead to the accumulation of signaling molecules with pivotal roles in improving chilling tolerance such as polyamines, nitric oxide, and proline; (5) alteration in phenylalanine ammonia-lyase and polyphenol oxidase enzyme activities; and (6) enhancement of sugar metabolism. In the present review, we have focused on the impacts of heat treatments on postharvest chilling tolerance and the mechanisms activated by this environment-friendly technology in fruits and vegetables.

Effects of Postharvest UV-C Treatment on Disease Resistance and Quality in Mandarin Fruit

Abstract:The rotting incidence,main quality parameters and flavonoid and total phenol contents were analyzed in harvested "honey" Mandarin fruit after irradiated with different doses of UV-C and stored at 15 ℃.The result showed that treatments with1.5 and 3 kJ/m2 doses of UV-C,delayed rotting infection by 6 and 12 days,and reduced rotting rate by 42.9% to 85.7% as compared to untreated fruits after 27 days of storage.1.5 and 3 kJ/m2 doses of UV-C irradiation reduced weight loss,increased SSC,SS/TA and vitamin C contents,and promoted synthesis and accumulation of flavonoid and total phenol,therefore maintained better quality and nutrition of Mandarin fruit.1.5 and 3 kJ/m2 doses of UV-C irradiation also improved the content of flavonoid and total phenol in fruit peel.These results demonstrated that UV-C treatment has the potential as a non-chemical tool for extending the shelf life of harvested Mandarin fruit.
Use of UV-C postharvest treatment for extending fresh whole tomato (Solanum lycopersicum, cv. Zinac) shelf-life

Abstract:The effect of UV-C treatments (0.32, 0.97, 2.56, 4.16 and 4.83 kJ.m−2 at 254 nm) on the physical-chemical properties [colour, texture, total phenolic content (TPC), weight loss (WL)], and mesophylic counts of whole tomato, was evaluated during 15 days at 10 °C. During storage, the Ctr samples acquired faster red colour than all UV-C samples (higher a* and lower °h values). Comparing texture of Ctr and UV-C samples at 15th storage day, an increase of 9 and 8 % on firmness of treated samples at low UV-C intensities (0.32 and 0.97 kJ.m−2, respectively) was observed. At the end of the storage, Ctr samples showed ca. 4 Log10 of mesophylic load, and the samples treated at 0.97 and 4.83 kJ.m−2 revealed the lowest microbial load (1.9 and 3.2 Log10, respectively). These results indicate that UV-C radiation, at an appropriate dose, combined with low storage temperature (10 °C) are an effective method to preserve the postharvest life of tomato, without adversely affecting quality parameters.

Abstract:Bell pepper is a model for studying the ripening process of non-climacteric fleshy fruit, during which numerous physiological changes occur. UV-C irradiation is an abiotic stress, which has an important influence on plant growth, development and ripening. We explored the influences of UV-C irradiation on physiological changes in bell pepper fruit during postharvest storage by measuring sensory quality, firmness and mass loss, and the underlying changes at the molecular level by combined transcriptome and metabolome analysis. Effects were detected on transcripts for numerous critical enzymes involved in fruit pigment accumulation, fruit texture change, fruit flavor and aroma production. Moreover, transcripts for several key enzymes and transcription factors involved in plant hormone pathways and their corresponding noncoding RNAs were identified and differential expression profiles were also analyzed. Components of a competing endogenous RNAs (ceRNA) regulatory network changed in response to UV-C irradiation and the relationships between them was analyzed. Thirty-one differentially expressed metabolites were identified. Combined transcriptome and metabolome analysis showed that the genes and metabolites that responded to UV-C were mainly involved in flavonoid, phenylalanine, tyrosine, tryptophan and phenylpropanoid biosynthesis. This study provides insights into bell pepper fruit physiology and the effects of UV-C irradiation on postharvest quality and enhances our understanding of the underlying mechanism whereby UV-C delays postharvest physiological changes in bell pepper fruit.

Abstract:Ultraviolet-B (UV-B) has been confirmed to be harmful to living organisms and it is of concern that the amount of UV-B radiation reaching the earth's surface is increasing because of the depletion of the stratospheric ozone layer. Effect of different levels of tea catechins on morphological damage and expression of chalcone synthase gene in Arabidopsis thaliana plants exposed to UV-B were investigated. Tea catechins at levels 250 - 1000 mgL(-1) screened off UV-B resulting in less expression of chalcone synthase gene and lighter stress of the UV-B exposed plant. The application of tea catechins was propitious to the recovery of the UV-B damaged plants, especially at concentration 250 mg L-1. The protective mechanism of tea catechins against UV-B stress is considered to be their sunscreen property instead of their free radical scavenging and antioxidant properties.
