Abstract:OBJECTIVE:Structure-similar ATP-binding cassette (ABC) transporter proteins, BbT1 in Beauveria bassiana (Bb) and IfT1 in Isaria fumosorosea (If), were characterized.METHODS:Various phenotypes including cellular antioxidant response, multidrug resistance and virulence were compared between wild-type strain and the constructed mutants DeltaBbT1, DeltaBbT1/BbT1 and DBbT1/IfT1.RESULTS:Compared with the wild-type, DeltaBbT1/BbT1 and DeltaBbT1/ IfT1 strains showing no significant changes in examined phenotypes, DeltaBbT1 became 27% to 2.1-fold less tolerant to the oxidative stress of 20-40 mmol/L H2O2 and 2-8 mmol/L menadione, 28% to 4.7-fold less resistant to carbendazim, itraconazole, dimetachlone, cycloheximide, ethirimol and 4-nitroquinoline N-oxide, and about 20% less virulent to the second-instar larvae of Spodoptera litura.CONCLUSION:The ABC transporter proteins BbT1 and IfT1 are similar not only in sequence structure but also in biological function, act as one of the determinants for the multidrug resistance of Bb and If, and also contribute to the fungal virulence due to involvement in cellular antioxidant response.
Mutation in Cyp51a and High Expression of Efflux Pump Gene of Aspergillus Fumigatus Induced by Propiconazole in Liquid Medium and Soil.

Abstract:Triazole resistance in Aspergillus fumigatus is a major cause of clinical inefficacy in the treatment of invasive aspergillosis (IA). The hypothesis that triazole fungicides have driven the development of resistance in A. fumigatus has garnered substantial attention due to the similar structure and global detection of antifungal resistant A. fumigatus (ARAF) isolates in the soil. However, there is little evidence linking the application of triazole fungicides to the emergence of ARAF in the soil. This study was conducted to test if the resistance in A. fumigatus and its associated mutations in cyp51A could be induced by propiconazole in liquid medium and soil. The results indicate that propiconazole can induce resistance by alteration of G138S in cyp51A, and the overexpression of cyp51A, AfuMDR3 and AfuMDR4. G138S in cyp51A was first detected in the soil and associated with resistance. The emergence of the ARAFs in the soil may depends upon the level of propiconazole, and the number of ARAFs in soil treated with propiconazole at 2- and 5-fold dose was much greater than those in soil treated at the recommended dosage. The current data indicate that propiconazole can induce triazole resistance in A. fumigatus and should be applied for agricultural purposes at levels at or below the recommended dosage to avoid the emergence of ARAF in the soil.

Abstract:Carbohydrate transporters are critical players mediating nutrient uptake during saprophytic and pathogenic growth for most filamentous fungi. For entomopathogenic fungi, such as Beauveria bassiana, assimilation of α-glucosides, in particular, trehalose, the major carbohydrate constituent of the insect haemolymph, has been hypothesized to represent an important ability for infectious growth within the insect hemocoel. In this study, a B. bassiana α-glucoside transporter homolog was identified and genetically characterized via generation of a targeted gene disruption mutant. Trehalose utilization was compromised in the mutant strain. In addition, inactivation of the α-glucoside transporter resulted in decreased conidial germination, growth, and yield on various carbohydrates (α-glucosides, monosaccharides and polyols) as compared to the wild-type strain. Insect bioassays revealed decreased mean lethal mortality time using both topical and intrahemocoel injection assays, although final mortality levels were comparable in both the mutant and wild type. Gene expression profiles showed altered expression of other putative transporters in the knockout mutant as compared to the wild type. These results highlighted complex sugar utilization and responsiveness in B. bassiana and the potential role for trehalose assimilation during fungal pathogenesis of insects.

Abstract:Resistance to solar ultraviolet (UV) irradiation is crucial for field-persistent control efficacies of fungal formulations against arthropod pests, because their active ingredients are formulated conidia very sensitive to solar UV wavelengths. This review seeks to summarize advances in studies aiming to quantify, understand and improve conidial UV resistance. One focus of studies has been on the many sets of genes that have been revealed in the postgenomic era to contribute to or mediate UV resistance in the insect pathogens serving as main sources of fungal insecticides. Such genetic studies have unveiled the broad basis of UV-resistant molecules including cytosolic solutes, cell wall components, various antioxidant enzymes, and numerous effectors and signaling proteins, that function in developmental, biosynthetic and stress-responsive pathways. Another focus has been on the molecular basis and regulatory mechanisms underlying photorepair of UV-induced DNA lesions and photoreactivation of UV-impaired conidia. Studies have shed light upon a photoprotective mechanism depending on not only one or two photorepair-required photolyases, but also two white collar proteins and other partners that play similar or more important roles in photorepair via interactions with photolyases. Research hotspots are suggested to explore a regulatory network of fungal photoprotection and to improve the development and application strategies of UV-resistant fungal insecticides. © 2021 Society of Chemical Industry.
Differential Contributions of Five ABC Transporters to Mutidrug Resistance, Antioxidion and Virulence of Beauveria Bassiana, an Entomopathogenic Fungus

Abstract:Multidrug resistance (MDR) confers agrochemical compatibility to fungal cells-based mycoinsecticdes but mechanisms involved in MDR remain poorly understood for entomopathogenic fungi, which have been widely applied as biocontrol agents against arthropod pests. Here we characterized the functions of five ATP-binding cassette (ABC) transporters, which were classified to the subfamilies ABC-B (Mdr1), ABC-C (Mrp1) and ABC-G (Pdr1, Pdr2 and Pdr5) and selected from 54 full-size ABC proteins of Beauveria bassiana based on their main domain architecture, membrane topology and transcriptional responses to three antifungal inducers. Disruption of each transporter gene resulted in significant reduction in resistance to four to six of eight fungicides or antifungal drugs tested due to their differences in structure and function. Compared with wild-type and complemented (control) strains, disruption mutants of all the five transporter genes became significantly less tolerant to the oxidants menadione and H₂O₂ based on 22-41% and 10-31% reductions of their effective concentrations required for the suppression of 50% colony growth at 25°C. Under a standardized spray, the killing actions of ΔPdr5 and ΔMrp1 mutants against Spodoptera litura second-instar larvae were delayed by 59% and 33% respectively. However, no significant virulence change was observed in three other delta mutants. Taken together, the examined five ABC transporters contribute differentially to not only the fungal MDR but antioxidant capability, a phenotype rarely associated with ABC efflux pumps in previous reports; at least some of them are required for the full virulence of B. bassiana, thereby affecting the fungal biocontrol potential. Our results indicate that ABC pump-dependent MDR mechanisms exist in entomopathogenic fungi as do in yeasts and human and plant pathogenic fungi.
[Intraspecific Variations of Paecilomyces Fumosoroseus in Temperature Fitness and Carbendazim Resistance].

Abstract:A total of six wild strains of fungal biocontrol agent Paecilomyces fumosoroseus were grown at 15 degrees C-35 degrees C to reveal their variations in colony growth, conidiation, and conidial viability. The results showed that the test three indices were the best at 25 degrees C, but differed significantly at different temperatures for a given strain or at a given temperature among test strains. The strains Pfr116 and Pfr6206 behaved better or more stably than other strains at 15 degrees C-35 degrees C. The sensitivities of the fungal strains to gradient concentrations of fungicide carbendazim were assayed at the optimal temperature, based on their colony growth diameters and the counts of colony forming units (CFU). The inhibitory effect of carbendazim concentration on CFU counts fitted well to a Logistic model, with high coefficients of determination being achieved for all the strains (r2 > or = 0.90). The fitted minimal inhibitory concentration (MIC) values showed a low carbendazim resistance of strains Pfr4205 and Pfr116 (MIC < or = 20.0 microg x ml(-1)), whereas the other four strains fell within medium resistance to the fungicide (20 < or = MIC < or = 100). The MIC of strain Pfr6206 (93.5 microg x ml(-1) was very close to the high resistance level of MIC > 100 microg x ml(-1). Therefore, strain Pfr6206 was satisfactory with both the temperature fitness and the carbendazim resistance, and could be considered as a candidate strain for a fungal formulation adapting to more variable crop systems against insect pests.
Identification and Characterization of Carbendazim-Resistant Isolates of Gibberella Zeae.

Abstract:Sensitivity of Gibberella zeae to carbendazim was determined by measuring mycelial growth in fungicide-amended media. Among 1,529 isolates tested, 31 isolates showed a high level of resistance (HR) to carbendazim (fungicide concentration that results in 50% inhibition of mycelial growth [EC50] of 10.35 to 30.26 mg a.i. liter-1) and 10 isolates were moderately resistant (MR) (EC50 of 4.50 to 7.28 mg a.i. liter-1). The remaining 1,488 isolates were sensitive to carbendazim and were unable to grow on potato dextrose agar amended with carbendazim at 2 mg a.i. liter-1. Analysis of DNA sequences of the β2-tubulin (Tub2) gene showed that all 10 MR isolates had a point mutation at codon 198 causing a replacement of glutamic acid by glutamine. At the codon position 167, the amino acid phenylalanine was replaced by tyrosine in 28 of 31 HR isolates. The remaining three HR isolates had a point mutation at codon 200 which converted phenylalanine to tyrosine. Based on these point mutations in the Tub2 gene, allele-specific polymerase chain reaction primers were developed for rapid detection of the point mutations. The rapid molecular method will be a valuable tool for the monitoring of carbendazim resistance in G. zeae. Additionally, deletion of the β1-tubulin gene (Tub1) in the HR isolate GJ33 resulted in increased resistance to carbendazim. These results indicate that Tub1 plays a role in the sensitivity of G. zeae to carbendazim.
Characteristics of Inositol Phosphorylceramide Synthase and Effects of Aureobasidin A on Growth and Pathogenicity of Botrytis Cinerea

Abstract:Inositol phosphorylceramide (IPC) synthase is the key enzyme with highly conserved sequences, which is involved in fungal sphingolipid biosynthesis. The antibiotic aureobasidin A (AbA) induces the death of fungi through inhibiting IPC synthase activity. The mutations of AUR1 gene coding IPC synthase in fungi and protozoa causes a resistance to AbA. However, the mechanism of AbA resistance is still elusive. In this paper, we generated two mutants of Botrytis cinerea with AbA-resistance, BcAUR1a and BcAUR1b, through UV irradiation. BcAUR1a lost an intron and BcAUR1b had three amino acid mutations (L197P, F288S and T323A) in the AUR1 gene. AbA strongly inhibits the activity of IPC synthase in wild-type B. cinerea, which leads to distinct changes in cell morphology, including the delay in conidial germination, excessive branching near the tip of the germ tube and mycelium, and the inhibition of the mycelium growth. Further, AbA prevents the infection of wild-type B. cinerea in tomato fruits via reducing oxalic acid secretion and the activity of cellulase and pectinase. On the contrary, AbA has no effect on the growth and pathogenicity of the two mutants. Although both mutants show a similar AbA resistance, the molecular mechanisms might be different between the two mutants.
Multi-Sited Mutations of Beta-Tubulin Are Involved in Benzimidazole Resistance and Thermotolerance of Fungal Biocontrol Agent Beauveria Bassiana

Abstract:Fungicide resistance and thermotolerance of biocontrol agents in mitosporic fungi are of merits for enhancing fungal formulations against insect pests in the field. Among 20 wild strains of Beauveria bassiana (a well-known fungal biocontrol agent) tested in this study, 19 were sensitive or highly sensitive to carbendazim (methyl 2-benzimidazole carbamate), a typical benzimidazole fungicide, despite low resistance found in one strain. Sequential mutagenesis of a carbendazim-sensitive wild strain [minimal inhibitory concentration (MIC) = 1.32 microg ml(-1)] under artificial selection pressure generated 11 mutants sharing a common MIC of > 1000 microg ml(-1) without visible variation in colony growth and conidiation capacity. This represents at least 758-fold enhancement of the resistance among the mutants. However, accompanied with the enhanced resistance, all the mutants became less thermotolerable. Stressed at 48 degrees C, conidial LT(50)s of the mutants varied from 1.8 to 9.6 min and were lower than the parental LT(50) (36 min). Moreover, the contents of hydrophobin-like proteins in conidial walls declined significantly among the mutants compared with that of the wild parent. Mutations commonly relating to benzimidazole resistance in fungi were located at Q134, F167 and/or E198 around the taxol-binding site of beta-tubulin by sequencing the beta-tubulin of the mutants. Also, mutations of other 37 amino acid residues in the sequences (each having one to five residues mutated) were found for the first time and they were diverse in spatial structure. All mutations restricted to the half of beta-tubulin close to alpha-tubulin were likely involved in variation in each of the traits concerned but their interactions were complicated.
Relationships between oxidative tolerance and polysaccharide content in aerial conidia of the entomopathogenic fungi

Abstract:Entomopathogenic hyphomycete has long been widely applied in biocontrol of insect pests due to easy production, formulation and environmental safety. This study was to illustrate the relationship between the oxidative tolerance and the polysaccharide content in aerial conidia of Beauveria bassiana and Isaria fumosorosa. Survival indices of 11 isolates were separately assessed as a ratio of the viability of conidia exposed to 10min stress with H2 O2 over that of unstressed conidia and fitted well to a survival model(r2>0.95). For a given isolate, the fitted model generated an LD50, the concentration for 50% viability loss under the stress. As a result, I. fumosorosea was more tolerant to oxidative stress than B. bassiana. The LD50 for B. bassiana and I. fumosorosea was correlated well to the conidial polysaccharide content of the respective strain. The conidial contents of B. bassiana could be modified by changing the concentration and type of sugar in media. However, the survival indices of isolate were still correlated well to the polysaccharide content from conidia produced on glucose-,sucrose-, or starch-based media. The relationship between conidial polysaccharide content and oxidative tolerance suggested a new way to search for fungal biocontrol agents with more environmental stability.
Fungal Insecticidal Activity Elevated by Non-Risky Markerless Overexpression of an Endogenous Cysteine-Free Protein Gene in Beauveria Bassiana.

Abstract:BACKGROUND Fungal insecticides are notorious for slow kill action, an intrinsic trait that can be improved by the genetic engineering of an exogenous or endogenous virulence factor. However, transgenic insecticides expressing exogenous toxin and herbicide-resistant marker genes may cause unexpected ecological risks and are hardly permitted for field release due to strict regulatory hurdles. It is necessary to improve biotechnology that can speed up fungal insect-killing action and exclude ecological risk source. RESULTS A markerless transformation system of Beauveria bassiana, a main source of wide-spectrum fungal insecticides, was reconstructed based on the fungal uridine auxotrophy (Delta ura3). The system was applied for overexpression of the small cysteine-free protein (120 amino acids) gene cfp previously characterized as a regulator of the fungal virulence and gene expression. Three cfp-overexpressed strains showed much faster kill action to Galleria mellonella larvae than the parental wild-type via normal cuticle infection but no change in vegetative growth and aerial condition. The faster kill action was achieved due to not only significant increases in conidial adherence to insect cuticle and total activity of secreted cuticle-degrading Pr1 proteases and of antioxidant enzymes crucial for collapse of insect immune defense but acceleration of hemocoel localization, proliferation in vivo and host death from mummification. CONCLUSION The markerless system is free of any foreign DNA fragment as a source of ecologic risk and provides a novel biotechnological approach to enhancing fungal insecticidal activity with non-risky endogenous genes and striding over the regulatory hurdles. A Conidial Protein (CP15) of Beauveria Bassiana Contributes to the Conidial Tolerance of the Entomopathogenic Fungus to Thermal and Oxidative Stresses

Abstract:Aerial conidia are central dispersing structures for most fungi and represent the infectious propagule for entomopathogenic fungus Beauveria bassiana, thus the active ingredients of commercial mycoinsecticides. Although a number of formic-acid-extractable (FAE) cell wall proteins from conidia have been characterized, the functions of many such proteins remain obscure. We report that a conidial FAE protein, termed CP15, isolated from B. bassiana is related to fungal tolerance to thermal and oxidative stresses. The full-length genomic sequence of CP15 was shown to lack introns, encoding for a 131 amino acid protein (15.0 kDa) with no sequence identity to any known proteins in the NCBI database. The function of this new gene with two genomic copies was examined using the antisense-RNA method. Five transgenic strains displayed various degrees of silenced CP15 expression, resulting in significantly reduced conidial FAE protein profiles. The FAE protein contents of the strains were linearly correlated to the survival indices of their conidia when exposed to 30-min wet stress at 48°C (r 2 = 0.93). Under prolonged 75-min heat stress, the median lethal times (LT50s) of their conidia were significantly reduced by 13.6–29.5%. The CP15 silenced strains were also 20–50% less resistant to oxidative stress but were not affected with respect to UV-B or hyperosmotic stress. Our data indicate that discrete conidial proteins may mediate resistance to some abiotic stresses, and that manipulation of such proteins may be a viable approach to enhancing the environmental fitness of B. bassiana for more persisting control of insect pests in warmer climates.
Molecular Mechanisms Underlying Metabolic Resistance to Cyflumetofen and Bifenthrin in Tetranychus urticae Koch on Cowpea

Abstract:Tetranychus urticae Koch (T. urticae) is one of the most tremendous herbivores due to its polyphagous characteristics, and is resistant to most acaricides. In this study, enzyme-linked immunosorbent assay (ELISA), transcriptome sequencing (RNA-seq) and quantitative real-time PCR (qRT-PCR) were carried out to analyze the mechanisms of T. urticae metabolic resistance to cyflumetofen and bifenthrin on cowpea. The enzyme activity of UDP-glucuronosyltransferases (UGTs) and carboxylesterases (CarEs) in the cyflumetofen-resistant (R_cfm) strain significantly decreased, while that of cytochrome P450 monooxygenases (P450s) significantly increased. Meanwhile, the activities of glutathione-S-transferases (GSTs), CarEs and P450s in the bifenthrin-resistant (R_bft) strain were significantly higher than those in the susceptible strain (Lab_SS). According to the Kyoto Encyclopedia of Genes and Genomes (KEGG) and Gene Ontology (GO) analyses, in the R_cfm mite strain, two carboxyl/cholinesterase (CCE) genes and two P450 genes were upregulated and one gene was downregulated, namely CYP392E7; in the R_bft mite strain, eleven CCE, nine UGT, two P450, four GST and three ABC genes were upregulated, while four CCE and three P450 genes were downregulated. Additionally, 94 differentially expressed genes (DEGs) were common to the two resistant groups. Specifically, TuCCE46 and TuCCE70 were upregulated in both resistant groups. Furthermore, the qRT-PCR validation data were consistent with those from the transcriptome sequencing analysis. Specifically, TuCCE46 (3.37-fold) was significantly upregulated in the R_cfm strain, while in the R_bft strain, TeturUGT22 (5.29-fold), teturUGT58p (1.74-fold), CYP392A11 (2.89-fold) and TuGSTd15 (5.12-fold) were significantly upregulated and TuCCE01 (0.13-fold) and CYP392A2p (0.07-fold) were significantly downregulated. Our study indicates that TuCCE46 might play the most important role in resistance to cyflumetofen, and TuCCE01, teturUGT58p, teturUGT22, CYP392A11, TuGSTd15, TuGSTm09 and TuABCG-13 were prominent in the resistance to bifenthrin. These findings provide further insight into the critical genes involved in the metabolic resistance of T. urticae to cyflumetofen and bifenthrin.

Carboxylesterase and Cytochrome P450 Confer Metabolic Resistance Simultaneously to Azoxystrobin and Some Other Fungicides in Botrytis cinerea

Abstract:Plant pathogens have frequently shown multidrug resistance (MDR) in the field, often linked to efflux and sometimes metabolism of fungicides. To investigate the potential role of metabolic resistance in B. cinerea strains showing MDR, the azoxystrobin-sensitive strain B05.10 and -resistant strain Bc242 were treated with azoxystrobin. The degradation half-life of azoxystrobin in Bc242 (9.63 days) was shorter than that in B05.10 (28.88 days). Azoxystrobin acid, identified as a metabolite, exhibited significantly lower inhibition rates on colony and conidia (9.34 and 11.98%, respectively) than azoxystrobin. Bc242 exhibited higher expression levels of 34 cytochrome P450s (P450s) and 11 carboxylesterase genes (CarEs) compared to B05.10 according to RNA-seq analysis. The expression of P450 genes Bcin_02g01260 and Bcin_12g06380, along with the CarEs Bcin_12g06360 in Saccharomyces cerevisiae, resulted in reduced sensitivity to various fungicides, including azoxystrobin, kresoxim-methyl, pyraclostrobin, trifloxystrobin, iprodione, and carbendazim. Thus, the mechanism of B. cinerea MDR is linked to metabolism mediated by the CarE and P450 genes.

Abstract:Fusarium culmorum is a fungal pathogen causing economically important diseases on a variety of crops. Fungicides can be applied to control this species with triazoles being the most efficient molecules. F. culmorum strains resistant to these molecules have been reported, but the underlying resistance mechanisms remain unknown. In this study, a tebuconazole-adapted F. culmorum strain was developed with a level of fitness similar to its parental strain. The adapted strain showed cross-resistance to all demethylation inhibitors (DMIs), but not to other classes of fungicides tested. RNA-Seq analysis revealed high transcriptomic differences between the resistant strain and its parental strain after tebuconazole treatment. Among these changes, FcABC1 (FCUL_06717), a pleiotropic drug resistance transporter, had a 30-fold higher expression level upon tebuconazole treatment in the adapted strains as compared to the wild-type strain. The implication of this transporter in triazole resistance was subsequently confirmed in field strains harboring distinct levels of sensitivity to triazoles. FcABC1 is present in other species/genera, including F. graminearum in which it is known to be necessary for azole resistance. No difference in FcABC1 sequences, including the surrounding regions, were found when comparing the resistant strain to the wild-type strain. Fusarium culmorum is therefore capable to adapt to triazole pressure by overexpressing a drug resistance transporter when submitted to triazoles and the same mechanism is anticipated to occur in other species.

Analysis of resistance risk and mechanism of the 14α-demethylation inhibitor ipconazole in Fusarium pseudograminearum

Abstract:Ipconazole is a broad-spectrum triazole fungicide that is highly effective against Fusarium pseudograminearum. However, its risk of developing resistance and mechanism are not well understood in F. pseudograminearum. Here, the sensitivities of 101 F. pseudograminearum isolates to ipconazole were investigated, and the average EC50 value was 0.1072 μg/mL. Seven mutants resistant to ipconazole were obtained by fungicide adaption, with all but one showing reduced fitness relative to the parental isolates. Cross-resistance was found between ipconazole and mefentrifluconazole and tebuconazole, but none between ipconazole and pydiflumetofen, carbendazim, fludioxonil, or phenamacril. In summary, these findings suggest that there is a low risk of F. pseudograminearum developing resistance to ipconazole. Additionally, a point mutation, G464S, was seen in FpCYP51B and overexpression of FpCYP51A, FpCYP51B and FpCYP51C was observed in ipconazole-resistant mutants. Assays, including transformation and molecular docking, indicated that G464S conferred ipconazole resistance in F. pseudograminearum.
Biological and Molecular Characterizations of Fluxapyroxad-Resistant Isolates of Botrytis Cinerea

Abstract:Gray mold caused by Botrytis cinerea leads to serious losses in various crops. Fluxapyroxad, a succinate dehydrogenase inhibitor (SDHI) fungicide, has been used to control gray mold for several years in China. In this study, we identified six fluxapyroxad-resistant (FluR) isolates from 96 isolates of B. cinerea collected in fields. Phenotypic assays showed that the FluR isolates exhibited defects in mycelial growth, conidiation, sclerotium formation, stress tolerance and virulence. Analysis of nucleotide sequences of succinate dehydrogenase subunit-coding genes revealed that various point mutations, including P225F, N230I, K283N and H272R in BcSdhB, G37S and P80H in BcSdhC, and V9A in BcSdhD, were associated with the resistance to fluxapyroxad in B. cinerea. Sensitivity assays showed that the FluR isolates also exhibited resistance to another SDHI boscalid, but there was no cross-resistance between fluxapyroxad and other fungicides with different modes of action. These results indicate that the resistance of B. cinerea to fluxapyroxad has occurred in fields, and further monitoring and management of the resistance to fluxapyroxad in B. cinerea should be carried out.
Induction and Characterization of Iprodione-resistant Mutants of Cladosporium Fulvum

Abstract:Sensitivity of three phenotypes of Cladosporium fulvum isolates,namely benzimidazole sensitive-diethofencarb resistant(BenS-DieR),benzimidazole resistant-diethofencarb sensitive(BenR-DieS)and benzimidazole resistant-diethofencarb resistant(BenR-DieR),to different classes of fungicides was determined.The results showed that there was no relationship between the sensitivity of C.fulvum to benzimidazole,diethofencarb and to other fungicides.Activity of mycelial growth inhibition of the 16 fungicides against C.fulvum was procymidonevinclozoliniprodionetebuconazolechlorothalonilpyrimethanilkresoxim-methylmancozeb8-hydroxyquinoline copper(Ⅱ) saltpropiconazoldifenoconazoleazoxystrobinmepronildimethomorphboscalidtriadimefon.Activity of spore germination inhibition was kresoxim-methylprocymidonechlorothalonilvinclozolinmepronil8-hydroxyquinoline copper(Ⅱ) saltiprodionemancozebazoxystrobinboscalidpyrimethaniltebuconazolepropiconazoldifenoconazoletriadimefondimethomorph.A total of 17 iprodione-resistant mutants was isolated through UV irradiation with a mutation frequency of 4.5×10-7.Among them,the percentage of low level resistant mutants,moderate level resistant mutants,and high level resistant mutants was 17.65%,70.59% and 11.75%,respectively.All mutants showed cross-resistance to procymidone and vinclozolin.No significant difference in sensitivity was detected between these mutants and their parent to other fungicides such as benzimidazoles,DMIs and QoIs.There was no significant difference in growth,sporulation and pathogenicity between mutants and their parent too.However,these mutants were more sensitivity to osmotic stress than their parent.
Multiple routes to fungicide resistance: Interaction of Cyp51 gene sequences, copy number and expression

Abstract:We examined the molecular basis of triazole resistance in Blumeria graminis f. sp. tritici (wheat mildew, Bgt), a model organism among powdery mildews. Four genetic models for responses to triazole fungicides were identified among US and UK isolates, involving multiple genetic mechanisms. Firstly, only two amino acid substitutions in CYP51B lanosterol demethylase, the target of triazoles, were associated with resistance, Y136F and S509T (homologous to Y137F and S524T in the reference fungus Zymoseptoria tritici). As sequence variation did not explain the wide range of resistance, we also investigated Cyp51B copy number and expression, the latter using both reverse transcription-quantitative PCR and RNA-seq. The second model for resistance involved higher copy number and expression in isolates with a resistance allele; thirdly, however, moderate resistance was associated with higher copy number of wild-type Cyp51B in some US isolates. A fourth mechanism was heteroallelism with multiple alleles of Cyp51B. UK isolates, with significantly higher mean resistance than their US counterparts, had higher mean copy number, a high frequency of the S509T substitution, which was absent from the United States, and in the most resistant isolates, heteroallelism involving both sensitivity residues Y136+S509 and resistance residues F136+T509. Some US isolates were heteroallelic for Y136+S509 and F136+S509, but this was not associated with higher resistance. The obligate biotrophy of Bgt may constrain the tertiary structure and thus the sequence of CYP51B, so other variation that increases resistance may have a selective advantage. We describe a process by which heteroallelism may be adaptive when Bgt is intermittently exposed to triazoles.
Procymidone Application Contributes to Multidrug Resistance of Botrytis cinerea

Abstract:The necrotrophic pathogen Botrytis cinerea infects a broad range of plant hosts and causes substantial economic losses to many crops. Although resistance to procymidone has been observed in the field, it remains uncertain why procymidone is usually involved in multidrug resistance (MDR) together with other fungicides. Nine mutants derived from the B. cinerea strain B05.10 through procymidone domestication exhibited high resistance factors (RFs) against both procymidone and fludioxonil. However, the fitness of the mutants was reduced compared to their parental strain, showing non-sporulation and moderate virulence. Furthermore, the RFs of these mutants to other fungicides, such as azoxystrobin, fluazinam, difenoconazole, and pyrimethanil, ranged from 10 to 151, indicating the occurrence of MDR. Transcriptive expression analysis using the quantitative polymerase chain reaction (qPCR) revealed that the mutants overexpressed ABC transporter genes, ranging from 2 to 93.7-fold. These mutants carried single-point mutations W647X, R96X, and Q751X within BcBos1 by DNA sequencing. These alterations in BcBos1 conferred resistance to procymidone and other fungicides in the mutants. Molecular docking analysis suggested distinct interactions between procymidone and Bos1 in the B. cinerea standard strain B05.10 or the resistant mutants, suggesting a higher affinity of the former towards binding with the fungicide. This study provides a comprehensive understanding of the biological characteristics of the resistant mutants and conducts an initial investigation into its fungicide resistance traits, providing a reference for understanding the causes of multidrug resistance of B. cinerea in the field.
