Spectroscopy studies on the interaction between bovine serum albumin and colloidal spheres self-assembled from azobenzene-containing amphiphilic diblock copolymers

Abstract:An amphiphilic azobenzene diblock copolymer {2-[4-(4-methoxy phenyl azo)phenoxy]hexyl acrylate co-acrylic acid} (PAzoM-b-PAA) was synthesized via reversible addition-fragmentation chain transfer polymerization. The diblock copolymer self-assembled into spheres with nanostructure in DMSO/H2O solution. The binding interaction of the PAzoM-b-PAA colloidal spheres with bovine serum albumin (BSA) was investigated by fluorescence spectra, ultraviolet visible (UV-Vis) spectra and transmission electron microscopy (TEM) techniques. Mechanism discussion results indicated that the azo polymer colloidal spheres caused the fluorescence quenching of BSA by a dynamic quenching procedure combined with the formation of the BSA spheres complex. TEM was used to confirm the adsorption of BSA Onto the micro-spheres. Quenching constants and the number of binding sites were determined according to the Stern-Volmer equation. The thermodynamic parameters Delta H-0, Delta S-0, and Delta G(0) (at 298, 309, and 314 K) were calculated, indicating that hydmphobic forces play a major role in PAzoM-b-PAA colloidal spheres BSA association. Synchronous fluorescence spectra were applied to investigate the conformation change of the BSA.
