Abstract:Objective: To study of protective effect of schisandrae chinenesis oil on the islet B cells of type 2 diabetic rats and explore its hypoglycemic effect and mechanism. Methods: The high-fat diet combined with the twice injection of low-dose STZ at the ideal interval was used to establish the experimental rat model of type 2 diabetes. The Wistar model rats were randomly divided into diabetic model group and schisandrae chinenesis oil treatment group, meanwhile control group and normal schisandrae chinenesis oil group were set up. After 6 weeks abdominal injection of rats with schisandrae chinenesis oil, the fasting blood glucose (FBG) and fasting plasma insulin level (FINS) in serum of the rats in various groups were examined) the malondialdehyde (MDA) content, superoxide dismutase (SOD) and catalase (CAT) activities in serum were measured; RT-PCR assay was used to examine the expressions of Bcl-2, Bax and Caspase 3 mRNA. Results: Compared with diabetic model group, the level of FBG in schisandrae chinenesis oil gourp was significantly decreased (P<0.05), the FINS level was markedly increased (P<0.05) s the serum MDA content, and the SOD and CAT activities were increased (P<0.05) s the expression of Bcl-2 mRNA was increased (P<0.05), the expression level of Caspase 3 mRNA was decreased (P<0.05), and the Bax mRNA expression had no change. However there were no differences in these indexes between control group and normal schisandrae chinenesis oil group. Conclusion: Schisandrae chinenesis oil can significantly reduce blood glucose by decreasing lipid peroxidation, increasing the antioxidant capacity of the diabetic rats, regulating apoptosis-related genes, and improving insulin secretion.

Abstract:Islet cell dysfunction in type 2 diabetes is primarily attributed to increased apoptosis of pancreatic β-Cells. The aim of the present study was to investigate the effects of Schisandrae chinensis oil on pancreatic β-Cells in type 2 diabetes mellitus rats and the associated molecular mechanisms of action. Wistar rats were randomly divided into diabetic rats and control rats, diabetic rats treated with Schisandrae chinensis oil (1 mg/kg), and control rats treated with Schisandrae chinensis oil. The serum fasting blood glucose, insulin, total cholesterol, and triglyceride levels along with MDA content, SOD and CAT activities in pancreatic tissues were measured. TUNEL was used to observe the apoptosis of rat pancreatic cells. Western blot was used to determine specific protein expression. The results showed that the oil significantly decreased fasting blood glucose, total cholesterol, triglyceride levels as well as the pancreatic MDA, but increased SOD and CAT activities. The protein expression of Bcl-2, PDX-1, GLUT-2, and GCK but not caspase 3 was significantly enhanced in the oil-treated rats compared with diabetic rats. However, Bax content was not significantly different between the control and DM groups. Schisandra chinensis oil improves pancreatic β-cell function by enhancing antioxidant potential of the pancreas, upregulating the expression of anti-apoptotic genes, increasing expression of glucose metabolism, and delaying islet cell apoptosis.

Abstract:Objective:To investigate the effect of the extract of Schisandra chinensis on the matrix metalloproteinases(MMPs) system in kidney tissue of the diabetic rats,and to explore its protective effect on the kidney tissue from the matrix degradation perspective.Methods:STZ was used to establish rat models of diabetes mellitus.A total of 45 diabetic rats were randomly divided into model group,extract of Schisandra chinensis group and Benazepril group,and there were 15 rats in each group.Another 15 rats were selected and used as normal control group.12 weeks after administration,the routine blood and urine biochemical indexes,the histological changes,blood glucose (BG),blood urea nitrogen(BUN),serum creatinine(Scr),high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterin(LDL-C),total cholesterol(T-CHO),and triglyceride(TG) levels,excretion rates of albuminuria and proteinuria of the rats in various groups were detected;the expression amounts of fibronectin (FN),type Ⅳ collagen (Col Ⅳ),and matrix metalloproteinase inhibitor metalloproteinase-2 (TIMP-2) in kidney tissue of the rats were detected by immunohistochemical method;the activity of matrix metalloproteinase-2 (MMP-2) was detected by zymography.Results:Compared with model group,the glomeruli matrix accumulation of the rats in extract of Schisandra chinensis group was significantly improved,the excretion rate of albuminuria,LDL-C level and serum MDA level were decreased(P<0.05),the activities of CAT(P<0.01)and SOD(P<0.05)in kidney tissue were increased,and the level of MDA in kidney tissue was decreased(P<0.05).The immunohistochemistry results showed that compared with model group,the expression amounts of FN,Col Ⅳ,and TIMP-2 in kidney tissue of the rats in extract of Schisandra chinensis group were significantly decreased.The zymography results showed that compared with model group,the activity of MMP-2 in kidney tissue of the rats in extract of Schisandra chinensis group was significantly increased(P<0.05).Conclusion:Extract of Schisandra chinensis has protective effect on the kidney tissue of the diabetic rats induced by STZ,and the mechanism may be related to the inhibition of oxidative stress and the improvement of MMP-2 activity as well as the inhibition of TIMP-2 expression which could improve the matrix degradation.

Abstract:OBJECTIVE:To study the effect of total polyphenol of Rhodiola sachalinensis on high fat diet-STZ induced type 2 diabetic rats.METHODS:36 wistar rats were randomly divided into 6 groups:normal control group(constant volume normal saline),model group(constant volume normal saline),Metformin hydrochloride tablet group(0.3 g·kg-1),R.sachalinensis total polyphenol high-dose,medium-dose and low-dose groups(100,50,25 g·kg-1).Those groups were given medicine intragastrically once a day for 21 days.12 h after last administration,blood samples were taken from tail vein.The blood glucose(BG) level was determined.Abstaining from food but not drink for 18 h,the activity of SOD and the content of MDA in pancreas cell mitochondria were determined.The anti-hyperglycemic effect of R.sachalinensis total polyphenol was evaluated.RESULTS:BG level of model group was significantly higher than that of normal control group(P0.01).Compared with model group,BG levels of R.sachalinensis total polyphenol low-dose,medium-dose and high-dose groups decreased significantly(P0.01),but there was no significant difference between R.sachalinensis total polyphenol middle-dose group and Metformin hydrochloride tablet group.Compared with normal control group,the content of MDA in pancreas cell mitochondria in model group increased significantly(P0.01),but SOD activity significantly reduced(P0.01).Compared with model group,the content of MDA in R.sachalinensis total polyphenol groups reduced significantly(P0.01),and SOD activity increased significantly(P0.01).CONCLUSION:Total polyphenol of R.sachalinensis have good anti-hyperglycemic effect,and it is probably due to antioxidant effect of polyphenols in it.

Abstract:Aim To study whether neferine influences urinary metabolites of type 2 diabetic mellitus rats.Methods The rats were fed with high-sucrose and high-fat chow for 4 weeks,then were injected intraperitoneal(ip) with STZ(30 mg·kg-1).One week later,rats with fasting blood sugar levels higher than 7.8 mmol·L-1 were defined as type 2 diabetes mellitus(T2DM) rats.Neferine treated T2DM rats were injected intraperitoneal(ip) with neferine(20 mg·kg-1·d-1) for 4 weeks.Fasting blood sugar of all rats was measured after 4 weeks.24 h urine samples from rats were collected and prepared.All the separations were performed on UPLC/QTOF-MS system.Results The blood glucose values in neferine treated T2DM rats were decreased in comparison with those in T2DM rats(P0.05).Dihydroxyfumaric acid in urinary metabolite profiles of neferine treated T2DM rats was decreased compared with that in T2DM group.Increase of PGI2,LXA4 and LXB4 was revealed in the urinary metabolite profiles of neferine treated T2DM group than that in T2DM group.Conclusions Neferine can decrease blood glucose and change urinary metabolite profiles of T2DM rats.The mechanism of neferine may be associated with antioxidant stress and anti-inflammatory effect.

Abstract:AIM: To study the effect of ligustrazine on the cardiacmyocyte lesion in rats with type 2 diabetes mellitus.METHODS: Male Wistar rats were injected with STZ via tail vein under high-glucose and high-fat feeding for 4 weeks to establish the animal model of type 2 diabetes mellitus.Ligustrazine at different doses was used to treat the diabetic rats.The body weight, blood glucose and the morphology of heart tissues were observed.The myocardial levels of IL-1β, IL-6 and TNF-α were detected by ELISA, and the protein expression of IKKβ and NF-κB in the myocardium was determined by Westeren blotting.RESULTS: Ligustrazine at high dose alleviated the body weight reduction and blood glucose elevation cause by diabetes, and reduced pro-inflammatory factors IL-1β, TNF-α and IL-6.Moreover, the protein expression of IKKβ and NF-κB was significant decreased by ligustrazine.CONCLUSION: Ligustrazine inhibits the myocardial inflammation caused by diabetes through anti-inflammatory pathway.

Abstract:Objective: To study the effect of Dtm volatile oil on type II diabetes(T2D) rats and its mechanism.Methods: T2D rat is established by streptozotocin(STZ 50mg/kg) and hyperotophic food,then observe the hypoglycemic effects.Fasting blood glucose(FBG),serum insulin level,fat content and 125I-insulin receptor to liver plasmamembrane of the T2D rats are measured.Results: Dtm volatile oil could significantly lower FBG,improve dyslipidemia,make insulin level norma1 and remarkably raise the level of the low-affinity insulin receptor.Compared with model group,they have a conspicuous difference(P﹤0.01).Conclusion: Dtm volatile oil has an obvious therapeatic effect on T2D rats.Its mechanism is related to improve the insulin receptor level and raise the insulin receptor number.

Abstract:【Objective】 To study the effect of licorice flavonoids(LF) on antioxidative ability in type 2 diabetic rats.【Methods】 The type 2 diabetic rat model was replicated by high-fat and high-sugar diet combined with low-dose streptozotocin(STZ) injection.Rats in control and model groups were administered with solvent(1.2-propanediol).Meanwhile,rats in control treatment group(CLF) and licorice flavonoids treatment group(LF) were administered with 300 mg/(kg·d) of LF.After 6 weeks,the levels of the FBG,T-AOC,SOD,GSH-PX and MDA in serum were examined.【Results】 FBG in LF group was significantly lower than those in DM group(P 0.01).Comparing with DM group,GSH-PX and SOD activities and T-AOC level in serum were much higher(P 0.01 or P 0.05 P 0.05);MDA level in serum in LF group was much lower(P 0.05).【Conclusion】 LF has the significant effect improving antioxidant ability in the type 2 diabetic rats.

Abstract:Objective To study the anti-diabetic effect and mechanism of Folium Clausenae Lansii(FCL) on streptozocin(STZ)-induced diabetic rats. Methods Rats were randomized into 6 groups, namely normal control group,model group,glibenclamide group,and high-,medium- and low-dose FCL groups. Except for the normal control group,rats in other groups were given intraperitoneal injection of STZ to induce type 2 diabetes,and then were given solvent,glibenclamide,high-,medium- and low-dose FCL respectively according to the grouping. Effects of FCL on fasting blood glucose(FBG),oral glucose tolerance test(OGTT),fasting insulin(FINS),lipid parameters, oxidative stress, and histological features of pancreatic tissue were investigated. Results FCL could decrease the content of FBG and blood lipid levels,increase oral glucose tolerance and FINS level,enhance serum superoxide dismutase(SOD),hydrogen peroxidase(CAT)and glutathione(GSH)activities,reduce malondialdehyde(MDA)and nitric oxide(NO)contents ,and relieve the pathological changes of pancreatic gland. Conclusion FCL has obvious anti-diabetic and anti-hyperlipidemia effects on diabetic rats, and the mechanism may be related with the enhancement of insulin secretion and antioxidant effect.

Abstract:Objective To develop a rat model of type 2 diabetic mellitus and study the protective effects of peanut oil on improvement and treatment of type 2 diabetic mellitus.Methods After 2 months of high-fat food feeding,type 2 diabetic mellitus models were prepared with low dose STZ intraperitoneal injection in 8 weeks old SD rats.Then they were treated with different dose of peanut oil.One month later,the rats were examined with Morris water maze test and then the levels of serum lipids,serum insulin and blood glucose were measured.Results The rat models were characterized by hyperglycemia,hyperlipemia and hyperinsulinemia which resembled the clinical phenotype of type 2 diabetes.The results of Morris water maze test : there was no difference among 4 groups(P>0.05).The levels of blood glucose、serum insulin and serum lipids were significantly lower in 5 mL peanut oil group than those of in diabetic model group(P<0.05).Conclusion The data of Morris water maze test suggests that there is no cognitive impairment in early stage of type 2 diabetic mellitus.The decrease of hyperglycemia,hyperlipemia and hyperinsulinemia with peanut oil indicates that peanut oil can improve and treat the type 2 diabetic mellitus.
Effect of Shuangyi Jiangtang Preseription on Metabolism of Blood Glucose and Blood Lipids in Type 2 Diabetic Rats

Abstract:Objective:To investigate the effect of the Shuangyi Jiangtang prescription on the blood glucose and blood lipids in type 2 diabetic rats.Method: rats model of type 2 diabetes mellitus was established by taking high fat diet for 4weeks and combined with STZ ip for 40 mg · kg-1,and the model rats were randomly divided into diabetes model group(T2DM),rosiglitazone maleate positivecontrol groups(RM,ig 0.29 mg · kg-1 ·d-1),Shuangyi Jiangtang prescription high dose group(SG,ig 4 000 mg · kg-1 · d-1),Shuangyi Jiangtang prescription low dose group(SD ig 1 000 mg · kg-1 · d-1),and the normal control group(NC).We had observed the general condition of rats during and after the whole treatment of 4 weeks,and tested glucose tolerance,glycated hemoglobin(HbA1c),blood lipids,and fasting serum insulin(INS).Result: In the group of SG,the rats' symptoms of polydipsia,polyuria and mental status were improved in some degree,fasting blood glucose(FBG),0.5 h blood glucose(BG) and 2 h BG were lower than the T2DM group,the fasting serum insulin level was higher than the T2DM group(P0.05).Compared with the model group,fasting blood glucose,glycosylated hemoglobin,lipids were improved in some degree,but there was no significant statistics difference.In the groups of SD and RM,the general condition of rats were bettered,and compared with the model group,the glucose tolerance,HbA1c and the blood lipid of the rats decreased significantly(P0.05),and the serum insulin increased significantly(P0.05),but between the two groups of SD and RM,there was no difference in those indexes.Conclusion: The Shuangyi Jiangtang prescrription can improve sugar,lipid metabolism and islet cells function of type 2 diabetic rats,the low-dose group has the best effect,and its efficacy is similar to the rosiglitazone maleate.

Abstract:Objective To study the effect of licorice flavonoids(LF) on fasting blood levels of glucose(FBG),lipids,insulin,adipokines and other biochemical indicators. Methods The type 2 diabetic rat model was replicated by feeding high-fat and high-sugar diet combined with low-dose streptozotocin(STZ) injection.The diabetic rats were divided into model group(DM),high-dose LF treatment group(HLF) and low-dose LF treatment group(LLF).Other 20 normal rats were divided into control group(Con) and control treatment group(CLF).Rats in different groups were administered LF(100mg or 300mg·kg-1·d-1) or solvent(propylene glycol) respectively.Five weeks later,the levels of fasting levels of glucose(FBG),triglycerides(TG),total cholesterol(TC),free fatty acids(FFA),high-density lipoprotein cholesterol(HDL-C),low-density lipoprotein cholesterol(LDL-C),total antioxidation capability(T-AOC),insulin(Ins),tumor necrosis factor alpha(TNF-α) and leptin were examined.Insulin sensitivity index(ISI),liver index,kidney index and the relative weight of abdominal adipose tissue were calculated. Results The high-dose LF(300mg·kg-1·d-1) treatment significantly decreased the levels of FBG,TG,TC,LDL-C,FFA,T-AOC,TNF-α,leptin,liver index and kidney index,and increased the levels of T-AOC,ISI and the relative weight of abdominal adipose tissue.Compared with high-dose treatment,low dose of LF was less effective. Conclusion LF significantly decreases the blood glucose level,improves the lipid metabolic disorders and ameliorates insulin resistance in type 2 diabetic rats.

Abstract:Objeitive: To evaluate the influence of Olea europaea L. leaf extracts on the blood glucose and the blood lipid of type 2 diabetic mellitus rats and discuss its mechanism. Methods: With metformin used as control, the influence of Olea europaea L. leaf extracts (500, 250, 125, 62. 5 mg/kgB W) was evaluated on blood glucose and blood lipid of type 2 diabetic mellitus rats induced by high-glucose-fat diet and STZ, through the determination of FBG, AUC, FINS, HOMA-IR, ISI, serum lipid and GSP. Results: Olea europaea L. leaf extracts (500, 250, 125 mg/kgB W) significantly decreased levels of FBG and AUC. Rats received daily treatment for 5 weeks, levels of serum TG, TCHO, LDL-C, NEFA, GSP and HOMA-IR were significantly lower, while serum HDL-C and ISI were higher, compared with the model group. Conclusions: Olea europaea L. leaf extracts has a significant effect of lowering blood sugar on diabetic mellitus rats, can improve the resistance of glucose and insulin resistance, and can adjustment blood lipid metabolismabnormality.

Abstract:【Objective】To explore the effect of 18β-glycyrrhetinic acid on fasting blood glucose(FBG),lipids and sensitivity of insulin in type 2 diabetic rats.【Methods】Type 2 diabetes model was replicated by high-fathigh-sugar diet combined with lower dose STZ injection.All rats were randomly divided into five groups: control group(CON),control 18β-glycyrrhetinic acid treatment group(CGA),diabetes group(DM),DM+18β-glycyrrhetinic acid treatment group(DGA) and DM+glibenclamide treatment group(DGL).They were orally administered with 5% DMSO,18β-glycyrrhetinic acid[100mg/(kg·d)] and glibenclamide [600μg/(kg·d)] respectively.After 45 days of continuous medication,detect fasting blood glucose(FBG),lipids(TG,TC,LDL-C,HDL-C) and insulin level in serum of all rats.【Results】18β-glycyrrhetinic acid treatment significantly decreased the levels of FBG,TG,TC,LDL-C and homeostasis model assessment of insulin resistance(HOMA-IR) in diabetic rats,and also increased anti atherosclerosis index(AAI).【Conclusion】18β-glycyrrhetinic acid has hypoglycemic and hypolipidemic effects,and it can also ameliorate insulin resistance.

Abstract:Background: Type II diabetes has become a serious threat to human health in recent years. Among adults above 20 years old in China, the prevalence rate of diabetes is 9.7%. Thus, it is imperative to study the mechanisms underlying type II diabetes to develop effective therapeutic treatments. Objective: To examine metabolic changes in a rat model of type II diabetes and explore mechanisms underlying the beneficial effects of traditional Chinese medicine (TCM) in this model. 120 rats were divided into four groups, including a control group, a high-fat diet group (high-fat diet and streptozotocin injection), a TCM group (high-fat diet, streptozotocin injection, followed by TCM administration), and a rosiglitazone maleate group (high-fat diet, streptozotocin injection, followed by rosiglitazone maleate administration). Metabolites in urine samples from 1-3 weeks (time point 1) and 4-6 weeks (time point 2) of drug administration were compared by gas chromatography-mass spectrometry (GC-MS). Results: Our results showed that in the high-fat diet group, at time point 2, the levels of dihydroxybenzoic acid, L-ascorbic acid, D-gluconic acid, octadecanoic acid, and glutaric acid in urine were significantly higher than at time point 1. In the TCM group, at time point 2, the urine levels of L-ascorbic acid were markedly lower than at time point 1. Conclusion: Our studies demonstrated that examining urine metabolic changes provided important insights into the mechanisms underlying type II diabetes as well as the therapeutic effects of TCM.

Abstract:The effect of Radix Scutellariae treated on type 2 diabetic rats has been investigated by a liquid chromatography coupled with tandem mass spectrometry (LC–MS/MS) based urinary quantitative approach. In this research, multiple reactions monitoring mode of MS/MS in LC–MS/MS analysis was used to quantitatively analyze the concentrations of 7 endogenous compounds in urine of normal control group, type 2 diabetic model group and Radix Scutellariae-treated group, and multivariate statistical analysis was utilized for MS data processing. The above-mentioned three groups can be distinguished via pattern recognition. The obtained results indicated that Radix Scutellariae affect the urinary metabolic profiling of type 2 diabetic rats on the polyol pathway, protein glycation reaction and amino acids metabolism pathway. According to these results, Radix Scutellariae should have the pharmacological effect on preventing or delaying the onset and progression of diabetes and its complications.
Hypoglycemic effect of fermented Inonotus obliquus on type II diabetic rats induced by streptozotocin

Abstract:To test effects of FIO on blood glucose, blood lipid, and antioxidation in type Ⅱ diabetes, the type Ⅱ diabetic rat model was established by high fat diet-feeding combined with intraperitoneal injection of STZ.The results showed that FIO significantly lowered FBG levels, promoted synthesis of HGln, and improved insulin resistance in the type Ⅱ diabetic rats, compared to untreated control diabetic animals.In addition, FIO not only significantly decreased TC, TG, FFA, LDL and MDA in serum, but also dramatically increased the HDL in serum and the activities of SOD in type Ⅱ diabetic rats.Our study showed that FIO significantly lowered high blood glucose, improved lipid metabolism and antioxidant ability.

Abstract:Type 2 diabetes mellitus (T2DM) is a metabolic disease characterized by hyperglycemia. The aim of this work was to investigate the effect of sciadonic acid (SA) on disorders of glucolipid metabolism and intestinal flora imbalance and to further investigate its potential molecular mechanism of anti-diabetes. The experimental data indicated that SA could alleviate hyperlipidemia, insulin resistance, oxidative stress, the inflammatory response, repair liver function damage, and promote glycogen synthesis caused by T2DM. SA could also activate the PI3K/AKT/GLUT-2 signaling pathway, promote glucose metabolism gene expression, and maintain glucose homeostasis. Furthermore, 16S rRNA analysis revealed that SA could reduce the Firmicutes/Bacteroidota (F/B) ratio; promote norank_f__Muribaculaceae, Allobaculum, Akkermansia, and Eubacterium_siraeum_group proliferation; increase the levels of major short-chain fatty acids (SCFAs), such as acetic acid, propionic acid, and butyric acid; and maintain the homeostasis of the intestinal flora. In conclusion, these results suggested that SA could reshape the structural composition of intestinal microbes, activate the PI3K/AKT/GLUT2 pathway, improve insulin resistance, and decrease blood glucose levels.
