Abstract:We report the first example of highly enantioselective organocatalytic synthesis of 3-difluoroalkyl substituted 3-hydroxyoxindoles. The total synthesis of the difluoro analogue of convolutamydine E was achieved by this method.

Abstract:Highly enantioselective synthesis of 1,3-bis(hydroxymethyl)-2-oxindoles was established through chiral Nd-III-N,N'-dioxide in up to 93% yield and >99% cc under mild conditions. The key features of this approach include using cheap and readily available formalin as a hydroxymethylation Cl unit and unprotected 3-substituted-2-oxindoles as nucleophiles directly. The initial C-aldol product from 3-substituted-2-oxindole was converted to the corresponding 1,3-bis(hydroxymethyl)-2-oxindole derivative immediately under the reaction conditions. Moreover, the catalyst was water-tolerant, and ligand and Nd(OTf)(3) could be easily recovered and reused with no loss in catalytic activity and enantioselectivity. The synthetic utility of this methodology was accomplished with the synthesis of key intermediates of physostigmine and coerulescine.
Catalytic asymmetric synthesis of 3-(alpha-hydroxy-beta-carbonyl) oxindoles by a Sc(III)-catalyzed direct aldol-type reaction.

Abstract:A direct catalytic asymmetric aldol-type reaction of 3-substituted-2-oxindoles with glyoxal derivatives and ethyl trifluoropyruvate, catalyzed by a chiral N,N'-dioxide-Sc(OTf)(3) (Tf = trifluoromethanesulfonyl) complex, has been developed that tolerates a wide range of substrates. The reaction proceeds in good yields and excellent enantioselectivities (up to 93% yield, 99:1 diastereomeric ratio (dr), and >99% enantiomeric excess (ee)) under mild conditions, to deliver 3-(alpha-hydroxy-beta-carbonyl) oxindoles with vicinal quaternary-tertiary or quaternary-quaternary stereocenters. Even with 1 mol % catalyst loading or on scaleup (10 mmol of starting material), maintenance of ee was observed, which showed the potential value of the catalyst system. In studies probing the reaction mechanism, a positive nonlinear effect was observed and Sc(III)-based enolate intermediates were detected by using ESIMS. On the basis of the experimental results and previous reports, a possible catalytic cycle was assumed.

Abstract:Asymmetric synthesis of 3-(3-indolomethyl)oxindoles through the addition of indole-substituted enolized ketoesters to 3-bromo-3-substituted oxindoles has been achieved using a N,N'-dioxide/Ho(III) complex. A number of 3-(3-indolomethyl)oxindoles, which may possess biological activity, were obtained in good yields with high diastereo- and enantioselectivities (up to 97% yield, >19 : 1 dr, 98% ee). Furthermore, time-dependent reversal of diastereoselectivity enabled access to optically active diastereomers. The product followed by facile transformations gave a new route into trigolute analogs.

Abstract:We describe here the first organocatalytic asymmetric synthesis of densely functionalized spiro-δ-lactam oxindoles featuring multiple stereogenic centers through a [3+3] annulation reaction. This protocol provides a novel, simple and efficient strategy for the synthesis of biologically important spiro-δ-lactam oxindoles with a range of substrates, excellent diastereoselectivities and enantioselectivities (up to >95:5 dr and 99% ee).

Abstract:Nickel‐catalyzed asymmetric intramolecular addition of aryl halides to α‐ketoamides has been achieved to afford chiral 3‐substituted‐3‐hydroxy‐2‐oxindoles in excellent yields and high enantioselectivities (up to 99% yield and 98% ee), which provides efficient access to valuable molecules containing 3‐hydroxy‐2‐oxindole core. The gram‐scale reaction proved the potential utility of the methodology.

Abstract:The direct asymmetric vinylogous Michael addition of 3,5-dialkyl-substituted 4-nitroisoxazoles with alpha,beta-unsaturated aldehydes catalyzed by a diphenylprolinol TBS ether organocatalyst is described, giving products with moderate to good yields and up to excellent enantioselectivities (96% ee). This approach provides an easy access to highly functionalized chiral isoxazole derivatives.

Abstract:An enantioselective intramolecular heterocyclization with in situ generated 3-hydroxyisoindolinone-derived N -acyliminium ions has been successfully accomplished. In the presence of a catalytic amount of a chiral phosphoric acid, functionalized 3,3-disubstituted isoindolinones bearing N -acyl- N , O -acetal moieties were obtained with good yields and a high level of stereocontrol (up to 98:2 er). This efficient method proceeds under mild conditions and exhibits broad scope with respect to both 3-hydroxyisoindolinones and hydroxyl partners. Abstract:An organocatalyzed asymmetric dearomative [3+2] annulation reaction of electron-deficient 2-nitrobenzo heteroarenes including 2-nitrobenzofurans, 2-nitrobenzothiophenes and 2-nitroindoles with 3-isothiocyanato oxindoles was developed. With a multiple hydrogen-bonding bifunctional thiourea as the catalyst, the reaction displays broad substrate scope and provides an access to structurally diverse polycyclic spirooxindoles containing three contiguous stereogenic centers in high yields with good diastereo- and enantioselectivities (up to 99% yield, 94:6 dr, and 97% ee). This organocatalysis protocol is able to afford the products with complementarily diastereoselectivity versus the previous work with transition metal catalysis.

Abstract:An organocatalytic and highly diastereo- and enantioselective reaction of 3-aminooxindoles with 2-enoylpyridines for the synthesis of chiral spiro[pyrrolidin-3,2'-oxindole] derivatives has been achieved. With the cinchonidine-based thiourea catalyst, the asymmetric Michael/cyclization reaction sequence of 3-aminooxindoles with 2-enoylpyridines followed by the dehydration and deprotection with concentrated hydrochloric acid provided a wide range of chiral 3',4'-dihydrospiro[indoline-3,2'-pyrrol]-2-ones, bearing two adjacent tri- and tetrasubstituted stereocenters, in moderate to good yields with overall excellent stereoselectivities. The synthetic application of this methodology was presented by the scale-up experiment and transformation of product 5a into the spiro-oxindole-based pyrrolidine 6. Furthermore, a plausible transition state for this cascade reaction sequence was also proposed. And this work will provide a new way to access chiral spiro[pyrrolidin-3,2'-oxindole] derivatives.

Abstract:Asymmetric construction of pharmacologically interesting tetrahydrofuranyl spirooxindole frameworks has been achieved through organocatalytic [3+2] annulations of the readily available 3‐hydroxyoxindoles and pyrrolidone‐derived cyclic ketolactams. A variety of chiral spiro tetrahydrofuranyl products, which contain four contiguous stereocenters including two tetrasubstituted carbon centers, have been rapidly synthesized with remarkable results (up to 99% yield, >95:5 dr, and 99:1 er). Synthetic derivatization of the hemiketal moiety enables the installation of various halogen atoms into the structurally complex molecules in a stereospecific manner. Preliminary screening of anticancer bioactivity was performed, and 4w showed obvious inhibitory capacity to the proliferation on a panel of cancer cell lines.

Abstract:A novel asymmetric (4 + 3) cycloaddition of indole-2,3-quinodimethanes in situ generated from 3-methyl-2-indolylmethanols with 3-indolylmethanols via chiral phosphoric acid catalysis has been established. The cycloaddition reaction exhibits a broad substrate scope affording the diverse enantioenriched cyclohepta fused diindoles in high yields with good enantioselectivities. Significantly, this work represents the first application of 3-methyl-2-indolylmethanols as 4C synthons instead of the commonly reported three-atom synthons in cycloaddition reactions.

Abstract:Asymmetric synthesis of enantioenriched C3-arylated oxindoles bearing tetrasubstituted stereocenter has been achieved using chiral N,N′-dioxide/transition metal complex which promote Friedel–Crafts alkylation of 3-bromo-3-substituted oxindoles with arenes and heteroarenes i.e., anisole derivatives, furans, pyrroles, 2-methoxythiophene, anilines, and phenols. A series of 3-aryl-indolinones with quaternary stereocenters have been obtained up to 99% yield, 99% ee. In addition, the rationale of the reactivity is based on the nucleophilicity parameters of arenes and heteroarenes were predicted from Mayr's Database. Also, a gram-scale synthesis has been achieved and this method has been applied to synthesis of bioactive compounds. Additionally, a plausible catalytic cycle has been proposed.

Abstract:An efficient organocatalytic conjugated addition reaction of pyrazol-3-ones with 3-trifluoroethylidene oxindoles has been developed for the synthesis of enantioenriched triflouromethylated indolin-2-ones bearing adjacent tertiary chiral centers in good yields and good to excellent diastereo- and enantioselectivities. Abstract:Chiral phosphoric acid-catalyzed enantioselective aza-Friedel-Crafts reaction of trifluoromethyl benzoxazinones with pyrroles is reported. Under mild conditions, a range of enantioenriched dihydrobenzoxazinones bearing trifluoromethylated quaternary stereocenters could be obtained in good to excellent yield and ee. A remarkable fluorine effect is observed, and preliminary mechanistic studies combined with theory calculations suggest that triple-hydrogen-bonding interactions hold the transition structure rigidly and allow the bulky substituents of the catalyst to influence the enantioselectivity.

Abstract:HHerein, we disclosed the asymmetric construction of an oxa‐quaternary stereocenter via an intramolecular oxa‐Michael (IOM) reaction in β‐substituted ortho‐hydroxymethyl chalcone by the formation of 1,1‐disubstituted‐1,3‐dihydroisobenzofuran using cinchona alkaloid‐based chiral amino‐squaramide catalyst. Both the (E‐ and Z)‐β‐substituted ortho‐hydroxymethyl chalcone provide (S)‐ and (R)‐enantiomers of the 1,1‐disubstituted‐1,3‐dihydroisobenzofuran with excellent stereospecificity. In general, excellent yields (up to 95%) and enantioselectivity (up to 98% ee) were obtained. Furthermore, the resulting 1,1‐disubstituted isobenzofuran or phthalan was converted to corresponding chiral 3,3‐disubstituted phthalides without losing the enantioselectivity. This methodology provides the core moiety of the (S)‐citalopram drug.

Abstract:The first organocatalytic enantioselective Michael/cyclization domino reaction between 3-amideoxindoles and α,β-unsaturated aldehydes is described. After sequential oxidation with pyridinium chlorochromate, a direct and one-pot preparation of highly sterically hindered spirocyclic oxindole-γ-lactams was achieved in 51-81% yields with 75-97% ee and ≤80/20 dr.
